The currently available neuroprotective drugs fail to achieve desired therapeutic efficacy mainly due to short circulation half-life and poor blood−brain barrier (BBB) permeability. For that, an edaravone-loaded pH/glutathione (pH/GSH) dual-responsive poly(amino acid) nanogel (NG/EDA) was developed to improve the neuroprotection of EDA. The nanogel was triggered by acidic and EDA-induced high-level GSH microenvironments, which enabled the selective and sustained release of EDA at the site of ischemic injury. NG/EDA exhibited a uniform sub-spherical morphology with a mean hydrodynamic diameter of 112.3 ± 8.2 nm. NG/EDA efficiently accumulated at the cerebral ischemic injury site of permanent middle cerebral artery occlusion (pMCAO) mice, showing an efficient BBB crossing feature. Notably, NG/EDA with 50 µM EDA significantly increased neuron survival (29.3%) following oxygen and glucose deprivation by inhibiting ferroptosis. In addition, administering NG/EDA for 7 d significantly reduced infarct volume to 22.2% ± 7.2% and decreased neurobehavioral scores from 9.0 ± 0.6 to 2.0 ± 0.8. Such a pH/GSH dual-responsive nanoplatform might provide a unique and promising modality for neuroprotection in ischemic stroke and other central nervous system diseases.展开更多

It is well known that the taken of aspirin for long-term increases the risk of serious gastrointestinal complications, therefore a controllable delivery of aspirin is of importance to lighten those side effects. This work is of interest because it provides a continuous route to prepare PLA-aspirin microspheres continuously with a high drug encapsulation efficiency.展开更多

Retrograde tracing, behavioral testing and histomorphometric analyses showed that compared with the empty PLGA conduit implantation group, the SPC implantation group had a larger number of growing and extending axons, a markedly increased diameter of regenerated axons and a greater thickness of the myelin sheath in the conduit. Furthermore, there was an increase in the size of the neuromuscular junction and myofiber diameter in the target muscle. These findings suggest that the novel artificial SPC nerve graft can promote axonal regeneration and remyelination in the transected peripheral nerve and can be used for repairing peripheral nerve injury.展开更多

A mechanism for the synthesis of P(AM-co-AA) hydrogel was proposed. To optimize the synthesis condition, the following parameters were examined in detail: the discharge voltage, discharge time, the content of the crosslinker, and the mass ratio of AM to AA. The results showed that the optimum pH range for cationic dyes removal was found to be 5.0-10.0. The P(AM-co-AA) hydrogel exhibits a very high adsorption potential and the ex- perimental adsorption capacities for Crystal violet (CV) and Methylene blue (MB) were 2974.3 mg/g and 2303.6 mg/g, respectively. The adsorption process follows a pseudo-second-order kinetic model. In addition, the adsorption mechanism of P(AM-co-AA) hydrogel for cationic dyes was also discussed.展开更多

However, isolation and puriifcation of Schwann cells are complicated by contamination with ifbroblasts. Current reported measures are mainly limited by either high cost or complicated procedures with low cell yields or purity. In this study, we collected dorsal root ganglia from neonatal rats from which we obtained highly puriifed Schwann cells using serum-free melanocyte culture medium. The purity of Schwann cells (〉95%) using our method was higher than that using standard medium containing fetal bovine serum. The obtained Schwann cells were implanted into poly(lactic-co-glycolic acid)/chi-tosan conduits to repair 10-mm sciatic nerve defects in rats. Results showed that axonal diameter and area were signiifcantly increased and motor functions were obviously improved in the rat sciatic nerve tissue. Experimental ifndings suggest that serum-free melanocyte culture medium is conducive to purify Schwann cells and poly(lactic-co-glycolic acid)/chitosan nerve conduits combined with Schwann cells contribute to restore sciatic nerve defects.展开更多

In this study, p-dioxanone (PDO) was employed to copolymerize with DL-lactide (LA) via ring-opening melt polymerization using Sn(Oct)2 as an initiator and ethylene glycol as a co-initiator. The obtained degradable macrodiols (HO-P(LA-co-PDO)-OH) were just such a copolymer consisting of PLA, GA and EG. 1HNMR was employed to characterize the copolymers, and the effect of PDO/LA molar ratios in the feedstock on the molecular weights of HO-P(LA-co-PDO)-OH was investigated by means of endhydroxyl analysis, 1H NMR or GPC-MALLs. The results confirmed the successful synthesis of HO-P(LA-co-PDO)-OH and revealed that one end-hydroxyl of the micarodiols was donated by LA or PDO and the other one by the co-initiator EG. In addition, the molecular weights of HO-P(LA-co-PDO)-OH increased with decreasing PDO/LA ratios.展开更多

The results show that PLA and ATPS were partially miscible, which was confirmed with the measurement of Tg by dynamic mechanical analysis(DMA) and differrential scanning calorimetry(DSC). The mechanical and thermal properties of the blends were improved. With increasing the ATPS content, the elongation at break and impact strength were increased. The elongation at break increased from 5% of neat PLA to 25% of the blend PLA/ATPS40. It was found that the cold crystallization behavior of PLA changed evidently by addition of ATPS. The cold crystallization temperature(Tcc) of each of PLA/ATPS blends was found to shift to a lower temperature and the width of exothermic peak became narrow compared with that of neat PLA. The thermogravimetry analysis(TGA) results showed that the peak of derivative weight for ATPS moved to higher temperature with increasing PLA content in PLA/ATPS blends. It can be concluded that PLA could increase the thermal stability of ATPS. The rheological measurement reveals the melt elasticity and viscosity of the blends decreased with the increased concentration of ATPS, which was favorable to the processing properties of PLA.展开更多

Three different carbonyl-bond orientations of side-groups resulted in the differences in depth of potential wells in their energetic contours for a meso or a racemic dyad. These discrepancies are interpreted as a result of various fine structures corresponding to grid search conformations as well as thereby different interactions. The analysis on the most stable conformations of PMAA confirmed that the ester groups are nearly perpendicular to the plane defined by the two adjacent skeletal bonds but may possibly change their relative orientations to meet the requirement of lower energy during the conformational state transition. For each polymer, two global energy maps of a meso and a racemic dyad were finally constructed from the superposition of energy data for the three kinds of side-group orientations by the Boltzmann factors. From an ensemble average, the proposed scheme with three rotational isomeric states (RIS) allowed us to access the experimentally unperturbed dimensions of PAA chain via the configurational statistical mechanics. Although the calculation was based on the short-range, local interactions, it was interested to note that the experimental characteristic ratios just fell within the range calculated for atactic chains.展开更多

Ibuprofen was loaded in the electrospun ultra-fine fibers. The results from environmental scanning electron microscopy (ESEM), wide angle X-ray diffraction (WAXD) and differential scanning calorimetry (DSC) demonstrated that ibuprofen could be perfectly entrapped in the fibers electrospun from PEG-PLLA using a-hydroxy octanoic acid or PEG-b-poly(a-hydroxy octanoic acid) (PEG-PHOA) as additives. Compared with electrospun PEG-PLLA fibers which entrapped 20 wt% ibuprofen, the PEG-PLLA electrospun fibers containing PEG-PHOA exhibited integral and robust after 1 week incubated in 37℃, pH 7.4 phosphate buffer solution with 10 μg/mL proteinase K. Compared with electrospun fibers without PEG-PHOA, the concentration ofproteinase K in release media had less effect on the release rate of ibuprofen. An unique release profile was found from PEG-PLLA fiber after the incorporation of PEG-PHOA. Enzyme degradation experiments demonstrated that PEG-PHOA but not a-hydroxy octanoic acid monomer was the crucial factor for integrity maintenance of the electrospun fibers, which may be due to the enzyme degradation tolerance property of the PEG-PHOA polymer additive.展开更多

The tissue engineering scaffold poly(D,L-lactide-co-glycolic acid) has good histocompatibility and can promote the growth of regenerating nerve fibers. The present study used small interfering RNA to silence Nogo-66 receptor gene expression in bone marrow mesenchymal stem cells and Schwann cells, which were subsequently transplanted with poly(D,L-lactide-co-glycolic acid) into the spinal cord lesion regions in rats. Simultaneously, rats treated with scaffold only were taken as the control group. Hematoxylin-eosin staining and immunohistochemistry revealed that at 4 weeks after transplantation, rats had good motor function of the hind limb after treatment with Nogo-66 receptor gene-silenced ceils prus the poly(O,L-lactide-co-glycolic acid) scaffold compared with rats treated with scaffold only, and the number of bone marrow mesenchymal stem cells and neuron-like cells was also increased. At 8 weeks after transplantation, horseradish peroxidase tracing and transmission electron microscopy showed a large number of unmyelinated and myelinated nerve fibers, as well as intact regenerating axonal myelin sheath following spinal cord hemisection injury. These experimental findings indicate that transplantation of Nogo-66 receptor gene-silenced bone marrow mesenchymal stem cells and Schwann cells plus a poly(D,L-lactide-co-glycolic acid) scaffold can significantly enhance axonal regeneration of spinal cord neurons and improve motor function of the extremities in rats following spinal cord injury.展开更多

It has been found to be a simple and effective approach to introduce specific recognition sites into synthetic polymers, namely, to create molecular imprinting polymers Remarkable features such as stability, ease of preparation and low cost, have made molecular imprinting polymers particularly attractive in chemical sensors, catalysis, drug delivery, and dedicated separations. Practical applications of molecular imprinting polymers require accessible sites, fast mass transfer, and quick binding. However, present techniques used to prepare molecular imprinting polymers most often result in materials exhibiting a high affinity and selectivity but a low capacity and poor site accessibility for the target molecules. It is also very difficult to remove the imprinted molecules located in these molecular imprinting polymers because the highly cross-linked structures do not allow the templates to move freely. To some extent, combining molecular imprinting technology with membrane separation and surface imprinting can overcome the shortcomings, such as mass transfer limitations and non-quantitative recovery of the template molecules seen for imprinted materials fabricated by conventional bulk methods. In that ease, it appears to us that molecular imprinting polymers with high surface area to volume ratios are particularly desirable for largescale applications. Eleetrospun nano and ultrafine fibrous membranes are the most suitable materials due to advantages such as: (1) large specific surfaces, providing relatively high imprinting sites per unit mass; (2) fine porous structures, resulting in the accessibility of imprinting sites and low diffusion resistance necessary for high efficiency; and (3) easy recoverability from practical operation or applicability for continuous usages. Therefore, in this work, we prepared a unique kind of imprinted material--molecularly imprinted fibrous membranes of poly ( acrylonitrile-co-acrylic acid) fabricated by means of an electrospinning process.展开更多

Combination of polymers modifies the physical properties of the conduits, altering their nerve-guidance properties. Conduits made from PLA-only or combined with other polymers have been used successfully for nerve repair, but their efficiency has not been compared. We compared the morphological and functional outcomes of peripheral nerve repair by using NGCs made of poly(lactic acid) and combined or not with polycaprolactone(PLA/PCL) or polyvinylpyrrolidone(PLA/PVP). To assess the functional recovery, we employed a mechanical hyperalgesia analysis, sciatic functional index(SFI), and electroneuromyography. The mechanical hyperalgesia analysis showed that the PLA group improved more rapidly than the PLA/PVP and PLA/PCL groups; similarly, in the electroneuromyography assay, the PLA group exhibited higher amplitude than the PLA/PCL and PLA/PVP groups. However, the SFI improvement rates did not differ among the groups. Morphologically, the PLA group showed more vascularization, while the nerve fiber regeneration did not differ among the groups. In conclusion, the PLA-only conduits were superior to the other NGCs tested for nerve repair.展开更多

文摘In this paper, poly (acrylamide-co-acrylic acid) (P(AM-co-AA)) hydrogel was pre- pared in an aqueous solution by using glow-discharge electrolysis plasma (GDEP) induced copoly- merization of acrylamide (AM) and acrylic acid (AA), in which N,N'-methylenebisacrylamide (MBA) was used as a crosslinker. In addition, the adsorption mechanism of P(AM-co-AA) hydrogel for cationic dyes was also discussed.

文摘The novel polymer metal chelate electrolytes(polychelates)were prepared by incorporation of cobalt sulfate(Co)into poly(acrylic acid)(PAA)host matrix.Quasi-solid state supercapacitor devices were fabricated using polychelates,PAA-Co X(X:3,5,7,and 10)where X represents the doping fraction(w/w)of Co in PAA.All polymer metal electrolytes were showed excellent bending-stretching properties, thermal stability and electrochemical durability with an optimum ionic conductivity of 3.15×10^(-4) S cm^(-1).Hierarchically porous activated carbon and nano-sized conductive carbon were used to form carbon composite symmetrical device electrodes. The electric double-layer capacitor(EDLC)and redox reactions of Co-incorporated polychelates at the interfaces of porous activated carbon provided an optimum specific capacitance of 341.33 F g^(-1) with a device of PAA-Co7,which is at least 15 times enhancement compared to the device of pristine PAA.The PAA-Co7 device also provided energy density of 21.25 Wh kg^(-1) at a power density of 117.69 W kg^(-1).A prolonged cyclic stability of the device exhibited superior capacitive performance after 10,000 charge-discharge cycles and the maintained 90%of its initial performance. In addition, the supercapacitor with a dimension of 1.5 cm×3 cm containing PAA-Co7 successfully operated the red-blue-green(RGB)LED light.

文摘Acidic poly(ionic liquid)s(PILs)with swelling ability were synthesized by free radical copolymerization of N-vinylimidazolium ionic liquids, divinylbenzene(DVB)and sodium acrylate(NaAA),and further acidification by sulfuric acid. The swelling ability of acidic PILs was greatly affected by cross-linker content and chain length of 3-alkyl-substituents on imidazolium.Cross-linked network structures could be observed from the cryogenic scanning electron microscopy(cryo-SEM)images of the swollen acidic PILs in formic acid.Acidic PILs with network structures in swollen state exhibited excellent activities in the esterification of cyclohexene and formic acid, and the catalytic activities were in positive correlation with their swelling abilities.Acidic PIL with 3-octyl-substituent and 2.5 mol%DVB(PIL-C8-2.5DVB-HSO4)had the highest swelling ability in formic acid and exhibited comparable catalytic activities with homogeneous catalysts such as sulfuric acid and p-toluenesulfonic acid.

文摘A novel conductive drug-loading system was prepared by using an improved emulsion electrostatic spinning method which contained polylactic acid (PLA),graphene oxide (GO),and nerve growth factor (NGF) coated with bovine serum albumin (BSA) nanoparticles.Firstly, the structure, mechanical properties, morphology and electrical conductivity of PLA/GO electro spun fiber membranes with different GO ratios were characterized.PLA/GO scaffolds can exhibit superior porosity, hydrophilic and biomechanical properties when the GO incorporation rate is 0.5%.The addition of GO in the PLA/GO electro spun fiber membranes can also create appropriate pH environment for the repair of injured nerve when the GO incorporation rate is above 0.5%.Secondly,PLA/GO/BSA/Genipin/NGF particles (with a ratio of BSA/NGF=3:1) prepared by modified emulsion electro spinning method will release more NGF than PLA/GO/NGF particles. In addition,PLA/0.5%GO/NGF scaffold can maintain its structure stability for at least 8 weeks observed by scanning electron microscope (SEM).Moreover, the degradation of PLA/0.5%GO/NGF scaffold is consistent with its weight loss. Finally,in vitro assay confirmes that PLA/GO composite scaffold exhibits low cytotoxicity to RSC96 cells.Cellular results have demonstrated that PLA/0.5%GO/NGF sustained-release drug sustained-release system with appropriate electrical stimulation (ES) can promote PC12 cell proliferation, and it can maintain its differentiation capability for at least 3 weeks. In conclusion,PLA/0.5%GO/NGF sustained-release drug sustained-release system can maintain its biological activity for at least 3 weeks and promote cell proliferation with appropriate ES.

文摘Schwann cells, nerve regeneration promoters in peripheral nerve tissue engineering, can be used to repair both the peripheral and central nervous systems. Experimental ifndings suggest that serum-free melanocyte culture medium is conducive to purify Schwann cells and poly(lactic-co-glycolic acid)/chitosan nerve conduits combined with Schwann cells contribute to restore sciatic nerve defects.

文摘Poly[(alanino ethyl ester)0.67 (glycino ethyl ester)0.33 phosphazene] (PAGP) was synthesized, and morphology and diameter of the electrospun PAGP nanofibers were systematically evaluated by using a cool field emission scanning electron microscope (SEM) with changing the important processing variables such as applied voltage, polymeric concentration, and ambient temperature. The average diameter of PAGP nanofibers was inversely proportional to the applied voltage, but increased with the increase of solution concentration. Lower environmental temperature was unfavorable due to the nanofibers conglutination.

文摘Integrating poly(lactic acid) (PLA), glycolic acid (GA) and ethylene glycol (EG) will hopefully result in a novel copolymer that combines such advantages as fastened and controllable release rate and improved flexibility together with good biocompatibility. In addition, the molecular weights of HO-P(LA-co-PDO)-OH increased with decreasing PDO/LA ratios.

文摘Blends of poly(lactic acid)(PLA) and thermoplastic acetylated starch(ATPS) were prepared by means of the melt mixing method. The rheological measurement reveals the melt elasticity and viscosity of the blends decreased with the increased concentration of ATPS, which was favorable to the processing properties of PLA.

文摘Poly(L-lactic acid)(PLLA)is a thermoplastic material with complete degradability, high biocompatibility and excellent mechanical properties. It can replace petroleum-based polymers are currently being used in the fields of packaging, agriculture, textiles, medical and so on.However,PLLA’s extremely flammability greatly limits its wider application. An bio-based flame retardant L-APP/PLLA composites was prepared by melt blending of the L-APP and PLLA.The morphology, impact properties, thermal properties and flame retardant properties of composites were investigated by field emission scanning electron microscope(SEM),impact tester, differential scanning calorimeter(DSC),thermogravimetric analyzer(TGA),limiting oxygen indexer(LOI)and horizontalvertical burning tester. The results showed that the degree of crystallization(X_(c))and LOI of L-APP/PLLA composites increased as increasing of L-APP content. What’s more, the impact strength first increased and then decreased, the glass transition temperature(T_(g))and melting temperature(T_(m))do not changed significantly. The impact strength of composites was 9.1 kJ/m^(2) at a 5 wt%loading for L-APP,which was the highest level. When the content of L-APP was 20%,the LOI was 30.8%,the Xc was 42.3%and the UL-94 level was V-0.This research can promote the value-added utilization of lignin and the application of PLLA in the fields of flame retardant materials.

文摘A full-relaxation optimization of molecule and the Dreiding force field are employed to obtain the geometry parameters and the conformational energy surfaces of meso or racemic dyad of poly(acrylic acid) (PAA) and poly(methacrylic acid) (PMAA). Although the calculation was based on the short-range, local interactions, it was interested to note that the experimental characteristic ratios just fell within the range calculated for atactic chains.

文摘Poly(a-hydroxy octanoic acid) was first used as an additive for the preparation of electrospun ultra-fine fibers of poly(ethylene glycol)-b-poly(L-lactide) (PEG-PLLA). Enzyme degradation experiments demonstrated that PEG-PHOA but not a-hydroxy octanoic acid monomer was the crucial factor for integrity maintenance of the electrospun fibers, which may be due to the enzyme degradation tolerance property of the PEG-PHOA polymer additive.

文摘Inhibition of neurite growth, which is in large part mediated by the Nogo-66 receptor, affects neural regeneration following bone marrow mesenchymal stem cell transplantation. These experimental findings indicate that transplantation of Nogo-66 receptor gene-silenced bone marrow mesenchymal stem cells and Schwann cells plus a poly(D,L-lactide-co-glycolic acid) scaffold can significantly enhance axonal regeneration of spinal cord neurons and improve motor function of the extremities in rats following spinal cord injury.
文摘IntroductionOver the past few decades, molecular imprinting has been described as a technology for preparing "molecular doors" which can be matched to "template keys". Therefore, in this work, we prepared a unique kind of imprinted material--molecularly imprinted fibrous membranes of poly ( acrylonitrile-co-acrylic acid) fabricated by means of an electrospinning process.

文摘Poly(lactic acid)(PLA)-containing nerve guidance conduits(NGCs) are currently being investigated for nerve repair as an alternative to autograft, which leads to permanent functional impairment in the territory innervated by the removed nerve. In conclusion, the PLA-only conduits were superior to the other NGCs tested for nerve repair.
