Background: Although BCG is the most widely administered vaccine in the world, there have never been as many cases of TB as there are now. Globally, more than 8.8 million people developed active TB and 1.4 million—ma...Background: Although BCG is the most widely administered vaccine in the world, there have never been as many cases of TB as there are now. Globally, more than 8.8 million people developed active TB and 1.4 million—many of them—died in 2010. It is estimated that half of pulmonary TB cases arise from latent Mtb infection, making the study of latency and reactivation of utmost importance. Methods: Widely administered BCG vaccines and a gene modified recombinant BCG (rBCG) strain, AERAS-422, were used as models to investigate the growth promoting function of resuscitation-promoting factors (Rpfs) in different bacilli culture phases. Different supernatant fractions were prepared by ultrafiltration, and the promoting function of each fraction containing secreted Rpf(s) was evaluated by growth curve monitoring and colony counting on 7H10 agar plates. Results: The promoting effect of culture supernatants was mainly associated with the high molecular weight fraction (>30 kDa), which stimulated bacterial growth, but did not extend the exponential phase of stimulated culture. Anti-RpfB antibody showed significant growth restriction of the tested cultures. When comparing rBCG cultures containing 7H9 medium, the 10 - 30 kDa fraction, or the >30 kDa fraction, only the >30 kDa fraction was displayed with down-regulation of the secretion of RpfC, D and E. In colony counting tests, the plates containing the >30 kDa fraction had total countable colony numbers 2 to 3 fold higher than the plates with the 10 - 30 kDa fraction, and colonies appeared one to two weeks earlier than on the regular plates. The potential applications of the prepared supernatant fractions containing RpfA and RpfB are discussed, which may include accelerating diagnosis of Mtb infection and future TB vaccine development.展开更多

The subject of the presented studies will be culture promotion of the region. One of the areas of promotion, little appreciated in Poland so far, is culture and culture heritage which are of greater and greater importance for the regional development. Culture heritage can be a resource to strengthen the region and improve the conditions for economic growth. Culture goods constitute an important factor in the development of socio-economic region. They increase tourist attractiveness contributing to the development of tourism and agro-tourism in the area. The rich history, multi-layered and diverse culture heritage, both tangible and intangible, can be one of the main assets of raising the competitiveness of the given region in the country and the European Union (EU). It should be assumed that in a market economy town or municipality operate in a manner similar to enterprise, they produce products and services offered to the internal market (municipality) as well as to external one (surrounding of the municipality). Competitive conditions require the use of all tools to increase the effectiveness of the operation, including marketing tools. Being a marketing issue a product, which is offered by a city/town, can be considered in two aspects: first as a mega-product (i.e., a city/town as a whole), second as a distribution of a mega-product for individual municipal sub-products. Presented concept of a product offered by a city in its two distinctions can be also applied to a municipality. Market-based approach in relation to territorial units is usually called "territorial marketing". Alternatively, they can be also called "spatial marketing" and "area marketing". Depending on the type of territorial units and the scope of marketing activities various concepts are used (e.g., urban marketing and municipal marketing). Currently in Poland these concepts are generally related to basic units of the administrative division of the country namely to urban and rural municipalities. Methods: It is an action-research study wit...Objective: The study aims to evaluate the use of a teaching method proposed by Paulo Freire, Culture Circles, in the education of teenagers multipliers on leprosy awareness. Methods: It is an action-research study with a qualitative approach developed in a public school in Pernambuco, Brazil. Five Culture Circles were conducted involving the participation of 26 teenagers. The followings were used as data collection tools: observation, field notes, photography and filming. Results: The educational intervention on health addressed the following topics: 1) Definition and transmission of leprosy;2) Characteristics and diagnosis of leprosy;3) Treatment of leprosy;4) Aesthetics, prejudice and mental health related to leprosy;and 5) Planning of educational activities for teenagers as health multipliers on leprosy awareness. The educational action on health provided this age group to perceive themselves, act as political subjects in the development of Culture Circles, and act as protagonists in the dissemination of knowledge on leprosy. Conclusions: This study highlights that the application of active methodologies, such as Culture Circles, is able to encourage the engagement of young people in community empowerment and bring together health professionals and the school community in an intersectoral work in order to develop action strategies involving the promotion of health in the context of neglected diseases such as leprosy.展开更多
