In this study,two indica varieties with different dormancy characteristics [4 K58( II-32 B dormant),4 K59( II-32 B) ] and their F2 seeds( C178,C179) obtained through hybridization with sterile line( II-32 A) were used as materials. Different aged seeds( 0,3,6 and 9 d) of these four varieties were acquired by artificial accelerated aging method. Effects of artificial aging on malondialdehyde( MDA) content and catalase( CAT),ascorbate peroxidase( APX),peroxidase( POD) and superoxide dismutase( SOD) activity were investigated. The results showed that with the prolongation of aging time,MDA contents of these four rice varieties increased significantly,while the activity of antioxidant enzymes decreased dramatically. Besides,MDA contents and SOD,CAT and APX activity of the two inbred lines were significantly lower than those of the hybrid varieties. In addition,the activity of antioxidant enzymes in 4 K58 and C178 was significantly lower than that in 4 K59 and C179,respectively. This study indicated that artificial aging treatment significantly inhibited the activity of antioxidant system in seeds,improved membrane lipid peroxidation degree,and thus aggravated the deterioration of seeds. In addition,it also suggested that rice seeds with dormant property were more intolerant to storage. 展开更多

Vegetation restoration and reconstruction are effective approaches to desertification control and achieving social and economic sustainability in desert areas.However,the self-succession ability of native plants during the later periods of vegetation restoration remains unclear.Therefore,this study was conducted to bridge the knowledge gap by investigating the regeneration dynamics of artificial forest under natural conditions.The information of seed rain and soil seed bank was collected and quantified from an artificial Caragana korshinskii Kom.forest in the Tengger Desert,China.The germination tests were conducted in a laboratory setting.The analysis of species quantity and diversity in seed rain and soil seed bank was conducted to assess the impact of different durations of sand fixation(60,40,and 20 a)on the progress of vegetation restoration and ecological conditions in artificial C.korshinskii forest.The results showed that the top three dominant plant species in seed rain were Echinops gmelinii Turcz.,Eragrostis minor Host.,and Agropyron mongolicum Keng.,and the top three dominant plant species in soil seed bank were E.minor,Chloris virgata Sw.,and E.gmelinii.As restoration period increased,the density of seed rain and soil seed bank increased first and then decreased.While for species richness,as restoration period increased,it gradually increased in seed rain but decreased in soil seed bank.There was a positive correlation between seed rain density and soil seed bank density among all the three restoration periods.The species similarity between seed rain or soil seed bank and aboveground vegetation decreased with the extension of restoration period.The shape of the seeds,specifically those with external appendages such as spines and crown hair,clearly had an effect on their dispersal,then resulting in lower seed density in soil seed bank.In addition,precipitation was a crucial factor in promoting rapid germination,also resulting in lower seed density in soil seed bank.Our findings provide valuable insights for guiding future interventions during the later periods of artificial C.korshinskii forest,such as sowing and restoration efforts using unmanned aerial vehicles. 展开更多

Seed vigor is a crucial trait for the direct seeding of rice.Here we examined the genetic regulation of seed vigor traits in rice,including germination index(GI)and germination potential(GP),using a genome-wide association study approach.One major quantitative trait locus,qGI6/qGP6,was identified simultaneously for both GI and GP.The candidate gene encoding the cytochrome c oxidase subunit 5B(OsCOX5B)was validated for qGI6/qGP6.The disruption of OsCOX5B caused the vigor traits to be significantly lower in Oscox5b mutants than in the japonica Nipponbare wild type(WT).Gene co-expression analysis revealed that OsCOX5B influences seed vigor mainly by modulating the tricarboxylic acid cycle process.The glucose levels were significantly higher while the pyruvic acid and adenosine triphosphate levels were significantly lower in Oscox5b mutants than in WT during seed germination.The elite haplotype of OsCOX5B facilitates seed vigor by increasing its expression during seed germination.Thus,we propose that OsCOX5B is a potential target for the breeding of rice varieties with enhanced seed vigor for direct seeding. 展开更多

Rice production generates a significant amount of agricultural waste. This study aimed to give results related to the existence of antioxidant phenols in agricultural waste of selected Northern Thai rice varieties. The antioxidant activities, contents of total flavonoids and phenolic compounds in the ethanolic rice husk extract were evaluated. The highest antioxidant activities were found in the variety PES1CMU, with 2,2’-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid and 2,2-diphenyl-1-picrylhydrazyl as 679.66 and 4.16 mmol/(L·g) trolox equivalent, respectively, ferric reducing antioxidant power as 0.87 mmol/(L·g) Fe2+, total phenolic content as 29.90 mmol/(L·g) gallic acid and total flavonoid content as 12.16 mg/g catechin equivalent. Polyphenol compounds were identified mainly by standard polyphenols using the liquid chromatography mass spectrometry, with the highest contents of phytic acid, o-coumaric acid, naringin and kaempferol. The non-glutenous and wetland ecotypes of rice husk samples were the richest in antioxidant activities and polyphenol contents characterized by using principal component analysis. The glutenous rice husk contained higher antioxidant activities than the rest. Interestingly, quercetin is a significant phenolic compound that positively correlated with the overall antioxidant activities of rice husk. This finding will be relevant for future application of rice husk antioxidant components in the production of functional ingredients as well as for the food and pharmaceutical industries. 展开更多

In order to investigate the effects of seed rate on root twining power and seedling quality of machine-transplanted super rice cultivars, Yongyou 12 and Zhongzheyou No.l, total 6 seed rates were arranged, and the growth characteris- tics, quality and root twining power of machine-transplanted rice seedlings were studied. The results showed that seed rate had great effects on root twining power and qualityof rice seedlings; with the increase of seed rate, the root twining power of rice seedlings was increased, but the quality of rice seedlings was significantly reduced; under the condition of high seed rate, rice seedlings were weak with less accumulate dry matter and nonuniform growth, and the proportion of weak seedlings was higher. When the seed rate of both rice cultivars was reduced by 10 g/tray, the average seedling height was increased by 0.5 cm, leaf age was increased by 0.1, stem base diameter was increased by 0.1 cm, and root number per plant was increased by 0.3. When the seed rate was increased by 10 g/tray, the accumulated dry matter per plant was reduced by 5.1 mg. With the increase of seed rate, the seedling quality of Zhongzheyou No.1 was decreased rapidly, and the proportion of weak seedlings was increased rapidly. However, Yongyou 12 has a larger elasticity in seed rate. When the seed rate reached to 50 g/tray, blanket seedlings of Yongy- ou 12 were nursed with uniform growth and higher quality, but the shoots were sparse with lower leaf area index. However, when the seed rate was up to 50 g/tray, blanket seedlings of Zhongzheyou No.1 were nursed with uniform growth and higher quality, as well as higher-density shoots. From a comprehensive point of view, the optimum seed rates of Yongyou 12 and Zhongzheyou No.1 were 65 and 50 g/tray, respectively. 展开更多

Nitrogen(N) and seeding rates are important factors affecting grain yield and N use efficiency(NUE) in directseeded rice. However, these factors have not been adequately investigated on direct-seeded and double-season rice(DDR) in Central China. The objective of this study was to evaluate the effects of various N and seeding rates on the grain yield and NUE of an ultrashort-duration variety grown under DDR. Field experiments were conducted in 2018 in Wuxue County and 2019 in Qichun County, Hubei Province, China with four N rates and three seeding rates.The results showed that the grain yield of the ultrashort-duration variety ranged from 6.32 to 8.23 t ha–1with a total growth duration of 85 to 97 days across all treatments with N application. Grain yield was increased significantly by N application in most cases, but seeding rate had an inconsistent effect on grain yield. Furthermore, the response of grain yield to the N rates was much higher than the response to seeding rates. The moderate N rates of 100–150 and 70–120 kg N ha–1in the early and late seasons, respectively, could fully express the yield potential of the ultrashort-duration variety grown under DDR. Remarkably higher N responses and agronomic NUE levels were achieved in the early-season rice compared with the late-season rice due to the difference in indigenous soil N supply capacity(INS) between the two seasons. Seasonal differences in INS and N response should be considered when crop management practices are optimized for achieving high grain yield and NUE in ultrashort-duration variety grown under DDR. 展开更多

As an abiotic stress,adverse germination temperatures cause serious disruptions in physiological and biochemical processes involved in seed germination.Using a factorial experiment,we examined the effects of different seed priming treatments on enzymatic and biochemical performances of rice seed germination under different temperatures.Each of the rice genotypes(Hashemi,Sadry-domsefid,IRON-70-7053-7 and NORIN-22)was primed with hydro-hardening,KCl,CaCl2 and ascorbic acid(AsA)and without a priming agent as a control at low(15℃),optimum(25℃)and high(35℃)germination temperatures.The results showed that the enzymatic and biochemical performances of all the rice genotypes were affected by the seed priming agents,especially under the low germination temperature.At 15℃,seed priming with AsA was found to be the best agent for the activities of amylase,α-amylase,catalase(CAT),peroxidase(POX),ascorbate peroxidase(APOX)and superoxide dismutase(SOD)as well as the content of soluble sugars in the NORIN-22 genotype,and for protease activity and soluble protein content in the IRON-70-7053-7 genotype.SOD at the low germination temperature and CAT,POX and protease at the optimum and high germination temperatures were the most important enzymes in occurrence of germination potential in terms of seedling length,vigor index,normal seedling rate and germination rate.Under the priming agents,the highest changes in normal seedling rate were observed at the low and optimum germination temperatures by AsA priming in the Hashemi and NORIN-22 genotypes,and at the high germination temperature under KCl priming in the Hashemi genotype. 展开更多

Drought stress is a serious threat to the germination of plant seeds and the growth of seedlings.Melatonin has been proven to play an important role in alleviating plant stress.However,its effect on seed germination under drought conditions is still poorly understood.Therefore,we studied the effects of melatonin on rice seed germination and physiological characteristics under drought stress.Rice seeds were treated with different concentrations of melatonin(i.e.,0,20,100,and 500μM)and drought stress was simulated with 5%polyethylene glycol 6000(PEG6000).The results showed that 100μM melatonin can effectively improve the germination potential,rate and index;the vigor index of rice seeds;and the length of the shoot and root.In addition,that treatment also increased the activity of superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT),and reduced the content of malondialdehyde(MDA).The grey relational grade between the shoot MDA content and the melatonin seed-soaking treatment was the highest,which could be useful for evaluating the effect of melatonin on drought tolerance.Two-way analysis of variance showed that the effect of single melatonin treatment on rice seeds was more significant than that of single drought stress and interaction treatment of drought and melatonin(p<0.05).The subordinate function results showed that 100μM melatonin significantly improved the germination and physiological indexes of rice seeds and effectively alleviated the adverse effects of drought stress on rice seedlings.The results helped to improve the understanding of the morphological and physiological involvement of melatonin in promoting seed germination and seedling development under drought stress. 展开更多

A Pleiotropic Drug Resistance Family Protein Gene Is Required for Rice Growth, Seed Development and Zinc Homeostasis

Zn deficiency in crops frequently occurs in many types of soils. It is therefore crucial to identify genetic resources linking Zn acquisition traits and development of crops with improved Zn-use efficiency for sustainable crop production. In this study, we functionally identified a rice uncharacterized ABCG(ATP-binding cassette G-subfamily) gene encoding a PDR20(pleiotropic drug resistance 20) metal transporter for mediation of rice growth, seed development and Zn accumulation. OsPDR20 was localized to the plasma membrane, but it was not transcriptionally induced under Zn deficiency, rather was sufficiently up-regulated under high level of Zn stress. Yeast(Saccharomyces cerevisiae) transformed with OsPDR20 displayed a relatively lower Zn accumulation with attenuated cellular growth, suggesting that OsPDR20 had an activity for Zn transport. Knocking-down OsPDR20 by RNA interference(RNAi) compromised rice growth with shorter plant height and decreased biomass in rice plantlets grown under hydroponic media. Zn concentration in the roots of OsPDR20 knocked-down rice lines declined under Zn deficiency, while they remained unchanged compared with the wild type under normal Zn supply. A rice lifelong field trial demonstrated that OsPDR20 mutation impaired the capacity of seed development, with shortened panicle and seed length, compromised spikelet fertility, and reduced grain number per plant or grain weight per unit area. Interestingly, OsPDR20 mutation elevated the accumulation of Zn in husk and brown rice over the wild type. Overall, this study pointed out that OsPDR20 is fundamentally required for rice growth and seed development through Zn transport and homeostasis. 展开更多

ObjectiveThis study aimed to investigate the effect of fulvic acid on the growth and yield components of direct seeding rice (Nanjing 44). MethodThe rice seeds were soaked in 0 (water as a control), 1, 2, 4 and 6 g/L fulvic acid (FA) before sowed. Then, the rice morphological indices, leaf chlorophyll content, photosynthesis parameters, root activity and chlorophyll fluorescence parameters were measured in the following field studies. ResultCompared with the control, the leaf area index, chlorophyll content, net photosynthetic rate, stomatal conductance and dry matter weight and some fluorescence parameters such as the maximum photochemical efficiency of photosystem II (PSII) (Fv/Fm), excitation energy capture efficiency of opened PSII reaction center (Fv’/Fm’) and efficiency of the open reaction centre (ΦPS II) increased by different levels at both jointing stage and heading stage of direct seeding rice, whose seeds were soaked by FA with different concentrations. High FA concentration (4-6 g/L) significantly increased the cultivated rice leaf area index, chlorophyll content, net photosynthetic rate, stomatal conductance, transpiration rate and dry matter weight by 10.32% -22.88% , 5.88% -13.11% , 12.16% -26.84% , 11.43% -88.46% , 10.63% -21.63% , 18.49% -19.68% , respectively, thereby improving the physiological function and light energy transform efficiency of rice at the growth stage. With FA concentration increasing, the yield, effective panicles, grain number per panicle and seed setting rate were increased significantly compared with the control by 17.52%-18.71%, 3.46%-3.85%, 6.30%- 6.51% and 7.82%-8.69% respectively. ConclusionSoaking rice seed with FA could be considered as an effective way to improve the rice competitiveness at early growth stage. 展开更多

The results show that when 100 kg of rice seeds were coated with 200 g of 35% thiamethoxam-prochloraz FS,the control effects to rice thrips were 93.0% and 84.7% 15 and 20 days after sowing, and the control effect to rice bakanae disease was 90.8% 30 days after sowing. As 100 kg of rice seeds were coated with 250 g of 35% thiamethoxam-prochloraz FS, the control effects to rice thrips were 98.2% and 94.9% 15 and 20 days after sowing, and the control ef- fect to rice bakanae disease was 94.6% 30 days after sowing. 35% thiamethoxam- prochloraz FS is safe to rice seedlings and can effectively promote the growth of rice seedlings. 展开更多

The insecure problems of seed production have seriously hampered the healthy and sustainable development of two-line hybrid rice.The safety issues on seed production of two-line hybrid rice and current research situation were pointed out in this paper.The three main reasons for unsafety in seed production of twoline hybrid rice were unsuitable site selection,high critical sterility-inducing temperature and the drift of critical temperature.In this paper,strategies and measures were put forward based on many years of practice.It could minimize the risk in seed production of two-line hybrid rice by selecting dual-purpose genic male sterile line with lower critical sterility-inducing temperature and long lower temperature resistant time.Based on the climate data and climatic demands of the "three safe-periods" in seed production,a new idea for determining appropriate bases and periods for seed production was proposed by using computer technology,which solved the aimless selection of sites and periods for the seed production of two-line hybrid rice.Besides,we established a system of single plant selection and identification method and original seed propagation with cyclic cold water,which could reduce the generation number of original seed propagation in seed production and avoid the drift of critical sterility-inducing temperature.This paper improved the seed production safety in the three aspects of seed nature,seed source and seed production site. 展开更多

The result showed that spraying 3. 375 L/hm2 48% bentazon aqua could control the self-pollinated fertilization rate below 1.0% and survival plant rate was 0. The spray of bentazon would not influence MC526 pollen vitality. Seed setting rate of female parent was not obvious different from that of control group with spraying bentazon aqua more than 3. 375 L/hm^2 from the sprouting appearing period to full heading stage. This demonstrated that bentazon aqua had no influence on seed setting rate during florescence of female parent. The weight ratio between male parent seeds and female parent seeds was 1 to 5, then seeds from parents with that weight ratio would be mixedly planted for seed production. With this weight ratio, relative proper structure of panicle and grain would be obtained ,panicle ratio was 1 to 4.3, glumous flowers ratio was 1 to 3.5. Cross seed-setting rate of female parent was 72.6% and seed production per hm^2 was 66 000 kg. 展开更多

[Method] Totaling 7 drug doses were set in the test and the effects were observed 5, 10, 15 and 30 d after drug application, with the remained weeds surveyed 15 and 30 d after drug application. [Result] The test showed pyrazosulfuron-ethyl·quinclorac 50%WP was safe to rice as treatment agent of stem and leaf, and rice yield increased dramatical y, with the growth rate in 2.1%-49.3%. After the drug application, the prevention effects on Echinochloa crusgal i, Ludwimlia prostrata, Monochoria vamli-nalis, and Zong grass were 92.2%, 96.6%, 93.5%, and 94.8%, and the effects on fresh weight were 94.8%, 97.1%, 93.3% and 94.9%, respectively. [Conclusion] Pyra-zosulfuron-ethyl·quinclorac 50% WP can be taken as treatment agents of stem and leaf of dry direct seeded rice. 展开更多

However, the high cost of seed production and the complicated procedures have become a bottleneck in the development of hybrid rice. The research progress on mixed sowing seed production techniques of hybrid rice was introduced from the aspects of rice resources creation, breeding, sowing seed technology research and cost benefit analysis. The production technology of the new mixed seeding combina- tion "Xinhunyou 6" was investigated, including the research and validation of benta- zon treatment period and dosage, mixing ratio of male and female parents, and the comparative test of different different sowing methods, which revealed that the mechanization technology of seed production of hybrid rice was mature and feasible and would be one of the most important development trend of technological devel- opment of hybrid rice production. 展开更多
