Circadian clock genes are crucial for generating and sustaining most rhythmic daily functions in the animal kingdom,which entrain the rhythms of biochemical,physiological,and behavioural processes.To better understand...Circadian clock genes are crucial for generating and sustaining most rhythmic daily functions in the animal kingdom,which entrain the rhythms of biochemical,physiological,and behavioural processes.To better understand the molecular oscillations of the circadian rhythms in darkbarbel catfish(Pelteobagrus vachellii),we isolated and characterized two circadian clock genes in P.vachellii,period 1(per1),and period 3(per3).The circadian clock gene per1 was found to encode a 1428-amino acid polypeptide,including PER-ARNT-SIM(PAS)dimerisation domains,a PAS-associated C-terminal motif(PAC),a short mutable domain(S/M),and a nuclear export signal(NES).The 4902-bp per3 cDNA includes an open reading frame encoding a 1292-amino acid residue polypeptide with a PER-ARNT-SIM(PAS)domain,cytoplasmic localisation domain(CLD),interaction site(TIS),and a nuclear localisation signal(NLS).The per1 and per3 gene was constitutively expressed in all examined tissues.Moreover,per1 expression within a light/dark cycles showed rhythmic expression in the diencephalon,brain,liver and intestine,with the acrophase at 15:15,12:52,7:51 and 12:55,respectively.Daily expression of per3 was rhythmic in the diencephalonbrain,liver and intestine,with the acrophase at 8:15,9:54,10:39,and 10:25 h,respectively.These findings expand our understanding of circadian mechanism at the molecular level in this species.展开更多
The purpose is to study the accuracy of ocean wave parameters retrieved from C-band VV-polarization Sentinel-1Synthetic Aperture Radar(SAR) images, including both significant wave height(SWH) and mean wave period...The purpose is to study the accuracy of ocean wave parameters retrieved from C-band VV-polarization Sentinel-1Synthetic Aperture Radar(SAR) images, including both significant wave height(SWH) and mean wave period(MWP), which are both calculated from a SAR-derived wave spectrum. The wind direction from in situ buoys is used and then the wind speed is retrieved by using a new C-band geophysical model function(GMF) model,denoted as C-SARMOD. Continuously, an algorithm parameterized first-guess spectra method(PFSM) is employed to retrieve the SWH and the MWP by using the SAR-derived wind speed. Forty-five VV-polarization Sentinel-1 SAR images are collected, which cover the in situ buoys around US coastal waters. A total of 52 subscenes are selected from those images. The retrieval results are compared with the measurements from in situ buoys. The comparison performs good for a wind retrieval, showing a 1.6 m/s standard deviation(STD) of the wind speed, while a 0.54 m STD of the SWH and a 2.14 s STD of the MWP are exhibited with an acceptable error.Additional 50 images taken in China's seas were also implemented by using the algorithm PFSM, showing a 0.67 m STD of the SWH and a 2.21 s STD of the MWP compared with European Centre for Medium-range Weather Forecasts(ECMWF) reanalysis grids wave data. The results indicate that the algorithm PFSM works for the wave retrieval from VV-polarization Sentinel-1 SAR image through SAR-derived wind speed by using the new GMF C-SARMOD.展开更多

This study examined the mechanism by which ACTH affects the hair cycle using mice d...Adrenocorticotropin hormone (ACTH), which is secreted in response to psychological stress, plays an important role in the hair cycle. This study examined the mechanism by which ACTH affects the hair cycle using mice deficient in melanocortin receptor-2(MC2R<sup>-/-</sup>), which is a main receptor for ACTH. We observed the hair cycle using female MC2R<sup>-/-</sup> mice at 15 weeks old and five days old to determine whether there were any age-dependent differences. The 15-week-old MC2R<sup>-/-</sup> mice showed the anagen phase for all mice. On the other hand, all of the MC2R<sup>+/+</sup> mice showed the telogen phase at the same age. Moreover, in the five-day-old mice, the hair growth of the MC2R<sup>-/-</sup> mice occurred earlier than in the MC2R<sup>+/+</sup> mice. Both the 15-week-old and five-day-old MC2R<sup>-/-</sup> mice had higher levels of ACTH and alpha-melanocyte stimulating hormone in the blood than did the MC2R<sup>+/+</sup> mice. In addition, in the 15-week-old MC2R<sup>-/-</sup> mice, the hair cycle shifted to the telogen phase following the administration of a cyclic guanosine monophosphate (cGMP) inhibitor and MC1R/MC5R inhibitor. In the five-day-old MC2R<sup>-/-</sup> mice, the hair growth was slowed by the administration of corticosterone. However, the mammalian cellular circadian rhythm in photobiomodulation remains unknown. A lot of evidence has shown that >20% of expressed mRNAs in bone stissues oscillate in a circadian manner. The aim of this paper is to investigate the mRNA expression of period 1 (per 1) in murine bone marrow-derived mesenchymal stem cells (BMSCs) which were irradiated by 635 nm red laser light. The cells were seeded in 35mm tissue-culture dishes at a density of 8 × 104 cells/dish and cultured in Dulbecco’s modi?ed Eagle’s medium (DMEM). BMSCs were irradiated once by 635 nm red light with radiation energies 0 J/cm2, 1 J/cm2, 4 J/cm2. mRNA expression of per 1 via Semi-Quantitative Real-time RT-PCR at 0h, 6 h, 12 h, 18 h, 24 h. The genes displayed a oscillatory period of nearly 24 hours. And 635 m laser light changed the mRNA expression of per 1. We conclude that red light irradiation can affect the circadian rhythm of BMSCs.展开更多
In the present paper, a general solution involv- ing three arbitrary functions for the generalized (2+1)- dimensional KdV-mKdV equation, which is derived from the generalized (1+1)-dimensional KdV-mKdV equa- tio...In the present paper, a general solution involv- ing three arbitrary functions for the generalized (2+1)- dimensional KdV-mKdV equation, which is derived from the generalized (1+1)-dimensional KdV-mKdV equa- tion, is first introduced by means of the Wiess, Tabor, Carnevale (WTC) truncation method. And then multi- symplectic formulations with several conservation laws taken into account are presented for the generalized (2+1)- dimensional KdV-mKdV equation based on the multi- symplectic theory of Bridges. Subsequently, in order to simulate the periodic wave solutions in terms of rational functions of the Jacobi elliptic functions derived from thegeneral solution, a semi-implicit multi-symplectic scheme is constructed that is equivalent 1:o the Preissmann scheme. From the results of the numerical experiments, we can con- clude that the multi-symplectic schemes can accurately sim- ulate the periodic wave solutions of the generalized (2+1)- dimensional KdV-mKdV equation while preserve approxi- mately the conservation laws.展开更多
To investigate the effect of two deoxyribozymes targeting period1 (per1) mRNA in vitro for exploring a novel gene therapy approach about circadian rhythm diseases, the specific deoxyribozymes targeting per1 were desig...To investigate the effect of two deoxyribozymes targeting period1 (per1) mRNA in vitro for exploring a novel gene therapy approach about circadian rhythm diseases, the specific deoxyribozymes targeting per1 were designed and synthesized chemically following MFold analysis according to its mRNA secondary structure. per1 RNA fragments were prepared by in vitro transcription of pcDNA3.1(+)-per1164:256. The cleavage reactions containing deoxyribozymes and per1 RNA fragments were performed under certain conditions. With the transfection tech- nique mediated by LipofectAMINETM, pcDNA3-per1 and DRz164 or DRz256 were introduced into NIH3T3 cells. The effects of deoxyribozymes on per1 were studied by reverse tran- script-polymerase chain reaction (RT-PCR) and flow cytometry (FCM). When deoxyribozymes and RNA transcripts were incubated under the adopted conditions at 37℃ for 2 h, about 63% of per1164:256 RNA transcripts were cleaved by DRz164 and about 50.5% by DRz256. After co- transfecting pcDNA3-per1 with DRz164 or DRz256, the expression of per1 mRNA was de- creased, as indicated by RT-PCR semi-quantity analysis. FCM analysis showed that Per1 protein was inhibited. Both DRz164 and DRz256 targeting per1 have the specific cleavage activity to- ward per1 mRNA in vitro and can highly block the expression of per1 gene in cellular milieu.展开更多

Under some transversalconditions and the non–twisted condition, the existence, coexistence and incoexistence of2–point–loop, 1–homoclinic orbit, simple 1–periodic orbit and 2–fold 1–periodic orbit, and thenumber of 1–periodic orbits are studied. Moreover, the bifurcation surfaces and existence regionsare given. Lastly, the above bifurcation results are applied to a planar system and an insidestability criterion is obtained.展开更多
The Mg96Y3Zn1 alloy processed by extrusion and equal channel angular pressing (ECAP) was investigated. It was found that the Mg96Y3Zn1 alloy processed by extrusion and ECAP obtained ultrafine grains and exhibited ex...The Mg96Y3Zn1 alloy processed by extrusion and equal channel angular pressing (ECAP) was investigated. It was found that the Mg96Y3Zn1 alloy processed by extrusion and ECAP obtained ultrafine grains and exhibited excellent mechanical properties. After ECAP, the average grain size of Mg96Y3Zn1 alloy was refined to about 400 nm. The highest strengths with yield strength of 381.45 MPa and ultimate tensile strength of 438.33 MPa were obtained after 2 passes at 623 K. The high strength of Mg96Y3Zn1 alloy was due to the strengthening by the grain refinement, the long period stacking (LPS) structure, solid solution, fine Mg24Y5 particles, and nano-scale precipitates. It was found that the elongation was decreased with pass number increasing. It was because that the cracks were preferentially initiated and propagated in the interior of X-phase during the tensile test.展开更多
