Antibiotic resistance poses a significant global health threat, necessitating a thorough understanding of its prevalence in various ecological contexts. Medicinal plants, renowned for their therapeutic properties, host endophytic bacteria that produce bioactive compounds. Understanding antibiotic resistance dynamics in these bacteria is vital for human health and antibiotic efficacy preservation. In this study, we investigated antibiotic resistance profiles in endophytic bacteria from five medicinal plants: Thankuni, Neem, Aparajita, Joba, and Snake plant. We isolated and characterized 113 endophytic bacteria, with varying resistance patterns observed against multiple antibiotics. Notably, 53 strains were multidrug-resistant (MDR), with 14 exhibiting extensive drug resistance (XDR). Thankuni-associated bacteria displayed 44% MDR and 11% XDR, while Neem-associated bacteria showed higher resistance (60% MDR, 13% XDR). Aparajita-associated bacteria had lower resistance (22% MDR, 6% XDR), whereas Joba-associated bacteria exhibited substantial resistance (54% MDR, 14% XDR). Snake plant-associated bacteria showed 7% MDR and 4% XDR. Genus-specific distribution revealed Bacillus (47%), Staphylococcus (21%), and Klebsiella (11%) as major contributors to MDR. Our findings highlight diverse drug resistance patterns among plant-associated bacteria and underscore the complexity of antibiotic resistance dynamics in diverse plant environments. Identification of XDR strains emphasizes the severity of the antibiotic resistance problem, warranting further investigation into contributing factors. 展开更多

Phyllosphere microorganisms are a crucial component of environmental microorganisms, highly influenced by host characteristics, and play a significant role in plant health and productivity.Nonetheless, the impact of host characteristics on shaping phyllosphere microbial communities of plants with different life forms remains ambiguous.Utilizing high-throughput sequencing technology, this study analyzed the diversity and community composition of phyllosphere epiphytic microorganisms(e.g.,bacteria and fungi)of various plant life forms in the hinterland of the Gurbantunggut Desert,Northwest China.Functional annotation of prokaryotic taxa(FAPROTAX)and fungi function guild(FUNGuild)were employed to assess the ecological functions of microorganisms and to investigate the role of stochastic and deterministic processes in shaping phyllosphere microbial communities.Result showed a diverse array of phyllosphere epiphytic microorganisms in the desert plants, with Proteobacteria,Cyanobacteria, and Actinobacteriota dominating bacterial community, while Ascomycota and Basidiomycota were prevalent in fungal community.Comparison across different plant life forms highlighted distinct microbial communities, indicating strong filtering effects by plant characteristics.FAPROTAX prediction identified intracellular parasites(accounting for 27.44%of bacterial community abundance),chemoheterotrophy(10.12%),and phototrophy(17.41%)as the main functions of epiphytic bacteria on leaves of different life form plants.FUNGuild prediction indicated that phyllosphere epiphytic fungi primarily served as Saprotrophs(81.77%),Pathotrophs(17.41%),and Symbiotrophs(0.82%).Co-occurrence network analysis demonstrated a predominance of positive correlations among different microbial taxa.Raup-Crick dissimilarity index analysis revealed that deterministic processes predominantly influenced phyllosphere bacterial and fungal community assembly.Variance partitioning analysis and random forest modeling suggested that plant leaf functional traits significantly impacted both bacterial and fungal community composition, with fungal community composition showing a closer association with leaf nutrients and physiology compared with bacterial community composition. The distinct responses of bacterial and fungal communities to plant traits were attributed to the differing properties of bacteria and fungi, such as bacteria having higher potential dispersal rates and broader ecological niches than fungi.Overall, the results indicate that phyllosphere bacterial and fungal communities undergo similar community assembly processes, with fungi being more influenced by plant characteristics than bacteria. These findings offer novel insights into the ecology of phyllosphere microbial communities of desert plants. 展开更多

Positive associations between alpine cushion plants and other species have been extensively studied. However, almost all studies have focused on the associations between macrofauna.Studies that have investigated positive associations between alpine cushion plants and rhizospheric microbes have been limited to the vegetation growing season. Here,we asked whether the positive effects that alpine cushion plants confer on rhizospheric microbe communities vary with seasons. We assessed seasonal variations in the bacterial diversity and composition in rhizosphere of two alpine cushion plants and surrounding bare ground by employing a high throughput sequencing method targeting the V3 region of bacterial 16 S rRNA genes.Soil properties of the rhizosphere and the bare ground were also examined. We found that cushion rhizospheres harbored significantly more C,N,S,ammonia nitrogen, and soil moisture than the bare ground.Soil properties in cushion rhizospheres were not notably different, except for soil pH.Bacterial diversities within the same microhabitats did not vary significantly with seasons. We concluded that alpine cushion plants had positive effects on the rhizospheric bacterial communities, even though the strength of the effect varied in different cushion species.Cushion species and the soil sulfur content were probably the major factors driving the spatial distribution and structure of soil bacterial communities in the alpine communities dominated by cushion plants. 展开更多

This article describes a coherent biocommunication categorization for the kingdoms of bacteria, fungi and plants. The investigation further shows that, besides biotic sign use in trans-,inter-and intraorganismic communication processes,a common trait is interpretation of abiotic influences as indicators to generate an appropriate adaptive behaviour.Far from being mechanistic interactions, communication processes within organisms and between organisms are sign-mediated interactions.Sign-mediated interactions are the precondition for every cooperation and coordination between at least two biological agents such as cells, tissues, organs and organisms.Signs of biocommunicative processes are chemical molecules in most cases. The signs that are used in a great variety of signaling processes follow syntactic(combinatorial) ,pragmatic(context-dependent) and semantic(contentspecific) rules. These three levels of semiotic rules are helpful tools to investigate communication processes throughout all organismic kingdoms. It is not the aim to present the latest empirical data concerning communication in these three kingdoms but to present a unifying perspective that is able to interconnect transdisciplinary research on bacteria, fungi and plants. 展开更多

Phenazines are secondary metabolites with broad spectrum antibiotic activity and thus show high potential in biological control of pathogens. In this study, we identified phenazine biosynthesis (phz) genes in two genome-completed plant pathogenic bacteria Pseudomonas syringae pv. tomato (Pst) DC3000 and Xanthomonas oryzae pv. oryzae (Xoo) PXO99A. Unlike the phz genes in typical phenazine-producing pseudomonads, phz homologs in Pst DC3000 and Xoo PXO99A consisted of phzC/D/E/F/G and phzC/E1/E2/F/G, respectively, and the both were not organized into an operon. Detection experiments demonstrated that phenazine-l-carboxylic acid (PCA) of Pst DC3000 accumulated to 13.4 IJg L-1, while that of Xoo PXO99A was almost undetectable. Moreover, Pst DC3000 was resistant to 1 mg mL-1 PCA, while Xoo PXO99A was sensitive to 50 IJg mL ~ PCA. Furthermore, mutation of phzF blocked the PCA production and significantly reduced the pathogenicity of Pst DC3000 in tomato, while the complementary strains restored these phenotypes. These results revealed that Pst DC3000 produces low level of and is resistant to phenazines and thus is unable to be biologically controlled by phenazines. Additionally, phz-mediated PCA production is required for full pathogenicity of Pst DC3000. To our knowledge, this is the first report of PCA production and its function in pathogenicity of a plant pathogenic P. syringae strain. 展开更多

A total of 5 biochemically characterized lipase positive bacterial strains were screened for Polyhydroxyalkanoates(PHA)production by Nile blue staining and confirmation was done by Sudan Black B.PHA production ability for all strains was optimized followed by time profiling calculation and comparison via using glucose and two plant oils i.e.,canola and mustard oil.Quantitative analysis showed that glucose can serve as a carbon source for maximum biomass(2.5 g/L CDW for strain 5)and PHA production(70.3%for strain 2).PHA produced by strain 2 was further analyzed for its chemical composition and type via Fourier Transform Infrared(FT-IR)spectroscopy.It revealed homopolymer(PHB)and copolymer(PHB-co-PHV)production of PHA(peaks at 1743 cm-1 and 2861 cm-1,respectively)with both canola and mustard oil unlike glucose which produced only homopolymer one i.e.,PHB(peaks at 1110 cm-1,1411 cm-1 and 1650 cm-1).Crystallinity of FT-IR analyzed PHA was calculated using mathematical formulas which showed decrease from glucose to canola to mustard oil. This study revealed that plant oils can serve as better carbon source to produce better quality(ductile and copolymer)PHA.Moreover,16S rRNA gene sequencing analysis showed that strain 1,strain 2,strain 3,strain 4 and strain 5 are Stenotrophomonas sp.N3,Exiguobacterium sp.N4,Exiguobacterium sp.Ch3,Cellulosimicrobium sp.A8 and Klebsiella sp.LFSM2,respectively. 展开更多

The bacteria and plants were associated to remove diesel oil pollutants from soil.Three efficiently degrading bacteria(named strains Q10,Q14 and Q18,respectively) were isolated. Two plants(alfalfa and Indian mustard) were selected to form the association.Biodegradation of diesel oil pollutants in soil was accelerated by bacteria-plants association. The main results are summarized as follows. The plants-bacteria association was more effective in biodegradation of diesel oil pollutants in soil than in respective experiments carried out with plants or bacteria alone.Strain Q18-Indian mustard association resulted in the maximum diesel oil reduction(69.18%).The activities of catalase and polyphenol oxidase in soil were enhanced and microbial populations in soil, especially in rhizosphere, were also stimulated in the treatment of bacteria-plant association.Overall, the soil conditions might be improved by alfalfa or Indian mustard to benefit the growth of bacteria, which resulted in degradation of diesel oil pollutants more effective by the bacteria-plant association. The bacteria-plants association may be a better approach to the removal of diesel oil pollutants from soil. 展开更多

Endophytic bacteria of halophytic plants play essential roles in salt stress tolerance. Therefore,an understanding of the true nature of plant-microbe interactions under extreme conditions is essential. The current study aimed to identify cultivable endophytic bacteria associated with the roots and shoots of Seidlitzia rosmarinus Ehrenb.ex Boiss.grown in the salt-affected soil in Uzbekistan and to evaluate their plant beneficial traits related to plant growth stimulation and stress tolerance.Bacteria were isolated from the roots and the shoots of S.rosmarinus using culture-dependent techniques and identified by the 16S rRNA gene.RFLP(Restriction Fragment Length Polymorphism)analysis was conducted to eliminate similar isolates. Results showed that the isolates from the roots of S.rosmarinus belonged to the genera Rothia,Kocuria,Pseudomonas,Staphylococcus,Paenibacillus and Brevibacterium. The bacterial isolates from the shoots of S.rosmarinus belonged to the genera Staphylococcus,Rothia,Stenotrophomonas,Brevibacterium,Halomonas,Planococcus,Planomicrobium and Pseudomonas, which differed from those of the roots.Notably,Staphylococcus,Rothia and Brevibacterium were detected in both roots and shoots, indicating possible migration of some species from roots to shoots. The root-associated bacteria showed higher levels of IAA(indole-3-acetic acid)synthesis compared with those isolated from the shoots,as well as the higher production of ACC(1-aminocyclopropane-1-carboxylate)deaminase.Our findings suggest that halophytic plants are valuable sources for the selection of microbes with a potential to improve plant fitness under saline soils. 展开更多

Poor soil is one of the agricultural world’s principal challenges, inciting the use of chemical fertilizer’s to improve overall soil quality. However, the use of chemical fertilizer has significant and cascading environmental consequences. Therefore, the use of beneficial microbes’ inoculation in treating poor soil is a considerably ecofriendly sustainable solution. In the current study, we supplemented nutrient-deprived soil with plant growth promoting bacteria (PGPB), Pseudomonas fluorescens. The bacterial inoculations of Pseudomonas fluorescenswere added to the poor soil following two days post-sowing of Zea mays var. amylacea and Pennisetumamericanum p. seedlings. Metabolite analyses were conducted two months after treatment for both shoots and roots using nuclear magnetic resonance method (NMR). The data indicated significant changes in 19 metabolites relative to control in both plants shoot and roots. Among these metabolites, 7 were upregulated in roots of Zea mays var. amylacea, and 9 metabolites were upregulated in roots of Pennisetum americanum p. The PGPB enhanced sugars (fructose, glucose, sucrose) and amino acids (glutamate, alanine and succinate) in roots, while down regulating in shoots of Pennisetum americanum p. The Pseudomonas fluorescens induced, predominantly,Aminoacyl-tRNA related metabolite, and Alanine, aspartate and glutamate metabolite biosynthesis in Zea mays var. amylacea), whereas PGPB induced metabolites in Pennisetum americanum p., dominated by up regulated carbohydrate related (starch and sucrose) metabolites. The difference in some metabolic response between the two plants indicated that PGPB influence has a species-specific manner. 展开更多

Plant pathogenic bacteria are recognized to be harmful microbes able to decrease the quantity and quality of crop production in the world. Punica granatum peel was screened for its potential use as biological control agent for plant pathogenic bacteria. P. granatum peel was successfully extract using n-hexane, methanol and ethyl acetate by maceration. The highest yield obtained by ethyl acetate showed that ethyl acetate extracted more compounds that readily soluble to methanol and n-hexane. For in-vitro antibacterial activity, three different species of plant pathogenic bacteria were used namely Erwinia carotovorum subsp. Carotovorum, Ralstonia solanacearum, and Xanthomonas gardneri. For all crude extracts, four different concentrations 25, 50, 100 and 200 mg/ml were used in cup-plate agar diffusion method. Streptomycin sulfate at concentration 30 μg/ml was used as positive control while each respective solvent used for peel extraction was used as negative control. The results obtained from in vitro studies showed only ethyl acetate extract possessed antibacterial activity tested on the plant pathogenic bacteria. Methanol and n-hexane did not show any antibacterial activity against plant pathogenic bacteria selected where no inhibition zones were recorded. R. solanacearum recorded the highest diameter of inhibition zones for all range of concentrations introduced followed by E. carotovorum subsp. Carotovorum and X. gardneri. For the minimum inhbitory concentration (MIC) and minimum bactericidal concentration (MBC), only the ethyl acetate extract was subjected to the assay as only ethyl acetate extract exhibited antibacterial activity. The minimum concentration of ethyl acetate extract that was able to inhibit plant pathogenic bacteria was recorded at a concentration of 3.12 mg/ml which inhibited R. solancearum and E. carotovorum subsp. Carotovorum, followed by X. gardneri at concentration 6.25 mg/ml. For the minimum bactericidal concentration (MBC), the results showed that at the concentration of 12.5 mg/ml, the extract was still capable of killing the pathogenic bacteria, R. solanacearum, and P. caratovora sub.sp. caratovora while for the bacteria X. gardneri, the concentration that was able to kill the bacteria was 25 mg/ml. The qualitative estimation of phytochemical constituents within P. granatum L. ethyl acetate peel extracts had revealed the presence of tannins, flavonoids, phenols alkaloid, Saponins, and terpenoids. This study has demonstrated that Ethyl Acetate peel extracts of P. granatum has significant antibacterial activity against pathogenic plant bacterial, and it could be of high agricultural value. 展开更多

Endophytic bacteria are promising bacterial fertilizers to improve plant growth under adverse environment. For ecological remediation of coastal wetlands,it was necessary to investigate the effect and interaction of endophytes on halophytes under saline-alkali stress. In this study,an endophytic bacterium strain HK1 isolated from halophytes was selected to infect Suaeda glauca under pH(7 and 8)and salinity gradient(150,300 and 450mmolL^(-1)).Strain HK1 was identified as Pantoea ananatis and it had ability to fix nitrogen, dissolve inorganic phosphorus and produce indole-3-aceticacid(IAA).The results showed that strain HK1 could promote the growth of S.glauca seedings when the salinity was less than 300mmolL^(-1),in view of longer shoot length and heavier fresh weight. The infected plants could produce more proline to decrease the permeability of cells, which content increased by 26.2%–61.1%compared to the non-infected group. Moreover, the oxidative stress of infected plants was relieved with the malondialdehyde(MDA)content decreased by 16.8%–32.9%,and the peroxidase(POD)activity and catalase(CAT)activity increased by 100%–500%and 6.2%–71.4%,respectively.Statistical analysis revealed that increasing proline content and enhancing CAT and POD activities were the main pathways to alleviate saline-alkali stress by strain HK1 infection, and the latter might be more important. This study illustrated that endophytic bacteria could promote the growth of halophytes by regulation of osmotic substances and strengthening antioxidant activities. This finding would be helpful for the bioremediation of coastal soil. 展开更多

Leucine rich repeats (LRRs) are present in over 14,000 proteins that have been identified in viruses, bacteria, archaea, and eukaryotes. Two to sixty-two LRRs occur in tandem forming an overall arc shaped domain. There are eight classes of LRRs. Plant specific LRRs (class: PS-LRR) had previously been recognized in only plant proteins. However, we find that PS-LRRs are also present in proteins from bacteria. We investigated the origin of bacterial PS-LRR domains. PSLRR proteins are widely distributed in most plants;they are found in only a few bacterial species. There are no PS-LRR proteins from archaea. Bacterial PS-LRRs in twenty proteins from eleven bacterial species (in the three phyla: Proteobacteria, Cyanobacteria, and Bacteroidetes) are significantly more similar to the PS-LRR class than to the other seven classes of LRR proteins. Not only amino acid sequences but also nucleotide sequences of the bacterial PS-LRR domains show highly significant similarity with those of many plant proteins. The program, EGID (Ensemble algorithm for Genomic Island Detection), predicts that Synechococcus sp. CYA_ 1022 came from another organism. Four bacterial PS-LRR proteins contain AhpC-TSA, IgA peptidase M64, the immunoglobulin domain, the Calx-b domain, and the He_PIG domain;these domains show no similarity with any eukaryotic (plant) proteins, in contrast to the similarities of their respective PS-LRRs. The present results indicate that horizontal gene transfer (HGT) of genes/gene fragments encoding PS-LRR domains occurred between bacteria and plants, and HGT among the eleven bacterial species, of the three phyla, as opposed to descent from a common ancestor. There is the possibility of the occurrence of one HGT event from plant to bacteria. A series of HGTs might then have occurred recently and rapidly among these eleven species of bacteria. 展开更多
