In this study, we analysis the the synthesis and storage sites of phenolic compound in E. purpurea. We used fluorescent microscopy, transmission electron microscopy, cytochemical and immunocytochemical localization to observe the distribution of phenolic compounds. Our results show that the phenolic compounds were mostly distributed in the cortex parenchyma cells, vascular parenchyma cells and pith parenchyma cells in the root and rhizome, and mainly present in the vacuoles, large intercellular spaces and their surrounding cell walls. No phenolic compounds were observed in the cytoplasm and the organelles. We concluded that the phenolic compounds were synthetized in the cortex parenchyma cells, vascular parenchyma cells and pith parenchyma cells in the root and rhizome, and stored in the vacuoles of parenchyma cells. The above results provided significantly cytological information for further approaching the metabolic regulation and transfer pathways of phenolic compounds in biochemistry and molecular biology.展开更多

[Methods]Volatile oil components from different sites of N. jatamansi were identified and isolated by GC-MS. [Results] There were 56 kinds of volatile oil components from different sites of N. jatamansi,in which 39 components from herb of N. jatamansi,39 components from root and rhizome of N. jatamansi, and there were 22 common components. But 1 component in higher concentration had obvious difference. [Conclusions] The herb of N. jatamansi could not completely replace " root and rhizome" of N. jatamansi as the medicine, which was consistent with prior detection result of each physicochemical index from different sites of N. jatamansi. The research could provide the reference for making quality standard of N. jatamansi medicine and rational basis for its further research and development.展开更多

In the study, we investigated whether BCC had effects on left ventricular hypertrophy (LVH) and myocardial fibrosis in spontaneous hypertensive rats (SHR). For SHR + BCC group, BCC (0.3 g/kg) was orally administered daily for 12 weeks. The SHR group and the Wistar Kyoto rats (WKY, normal control) group, the equal volume of 5‰ CMC-Na distilled water. After 12 weeks, left ventricle was segregated from each rat in the groups, and the left ventricle weight/body weight (LVW/BW) calculated. The volume fraction of collagen (VFC) in myocardium and the diameter of cardiac muscle cell (DCMC) were examined by histological staining. Biochemical indicators of blood sample such as Angiotensin II (Ang II), Aldosterone (ALD), Hyaluronic Acid (HA), Laminin (LN), Procollagen III (PC III) and Collagen type IV (CIV) levels were detected by using radioimmunoassay (RIA). And also NOS and iNOS levels were measured by means of ultra-violet spectroscopy (UV). The results shown that in SHR + BCC group, the LVW/BW, DCMC and VFC decreased significantly versus SHR group, the same as biochemical indicators except NOS and iNOS. All of above index was similar to WKY group. Statistically significant correlations were found among the plasma Ang II level, the mean systolic blood pressure (SBP), and the NOS level of the three groups. Our study indicates that the BCC can control the LVH and myocardial fibrosis in SHR.展开更多

Methods: Subconfluent HaCaT cells were sham or UVB irradiated and treated with above TCM agents. The damage degree of HaCaT cells was observed by a light microscop. Cell growth was recorded by cell count and cellular activity was detected by MTT method. The secretion amount of IL-6 and TNF-α was measured by ELISA. Results: The irradiation damage of HaCaT cells was depended on the irradiated dosages and cellular activity was reduced by 36%-80%, with a maximum decrease over 90% after 72 h. The intervention of the above drugs may increase the cellular activity by 10%-72%. The photo-protective efficiency was more apparent in EGCG (from 1.19±0.07 to 1.28±0.06, P<0.01) than that in hydroxychloroqine (from 0.43±0.04 to 0.96±0.04, P<0.05). The other two tested drugs also showed photo-protective effect(from 0.44±0.07 to 1.21±0.02, P<0.05). As to cytokine secretion, EGCG could decline the secretion amount of IL-6 and TNF-α apparently. Hydroxychloroqine and baikal skullcap root baikal skullcap root could only reduce the secretion of IL-6. The secretion of IL-6 and TNF-α could not be inhibited by szechwan lovge rhizome. Conclusion: The injury effect of UVB irradiation on cultured keratinocytes is dose-dependent and the tested drugs have photo-protective potency. Inhibition of cytokine secretion may be one of the mechanisms related to reducing the damage effect of UVB irradiation.展开更多

The species is threatened in the natural habitat due to unsustainable harvesting of rhizome for medicine and habitat destruction. The species has very poor adaptive power to the synthetic environment and fails to grow and propagate. Present study was undertaken to develop suitable low cost propagation technique and produce clonal planting materials through rhizome splitting and root cutting. Rhizomes and roots were cut into segments and sowed in the raised soil bed prepared by mixing decayed wood powder, sand and top black soil at 1:1:3 ratios in a shaded poly house. Of the sowed rhizome segments ~55% segments remained recalcitrant to morphogenetic response in the first year of the study and the response improved in the subsequent years. In the third year ~51% rhizome segments responded positively. The horizontal root cuts mostly remained morphogenetically dormant but inclined root cuts exhibited better response. The morphogenetic response from the roots was comparatively very poor compare to rhizome segments. The plantlets formed both rhizome segments and root cuttings were maintained for three years in the bed/pots followed by transferring in the cultivated plots.展开更多

Methods: Eight 8-week-old male C57BL/6J mice were selected as the control group. Forty 8-week-old male apoE KO mice were randomly divided into the model group, apoE KO + intraperitoneal injection immunoglobulin group (IVIG), apoE KO + simvastatin group (Sm), apoE KO + high dosage of Xiongshao Capsule (XSC,芎芍胶囊) group (XSCH), and apoE KO + low dosage of XSC group (XSCL), 8 mice in each group. Mice in the control group were put on a normal diet, and others were fed with a high-fat diet. After 10-week different interventions, monocyte CD16 expression was detected by flow cytometry, aortic matrix metalloproteinase-9 (MMP-9) mRNA expression was detected using reverse transcription- polymerase chain reaction, and serum tumor necrosis factor (TNF)-a level was detected using enzyme-linked immunosorbent assay. Results: Compared with the control group, monocyte CD16 expression, aortic MMP-9 mRNA expression, and serum TNF-a level in the model group increased obviously (P〈0.01). Injections of apoE KO mice with intraperitoneal immunoglobulin during a 5-day period significantly reduced the monocyte CD16 expression, aortic MMP-9 mRNA expression, and serum TNF-a level (P〈0.01 or 0.05) over a 10-week period of high-fat diet. Indices above in the Sm group, XSCH group, and XSCL group decreased in a different degree. Of them, the aortic MMP-9 mRNA expression in XSCH group was lower than that in Sm group (P〈0.05) and the monocyte CD16 expression and serum TNF-a level showed no significant difference between XSCH group and Sm group (P〉0.05). Correlation analyses suggested positive correlation between monocyte CD16 expression and aortic MMP-9 mRNA expression or serum TNF-a level in IVIG group, XSCH group, and XSCL group. Conclusions: Fc γ/R IliA mediates systemic inflammation in the progression of coronary heart disease with blood stasis syndrome. XSC could stabilize atherosclerotic plaque by suppressing inflammation and its target was relative with Fc γ RIIIA.展开更多
