A water soluble crude polysaccharide has been isolated from the fruiting body of cultivated Cordyceps militarvs(L.) Link. Thei...A water soluble crude polysaccharide has been isolated from the fruiting body of cultivated Cordyceps militarvs(L.) Link. The GC analysis indicated that it contained the monosaccharides of Gal, Man, Glc and GlcA. Their molar ratio is 1 00∶1 58∶7 89∶0 19. The crude polysaccharide was fractionated with ethanol to get three fractions, which were CM Ⅰ, CM Ⅱ and CM Ⅲ. CM Ⅱ was further purified to get one homogeneous fraction named CMB. The molecular weight was estimated to be 60000 by means of gel filtration. CMB is a greyish white powder. The GC analysis indicated that it was composed of Gal, Man, Glc and the molar ratio is 1 00∶1 38∶5 10. Partial hydrolysis with an acid, periodate oxidation, Smith degradation, IR, GC and methylation analyses, GC/MS analyses were used for the structure analyses of CMB. The results reveal that CMB is of a lower branched structure. The main chain is composed of (1→6) linked Man, (1→6) linked Glc, (1→4) linked Glc and (1→4) linked Gal. Some of (1→4) linked Glc and all (1→4) linked Gal residues are substituted at 3 O and 6 O respectively. (1→4) Linked Glc residues without branch probably exist with other monosaccharides alternately. On an average, there is one branch every ten hexose residues. The side chain is composed of major (1→4) linked Glc and minor (1→6) linked Glc. The nonreduced end is composed of Gal and Glc. The main glycosidic link of CMB is α configuration.展开更多

This plant has good potential as a donor plant in breeding programme for Phytophthora to foot rot resistance in the cultivated species, P. nigrum. This project was carried out to reveal the optimum time to collect pollen of P. colubrinum for artificial pollination. Through this study, anther dehiscence was proved occurred around 9 am. Based on the result obtained from study on the time of anther dehiscence, the pollen was classified into five stages for testing on the viability, i.e., stage 1, before anther dehiscence, i.e., around 7 am (pollen obtained by crushing on the anther); stage 2: immediately after anther dehiscence, i.e., around 9 am; stage 3: two hours after anther dehiscence, i.e., around 11 am; stage 4: four hours after anther dehiscence, i.e., around 1 pm; and stage 5: six hours after anther dehiscence, i.e., around 3 pm. Data obtained from pollen germination and pollen tube growth study showed that pollen of P. colubrinum was the most viable at two hours after anther dehiscence and onward. It was also proved that the viability can be maintained up to at least eight hours. Pollen obtained before anther dehiscence by crushing on the anther was found less viable with lower percentage of pollen germination and retarded pollen tube growth. In other words, the experiment enlightened that artificial pollination between P. nigrum L. and P. colubrinum Link was ideally to be carded out approximately starting from 11 am until 5 pm, considering the viability of pollen ofP. colubrinum.展开更多
Multilayer film was fabricated on an electrode surface by alternate layer-by-layer(LBL) adsorption of polycationic redox polymer(PEI-Fc) and dye-linked L-proline dehydrogenase(L-proDH).The electrochemistry of th...Multilayer film was fabricated on an electrode surface by alternate layer-by-layer(LBL) adsorption of polycationic redox polymer(PEI-Fc) and dye-linked L-proline dehydrogenase(L-proDH).The electrochemistry of the PEI-Fc/L-proDH multilayer modified electrode was investigated by cyclic voltammetry,and the enzyme catalysis mediated by the redox polymer was studied in a solution containing L-proline.It was observed that electron communication between L-proDH and the electrode was achieved with the help of PEI-Fc.A mathematical expression for the current response was evaluated based on the Michaelis-Menten kinetics mode,and the calculated currents fitted well with the experimental data.The kinetic analysis indicates that only a small fraction of the immobilized enzyme was efficiently electrically wired by the redox polymer.展开更多
