In previous study we reported that pretreatment with plasmolysis enhanced somatic embryo formation in hypocotyls of Eleutherococcus senticosus.In the present study,the expression level of callose synthase gene in embryos of E.senticosus in response to 2,4-D,sucrose and mannitol treatments was analyzed by RT-PCR.The results show that plasmolysis pretreatment using sucrose and mannitol significantly promoted the expression of callose synthase gene.Also,the thicker cell walls of explant plasmolyzed compared with controls were observed during the somatic embryogenesis.We suggest that the callose may make the cells in epidermis separate from neighboring cells and then develop into embryogenic potential cells. 展开更多

Effects of Low Temperature on Freezing Injury of Various Winter Wheat Cultivars at Different Sowing Time 被引量:1

Freezing injury is one of the major disasters for the production of winter wheat in the North China Plain, which leads to a significant decrease of wheat yield. This study was conducted to investigate the impact of subfreezing temperature on freezing injury of various winter wheat cultivars at different sowing time. Three wheat cultivars, including Zhengmai 9023, Wanmai 48 and Wanmai 50, were sowed on 25 September and 5 October, respectively. Plant anatomy was applied to investi- gate the impact of subfreezing temperature on cells of three wheat cultivars, results showed that severe plasmolysis occurred in wheat sowed earlier suddenly encoun- tered subfreezing temperature without cold acclimation, compared with wheat sowed at proper sowing time. The degree of plasmolysis of different cultivars and tissues of wheat had significant differences and showed positive correlation with subfreezing temperature. Wanmai 50 had the highest cold resistance compared with Zhengmai 9023 and Wanmai 48, and there was no significant difference between Zhengmai 9023 and Wanmai 48. This study concluded that wheat cold resistance may be im- proved by adopting proper cultivars and sowing dates. 展开更多

Alterations of Intracellular Ca^(2+) Concentration and Ultrastructure in Spruce Apical Bud Cells during Seasonal Transition 被引量:4

Potassium antimonite was used to localize Ca2+ in the apical bud cells of spruce from July 1999 to May 2000. During the period of active growth (July 14), Calcium precipitates, an indication of Ca2+ localization, were mainly distributed in vacuoles, intercellular spaces and cell walls. Few Ca2+ deposits localized in the cytosol and nucleus, showing a low level of the cytosolic and nuclear Ca2+ concentration in the warm summer. In August, some Ca2+ deposits appeared in the cytosol and nuclei, indicating that Ca2+ influx occurred in the cytosol and nucleus as the day length became shorter. From September to November, high levels of the cytosolic and nuclear Ca2+ remained. During the mid-winter (December and January), the distribution of Ca2+ deposits and the ultrastructures in the cells were altered dramatically. Plasmolysis occurred in many cells due to the protoplasmic dehydration. In addition plasmalemma invagination and nuclear chromatin aggregation also occurred. A large number of Ca2+ deposits appeared in the space between the plasmalemma and the cell wall. And also some Ca2+ deposits were distributed in the plastids. However, few Ca2+ deposits were observed in the cytosol and nuclei. By spring of the next year (May), when plants were de-acclimated and resumed active growth, Ca2+ subcellular localization essentially restored to that observed in July of the last year, i.e., the cells contained low cytosolic and nuclear Ca2+ concentrations; Ca2+ deposits were mainly distributed in the vacuoles, cell walls and intercellular spaces. The relationships between the seasonal changes of intracellular Ca2+ concentration and the development of dormancy/cold acclimation, as well as plasmolysis associated with dormancy and cold hardiness were discussed. 展开更多

The carbon dioxide(CO_(2))conversion to useable compounds remains a great contest to scientists,engineers,and environmentalists with regard to the reverse of the oxidative degradation of organics.This conversion is essential for the development of complementary fuels and raw materials for various industries,which in turn will help in avoiding the drastic increase in tropospheric temperature due to greenhouse effect leading to global warming.The solar energy is the earth's essential power source along with the other various forms of energy for example fossil fuels,hydropower,wind,and biomaterials,etc.The final goal is to establish the artificial photosynthesis,which can be replicated thru various chemical reduction techniques of CO_(2)by employing appropriate photo-,thermal-and electro-catalysts in order to produce different one carbon atom(C_(1))and higher carbon atoms containing products.Besides,the utilization of clean and sustainable CO_(2)towards high-value products is of great interest today due to the recognized environmental worries and subsequent lessening of the fossil fuels utilization load to meet the energy demand of mankind.This way,solar energy can directly and/or indirectly be altered and stored in chemical energy form for industrial as well as societal applications.In this article our endeavor is to summarize the advances in CO_(2)chemical reduction research area till date especially in free radical-based methods such as electrochemical,photochemical and plasma chemical for the development of carbon species up to two carbon(C_(2))atoms containing products perceived in the chemical reduction of CO_(2).The author hopes that this piece of work will be helpful to researchers and readers who are focused on the field of CO_(2). 展开更多
