In order to investigate the regulative function of telom erase and phosphorylated (acti- vated) extracelluar regulated protein kinase (ERK) 1and 2 in the leukemic cell lines HL - 6 0 and K5 6 2 proliferation inhibit...In order to investigate the regulative function of telom erase and phosphorylated (acti- vated) extracelluar regulated protein kinase (ERK) 1and 2 in the leukemic cell lines HL - 6 0 and K5 6 2 proliferation inhibition and apoptosis,three chemotherapeutic drugs Harringtonine(HRT) , Vincristine(VCR) and Etoposide(Vp16 ) were selected as inducers.The proliferation inhibition rate was detected by MTT m ethod,the cell cycle and cell apoptosis was analyzed by flow cytometry and the telom erase activity was detected by the telom eric repeat am plification protocol(TRAP) assay and bioluminescence analysis method.The phosphorylated ERK 1/ 2 protein expression was detected by western blot method.The results showed that HRT,VCR and Vp16 could inhibit cell proliferation,induce apoptosis,inhibit telomerase activity and down- regulate the protein expres- sion of phosphorylated ERK.Itwas suggested that ERK signal transduction pathway was involved in the down- regulation of telomerase activity and the onset of apoptosis in the leukem ic cells treat- ed by HRT,VCR and Vp16 .展开更多
Objective:To investigate the expression levels and clinical prognosis of tyrosine protein kinase-7(PTK7),human epidermal growth factor receptor 2(HER-2)and minichromosome maintenance protein 5(Mcm5)in esophageal squam...Objective:To investigate the expression levels and clinical prognosis of tyrosine protein kinase-7(PTK7),human epidermal growth factor receptor 2(HER-2)and minichromosome maintenance protein 5(Mcm5)in esophageal squamous cell carcinoma(ESCC)value.Methods:180 patients with ESCC were enrolled as the study subjects.Tumor tissues were collected and 100 paracancerous tissues were randomly collected.The expression levels of PTK7,HER-2 and Mcm5 proteins were detected by immunohistochemical staining,and their clinical pathological features and prognosis.Relationship.Results:The positive rates of PTK7,HER-2 and Mcm5 in ESCC tissues were significantly higher than those in adjacent tissues(P<0.05).The positive rate of PTK7 in lymphatic metastasis was significantly higher than that in lymphatic metastasis(P<0.05).The tumor body diameter was greater than 2 cm,the TNM stage was stage III and IV,and the HER-2 positive rate of ESCC tissues with lymphatic metastasis was significantly increased(P<0.05).The positive rate of Mcm5 in stage III and IV and combined lymphatic metastasis ESCC was higher in TNM stage(P<0.05).The expression level of PTK7 in ESCC tissues was positively correlated with lymphatic metastasis(P<0.05).HER-2 was positively correlated with TNM stage and lymphatic metastasis(P<0.05).Mcm5 was positively correlated with tumor size,TNM stage and lymphatic metastasis(P<0.05).The tumor-free survival of PTK7,HER-2,and Mcm5-positive ESCC patients was significantly shorter than that of the negative group(P<0.05).Conclusion:The expression levels of PTK7,HER-2,and Mcm5 were significantly up-regulated in ESCC tissues,and high levels of HER-2 and Mcm5 predicted more severe pathological stage and lymphatic metastasis,and PTK7,HER-2,Mcm5 and worse.Prognosis is related.展开更多
In order to add value to potato peels and also curb their environmental pollution problems, this study investigated the protein enrichment of potato peels with Saccharomyces cerevisiae via Solid-State Fermentation (SS...In order to add value to potato peels and also curb their environmental pollution problems, this study investigated the protein enrichment of potato peels with Saccharomyces cerevisiae via Solid-State Fermentation (SSF). SSF is a fermentation process which involves solid matrix and is carried out in absence or near absence of free water. SSF of potato peel mashed was carried out with S. cerevisiae at 30°C, pH of 5.5, moisture adjustment between 40 and 90%, addition of ammonium sulphate and urea salts as nitrogen supplements for the microorganisms for 3 days. The results showed that the percentage crude protein content of all the fermented samples increased significantly when compared with the unfermented sample. 40% moisture content adjustment and ammonium sulphate as nitrogen source gave the best result. The crude protein increased from 12.5% to 21.86%, which is 74.88% increment for ammonium sulphate supplementation, and 12.5% to 18.42%, which is 47% increment for urea supplementation. Therefore, the fermented peels could serve as good source of cheap protein enriched feed for livestock.展开更多
