Efficacy of recombinant human epidermal growth factor plus sodium hyaluronate eye drops in diabetic dry eye post-cataract surgery

BACKGROUND Dry eye syndrome(DES)after diabetic cataract surgery can seriously affect the patient’s quality of life.Therefore,effective alleviation of symptoms in patients with this disease has important clinical significance.AIM To explore the clinical effect of recombinant human epidermal growth factor(rhEGF)plus sodium hyaluronate(SH)eye drops on DES after cataract surgery in patients with diabetes.METHODS We retrospectively evaluated 82 patients with diabetes who experienced DES after cataract surgery at Tianjin Beichen Hospital,Affiliated Hospital of Nankai University between April 2021 and April 2023.They were classified into an observation group(42 cases,rhEGF+SH eye drops)and a control group(40 cases,SH eye drops alone),depending on the different treatment schemes.The therapeutic efficacy,dry eye symptom score,tear film breakup time(TFBUT),basic tear secretion score[assessed using Schirmer I test(SIt)],corneal fluorescein staining(FL)score,tear inflammatory markers,adverse reactions during treat-ment,and treatment satisfaction were compared between the two groups.RESULTS Therapeutic efficacy was higher in the observation group compared with the control group.Both groups showed improved TFBUT and dry eye,as well as improved SIt and FL scores after treatment,with a more pronounced improvement in the observation group.Although no marked differences in adverse reactions were observed between the two groups,treatment satisfaction was higher in the observation group.CONCLUSION rhEGF+SH eye drops rendered clinical benefits to patients by effectively ameliorating dry eye and visual impairment with favorable efficacy,fewer adverse reactions,and high safety levels.Thus,this treatment should be promoted in clinical practice. 展开更多

Objective:To evaluate the therapeutic effect of recombinant bovine basic fibroblast growth factor(rbFGF)eye gel combined with tobramycin-dexamethasone(TOB-Dex)eye drops on dry eye syndrome(DES)after cataract surgery.Methods:86 patients with DES after cataract surgery,admitted from November 2021 to November 2023,were randomly divided into groups.The observation group included 43 patients treated with rbFGF eye gel combined with TOB-Dex eye drops.The reference group included 43 patients treated with TOB-Dex eye drops alone.Multiple indicators,including total effective rate and clinical symptom scores,were compared between the two groups.Results:The total effective rate in the observation group was higher than in the reference group(P<0.05).Before treatment,there were no differences in clinical symptom scores,serum factors,or disease severity scores between the two groups(P>0.05).Three weeks after treatment,the observation group had lower clinical symptom scores,serum factors,and disease severity scores compared to the reference group(P<0.05).The adverse reaction rate in the observation group was lower than in the reference group(P<0.05).Conclusion:rbFGF eye gel combined with TOB-Dex eye drops can improve the clinical efficacy for patients with DES after cataract surgery,alleviate disease symptoms,reduce inflammatory responses,and have fewer adverse reactions. 展开更多

Objective To evaluate the effects of transforming growth factor β(TGF-β) and recombinant human bone morphogenetic protein 2 (rhBMP2) on human periodontal ligament fibroblasts (HPDLFs). Methods HPDLFs were done primary culture to detect the distinct concentrations of TGF-P and rhBMF2 on its proliferation, alkaline phosphatase (ALP) activity, osteocalcin (OC) synthesis and formation of the minerali-zed nodules, respectively. Results TGF-β (5~100ng/ml) significantly stimulated the proliferation of HPDLFs. The ALP activity of HPDLFs was evaluated evidently by 5ng/ml TGF-β. TGF-β( 0. 5 ~ 100ng/ml) had no effects on OC synthesis and formation of the mineralized nodules of HPDLFs. rhBMP2 (0. 25~2mg/ ml) had no remarkable effect on the proliferation of HPDLFs. The ALP activity, OC synthesis and forma-tion of the mineralized nodules of HPDLFs were significantly stimulated by 0. 5~ 2mg /ml rhBMP2. Conclusion The effects of TGF-β and rhBMP2 on HPDLFs are dose-dependent. TGF-P can stimulate HPDLFs to express the early marker of osteoblastic phenotype, and it lacks the ability to promote maturation of the osteogenic phenotype. rhBMP2 can not only stimulate the expression but also promote the maturation of osteoblas-tic phenotype of HPDLFs. 展开更多

AIM: To describe the significant over-expression of fibroblast growth factor receptor 3 (FGFR3), which is a signal transduction and cell proliferation related gene in hepatocellular carcinoma (HCC).METHODS: Following DNA microarray, Northern blot and quantitative real-time PCR were employed to confirm FGFR3 expression difference in HCC tissues and surrounding non-neoplastic liver tissue. FGFR3 expression levels were further determined by immunohistochemical study in 43 cases of HCC.RESULTS: Northern blot results showed the significant over-expression of FGFR3 in HCC tissues, which was consistent with that from DNA microarray. Quantitative real-time PCR demonstrated that the mean ratio of FGFR3 mRNA to glyceraldehyde-3-phosphate dehydrogenase (GADPH) mRNA in HCC tissue was 0.250, whereas the ratio in non-neoplastic liver tissue was 0.014. Statistical analyses of 43 cases of HCC revealed that HCC scored higher than the matched non-neoplastic liver tissues.Examination of clinicopathological features revealed a strong correlation of over-expression of FGFR3 with poor tumor differentiation and high nuclear grade.CONCLUSION: Over-expression of FGFR3 may play an important role in liver carcinogenesis. FGFR3 may be an ideal candidate as a molecular marker in the diagnosis of HCC and a potential therapeutic target. 展开更多

In this study, 100 tJg/L Noggin or 20 IJg/L basic fibroblast growth factor in serum-free neural induction medium was used to differen- tiate human embryonic stem cells H14 into neural precursors using monolayer differentiation. Two weeks after induction, significantly higher numbers of neural rosettes formed in the Noggin-induced group than the basic fibroblast growth factor-induced group, as detected by phase contrast micro- scope. Immunofluorescence staining revealed expression levels of Nestin, [3-111 Tubulin and Sox-1 were higher in the induced cells and reverse-transcription PCR showed induced cells expressed Nestin, Sox-1 and Neurofilament mRNA. Protein and mRNA expression in the Noggin-induced group was increased compared with the basic fibroblast growth factor-induced group. Noggin has a greater effect than basic fibroblast growth factor on the induction of human embryonic stem cell differentiation into neural precursors by monolayer differentiation, as Noggin accelerates and in- creases the differentiation of neural precursors. 展开更多

AIM:To characterize changes of corneal nerve morphology and tear indices in patients with neurotrophic keratitis(NK)treated with recombinant human nerve growth factor(rhNGF).METHODS:In a prospective observational study,six patients(nine eyes)were locally treated with rhNGF.Visual acuity,corneal fluorescein staining score,the heights of the tear river,lipid layer thickness(LLT),tear ferning(TF)test,conjunctival impression cytology(CIC)examination,the densities of cornea subbasal nerve fibers were determined before and after treatment.RESULTS:Compared with baseline,there was a significant difference in corneal fluorescence staining scores(P<0.01);all patient corneal epithelial defects recovered completely within 8wk,but there was no significant improvement in the height of the tear river(P=0.202).LLT was significantly increased when compared with baseline(P=0.042);however,the function of conjunctival goblet cells and mucin content did not significantly improve using the TF test and CIC examination(P=0.557,P=0.539).After 8wk of treatment,the average corneal subbasal nerve fiber density increased significantly(P<0.01),as did the number of corneal nerve fiber branches(P=0.001).CONCLUSION:RhNGF can increase the density of corneal subbasal nerve fibers,promote the healing of persistent corneal epithelial defects and corneal ulcers in patients with NK,also improving tear function partially. 展开更多

AIM: Epidermal growth factor (EGF) plays an important role in the regulation of gastrointestinal tissue growth and development, and it can stimulate epithelial proliferation, cell differentiation and growth. It has been established that the EGF can promote gastric cytoprotection and ulcer healing. But the potential ability of EGF to regulate the gastric cancer growth is unknown. This study is to investigate the influence of EGF on human gastric cancer cell and the implanted tumor growth of nude mice. METHODS: The cell growth rates of human gastric adenocarcinoma cell lines MKN-28, MKN-45, SGC-7901 and normal human gastric epithelial cells 3T3 were assessed when incubated with recombinant human EGF (rhEGF, 0.05, 0.1, 0.5, 1.0, 10, 50, 100 mg.L(-1)) using MTT method. The cells of MKN-28, MKN-45, SGC-7901 (gastric cancer tissue 1.5mm(3)) were implanted in the BALB/cA nude mice for 10 days.The EGF was given intraperitoneally (15, 30, 60 microg.kg(-1)) for 3 weeks. The body weights of the tumor-bearing animals and their tumor mass were measured afterwards to assess the mitogenic effect of rhEGF in the nude mice. RESULTS: Within the concentration range of 0.05-100mg.L(-1), rhEGF could increase the cell growth of normal 3T3 cells (cell growth rate 100% vs 102.8%, P【0.05), but partially restrain the gastric cancer cell growth. The latter effect was related to cell differentiation. In 15-60 microg/kg rhEGF groups, the mean implanted tumor mass of MKN-28 cell were 1.75 g, 1.91 g, 2.08 g/NS group 1.97 g (P】0.05), the mean tumor mass of SGC-7901 cell were 1.53 g, 1.07 g, 1.20 g/NS group 1.07 g (P】0.05), and for MKN-45 cell, the tumor mass were respectively 1.92 g, 1.29 g, 1.77 /NS group 1.82 g (P】0.05). So rhEGF had no obvious effect on implanted MKN-28, SGC-7901 and MKN-45 tumor growth. CONCLUSION: EGF has no stimulating effect on the human gastric cancer cell growth neither in vitro nor in vivo. 展开更多

Objective: To observe the antitumor effect and mechanism of recombinant human endostatin(Endostar) injection in tumor combined with intraperitoneal injection of cisplatin on subcutaneous transplanted Lewis lung cancer in rats. Methods: A total of 30 C57 rats were selected, and the monoplast suspension of Lewis lung cancer was injected into the left axilla to prepare the subcutaneous transplanted tumor models in the axilla of right upper limb. The models were randomly divided into Groups A, B, and C. Medication was conducted when the tumor grew to 400 mm3. Group A was the control group without any interventional treatment. Group B was injected with Endostar 5 mg.kg-1.d for 10 d. Group C was given the injection of Endostar 5 mg.kg-1.d combined with intraperitoneal injection of cisplatin 5 mg.kg-1.d for 10 d. All the rats in three groups were executed the day after the 10-d medication and the tumor was taken off for measurement of volume and mass changes and calculation of antitumor rate, after which the vascular endothelial growth factor(VEGF) concentration in rats' plasma was determined by ELISA. The tumor tissues were cut for the preparation of conventional biopsies. After hematoxylin-eosin staining, the pathologic histology was examined to observe the structures of tumor tissues, VEGF score and microvessel density(MVD) in each group. Results: The volume and mass of tumor in Groups B and C were significantly lower than Group A(P < 0.05) while the tumor volume and mass in Group C were significantly lower than Group B(P < 0.05). The antitumor rate in Group C was significantly higher than Group B(P < 0.05), but the tumor VEGF score, MVD and plasma VEGF level in Group C were significantly lower than Groups A and B(P < 0.05). In Group B, the tumor VEGF score, MVD and plasma VEGF level were significantly lower than Group A(P < 0.05). The microscopic image of Group C showed that its number of active tumor cells and the blood capillary around tumor was significantly smaller than that of Groups A and B, and meanwhile atrophy and liquefactive necrosis were seen in local tumor. Conclusions: Endostar injection combined with intraperitoneal injection of cisplatin is effective in reducing tumor VEGF score and MVD of transplanted tumor tissues in rats with Lewis lung cancer to obstruct the nutrient supply of tumor cells and kill tumor cells, so that the inhibition of tumor cell proliferation and metastasis can be achieved with a remarkable effect. 展开更多

Objective:To investigate the effect of radiotherapy plus recombinant human endostatin(RHendostatin) on esophageal cancer and its mechanism.Methods:A total of SO nudemice were equally randomized into control group,radiotherapy group,and combined therapy group Ⅰ,Ⅱ,and Ⅲ after inoculating with Ecal09 cell suspension(1×107 cells/mL).On the day of grouping,control group and radiotherapy group were injected normal saline,while radiotherapy group and 3 combined therapy groups received radiotherapy;besides,combined therapy group Ⅰ,Ⅱ,and Ⅲ was injected RH-endostatin of 2.5,5,10 mg/kg respectively.After 3-week therapy,the tumors of each group were collected and microvessel density and VEGF expression in tumors were determined.In vitro,Eca109 cells were divided into control group,radiotherapy group,and combined therapy group.Forty-eight hours after treatment cell cycle distribution and apoptosis rate were detected,and the activity of VEGF signal paths was semiquantitatively analyzed.Results:Since the 6th day of treatment,the relative tumor proliferation rate of combined therapy group Ⅱ was lower than radiotherapy group(P<0.05) and 40%since the 15 th day.Average microvessel density and EGFR expression in combined therapy group Ⅱ were lower than radiotherapy group(P<0.05).In vitro,the cell percentage in S and G2/M phase of combined therapy group cells was lower than that in radiotherapy group cells,while the apoptosis rate and the expression of VEGF,AKT,p-AKT,ERK1/2 and p-ERKl/2 in combined group were higher than that in radiotherapy group(P<0.05).Conclusions:RH-endostatin promotes the efficacy of radiotherapy on esophageal cancer,which may be partly realized by inhibiting the activity of VEGF related signal paths. 展开更多

目的探讨复方多粘菌素B软膏(TAO)联合重组牛碱性成纤维细胞生长因子(rh-bFGF)凝胶治疗深Ⅱ度烧伤患者的疗效。方法选取2022年1-11月该院收治的深Ⅱ度烧伤患者60例,采用随机数字表法分观察组和对照组,每组30例。对照组给予TAO治疗,观察... 目的探讨复方多粘菌素B软膏(TAO)联合重组牛碱性成纤维细胞生长因子(rh-bFGF)凝胶治疗深Ⅱ度烧伤患者的疗效。方法选取2022年1-11月该院收治的深Ⅱ度烧伤患者60例,采用随机数字表法分观察组和对照组,每组30例。对照组给予TAO治疗,观察组加用rh-bFGF凝胶治疗。对比2组患者临床疗效、症状消失时间、创面愈合时间、换药疼痛、炎症因子、并发症发生率及生活质量。结果观察组患者总有效率明显高于对照组的,并发症发生率明显低于对照组,中、重度疼痛程度明显低于对照组,肿胀消失时间、红斑消失时间、渗液消失时间、水疱消失时间、疼痛消失时间、创面愈合时间均明显短于对照组,差异均有统计学意义(P<0.05)。2组治疗前炎症因子、生活质量比较,差异均无统计学意义(P>0.05);观察组患者治疗后降钙素原、C反应蛋白水平均明显低于对照组,精简烧伤健康量表中的基本生活能力维度、手功能维度、情感维度、人际关系维度、性生活维度、体热维度、热敏感维度、配合治疗维度、工作维度评分均明显高于对照组,差异均有统计学意义(P<0.05)。结论TAO联合rh-bFGF凝胶治疗深Ⅱ度烧伤患者中疗效显著,能加快症状消失及创面愈合,减轻换药疼痛与炎症反应,降低并发症发生率,提高患者生活质量。 展开更多

Fibroblast growth factor 1(FGF1)regulates vertebrate cell growth,proliferation and differentiation,and energy metabolism.In this study,we cloned rainbow trout(Oncorhynchus my kiss)fgf1 and fgf1a,pre-pared their recombinant proteins(rFGF1 and rFGF1a),and described the molecular mechanisms by which they improve glycolipid metabolism in carnivorous fish.A 31-d feeding trial was conducted to investigate whether they could enhance glycolipid metabolism in rainbow trout on high-carbohydrate diets(HCD).A total of 720 rainbow trout(8.9±0.5 g)were equally divided into 4 groups:the chow diet(CD)group injected with PBS,the HCD group injected with PBS,the HCD group injected with rFGF1(400 ng/g body weight),and the HCD group injected with rFGF1a(400 ng/g body weight).The results showed that short-term HCD had a significant positive effect on the specific growth rate(SGR)of rainbow trout(P<0.05).However,it led to an increase in crude fat,serum triglyceride(TG)and glucose content,as well as serum glutamic pyruvic transaminase(GPT)and glutamic oxalacetic transaminase(GOT)contents(P<0.05),suggesting a negative health effect of HCD.Nevertheless,rFGF1 and rFGF1a showed beneficial therapeutic effects.They significantly reduced the crude fat content of the liver,serum TG,GOT,and GPT contents caused by HCD(P<0.05).The upregulation in atgl,hsl,and acc2 mRNAs implied the promotion of TG catabolism.Moreover,rFGF1 and rFGF1a contributed to promoting lipolysis by activating the AMPK pathway and reducing lipid accumulation in the liver caused by HCD.In addition,the rFGF1 and rFGF1 a-treated groups significantly reduced serum glucose levels and elevated hepatic glycogen content under HCD,and increased glucose uptake by hepatocytes.We observed a decrease in mRNA levels for pepck,g6pase,and pygl,along with an increase in mRNA levels for gys,glut2,and gk in the liver.Furthermore,these proteins regulated hepatic gluconeogenesis and glycogen synthesis by increasing the phosphorylation level of AKT,ultimately leading to an increase in GSK3βphosphorylation.In conclusion,this study demonstrates that rFGF1 and rFGF1a can enhance lipolysis and glucose utili-zation in rainbow trout by activating the AMPK pathway and AKT/GSK3βaxis. 展开更多

Objective:: To investigate the effects of recombinant human basic fibroblast growth factor (rhbFGF) on the cell proliferation during mandibular fracture healing in rabbits. Methods: The complex of rhbFGF and bovine type I collagen was implanted into the mandibular fracture site under periosteum of the animal. The whole mandible was harvested at 7, 14, 28, 56 and 84 days respectively after operation. The expression of proliferating cell nuclear antigen (PCNA) in callus was examined with immunohistochemical staining. Results: PCNA-positive cells in callus in the rhbFGF-treated group on days 7 and 14 were more than that in the control group (P< 0.01 ). Conclusions: It indicates that rhbFGF can stimulate cell proliferation during mandibular fracture healing in rabbits. 展开更多

The membranous complex of rhbFGF and bovine type I collagen was prepared and implanted into the rabbit mandible fracture site under periosteum. The animals were sacrificed on 7, 14, 28, 56 and 84 days respectively after operation and the whole mandibles were harvested. The expression of factor Vm related antigen (F8-RA) in callus was examined with immimohistochemical staining.Results: The amounts of microvascular formation in calluses in the rhbFGF-treating group on days 7, 14, 28 and 56 were more than those of the control group (P < 0.01).Conclusions: The results indicated that rhbFGF could stimulate microvascular formation during mandible fracture healing in rabbits. 展开更多

目的探讨重组人碱性成纤维细胞生长因子(rh-bFGF)联合胰岛素强化治疗深Ⅱ度烧伤伴应激性高血糖的效果及对机体代谢、免疫指标的影响。方法选取我院2017年2月—2019年2月收治的深Ⅱ度烧伤伴应激性高血糖97例,采用随机数字表法分为观察组4... 目的探讨重组人碱性成纤维细胞生长因子(rh-bFGF)联合胰岛素强化治疗深Ⅱ度烧伤伴应激性高血糖的效果及对机体代谢、免疫指标的影响。方法选取我院2017年2月—2019年2月收治的深Ⅱ度烧伤伴应激性高血糖97例,采用随机数字表法分为观察组48例和对照组49例,观察组在基础治疗上给予rh-bFGF联合胰岛素强化治疗,对照组在基础治疗上给予rh-bFGF联合中性胰岛素治疗。术后14d观察两组临床疗效,比较两组创面愈合时间,治疗前及治疗后第3、7、14日两组血糖、前白蛋白、尿3-甲基组氨酸(3-MH)和CD3+、CD4+、CD8+、CD4+/CD8+值,以及观察两组治疗期间不良反应发生情况。结果治疗后观察组总有效率为91.67%高于对照组的73.47%(P<0.05)。观察组创面愈合时间为(19.83±2.21)d短于对照组的(23.77±2.96)d,差异有统计学意义(P<0.05)。治疗后两组空腹血糖变化较平稳,均较治疗前降低(P<0.05),且观察组治疗后3、7、14d空腹血糖均低于对照组(P<0.05)。治疗后两组前白蛋白水平均升高,且观察组治疗后3、7、14d前白蛋白水平亦高于对照组,差异均有统计学意义(P<0.05)。治疗后观察组尿3-MH水平降低,且观察组治疗后3、7、14d尿3-MH水平低于对照组,差异均有统计学意义(P<0.05)。两组治疗后CD3+、CD4+、CD8+、CD4+/CD8+水平均较治疗前改善(P<0.05);观察组治疗后同时间点CD4+/CD8+水平较对照组改善明显(P<0.05)。两组治疗期间均未出现严重不良反应。结论rh-bFGF联合胰岛素强化治疗深Ⅱ度烧伤伴应激性高血糖可显著提高临床效果,缩短创面愈合时间,并可纠正机体代谢紊乱状态,改善机体免疫功能,且治疗安全性较好。 展开更多

METHODS: Enucleated eyes were subjected to immunostaining for bFGF protein. Human cataract LECs were cultured in vitro, and treated with bFGF for 48 hr. Proliferation was estimated by the positive area ratio of proliferating cell nuclear antigen (PCNA) in immunohistochemistry. 目的:观察重组人碱性成纤维细胞生长因子(recombinant human basic fibroblast growth factor,rh-bFGF)联合局部外用胰岛素治疗糖尿病深Ⅱ度烧伤的临床价值。方法:选取2017年5月-2018年5月笔者科室收治的80例糖尿病深Ⅱ度烧伤患者,按随... 目的:观察重组人碱性成纤维细胞生长因子(recombinant human basic fibroblast growth factor,rh-bFGF)联合局部外用胰岛素治疗糖尿病深Ⅱ度烧伤的临床价值。方法:选取2017年5月-2018年5月笔者科室收治的80例糖尿病深Ⅱ度烧伤患者,按随机数表法分为观察组与对照组,每组40例。对照组采用rh-bFGF治疗,观察组采用rh-bFGF联合局部外用胰岛素治疗。比较两组创面愈合时间、治疗2周与3周的创面愈合率、治疗前、治疗2周时的创面缺氧诱导因子1α(hypoxia inducible factor-1α,HIF-1α)、血管内皮生长因子(vascular endothelial growth factor,VEGF)及微小RNA21表达,温哥华瘢痕量表(Vancouver Scar Scale,VSS)评价治疗后3个月与6个月的瘢痕增生程度,记录治疗期间不良反应。结果:观察组创面愈合时间(16.82±3.22)d,少于对照组的(21.91±4.01)d,治疗2周、3周的愈合率高于对照组,差异有统计学意义(P<0.05)。治疗2周后,两组创面VEGF、微小RNA21表达均较治疗前增强、HIF-1α表达减弱,差异有统计学意义(P<0.05)。观察组创面VEGF蛋白的阳性表达细胞数高于对照组,HIF-1α蛋白的阳性表达细胞数、微小RNA21的Ct值低于对照组,差异有统计学意义(P<0.05)。观察组治疗后3个月与6个月的VSS总评分分别为(3.94±0.63)分和(4.99±0.79)分,低于对照组(4.57±0.71)分和(5.70±0.82)分,差异有统计学意义(P<0.05)。两组治疗期间均未见严重感染情况。结论:重组人碱性成纤维细胞生长因子联合局部外用胰岛素治疗糖尿病深Ⅱ度烧伤可下调HIF-1α、上调VEGF,缩短创面愈合时间,减轻瘢痕形成,为安全可靠的治疗方案,值得推广。 展开更多
