A cold-adapted gram-negative bacterium, hydrocarbon-degrading Pseudoalteromonas sp. NJ289, was isolated from sea-ice of the Antarctica region, and sequenced the whole genome through the next generation sequencing platform. The assembly yielded three contigs representing two chromosomes and one plasmid with the sizes of 3.2 Mb, 636 kb and 1.8 kb, respectively. The G+C contents of genome were 40.83% and included 3 589 ORFs. Functional annotation indicated some potential roles in enzymatic activity and environmental adaptability. This study may help for understanding the population diverse, evolutionary ecology and the microbial interaction.展开更多
In order to understand molecular characterization of Citrus tatter leaf virus(CTLV) isolated from China,full-length cDNAs of CTLV-MTH and CTLV-XHC from Citrus reticulata and Citrus sinensis were cloned and sequenced...In order to understand molecular characterization of Citrus tatter leaf virus(CTLV) isolated from China,full-length cDNAs of CTLV-MTH and CTLV-XHC from Citrus reticulata and Citrus sinensis were cloned and sequenced based on whole-genome amplification by RT-PCR.The complete nucleotide sequences of CTLV-MTH and CTLV-XHC were determined to be 6497 nucleotides in length and shared 79.9-91.0%and 78.8-98.0%nucleotide sequence identity,respectively,with other Apple stem grooving virus(ASGV) or CTLV strains available in GenBank.Unexpectedly,CTLV-MTH showed the highest nucleotide sequence identity(91%) with an apple isolate of ASGV,followed by 86.5%with ASGV-HH and 85.7%with ASGV-CHN.Furthermore,CTLV-MTH and three ASGV strains were grouped to a separate cluster in the phylogenetic tree,suggesting it has a closer relationship to ASGV than to CTLV.Therefore,it can be concluded roughly that CTLV may be not a distinct strains of ASGV.We proposed that Citrus tatter leaf virus should be renamed Apple stem grooving virus.展开更多

The Oreta fuscopurpure...In this study, the first complete mitochondrial genome(mitogenome) of lepidopteran family Drepanidae(superfamily Drepanoidea) was reported with the notes about its phylogenetic implications. The Oreta fuscopurpurea mitogenome is 15,564 bp in length, including 13 protein-coding genes, two ribosomal RNAs(lrRNA and srRNA), 22 transfer RNAs and a non-coding control region(AT-rich region), with a 79.6% A+T content. The gene orientation and arrangement of the mitogenome are the same as other sequenced lepidopterans. All protein-coding genes usually start with the common ATN codon except for COI gene, which used CGA as the initial codon; eight PCGs use a typical stop codon of TAA, whereas the remaining PCGs use incomplete stop codon of T or TA. All tRNAs have the typical clover-leaf structure with the exception of t RNA^(Ser)(AGN). The lrRNA and sr RNA genes are 1,409 bp and 778 bp in size respectively, with the former harboring one(TA)_(13) microsatellite-like repeat and an 17 bp insertion. The 20 intergenic spacers totaling of 184 bp and 8 overlapping sequences totaling of 25 bp are scattered throughout the whole mitogenome. The 526 bp AT-rich region contains some structures characteristic of lepidopterans, such as the motif ATAGA preceded by an 19 bp poly-T stretch, a tRNA-like and a sterm-loop structures. Phylogenetic analysis of the Oreta fuscopurpurea with other 47 insect species covering 20 lepidopteran families were conducted based on the sequence data of the 13 mitogenomic protein coding genes with maximum likelihood(ML) and Bayesian inference(BI) methods, and the results showed distinctly that the superfamily Drepanoidea was sister to the clade of(Bombycoidea, Lasiocampoidea) +(Noctuoidea, Geometroidea).展开更多
In this study, China isolate HB of pseudorabies virus(PRV) was confirmed and genotypically characterized by amplifying and sequencing of partial UL34, a conservative gene involved in the egress of nucleocapsids from...In this study, China isolate HB of pseudorabies virus(PRV) was confirmed and genotypically characterized by amplifying and sequencing of partial UL34, a conservative gene involved in the egress of nucleocapsids from the nucleus, for phylogenetic analysis. The open reading frame(orf) of UL34 of PRV HB isolate is composed of 786 nucleotides, which encoded 262 amino acids. In addition, a potential transmembrane domain(241-260 aa) and 11 potential phosphorylation sites were also found in the UL34 of PRV HB isolate. Multiple amino acids alignment indicated that UL34 proteins of PRV strains derived from different geographic origins were highly conservative, but some mutations were also found. Phylogenetic analysis based on UL34 protein indicated that PRV HB strain was evolutionarily distinct from other recent China strains sequenced so far, forming a single clade within the phylogeny. Moreover, PRV HB isolate had close evolutionary relationship with Bo HV-1 and Bo HV-5 within the Alphaherpesvirinae. Taken together, these results indicated that PRV strains were in the progress of evolution. This study has expanded the knowledge of genetic profiles of PRV strains.展开更多
