Serum HA, type IV collagen, and LN were measured by ELISA. The liver/weight index and morphological changes were examined under electron microscope on d 7, 14, 21, and 28 by immunohistochemical alpha smooth muscle actin α-SMA staining as well as Sirius-red and HE staining.RESULTS: The levels of serum HA, type IV collagen and LN significantly increased from d 7 to d 28 (P = 0.043). The liver/weight index increased on d 7 and decreased on d 28. In the model group, the rat liver stained with lie and Sirius-red showed evident hemorrhage and necrosis in the central vein of hepatic 10 Iobules on d 7. Thin fibrotic septa were formed joining central areas of the liver on d 14. The number of α-SMA positive cells was markedly increased in the model group. Transitional hepatic stellate cells were observed under electron microscope. All rats in the model group showed micronodular fibrosis in the hepatic parenchyma and a network of α-SMA positive cells. Typical myofibroblasts were embedded in the core of a fibrous septum. Compared to the control group, the area-density percentage of collagen fibrosis and pathologic grading were significantly different in the model group (P〈0.05) on different d (7, 14, and 28). The area-density percentage of collagen fibrosis in hepatic tissue had a positive correlation with the levels of serum HA, LN, and type IV collagen.CONCLUSION: The morphological and serum HA, type IV collagen, and LN are changed in DMN-induced liver fibrosis in rats.展开更多

We explore the m...Objective: To study the effect of Biejiajian Oral Liquid (BOL) on the rennin angiotensin aldosterone system (RAAS) in plasma of hepatic fibrosis rats and in hepatic stellate cell (HSC) of normal rats. We explore the mechanism of BOL on inhibiting the activation of HSC and illustrate its mechanism of anti-hepatic fibrosis further. Methods: SD Rats were divided into 5 groups randomly: normal control group, model group, Enalapril group and BOL groups with different concentration (2.0 g/ml or 1.0 g/ml). Every group was administered with CCl4 and olive oil solution to induce hepatic fibrosis except normal one. Each group was treated with the respective drug for 5 weeks and then got the blood. The level of renin, angiotensin II and aldosterone in the plasma of liver fibrosis rats were detected by the radioimmunoassay. By using reverse transcription-polymerase chain reaction (RT-PCR) to measure the gene expression of the rennin, ACE, angiotensinogen, AT1R and ALD. The AT1R gene expression in normal HSC was determined by the immunohistochemical measurement. Results: BOL could effectively reduce the activity of the PRA, AngIIand ALD, which showed a significant effect on the inhibition of the AngII (P < 0.01). Meanwhile, compared with the normal control group, there was a notable inhibitory action on the PRA of HSC which was administrated by serum containing BOL (P < 0.05). And yet, drug applied group showed no difference with the model group for other factors of the RAAS. Conclusion: BOL can inhibit the expression of RAAS in the rat plasma and can inhibit the expression of the mRNA of renin in the normal HSC, which could be the mechanism of anti-hepatic fibrosis.展开更多

According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (...According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (emulsion of aimed antigen and Freund's incomplete adjuvant) were followed every 2-4 weeks. In general,high titer of the corresponding polyclonal antisera will be observed after 4-5 injections of sample B in 3-4months. This report presents a simply modified procedure that was able to stimulate the antisera formation in one month and achieve enough avidity to satisfy either Western blot or immunohistochemistry analysis.It just applied an additional injection of the sample A to the rabbit at the 3rd day after the primary immunization injection. You could gain the high titer of the antisera right after the first sample B injection in one month. This method has produced the desired results in three different recombinant antigens with different molecular weight (5.9 KD-55 KD) expressed from prokaryotic or eukaryotic cells.展开更多
