AIM To propose a hypothesis defining theabsorption,distribution,metabolism andelimination of traditional Chinese recipe(TCR)-component in blood of healthy subjects andpatients,and estimate its correctness.METHODS The pharmacokinetics(PK)of samedose of drug was studied in the animal model oftraditional Chinese syndrome(S)and healthyanimals.The classification,terminology,concept and significance of the hypothesis wereset forth with evidence provided in the presentstudy.The hypotheses consisted of traditionalChinese syndrome PK(S-PK)and traditionalChinese recipe PK(R-PK).Firstly,the observedtetramethylpyrazine(TMP)PK in healthy,chronically reserpinized rats(rat model ofspleen deficiency syndrome,RMSDS)andRMSDS treated with Sijunzi decoction(SJZD)forconfirmation were used to verify S-PK; secondly,the ferulic acid(FA)PK in healthy andhigh molecular weight dextran(HMWD)-inducedrabbit model with blood stasis syndrome(RDBSS)was also used to verify S-PK;andlastly,TMP PK parameters in serum of healthyrats after orally taken Ligusticum wallichii(LW),LW and Salvia miltiorrhiza(LW&SM)decoctions were compared to verify R-PK.RESULTS The apparent first-order absorption[Ka,(13.61±2.56)h<sup>-1</sup>,area under the blooddrug concentration-time curve[AUC,(24.88±9.76)μg.h<sup>-1</sup>mL<sup>-1</sup>],maximum drug concentration[C<sub>max</sub>,(4.82±1.23)μg·mL<sup>-1</sup>]of serum TMP inRMSDS were increased markedly(P【0.05)compared with those[Ka=(5.41±1.91)h<sup>-1</sup>,AUC=(5.20±2.57)μg·h<sup>-1</sup>·mL<sup>-1</sup>,C<sub>max</sub>=(2.33±1.77)μg·mL<sup>-1</sup>]of healthy rats(HR).Theapparent first-order rate constant for α and βdistribution phase[α=(0.38±0.09)h<sup>-1</sup>,β=(0.06±0.03)h<sup>-1</sup>,the apparent first-orderintercompartmental transfer rate constants[K<sub>10</sub>=(0.24±0.07)h<sup>-1</sup>,K<sub>12</sub>=(0.11±0.02)h<sup>-1</sup>,K<sub>21</sub>=(0.11±0.02)h<sup>-1</sup>]of serum TMP in RMSDS weredecreased significantly(P【0.01)comparedwith those[K<sub>10</sub>=(0.88±0.20)h<sup>-1</sup>,K<sub>12</sub>=(1.45±0.47)h<sup>-1</sup>,K<sub>21</sub>=(0.72±0.22)h<sup>-1</sup>]of HR.However,no apparent differences occurredbetween HR and RMSDS treated with SJZD.Theserum FA concentration and its AUC[(5.6690±2.3541)μg·h<sup>-1</sup>·mL<sup>-1</sup>] in RMBSS were also higherthan those[AUC=(2.7566±0.8232)μg·h<sup>-1</sup>·mL<sup>-1</sup>]of healthy rabbits(P【0.05).The Ka(11.51±2.82)h<sup>-1</sup>,AUC(0.84±0.17)μg·h<sup>-1</sup>·mL<sup>-1</sup>of LW & SM-derived TMP in serum weremuch lower(P【0.05)than those[Ka=(19.58±4.14)h<sup>-1</sup>,AUC=(1.27±0.26)μg·h<sup>-1</sup>·mL<sup>-1</sup>]ofLW-derived TMP in serum after oral decoctions.CONCLUSION The SDS and blood stasissyndrome state could affect significantly thepharmacokinetic parameters of drugs and theabnormal SDS pharmacokinetic parameters couldbe normalized by SJZD.The combination ofChinese medicine in TCR could reciprocallyaffect the pharmacokinetic parameters of othercomponents absorbed into the systemiccirculation.These results support the S-and R-PK hypothesis. 展开更多

Exploring the therapeutic potential of Qi Teng Mai Ning recipe in ischemic stroke and vascular cognitive impairment

Background:This study aims to explore the therapeutic effects of the Qi Teng Mai Ning recipe on ischemic stroke and vascular cognitive impairment through its potential to modulate cellular autophagy,with a focus on identifying its active ingredients and their target proteins.Methods:The study began with the identification of active ingredients in the Qi Teng Mai Ning recipe.It proceeded to screen the gene expression omnibus database for ischemic stroke and vascular cognitive impairment-associated differentially expressed mRNAs and to identify cellular autophagy-related proteins via the Human Autophagy Database.These proteins were annotated with Gene Ontology and Kyoto Encyclopedia of Genes and Genomes functions and subjected to molecular docking with the recipe’s core active ingredients.In vitro cell experiments were conducted on hypoxic HT22 cells,involving CCK8 assay,lentiviral transfection to silence autophagy related 9B(ATG9B),immunofluorescence staining,and qPCR validation to investigate the effects of the recipe on autophagy.Results:The analysis identified 104 active ingredients targeting 408 proteins and forming a complex ingredient-target network.Intersecting 55 ischemic stroke-related and 909 vascular cognitive impairment-related differentially expressed mRNAs revealed 14 co-expressed mRNAs.Molecular docking showed quercetin,kaempferol,myrcene,and conferone as key ingredients targeting autophagy-related proteins.Cellular experiments indicated that the recipe significantly enhanced cell viability under hypoxic conditions,reduced apoptosis,and modulated the expression of autophagy-related factors,thereby decreasing apoptosis rates in HT22 cells.Conclusion:The Qi Teng Mai Ning recipe offers protective effects against ischemic stroke and vascular cognitive impairment by modulating autophagy-related proteins.Its efficacy highlights the potential of traditional Chinese medicine in treating these conditions,though further research is needed to fully understand its mechanisms and clinical applications. 展开更多

BACKGROUND Chronic heart failure is a complex clinical syndrome.The Chinese herbal compound preparation Jianpi Huatan Quyu recipe has been used to treat chronic heart failure;however,the underlying molecular mechanism is still not clear.AIM To identify the effective active ingredients of Jianpi Huatan Quyu recipe and explore its molecular mechanism in the treatment of chronic heart failure.METHODS The effective active ingredients of eight herbs composing Jianpi Huatan Quyu recipe were identified using the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform.The target genes of chronic heart failure were searched in the Genecards database.The target proteins of active ingredients were mapped to chronic heart failure target genes to obtain the common drugdisease targets,which were then used to construct a key chemical componenttarget network using Cytoscape 3.7.2 software.The protein-protein interaction network was constructed using the String database.Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were performed through the Metascape database.Finally,our previously published relevant articles were searched to verify the results obtained via network pharmacology.RESULTS A total of 227 effective active ingredients for Jianpi Huatan Quyu recipe were identified,of which quercetin,kaempferol,7-methoxy-2-methyl isoflavone,formononetin,and isorhamnetin may be key active ingredients and involved in the therapeutic effects of TCM by acting on STAT3,MAPK3,AKT1,JUN,MAPK1,TP53,TNF,HSP90AA1,p65,MAPK8,MAPK14,IL6,EGFR,EDN1,FOS,and other proteins.The pathways identified by KEGG enrichment analysis include pathways in cancer,IL-17 signaling pathway,PI3K-Akt signaling pathway,HIF-1 signaling pathway,calcium signaling pathway,cAMP signaling pathway,NF-kappaB signaling pathway,AMPK signaling pathway,etc.Previous studies on Jianpi Huatan Quyu recipe suggested that this Chinese compound preparation can regulate the TNF-α,IL-6,MAPK,cAMP,and AMPK pathways to affect the mitochondrial structure of myocardial cells,oxidative stress,and energy metabolism,thus achieving the therapeutic effects on chronic heart failure.CONCLUSION The Chinese medicine compound preparation Jianpi Huatan Quyu recipe exerts therapeutic effects on chronic heart failure possibly by influencing the mitochondrial structure of cardiomyocytes,oxidative stress,energy metabolism,and other processes.Future studies are warranted to investigate the role of the IL-17 signaling pathway,PI3K-Akt signaling pathway,HIF-1 signaling pathway,and other pathways in mediating the therapeutic effects of Jianpi Huatan Quyu recipe on chronic heart failure. 展开更多

This paper performed an objective analysis of the historical role played by Zhiyi Tongzhi Recipe in the history of infectious diseases in China, and summed up the experience and lessons of the application of Zhiyi Tongzhi Recipe. Xiongdankaiming tablet (XDKMT), a well-known compound in traditional Chinese medicine, is widely used for the treatment of acute iridocyclitis and primary open-angle glaucoma. In this paper, accurate and reliable methods were developed for the identification of 20 constituents using high-performance liquid chromatography with photodiode array and electron spray ionization-mass spectrometry (HPLC-PDA/ESI-MSn ), and determination of nine of the constituents (chlorogenic acid, gentiopicroside, isochlorogenic acid B, diosmetin-7-O-β-D-glucopyranoside, apigenin, diosmetin, tauroursodeoxycholic acid, acacetin, and taurochenodeoxycholic acid) was developed using HPLC with ultraviolet absorption detector and evaporative light scattering detector (HPLC-UV/ELSD) for the first time. The best results were obtained on a Zorbax SB-C18 column with gradient elution using water (0.1% formic acid) (A) and methanol (0.1% formic acid) (B) at a flow rate of 0.7 ml/min. Tauroursodeoxycholic acid and taurochenodeoxycholic acid, owing to their low UV absorption, were detected by ELSD. The other seven compounds were analyzed by HPLC-UV with variable wavelengths. The calibration curves of all nine constituents showed good linear regression (R2 >0.9996) within the linearity ranges. The limits of detection and quantification were in the ranges of 0.0460-9.90 μg/ml and 0.115-24.8 μg/ml, respectively. The accuracy, in terms of recovery, varied from 95.3% to 104.9% with relative standard deviations (RSDs) less than 4.4%. Precision (with the intra- and inter-day variations less than 4.4%) was also suitable for its intended use. The developed method was successfully applied for the analysis of major components in XDKMT, which provides an appropriate method for the quality control of XDKMT. 展开更多

Erhuang capsule, a typical formula based on traditional Chinese medicine theory, is widely used to ameliorate multiple sclerosis, inflammation and side effects of glucocortJcoid treatment. Oligodendrocyte precursor cells are neural stem cells that are important for myelin repair and regeneration. In the present study, Erhuang capsule effectively improved clinical symptoms and neurological function scores, reduced mortality and promoted recovery of neurological functions of mice with experimental autoimmune encephalomyelitis. The mechanism of action involved significant increases in oligodendrocyte precursor cell proliferation in specific regions of the brain and spinal cord, increased oligodendrocyte lineage gene 2 expression and enhanced oligodendrocyte precursor cell differentiation. 展开更多

There are few effective therapies. Chinese medicine formula, the main pattern of traditional Chinese medicine(TCM) in clinic, applies a multi-component, multi-target and complicated approach in the treatment of diseases, and certainly shows good comprehensive therapeutic effects on fibrosis. In this review, the clinical study, effects and mechanism of action of Fuzheng Huayu recipe in the treatment of liver, pulmonary, and renal fibrosis was analyzed and evaluated. Meanwhile, based on the understanding of TCM pathogenesis for liver cirrhosis, this review will also briefly introduce the research of different classical Chinese medicine formulae with various functions in the treatment of experimental liver fibrosis induced by different methods, including comparing the efficacy, analyzing the action characteristics and mechanism of effective formulae, exploring pathological and biological basis of TCM diagnostic and therapeutic pattern, which will contribute to the research of TCM in the treatment of organ fibrosis. 展开更多

目的观察补肾活血方对肾虚血瘀型卵巢储备功能下降(DOR)患者的治疗效果。方法选取北京妇产医院中医科2021年1月至2022年12月就诊的180例诊断为DOR的肾虚血瘀型患者作为研究对象,采用计算机随机分组法分为雌二醇片/雌二醇地屈孕酮片组(... 目的观察补肾活血方对肾虚血瘀型卵巢储备功能下降(DOR)患者的治疗效果。方法选取北京妇产医院中医科2021年1月至2022年12月就诊的180例诊断为DOR的肾虚血瘀型患者作为研究对象,采用计算机随机分组法分为雌二醇片/雌二醇地屈孕酮片组(西药组)、补肾活血方组(中药组)和补肾活血方联合雌二醇片/雌二醇地屈孕酮片组(联合组),各60例。观察三组治疗前后卵泡刺激素(FSH)、黄体生成素(LH)、雌二醇(E_(2))、抗米勒管激素(AMH)、窦卵泡数(AFC)、卵巢体积、卵巢动脉和子宫动脉阻力指数(RI)、子宫内膜厚度的变化,比较三组中医证候评分及疗效。结果11例患者(4例来自西药组,3例来自中药组,4例来自联合组)退出研究。6个月后,三组FSH和LH均显著降低,E_(2)和AMH升高(P<0.05);西药组和联合组较中药组FSH降低更显著,E_(2)升高更显著(P<0.01)。治疗后,中药组和联合组卵巢体积增加(P<0.05),AFC显著增加(P<0.01),卵巢动脉RI显著降低(P<0.01);西药组卵巢体积增加不显著(P>0.05),AFC显著增加(P<0.05),卵巢动脉RI显著降低(P<0.05);三组子宫动脉RI均无显著改善(P>0.05)。三组子宫内膜厚度均显著增加(P<0.05),西药组和联合组较中药组增加更显著(P<0.01)。与治疗前相比,三组治疗后中医证候评分均显著降低(P<0.05),且中药组和联合组中医证候评分降低的程度大于西药组(P<0.05)。西药组总有效率为71.43%,中药组为82.46%,联合组为85.96%;联合组的疗效优于西药组(P<0.05)。结论补肾活血方能够通过平衡激素水平、促进排卵和修复子宫内膜对DOR患者产生良好的治疗效果,联合应用雌二醇片/雌二醇地屈孕酮片可能是治疗DOR的一种行之有效的治疗策略。 展开更多

目的 探究补肾健脾膏方治疗脾虚厌食症及其对中医证候和生长发育的影响。方法 选取2021年10月-2023年10月我院收治的106例脾虚厌食症患儿为研究对象,使用信封法将患儿分为研究组和常规组,各53例。常规组接受厌食症常规药物治疗,研究组... 目的 探究补肾健脾膏方治疗脾虚厌食症及其对中医证候和生长发育的影响。方法 选取2021年10月-2023年10月我院收治的106例脾虚厌食症患儿为研究对象,使用信封法将患儿分为研究组和常规组,各53例。常规组接受厌食症常规药物治疗,研究组在常规药物基础上联用补肾健脾膏方治疗,治疗周期均为4周。比较2组患儿的治疗有效率、中医证候积分、日摄食量、体质量指数(BMI)、胃肠道功能[胃泌素(GA)、胃动素(MLT)、血管活性肠肽(VIP)]、血清微量元素[血清铁(SF)、血清锌(Zn)、血清钙(Ca)]、药物不良反应、随访预后6个月内患儿的身高变化及预后复发率。结果 研究组的治疗有效率为96.23%,常规组为84.91%,研究组高于常规组(P<0.05);研究组主症、次症的中医证候积分均低于常规组(P<0.05);研究组治疗后的日摄食量和BMI均高于常规组(P<0.05);研究组GA、MLT水平高于常规组,VIP水平低于常规组(P<0.05);研究组SF、Zn、Ca均高于常规组(P<0.05);2组的不良反应发生率比较无统计学意义(P>0.05);研究组预后6个月的身高高于常规组(P<0.05);研究组患儿的复发率为7.55%,常规组患儿的复发率为22.64%,研究组低于常规组(P<0.05)。结论 补肾健脾膏方可有效改善脾虚厌食症患儿的厌食症状,增加每日摄食量,改善胃肠道功能,促进微量元素吸收,保障患儿正常生长发育。 展开更多

目的 探讨小针刀联合补肾活血祛痛方治疗腰椎间盘突出症(LDH)的临床效果。方法 选取2016年7月—2018年4月沈阳市中医院收治的135例患者作为研究对象,将所有患者随机分为甲组(小针刀联合补肾活血祛痛方)、乙组(补肾活血祛痛方)和丙组(小... 目的 探讨小针刀联合补肾活血祛痛方治疗腰椎间盘突出症(LDH)的临床效果。方法 选取2016年7月—2018年4月沈阳市中医院收治的135例患者作为研究对象,将所有患者随机分为甲组(小针刀联合补肾活血祛痛方)、乙组(补肾活血祛痛方)和丙组(小针刀),观察与对比3组患者的腰痛分级、治疗前后视觉模拟量表(VAS)评分和总体效果。结果 甲组总有效率为97.78%(44/45),高于乙组的88.89%(40/45)、丙组的82.22%(37/45),差异具有统计学意义(P<0.05);甲组治疗后VAS评分下降程度明显大于乙组和丙组;甲组治疗后Denis腰痛分级明显优于乙组和丙组,差异具有统计学意义(P<0.05)。结论对LDH患者进行小针刀联合补肾活血祛痛方治疗可以有效缓解患者的疼痛程度、缩短患者的住院时间、提升患者的生活质量,同时患者腰椎功能可以得到快速恢复,值得临床应用与推广。 展开更多

目的探讨疏肝和胃方通过抑制核因子-κB(NF-κB)通路调控M1巨噬细胞极化对反流性食管炎(RE)大鼠食管黏膜炎症的影响。方法将40只SD雄性大鼠随机分为假手术组(8只)和造模组(32只)。假手术组暴露食管下段及幽门,造模组予下食管括约肌切开... 目的探讨疏肝和胃方通过抑制核因子-κB(NF-κB)通路调控M1巨噬细胞极化对反流性食管炎(RE)大鼠食管黏膜炎症的影响。方法将40只SD雄性大鼠随机分为假手术组(8只)和造模组(32只)。假手术组暴露食管下段及幽门,造模组予下食管括约肌切开术加十二指肠半结扎术。术后将造模成功的大鼠随机分为模型组、疏肝和胃方低剂量组、疏肝和胃方高剂量组和雷贝拉唑组。假手术组和模型组予蒸馏水灌胃,疏肝和胃方低、高剂量组分别予以10.49 g·kg^(-1)·d^(-1)、20.98 g·kg^(-1)·d^(-1)疏肝和胃方灌胃,雷贝拉唑组予雷贝拉唑(2.06 mg·kg^(-1)·d^(-1))灌胃,灌胃持续2周。采用苏木精-伊红(HE)染色观察食管组织病理学改变;采用Western blot法检测核因子-κB p65(NF-κB p65)、NF-κB抑制蛋白α(IκBα)的磷酸化情况,以及白细胞分化抗原86(CD86)、甘露糖受体(CD206)的表达;采用实时荧光定量逆转录聚合酶链式反应(RT-qPCR)法检测肿瘤坏死因子-α(TNF-α)、白介素-1β(IL^(-1β))、转化生长因子-β(TGF-β)mRNA表达。结果HE染色结果显示,模型组鳞状上皮增生,炎症细胞浸润;疏肝和胃方低、高剂量组和雷贝拉唑组组织结构正常,基底层血管湖少且小,无炎症细胞浸润。与假手术比较,模型组CD86、磷酸化IκBα(pIκBα)/IκBα、核因子-κB磷酸化p65(NF-κB pp65)/p65蛋白,以及TNF-α、IL^(-1β)mRNA表达升高(P<0.05)。与模型组比较,疏肝和胃方低、高剂量组和雷贝拉唑组CD86蛋白以及TNF-α、IL^(-1β)mRNA表达下降(P<0.05),疏肝和胃方高剂量组和雷贝拉唑组pIκBα/IκBα、pp65/p65表达下降(P<0.05),但疏肝和胃方低剂量组与模型组pIκBα/IκBα、pp65/p65的表达差异无统计学意义(P>0.05)。与假手术组比较,模型组CD206表达减少(P<0.05),TGF-βmRNA表达呈下降趋势但差异无统计学意义(P>0.05),疏肝和胃方低、高剂量组和雷贝拉唑组CD206表达升高(P<0.05)。与模型组比较,疏肝和胃方高剂量组和雷贝拉唑组TGF-βmRNA表达升高(P<0.05)。结论疏肝和胃方可通过抑制RE大鼠的M1巨噬细胞极化来改善食管黏膜炎症,其作用机制可能与抑制NF-κB信号通路活化有关。 展开更多

目的:研究扶阳保肾方对糖尿病肾病合并Ⅳ型心肾综合征患者血清指标的影响。方法:选取120例糖尿病肾病合并Ⅳ型心肾综合征心肾阳虚证患者进行试验,按随机数字表法分为观察组与对照组,每组60例。两组均进行运动、戒烟、吸氧等常规治疗。... 目的:研究扶阳保肾方对糖尿病肾病合并Ⅳ型心肾综合征患者血清指标的影响。方法:选取120例糖尿病肾病合并Ⅳ型心肾综合征心肾阳虚证患者进行试验,按随机数字表法分为观察组与对照组,每组60例。两组均进行运动、戒烟、吸氧等常规治疗。对照组进行常规西药治疗,包括口服缬沙坦胶囊、胰岛素制剂;观察组在对照组基础上使用扶阳保肾方进行治疗。观察两组治疗效果,记录两组治疗前后中医证候积分,测定两组治疗前后超敏C反应蛋白(Hs-CRP)、肌酐(CREA)、尿素氮(BUN)、血清β_(2)微球蛋白(β_(2)-MG)、脑钠肽(BNP)水平,观察两组不良反应发生率。结果:观察组治疗效果优于对照组。治疗后两组患者各项中医证候积分下降,且观察组心悸、胸闷气短、下肢浮肿、身寒肢冷评分低于对照组,差异有统计学意义(均P<0.05)。治疗后观察组BUN、CREA、β_(2)-MG、Hs-CRP、BNP低于对照组,差异有统计学意义(均P<0.05)。两组不良反应比较,差异无统计学意义(P>0.05)。结论:扶阳保肾方治疗糖尿病肾病合并Ⅳ型心肾综合征效果较好,对肾功能改善明显,且能降低患者血清BUN、CREA水平及炎症反应,未增加患者不良反应。 展开更多

[目的]总结浙江省名中医丁彩飞教授治疗多囊卵巢综合征合并自身免疫性甲状腺炎的经验。[方法]通过门诊跟师,系统收集丁教授治疗多囊卵巢综合征合并自身免疫性甲状腺炎的医案,总结分析丁教授治疗多囊卵巢综合征合并自身免疫性甲状腺炎的病因病机、辨治思路、用药特色,并附一验案佐证。[结果]丁教授提出,“痰”邪是多囊卵巢综合征合并自身免疫性甲状腺炎的病机关键,重视“肝、脾、肾”三脏的功能恢复,疾病可分为脾虚肝郁、痰瘀互结、肝肾亏虚三类证型,治疗上提倡“健脾化痰,疏肝理气,补肾活血”,配合中医外治、情绪调摄等,以获良效。所选验案,丁教授辨为痰瘀互结、脾虚湿盛证,治以健脾化湿、活血化瘀,以五苓散化裁治疗,取得较好疗效,促甲状腺素恢复正常水平,月经按时来潮。[结论]丁教授根据多囊卵巢综合征合并自身免疫性甲状腺炎的病理特点和临床特征,贯通中西,内服外治,以获得调经种子,改善内分泌代谢状态的治疗效果,为中西医结合治疗多囊卵巢综合征合并自身免疫性甲状腺炎提供了参考。 展开更多

Objectives: Ziyin Huatan Recipe(ZYHT), a traditional Chinese medicine comprised of Lilii Bulbus, Pinelliae Rhizoma, and Hedyotis Diffusa, has shown promise in treating gastric cancer(GC). However, its potential mechanism has not yet been clearly addressed. This study aimed to predict targets and molecular mechanisms of ZYHT in treating GC by network pharmacology analysis and to explore the role of ZYHT in GC both in vitro and in vivo.Methods: Targets and molecular mechanisms of ZYHT were predicted via network pharmacology analysis. The effects of ZYHT on the expression of metastasis-associated targets were further validated by Western blot and quantitative real-time polymerase chain reaction. To explore the specific molecular mechanisms of the effects of ZYHT on migration and invasion, the runt-related transcription factor 3(RUNX3) gene was knocked out by clustered regularly interspaced short palindromic repeats/Cas9, and lentiviral vectors were transfected into SGC-7901 cells. Then lung metastasis model of GC in nude mice was established to explore the anti-metastasis effect of ZYHT. Western blot and immunohistochemistry were used to explore the impact of ZYHT on the expression of metastasis-related proteins with or without RUNX3 gene.Results: The network pharmacology analysis showed that ZYHT might inhibit focal adhesion, migration,invasion and metastasis of GC. ZYHT inhibited the proliferation, migration and invasion of GC cells in vitro via regulating the expression of metastasis-associated targets. Knocking out RUNX3 almost completely reversed the cell phenotypes(migration and invasion) and protein expression levels elicited by ZYHT.In vivo studies showed that ZYHT inhibited the metastasis of GC cells to the lung and prolonged the survival time of the nude mice. Knocking out RUNX3 partly reversed the metastasis of GC cells to the lung and the protein expression levels elicited by ZYHT.Conclusion: ZYHT can effectively inhibit the invasion and migration of GC in vitro and in vivo, and its molecular mechanism may relate to the upregulation of RUNX3 expression. 展开更多

自拟芪参消疲方治疗肺癌癌因性疲乏的临床疗效观察

目的:观察芪参消疲方对肺脾两虚型肺癌癌因性疲乏患者的临床疗效。方法:将90例患者根据病情分为对照组、治疗组1、治疗组2各30人。观察3组患者治疗前后Piper评分、中医证候积分、血常规、EORTC QLQ-C30评分、BFI评分等疗效指标以及安全... 目的:观察芪参消疲方对肺脾两虚型肺癌癌因性疲乏患者的临床疗效。方法:将90例患者根据病情分为对照组、治疗组1、治疗组2各30人。观察3组患者治疗前后Piper评分、中医证候积分、血常规、EORTC QLQ-C30评分、BFI评分等疗效指标以及安全性指标。结果:3组患者Piper疲乏评分、中医症候积分、QLQ-C30评分、BFI评分,治疗后较治疗前均有所改善,但治疗组1与治疗组2均优于对照组。对照组总有效率为36.67%,治疗组1总有效率为93.33%,治疗组2总有效率为76.67%。对照组血常规未见明显变化,治疗组1白细胞、红细胞、血红蛋白较前明显升高,治疗组2红细胞、血红蛋白、红细胞压积、血小板较前明显降低。且治疗组1红细胞高于治疗组2及对照组、血红蛋白高于治疗组2,治疗组2红细胞、红细胞压积均低于对照组。结论:芪参消疲方能够有效减轻肺脾两虚型肺癌癌因性疲乏患者的疲乏症状、改善临床指标,减少联合抗肿瘤治疗引起的不良反应的发生,且安全有效,值得临床推广。 展开更多

[目的]研究清肝活血方(QGHXR)通过调控半胱天冬酶(Caspase)-4/Caspase-3/GSDME细胞焦亡通路防治酒精性肝病(ALD)的作用机制。[方法]将18只C57BL/6小鼠随机分为对照组、模型组和QGHXR组复制ALD小鼠模型。检测血清肝功能、血脂水平;苏木精-伊红(HE)染色及油红O染色观察肝脏病理改变及肝脏脂质沉积状况;实时荧光定量逆转录聚合酶链反应(qRT-PCR)、蛋白免疫印迹(Western Blot)法及免疫荧光染色检测Caspase-4/Caspase-3/GSDME通路相关基因及蛋白表达情况。[结果]与空白对照组比较,模型组小鼠肝组织三酰甘油(TG)水平明显升高(P<0.01),血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)水平升高(P<0.05),血清白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平升高(P<0.05);肝脏组织出现病理改变及脂质沉积现象;肝组织IL-1B、IL-6、TNF-α、NLRP3 mRNA表达水平升高。与模型组比较,QGHXR组小鼠肝组织TG水平明显降低(P<0.01),血清ALT、AST、总胆红素(TBIL)、总胆汁酸(TBA)水平降低(P<0.05),血清TG、IL-6、TNF-α水平降低(P<0.05)以及血清高密度脂蛋白(HDL)水平升高(P<0.01);血清中血脂、肝脏病理改变及脂质沉积现象缓解;GsdmeN、Caspase-4、Caspase-3和C-Caspase-3基因及蛋白水平降低(P<0.05)。[结论]QGHXR可通过调节Caspase-4/Caspase-3/GSDME介导的细胞焦亡通路改善ALD小鼠肝损伤和脂质沉积。 展开更多
