To this end, a cross...The aim of the study was to assess feeding practices and the use of lysine and methionine in pig rationing on intensified and semi-intensive pig breeding in the Koudougou and Bobo-Dioulasso areas. To this end, a cross-sectional survey was carried out on 87 breeding in these towns. A Discriminant Factorial Analysis (DFA) confirming a k-means classification of the data collected was used to retain 71 breeding divided into three breeding classes: Class A (32.4% of breeding), Class B (14.08%) and Class C (53.52%). The results show that the majority of pig breeders were men between the ages of 36 and 59. Average herd sizes were 35 ± 28;79 ± 42 and 89 ± 21 pigs for Classes A, B and C respectively. The main breeds of pig found on the breeding were crossbred, Large white, local, Landrace and Duroc. Class A (26.1%), B (30%) and C (15.8%) breeders were familiar with both lysine and methionine. Class A breeders distributed feed staggered (65.2%) and in rations (34.8%). Lysine (13%) and methionine (8.7%) were purchased at 5250 FCFA/kg. Those in class B distributed feed staggered (50%) and in the form of rations (50%), in which they incorporated lysine (30%) and methionine (30%) purchased at a cost of 2500 FCFA/kg and 3000 FCFA/kg respectively. Rationing and staggered feeding were practiced by 23.7% and 76.3% of Class C breeders respectively. Only lysine purchased at 3400 FCFA/kg was incorporated into rations by 10.5% of breeders. The high cost of lysine and methionine was incriminated by Class A (100%), B (33.3%) and C (50%) breeders. In conclusion, intensive pig breeding, the practice of rationing and the incorporation of the amino acids lysine and methionine are of ascending importance from classes C, A to B. The high cost of feedstuffs, particularly lysine and methionine, compromises their use in rations, which could have a negative impact on expected breeding performance. The screening and use of feeds rich in and/or enriched with these amino acids, through the development or adaptation of technologies, could improve the efficiency of rations and the productivity of intensive pig breeding in Burkina Faso.展开更多
Background:The pig is an economically important livestock species and is a widely applied large animal model in medical research.Enhancers are critical regulatory elements that have fundamental functions in evolution,...Background:The pig is an economically important livestock species and is a widely applied large animal model in medical research.Enhancers are critical regulatory elements that have fundamental functions in evolution,development and disease.Genome-wide quantification of functional enhancers in the pig is needed.Results:We performed self-transcribing active regulatory region sequencing(STARR-seq)in the porcine kidney epithelial PK15 and testicular ST cell lines,and reliably identified 2576 functional enhancers.Most of these enhancers were located in repetitive sequences and were enriched within silent and lowly expressed genes.Enhancers poorly overlapped with chromatin accessibility regions and were highly enriched in chromatin with the repressive histone modification H3K9me3,which is different from predicted pig enhancers detected using ChIP-seq for H3K27ac or/and H3K4me1 modified histones.This suggests that most pig enhancers identified with STARR-seq are endogenously repressed at the chromatin level and may function during cell type-specific development or at specific developmental stages.Additionally,the PPP3CA gene is associated with the loin muscle area trait and the QKI gene is associated with alkaline phosphatase activity that may be regulated by distal functional enhancers.Conclusions:In summary,we generated the first functional enhancer map in PK15 and ST cells for the pig genome and highlight its potential roles in pig breeding.展开更多

[Method] Pig behavior's statistical analysis under both microbial fermentation bed pig breeding mode and traditional rearing mode was se lected to study the effect of different breeding environment on pig's growth [ Result] The results showed that the duration of piglets lying and sleeping behavior accounted for the highest proportion of tested behavior, up to more than 70% ,followed by feeding and drinking which reached 15.96% (under microbial fermentation bed breeding mode) and 9.33% (under traditional rearing mode) respectively. Compared with traditional intensive rearing mode, piglet exploratory behavior's occurrence probability and duration increased significantly, by 28.62% and 12.21% respectively. Fighting behavior's occurrence probability and duration decreased significantly, by 49.83% and 91.26% respectively. Nutrient index and health index were introduced to evaluate piglet's general heath condition under microbial fermentation bed breeding mode, and results indicated that the health index of piglets under microbial fermentation bed breeding mode had slight difference from that under traditional mode, namely 10.83 and 10.03 respectively. But health index of piglets under microbial fermentation bed breeding mode was 245.12, much higher than 21.96 under traditional mode. This secretion differs from milk and represents an essential vehicle of passive immunity,prebiotic compounds and growth factors involved in intestinal development. Most of the literature concerning colostrum composition refers mainly to human and cow; and little is known about pig colostrum metabolome and how it varies between pig breeds and different farrowing parity. Thus, the aim of the present research is to provide new information about pig colostrum composition and the associations between metabolites, the sows' breed and the survival and growth rates of their litters.Results: Colostrum samples were gathered from 58 parturitions of sows belonging to three different breeds chosen for their importance in Italian heavy pig production: 31 Large White, 15 Landrace and 12 Duroc respectively. The defatted and ultrafiltered colostrum samples were analysed using1 H–NMR spectroscopy. Principal Components Analysis(PCA) was assessed on the obtained spectra. In addition, using a Stepwise Regression and a Linear Regression analyses the metabolites named after the signals assignment were tested for their associations with piglets' performances.Twenty-five metabolites were identified, comprehending monosaccharides, disaccharides(such as lactose), organic acids(lactate, citrate, acetate and formate), nitrogenous organic acids(such as creatine) and other compounds,including nucleotides. PCA results evidence a clustering due to breed and season effects. Lactose was the main compound determining the assignment of the samples into different clusters according to the sow breed. Furthermore, some metabolites showed to be associated with piglets' performance and survival traits: acetate and taurine were positively related to litter weight gain and piglets' survival rate, respectively, while dimethylamine and cis-aconitate were linked to new-borns' impaired ability to survive.Conclusions: The results obtained suggest that colostrum composition is affected by breed, which, together with environmental conditions, may cause changes in colostrum metabolites content with possible consequences on piglets' performances. Among the identified metabolites, acetate, taurine, dimethylamine and cis-aconitate showed consistent associations with piglets' survival rate and litter weight gain, implying that these compounds may affect new-borns' ability to survive.展开更多

Background:A large number of pig breeds are distributed around the world,their features and characteristics vary among breeds,and they are valuable resources.Understanding the underlying genetic mechanisms that explai...Background:A large number of pig breeds are distributed around the world,their features and characteristics vary among breeds,and they are valuable resources.Understanding the underlying genetic mechanisms that explain across-breed variation can help breeders develop improved pig breeds.Results:In this study,we performed GWAS using a standard mixed linear model with three types of genome variants(SNP,InDel,and CNV)that were identified from public,whole-genome,sequencing data sets.We used 469 pigs of 57 breeds,and we identified and analyzed approximately 19 million SNPs,1.8 million InDels,and 18,016 CNVs.We defined six biological phenotypes by the characteristics of breed features to identify the associated genome variants and candidate genes,which included coat color,ear shape,gradient zone,body weight,body length,and body height.A total of 37 candidate genes was identified,which included 27 that were reported previously(e.g.,PLAG1 for body weight),but the other 10 were newly detected candidate genes(e.g.,ADAMTS9 for coat color).Conclusion:Our study indicated that using GWAS across a modest number of breeds with high density genome variants provided efficient mapping of complex traits.展开更多
Background Long-term natural and artificial selection has resulted in many genetic footprints within the genomes of pig breeds across distinct agroecological zones.Nevertheless,the mechanisms by which these signatures...Background Long-term natural and artificial selection has resulted in many genetic footprints within the genomes of pig breeds across distinct agroecological zones.Nevertheless,the mechanisms by which these signatures contribute to phenotypic diversity and facilitate environmental adaptation remain unclear.Results Here,we leveraged whole-genome sequencing data from 82 individuals from 6 domestic pig breeds originating in tropical,high-altitude,and frigid regions.Population genetic analysis suggested that habitat isolation significantly shaped the genetic diversity and contributed to population stratification in local Chinese pig breeds.Analysis of selection signals revealed regions under selection for adaptation in tropical(55.5 Mb),high-altitude(43.6 Mb),and frigid(17.72 Mb)regions.The potential functions of the selective sweep regions were linked to certain complex traits that might play critical roles in different geographic environments,including fat coverage in frigid environments and blood indicators in tropical and high-altitude environments.Candidate genes under selection were significantly enriched in biological pathways involved in environmental adaptation.These pathways included blood circulation,protein degradation,and inflammation for adaptation to tropical environments;heart and lung development,hypoxia response,and DNA damage repair for high-altitude adaptation;and thermogenesis,cold-induced vasodilation(CIVD),and the cell cycle for adaptation to frigid environments.By examining the chromatin state of the selection signatures,we identified the lung and ileum as two candidate functional tissues for environmental adaptation.Finally,we identified a mutation(chr1:G246,175,129A)in the cis-regulatory region of ABCA1 as a plausible promising variant for adaptation to tropical environments.Conclusions In this study,we conducted a genome-wide exploration of the genetic mechanisms underlying the adaptability of local Chinese pig breeds to tropical,high-altitude,and frigid environments.Our findings shed light on the prominent role of cis-regulatory elements in environmental adaptation in pigs and may serve as a valuable biological model of human plateau-related disorders and cardiovascular diseases.展开更多
[Objectives]This study was conducted to breed special pig breeds resistant to diarrhea by using modern biotechnology.[Methods]From Guizhou local breeds,such as Nuogu pigs,Kele pig,Yorkshire pigs and Duroc pigs,190 sam...[Objectives]This study was conducted to breed special pig breeds resistant to diarrhea by using modern biotechnology.[Methods]From Guizhou local breeds,such as Nuogu pigs,Kele pig,Yorkshire pigs and Duroc pigs,190 samples were collected for the analysis of anti-diarrhea gene.[Results]The anti-diarrhea genotype frequency of Kele pigs was 70.00%,which was higher than that of Nuogu pigs(67.37%)and Yorkshire pigs(Yorkshire pigs and Duroc pigs)(50.59%).The favorable anti-diarrhea gene of all Nuogu pigs,Kele pigs,and Yorkshire pigs and Duroc pigs was G,with gene frequencies of 0.7355,0.8368 and 0.8500,respectively,and the frequencies of allele A were 0.2645,0.1632 and 0.1500,respectively.In the process of generation selection,combination selection of GG♂×GG♀,GG♂×GA♀,GA♂×GG♀and GA♂×GA♀was conducted,and GG individuals were selected while gradually phasing out GA and AA individuals.The anti-diarrhea genotypes of 98 pigs in the offspring were tested,and it was found that the frequency of genotype GG was greatly improved,and the frequencies in Nuogu pigs,Kele pigs,Yorkshire pigs and Duroc pigs were increased to 73.91%,81.82%,85.25%and 66.67%respectively,thus forming a special anti-diarrhea breed.[Conclusions]This study provides a basis for selecting excellent breeding pigs,establishing core populations and screening resistance genes in the core populations and their offspring.展开更多
To assess the genetic diversity between randomly and selectively bred populations,we sequenced 438 bp of the mitochondrial DNA control region from 102 pigs.These samples represented four native pig breeds,one nucleus ...To assess the genetic diversity between randomly and selectively bred populations,we sequenced 438 bp of the mitochondrial DNA control region from 102 pigs.These samples represented four native pig breeds,one nucleus and one conservation herd from Yunnan,China.Twenty haplotypes with sixteen polymorphic sites were identified.The number of haplotypes in the nucleus herd of Saba pig and the conservation herd of Banna miniature pig were restricted to three and one,respectively,while the randomly bred pig populations exhibited over six haplotypes.Notably,haplotype diversity in randomly bred populations was significantly greater than the selectively bred populations(h=0.732 vs.0.425 and 0,exact test,P≤0.0036).These findings demonstrate that selective breeding generated low genetic diversity compared to randomly bred pig breeds.A timely intervention and well programmed breeding approach would stop further genetic diversity reduction in the nucleus and conservation herds of native pig breeds.Otherwise,selective breeding would dramatically reduce genetic diversity in only several years,indicating that sharp contradictions exist between breeding,conservation and genetic diversity.Genetic relationships are discussed based on net genetic distances among pig populations.展开更多

However, fiber has to be included in the diet to maintain normal physiological functions in the digestive tract. Moreover, the negative impact of dietary fiber will be determined by the fiber properties and may differ considerably between fiber sources. Various techniques can be applied to enhance nutritional value and utilization of available feed resources. In addition, the extent of fiber utilization is affected by the age of the pig and the pig breed. The use of potential prebiotic effects of dietary fiber is an attractive way to stimulate gut health and thereby minimize the use of anti-microbial growth promoters. Inclusion of soluble non-starch polysaccharides (NSP) in the diet can stimulate the growth of commensal gut microbes, inclusion of NSP from chicory results in changes in gut micro-environment and gut morphology of pigs, while growth performance remains unaffected and digestibility was only marginally reduced. The fermentation products and pH in digesta responded to diet type and were correlated with shifts in the microbiota. Interestingly, fiber intake will have an impact on the expression of intestinal epithelial heat-shock proteins in the pig. Heat-shock proteins have an important physiological role in the gut and carry out crucial housekeeping functions in order to maintain the mucosal barrier integrity. Thus, there are increasing evidence showing that fiber can have prebiotic effects in pigs due to interactions with the gut micro-environment and the gut associated immune system.展开更多
Background:A growing body of evidence has revealed that the mammalian genome is organized into hierarchical layers that are closely correlated with and may even be causally linked with variations in gene expression.Re...Background:A growing body of evidence has revealed that the mammalian genome is organized into hierarchical layers that are closely correlated with and may even be causally linked with variations in gene expression.Recent studies have characterized chromatin organization in various porcine tissues and cell types and compared them among species and during the early development of pigs.However,how chromatin organization differs among pig breeds is poorly understood.Results:In this study,we investigated the 3D genome organization and performed transcriptome characterization of two adipose depots(upper layer of backfat[ULB]and greater omentum[GOM])in wild boars and Bama pigs;the latter is a typical indigenous pig in China.We found that over 95%of the A/B compartments and topologically associating domains(TADs)are stable between wild boars and Bama pigs.In contrast,more than 70%of promoterenhancer interactions(PEIs)are dynamic and widespread,involving over a thousand genes.Alterations in chromatin structure are associated with changes in the expression of genes that are involved in widespread biological functions such as basic cellular functions,endocrine function,energy metabolism and the immune response.Approximately 95%and 97%of the genes associated with reorganized A/B compartments and PEIs in the two pig breeds differed between GOM and ULB,respectively.Conclusions:We reported 3D genome organization in adipose depots from different pig breeds.In a comparison of Bama pigs and wild boar,large-scale compartments and TADs were mostly conserved,while fine-scale PEIs were extensively reorganized.The chromatin architecture in these two pig breeds was reorganized in an adipose depotspecific manner.These results contribute to determining the regulatory mechanism of phenotypic differences between Bama pigs and wild boar.展开更多
Background:Oligosaccharides(OS)are indigestible carbohydrates naturally found in milk.The composition of porcine colostrum OS may influence the growth and the health of the neonate and consuming optimal concentrations...Background:Oligosaccharides(OS)are indigestible carbohydrates naturally found in milk.The composition of porcine colostrum OS may influence the growth and the health of the neonate and consuming optimal concentrations of OS may reduce piglet susceptibility to illness.In this manner,targeted supplementation of animal feed with OS is being explored as a health management tool in the livestock industry.The variation in OS composition between different breeds of pig and its association with the litter performance is currently unknown.The aim of this study was to characterize the colostrum OS composition from sows of different breed and parity and correlate this data with sow maternal traits.Methods:Eighty-three colostrum samples from parities 1 to 8 were gathered from 3 different breeds of sow:44 Large White sows,27 Landrace sows and 12 Duroc sows.Samples were taken between the birth of the first and the last piglet from sows that were not pharmacologically induced to farrow.OS were purified from the samples and analysed by MALDI-ToF mass spectrometry(21 OS compositions detected).The farrowing season and the maternal data were recorded for each sow,including the number of live piglets and the litter body weight at birth,at day(d)3 and at weaning.Results:Five OS compositions,including isomers of the bifidogenic Sialyllactose,Lacto-N-Tetraose and Lacto-NHexaose series,were detected in all the samples.Twelve other OS were identified in at least 50%of samples,and their abundances were affected by breed(P<0.05;6 of 12),marginally affected by season(P<0.10;3 of 12)and never by parity number.The abundances of each OS component were standardized by Z-score scaling(μ=0 and SD=1),transformed by principal component analysis,and four similarity clusters were generated.Cluster membership was associated with litter weight gain within 3 days(P=0.063)and at weaning(P<0.05),but not with piglet mortality within 3 days.Conclusions:OS composition of colostrum may partially explain the variability in maternal performance within and between different breeds of sow.The obtained OS data can provide useful information for the development of novel prebiotic food supplements for suckling and weaning pigs.展开更多

Among the pathogens, gram negative bacteria not only cause inflammation, but also cause different diseases and make the pigs more susceptible to virus infection. Vaccination, medication and elimination of sick pigs are major strategies of controlling disease. Genetic methods, such as selection of disease resistance in the pig, have not been widely used. Recently, the completion of the porcine whole genome sequencing has provided powerful tools to identify the genome regions that harboring genes controlling disease or immunity. Immunogenornics, which combines DNA variations, transcriptorne, immune response, and QTL mapping data to illustrate the interactions between pathogen and host immune system, will be an effective genomics tool for identification of disease resistance genes in pigs. These genes will be potential targets for disease resistance in breeding programs. This paper reviewed the progress of disease resistance study in the pig focusing on Gram-negative bacilli. Major porcine Gram-negative bacilli and diseases, suggested candidate genes/pathways against porcine Gram-negative bacilli, and distributions of QTLs for immune capacity on pig chromosomes were summarized. Some tools for immunogenomics research were described. We conclude that integration of sequencing, whole genome associations, functional genomics studies, and immune response information is necessary to illustrate molecular mechanisms and key genes in disease resistance.展开更多

In this study, qPCR was used to detect the expression of the BPI gene in twelve tissues of Meishan piglets from birth to weaning. BPI gene overexpression, bacterial adhesions count and indirect immunofluorescence were applied to analyze the relationship between BPI gene expression and the infectivity of Escherichia coli and Salmonella. The results showed that the BPI gene was expressed highly in duodenum, jejunum and ileum (fold changes of relative expression levels were more than 10 000, 500 and 200, respectively). The expression of the BPI gene at 35 days of age was significantly higher (P<0.01) than that at all other days. Transcription of the BPI gene was up-regulated 2 401-fold in porcine intestinal epithelial (IPEC-J2) cells that were transfected with the BPI gene overexpression lentivirus (IPEC-J2-BPI), and significantly higher (P<0.01) than that in negative control cells (IPEC-J2-NC). Protein expression levels in IPEC-J2-BPI cells were also increased. When IPEC-J2 cells were incubated with E. coli and Salmonella, respectively, for 2, 4, 6, 8, 10 and 12 h, the number of bacterial adhesions in IPEC-J2-BPI cells was significantly less (P<0.05) than that in IPEC-J2-NC cells. The results of indirect immunofluorescence analysis showed that the number of bacterial adhesions in IPEC-J2-BPI cells was significantly less (P<0.01) than that in IPEC-J2-NC cells. These results demonstrated that the BPI gene might play an important role in regulating weaning stress especially intestinal-mediated immune response. Overexpression of the BPI gene at the cellular level could significantly enhance the anti-bactericidal ability against Gram-negative bacteria such as E. coli and Salmonella. This has important biological significance in piglet resistance to bacterial diarrhea.展开更多

The results show that the majority of pig breeders were men between the ages of 36 and 59. Average herd sizes were 35 ± 28;79 ± 42 and 89 ± 21 pigs for Classes A, B and C respectively. The main breeds of pig found on the breeding were crossbred, Large white, local, Landrace and Duroc. Class A (26.1%), B (30%) and C (15.8%) breeders were familiar with both lysine and methionine. Class A breeders distributed feed staggered (65.2%) and in rations (34.8%). Lysine (13%) and methionine (8.7%) were purchased at 5250 FCFA/kg. Those in class B distributed feed staggered (50%) and in the form of rations (50%), in which they incorporated lysine (30%) and methionine (30%) purchased at a cost of 2500 FCFA/kg and 3000 FCFA/kg respectively. Rationing and staggered feeding were practiced by 23.7% and 76.3% of Class C breeders respectively. Only lysine purchased at 3400 FCFA/kg was incorporated into rations by 10.5% of breeders. The high cost of lysine and methionine was incriminated by Class A (100%), B (33.3%) and C (50%) breeders. In conclusion, intensive pig breeding, the practice of rationing and the incorporation of the amino acids lysine and methionine are of ascending importance from classes C, A to B. The high cost of feedstuffs, particularly lysine and methionine, compromises their use in rations, which could have a negative impact on expected breeding performance. The screening and use of feeds rich in and/or enriched with these amino acids, through the development or adaptation of technologies, could improve the efficiency of rations and the productivity of intensive pig breeding in Burkina Faso.

文摘Background:The pig is an economically important livestock species and is a widely applied large animal model in medical research.Enhancers are critical regulatory elements that have fundamental functions in evolution,development and disease.Genome-wide quantification of functional enhancers in the pig is needed.Results:We performed self-transcribing active regulatory region sequencing(STARR-seq)in the porcine kidney epithelial PK15 and testicular ST cell lines,and reliably identified 2576 functional enhancers.Most of these enhancers were located in repetitive sequences and were enriched within silent and lowly expressed genes.Enhancers poorly overlapped with chromatin accessibility regions and were highly enriched in chromatin with the repressive histone modification H3K9me3,which is different from predicted pig enhancers detected using ChIP-seq for H3K27ac or/and H3K4me1 modified histones.This suggests that most pig enhancers identified with STARR-seq are endogenously repressed at the chromatin level and may function during cell type-specific development or at specific developmental stages.Additionally,the PPP3CA gene is associated with the loin muscle area trait and the QKI gene is associated with alkaline phosphatase activity that may be regulated by distal functional enhancers.Conclusions:In summary,we generated the first functional enhancer map in PK15 and ST cells for the pig genome and highlight its potential roles in pig breeding.

文摘[Objective] This experiment aimed to compare piglets behavior under microbial fermentation bed breeding mode and traditional rearing mode. [Method] Pig behavior's statistical analysis under both microbial fermentation bed pig breeding mode and traditional rearing mode was se lected to study the effect of different breeding environment on pig's growth [ Result] The results showed that the duration of piglets lying and sleeping behavior accounted for the highest proportion of tested behavior, up to more than 70% ,followed by feeding and drinking which reached 15.96% (under microbial fermentation bed breeding mode) and 9.33% (under traditional rearing mode) respectively. Compared with traditional intensive rearing mode, piglet exploratory behavior's occurrence probability and duration increased significantly, by 28.62% and 12.21% respectively. Fighting behavior's occurrence probability and duration decreased significantly, by 49.83% and 91.26% respectively. Nutrient index and health index were introduced to evaluate piglet's general heath condition under microbial fermentation bed breeding mode, and results indicated that the health index of piglets under microbial fermentation bed breeding mode had slight difference from that under traditional mode, namely 10.83 and 10.03 respectively. But health index of piglets under microbial fermentation bed breeding mode was 245.12, much higher than 21.96 under traditional mode. [ Conclusion] Piglets would be healthier under microbial fermentation bed breeding mode.

文摘According to the relevant statistical data in National Agricultural cost-benefit Data Compilation(2001- 2013),we use the modified entropy weight- TOPSIS model to research the cost-benefit status of scale pig breeding in Shandong Province from the perspective of comparing it with that of the entire country and 9 other main pig producing areas. The results show that compared with the national average,the cost-benefit ratio of small scale pig breeding in Shandong Province is lower,while the cost-benefit ratio of medium scale and large scale pig breeding is significantly improved; the cost-benefit ratio of small scale pig breeding in Shandong Province is basically the same as that in 9 other main pig producing areas,while the cost-benefit ratio of medium scale and large scale pig breeding is higher; the output value of main products and the purchase price of piglet are two major bottlenecks restricting the cost-benefit improvement of scale pig breeding in Shandong Province.

The scale expansion of recent decade intensified the fluc- tuation of pig price in the short term but will stabilize the price in the long term. Therefore. it is necessary to expand breeding scale for maintaining long-term price stability and the optimal scale should be chosen according to the circumstances to ensure the social benefit maximization.
文摘Based on the general equilibrium theory of microeconomics,this study first analyzed the causes of sharp fluctuations in live pig prices,and then explored the financial capabilities of enterprises during the sharp fluctuations of live pig prices by using the financial data of 4 typical top listed enterprises from 2018 to 2021.By comparing the changes in the capabilities of enterprises,the impact of price on the financial capability of enterprises and differences were identified.The research results showed that the price of live pigs played a decisive role in enterprise profits,and there were huge differences in the fluctuation period.In the sharp increase period of price,price temptation is easy to cause enterprises to over-invest,resulting in excessive growth of enterprise assets,and increasing the business risk of enterprises.Based on the above conclusions,some policy suggestions were put forward to promote the stable development of industry from the three levels of enterprises,industries and government departments.
文摘Background: Colostrum is the first secretion produced by mammary glands during the hours immediately preceding and succeeding parturition. This secretion differs from milk and represents an essential vehicle of passive immunity,prebiotic compounds and growth factors involved in intestinal development. Most of the literature concerning colostrum composition refers mainly to human and cow; and little is known about pig colostrum metabolome and how it varies between pig breeds and different farrowing parity. Thus, the aim of the present research is to provide new information about pig colostrum composition and the associations between metabolites, the sows' breed and the survival and growth rates of their litters.Results: Colostrum samples were gathered from 58 parturitions of sows belonging to three different breeds chosen for their importance in Italian heavy pig production: 31 Large White, 15 Landrace and 12 Duroc respectively. The defatted and ultrafiltered colostrum samples were analysed using1 H–NMR spectroscopy. Principal Components Analysis(PCA) was assessed on the obtained spectra. In addition, using a Stepwise Regression and a Linear Regression analyses the metabolites named after the signals assignment were tested for their associations with piglets' performances.Twenty-five metabolites were identified, comprehending monosaccharides, disaccharides(such as lactose), organic acids(lactate, citrate, acetate and formate), nitrogenous organic acids(such as creatine) and other compounds,including nucleotides. PCA results evidence a clustering due to breed and season effects. Lactose was the main compound determining the assignment of the samples into different clusters according to the sow breed. Furthermore, some metabolites showed to be associated with piglets' performance and survival traits: acetate and taurine were positively related to litter weight gain and piglets' survival rate, respectively, while dimethylamine and cis-aconitate were linked to new-borns' impaired ability to survive.Conclusions: The results obtained suggest that colostrum composition is affected by breed, which, together with environmental conditions, may cause changes in colostrum metabolites content with possible consequences on piglets' performances. Among the identified metabolites, acetate, taurine, dimethylamine and cis-aconitate showed consistent associations with piglets' survival rate and litter weight gain, implying that these compounds may affect new-borns' ability to survive.
文摘The method of PCR RFLP was used to analyze the effects of ESR and FSH β genotypes on the litter size,especially the corresponding changes with the difference of genotype distributions in the Min,Sanjiang and Junmu I pigs.The results show that the ESR and FSH β genes are the major genes of litter size;the heterozygous genotype for ESR as well as for FSH β locus has a more litter size than others.However,the combined genotype ABDD means a more litter size than other genotypes,the frequencies of the heterozygous genotypes in Sanjiang and Junmu I are lower than in Min which maybe one of the most important reasons for the little litter sizes of those two breeds.

文摘Background:A large number of pig breeds are distributed around the world,their features and characteristics vary among breeds,and they are valuable resources.Understanding the underlying genetic mechanisms that explain across-breed variation can help breeders develop improved pig breeds.Results:In this study,we performed GWAS using a standard mixed linear model with three types of genome variants(SNP,InDel,and CNV)that were identified from public,whole-genome,sequencing data sets.We used 469 pigs of 57 breeds,and we identified and analyzed approximately 19 million SNPs,1.8 million InDels,and 18,016 CNVs.We defined six biological phenotypes by the characteristics of breed features to identify the associated genome variants and candidate genes,which included coat color,ear shape,gradient zone,body weight,body length,and body height.A total of 37 candidate genes was identified,which included 27 that were reported previously(e.g.,PLAG1 for body weight),but the other 10 were newly detected candidate genes(e.g.,ADAMTS9 for coat color).Conclusion:Our study indicated that using GWAS across a modest number of breeds with high density genome variants provided efficient mapping of complex traits.

文摘Background Long-term natural and artificial selection has resulted in many genetic footprints within the genomes of pig breeds across distinct agroecological zones.Nevertheless,the mechanisms by which these signatures contribute to phenotypic diversity and facilitate environmental adaptation remain unclear.Results Here,we leveraged whole-genome sequencing data from 82 individuals from 6 domestic pig breeds originating in tropical,high-altitude,and frigid regions.Population genetic analysis suggested that habitat isolation significantly shaped the genetic diversity and contributed to population stratification in local Chinese pig breeds.Analysis of selection signals revealed regions under selection for adaptation in tropical(55.5 Mb),high-altitude(43.6 Mb),and frigid(17.72 Mb)regions.The potential functions of the selective sweep regions were linked to certain complex traits that might play critical roles in different geographic environments,including fat coverage in frigid environments and blood indicators in tropical and high-altitude environments.Candidate genes under selection were significantly enriched in biological pathways involved in environmental adaptation.These pathways included blood circulation,protein degradation,and inflammation for adaptation to tropical environments;heart and lung development,hypoxia response,and DNA damage repair for high-altitude adaptation;and thermogenesis,cold-induced vasodilation(CIVD),and the cell cycle for adaptation to frigid environments.By examining the chromatin state of the selection signatures,we identified the lung and ileum as two candidate functional tissues for environmental adaptation.Finally,we identified a mutation(chr1:G246,175,129A)in the cis-regulatory region of ABCA1 as a plausible promising variant for adaptation to tropical environments.Conclusions In this study,we conducted a genome-wide exploration of the genetic mechanisms underlying the adaptability of local Chinese pig breeds to tropical,high-altitude,and frigid environments.Our findings shed light on the prominent role of cis-regulatory elements in environmental adaptation in pigs and may serve as a valuable biological model of human plateau-related disorders and cardiovascular diseases.

文摘[Objectives]This study was conducted to breed special pig breeds resistant to diarrhea by using modern biotechnology.[Methods]From Guizhou local breeds,such as Nuogu pigs,Kele pig,Yorkshire pigs and Duroc pigs,190 samples were collected for the analysis of anti-diarrhea gene.[Results]The anti-diarrhea genotype frequency of Kele pigs was 70.00%,which was higher than that of Nuogu pigs(67.37%)and Yorkshire pigs(Yorkshire pigs and Duroc pigs)(50.59%).The favorable anti-diarrhea gene of all Nuogu pigs,Kele pigs,and Yorkshire pigs and Duroc pigs was G,with gene frequencies of 0.7355,0.8368 and 0.8500,respectively,and the frequencies of allele A were 0.2645,0.1632 and 0.1500,respectively.In the process of generation selection,combination selection of GG♂×GG♀,GG♂×GA♀,GA♂×GG♀and GA♂×GA♀was conducted,and GG individuals were selected while gradually phasing out GA and AA individuals.The anti-diarrhea genotypes of 98 pigs in the offspring were tested,and it was found that the frequency of genotype GG was greatly improved,and the frequencies in Nuogu pigs,Kele pigs,Yorkshire pigs and Duroc pigs were increased to 73.91%,81.82%,85.25%and 66.67%respectively,thus forming a special anti-diarrhea breed.[Conclusions]This study provides a basis for selecting excellent breeding pigs,establishing core populations and screening resistance genes in the core populations and their offspring.

文摘To assess the genetic diversity between randomly and selectively bred populations,we sequenced 438 bp of the mitochondrial DNA control region from 102 pigs.These samples represented four native pig breeds,one nucleus and one conservation herd from Yunnan,China.Twenty haplotypes with sixteen polymorphic sites were identified.The number of haplotypes in the nucleus herd of Saba pig and the conservation herd of Banna miniature pig were restricted to three and one,respectively,while the randomly bred pig populations exhibited over six haplotypes.Notably,haplotype diversity in randomly bred populations was significantly greater than the selectively bred populations(h=0.732 vs.0.425 and 0,exact test,P≤0.0036).These findings demonstrate that selective breeding generated low genetic diversity compared to randomly bred pig breeds.A timely intervention and well programmed breeding approach would stop further genetic diversity reduction in the nucleus and conservation herds of native pig breeds.Otherwise,selective breeding would dramatically reduce genetic diversity in only several years,indicating that sharp contradictions exist between breeding,conservation and genetic diversity.Genetic relationships are discussed based on net genetic distances among pig populations.
文摘Dietary fiber is associated with impaired nutrient utilization and reduced net energy values. However, fiber has to be included in the diet to maintain normal physiological functions in the digestive tract. Moreover, the negative impact of dietary fiber will be determined by the fiber properties and may differ considerably between fiber sources. Various techniques can be applied to enhance nutritional value and utilization of available feed resources. In addition, the extent of fiber utilization is affected by the age of the pig and the pig breed. The use of potential prebiotic effects of dietary fiber is an attractive way to stimulate gut health and thereby minimize the use of anti-microbial growth promoters. Inclusion of soluble non-starch polysaccharides (NSP) in the diet can stimulate the growth of commensal gut microbes, inclusion of NSP from chicory results in changes in gut micro-environment and gut morphology of pigs, while growth performance remains unaffected and digestibility was only marginally reduced. The fermentation products and pH in digesta responded to diet type and were correlated with shifts in the microbiota. Interestingly, fiber intake will have an impact on the expression of intestinal epithelial heat-shock proteins in the pig. Heat-shock proteins have an important physiological role in the gut and carry out crucial housekeeping functions in order to maintain the mucosal barrier integrity. Thus, there are increasing evidence showing that fiber can have prebiotic effects in pigs due to interactions with the gut micro-environment and the gut associated immune system.

文摘Background:A growing body of evidence has revealed that the mammalian genome is organized into hierarchical layers that are closely correlated with and may even be causally linked with variations in gene expression.Recent studies have characterized chromatin organization in various porcine tissues and cell types and compared them among species and during the early development of pigs.However,how chromatin organization differs among pig breeds is poorly understood.Results:In this study,we investigated the 3D genome organization and performed transcriptome characterization of two adipose depots(upper layer of backfat[ULB]and greater omentum[GOM])in wild boars and Bama pigs;the latter is a typical indigenous pig in China.We found that over 95%of the A/B compartments and topologically associating domains(TADs)are stable between wild boars and Bama pigs.In contrast,more than 70%of promoterenhancer interactions(PEIs)are dynamic and widespread,involving over a thousand genes.Alterations in chromatin structure are associated with changes in the expression of genes that are involved in widespread biological functions such as basic cellular functions,endocrine function,energy metabolism and the immune response.Approximately 95%and 97%of the genes associated with reorganized A/B compartments and PEIs in the two pig breeds differed between GOM and ULB,respectively.Conclusions:We reported 3D genome organization in adipose depots from different pig breeds.In a comparison of Bama pigs and wild boar,large-scale compartments and TADs were mostly conserved,while fine-scale PEIs were extensively reorganized.The chromatin architecture in these two pig breeds was reorganized in an adipose depotspecific manner.These results contribute to determining the regulatory mechanism of phenotypic differences between Bama pigs and wild boar.

文摘Background:Oligosaccharides(OS)are indigestible carbohydrates naturally found in milk.The composition of porcine colostrum OS may influence the growth and the health of the neonate and consuming optimal concentrations of OS may reduce piglet susceptibility to illness.In this manner,targeted supplementation of animal feed with OS is being explored as a health management tool in the livestock industry.The variation in OS composition between different breeds of pig and its association with the litter performance is currently unknown.The aim of this study was to characterize the colostrum OS composition from sows of different breed and parity and correlate this data with sow maternal traits.Methods:Eighty-three colostrum samples from parities 1 to 8 were gathered from 3 different breeds of sow:44 Large White sows,27 Landrace sows and 12 Duroc sows.Samples were taken between the birth of the first and the last piglet from sows that were not pharmacologically induced to farrow.OS were purified from the samples and analysed by MALDI-ToF mass spectrometry(21 OS compositions detected).The farrowing season and the maternal data were recorded for each sow,including the number of live piglets and the litter body weight at birth,at day(d)3 and at weaning.Results:Five OS compositions,including isomers of the bifidogenic Sialyllactose,Lacto-N-Tetraose and Lacto-NHexaose series,were detected in all the samples.Twelve other OS were identified in at least 50%of samples,and their abundances were affected by breed(P<0.05;6 of 12),marginally affected by season(P<0.10;3 of 12)and never by parity number.The abundances of each OS component were standardized by Z-score scaling(μ=0 and SD=1),transformed by principal component analysis,and four similarity clusters were generated.Cluster membership was associated with litter weight gain within 3 days(P=0.063)and at weaning(P<0.05),but not with piglet mortality within 3 days.Conclusions:OS composition of colostrum may partially explain the variability in maternal performance within and between different breeds of sow.The obtained OS data can provide useful information for the development of novel prebiotic food supplements for suckling and weaning pigs.

文摘Over the past years, infectious disease has caused enormous economic loss in pig industry. Among the pathogens, gram negative bacteria not only cause inflammation, but also cause different diseases and make the pigs more susceptible to virus infection. Vaccination, medication and elimination of sick pigs are major strategies of controlling disease. Genetic methods, such as selection of disease resistance in the pig, have not been widely used. Recently, the completion of the porcine whole genome sequencing has provided powerful tools to identify the genome regions that harboring genes controlling disease or immunity. Immunogenornics, which combines DNA variations, transcriptorne, immune response, and QTL mapping data to illustrate the interactions between pathogen and host immune system, will be an effective genomics tool for identification of disease resistance genes in pigs. These genes will be potential targets for disease resistance in breeding programs. This paper reviewed the progress of disease resistance study in the pig focusing on Gram-negative bacilli. Major porcine Gram-negative bacilli and diseases, suggested candidate genes/pathways against porcine Gram-negative bacilli, and distributions of QTLs for immune capacity on pig chromosomes were summarized. Some tools for immunogenomics research were described. We conclude that integration of sequencing, whole genome associations, functional genomics studies, and immune response information is necessary to illustrate molecular mechanisms and key genes in disease resistance.

文摘The bactericidal/permeability increasing protein (BPI) has an important function of nonspecific killing of Gram-negative bacteria. In this study, qPCR was used to detect the expression of the BPI gene in twelve tissues of Meishan piglets from birth to weaning. BPI gene overexpression, bacterial adhesions count and indirect immunofluorescence were applied to analyze the relationship between BPI gene expression and the infectivity of Escherichia coli and Salmonella. The results showed that the BPI gene was expressed highly in duodenum, jejunum and ileum (fold changes of relative expression levels were more than 10 000, 500 and 200, respectively). The expression of the BPI gene at 35 days of age was significantly higher (P<0.01) than that at all other days. Transcription of the BPI gene was up-regulated 2 401-fold in porcine intestinal epithelial (IPEC-J2) cells that were transfected with the BPI gene overexpression lentivirus (IPEC-J2-BPI), and significantly higher (P<0.01) than that in negative control cells (IPEC-J2-NC). Protein expression levels in IPEC-J2-BPI cells were also increased. When IPEC-J2 cells were incubated with E. coli and Salmonella, respectively, for 2, 4, 6, 8, 10 and 12 h, the number of bacterial adhesions in IPEC-J2-BPI cells was significantly less (P<0.05) than that in IPEC-J2-NC cells. The results of indirect immunofluorescence analysis showed that the number of bacterial adhesions in IPEC-J2-BPI cells was significantly less (P<0.01) than that in IPEC-J2-NC cells. These results demonstrated that the BPI gene might play an important role in regulating weaning stress especially intestinal-mediated immune response. Overexpression of the BPI gene at the cellular level could significantly enhance the anti-bactericidal ability against Gram-negative bacteria such as E. coli and Salmonella. This has important biological significance in piglet resistance to bacterial diarrhea.
