Sequence analysis has revealed a DNA motif (GGGGAATTTGCT) similar to NF-кB consensus sequence resides in the negative regulatory element (-3028bp~ -2902bp, termed ε-NRAII) 5' to the cap site of this gene. NRF DNA fragment (-3010bp~ -2986bp) containing the NF-кB motif similar sequence was synthesized and used in electrophoresis mobility shift assay (EMSA) and competitive analysis. Data showed that a protein factor from nuclear extracts of K562 cells specifically interacted with NRF DNA fragment. The synthetic NF DNA fragment (containing NF-кB consensus sequence) could competed for the protein binding, but MNF DNA fragment (mutated NF-кB motif) could not, suggesting that the binding protein is a member of NF-кB/Rel family. Western blot assay demonstrated that the molecular weight of NF-кB protein in the nuclei of K562 cells is 50ku. We suggested that NF-кB p50 may play an important role in the regulation of human c-globin gene expression.展开更多

The results show that it is necessary to consider the nonlinear property of soil and to set interface elements between piles and soil in analysis of the negative skin friction of piles. Among a pile group, the pile located above the tunnel centerline bears larger downdrag than the ones far away from tunnel centerline.展开更多

Compared with other mantle-originated rocks, these basalts have lower PGE contents and wider ranges of primitive mantle-normalized ratios of palladium content to iridium one, palladium content to platinum one and palladium content to gold one exhibiting relative platinum and iridium depletion. Characteristics of PGE patterns indicated that the studied Mariana Trough basalts originated from low partial melting, and the MORB mantle beneath the spreading center had been contaminated by the are-island mantle. In the aspect of trace elements, Mariana Trough basalts showed the enrichment of LILE, lead and LREE, indicating that they had been influenced by subduetion compositions. All these demonstrated that Mariana Trough basalts are products of partial melting from a mixed mantle ( the contamination of MORB mantle by are-island mantle).展开更多

According to the characteristics of this method, however, it has been found out that the deficiency of the algorithms is to easily cause inaccurate simulation in the vicinity of free surface owing to the penalty function with only one controlling penalty parameter. Therefore, an improved cut-off pressure method was presented by constructing penalty function with two controlling penalty parameters. The improved algorithms were illustrated to be able to improve the simulation accuracy and raise the convergence speed through considering the effects of the partially saturated or unsaturated elements more reasonably and calculating the nodal external forces more accurately. Finally, the improved cut-off negative pressure method was applied to the simulation of unsteady seepage flow with free surface, and the corresponding program codes were developed with validation of some cases.展开更多
