Rock Engravings and Paintings: Rethinking of the Cupules, Gongs, and Grinding Hollows of Siuyu and Ughaugha in Singida East (Tanzania)

Our recent archaeological expeditions in the Siuyu and Ughaugha wards of Singida eastern province have recuperated the very compelling rock engravings (petroglyphs) that weren’t reported in Tanzania before. Archaeologically, Singida’s fame emanated from her endowments of rock paintings. However;during this research, we did discover rock cupules, gongs, and grinding hollows art together with rock paintings (pictographs) that have opened up a new avenue for rock art studies in Tanzania. We carried out intensive site surveys around four sites namely Siuyu, Ngaghe, Misimbwa and Ughaugha B with the aims of solely examining, recording, and documenting archaeological artifacts on the surface in order to research the cultural and behavioural patterns of early humans in the Singida region. Surveys also enabled us to locate suitable areas where future excavations would be established. In some cases, ethnographic enquiries were employed to get supplementary information on the present-day use of rock art sites. Besides, we interviewed local people to assess their general understanding of the presence of more sites or helping in locating, and discovering new sites. Through ethnographic inquiries, we discovered several sites with substantial number of rock engravings that were never document by any researcher. On top of that, this study reports other archaeological potentials of the region by describing in detail their significances for future research undertakings. Additionally, this paper article reports on the occurrence of Middle Stone Age (MSA) archaeological assemblages from the open-air site and the presence of grinding hollows and rock cupules (Siuyu complexes) in central Tanzania for the first time. 展开更多

A multi-mineral model for predicting petrophysical properties of complex metamorphic reservoirs:Case study of the Bozhong Depression,Bohai Sea

Interpreting reservoir properties through log data and logging responses in complex strata is critical for efficient petroleum exploitation,particularly for metamorphic rocks.However,the unsatisfactory accuracy of such interpretations in complex reservoirs has hindered their widespread application,resulting in severe inconvenience.In this study,we proposed a multi-mineral model based on the least-square method and an optimal principle to interpret the logging responses and petrophysical properties of complex hydrocarbon reservoirs.We began by selecting the main minerals based on a comprehensive analysis of log data,X-ray diffraction,petrographic thin sections and scanning electron microscopy(SEM)for three wells in the Bozhong 19-6 structural zone.In combination of the physical properties of these minerals with logging responses,we constructed the multi-mineral model,which can predict the log curves,petrophysical properties and mineral profile.The predicted and measured log data are evaluated using a weighted average error,which shows that the multi-mineral model has satisfactory prediction performance with errors below 11%in most intervals.Finally,we apply the model to a new well“x”in the Bozhong 19-6 structural zone,and the predicted logging responses match well with measured data with the weighted average error below 11.8%for most intervals.Moreover,the lithology is dominated by plagioclase,K-feldspar,and quartz as shown by the mineral profile,which correlates with the lithology of the Archean metamorphic rocks in this region.It is concluded that the multi-mineral model presented in this study provides reasonable methods for interpreting log data in complex metamorphic hydrocarbon reservoirs and could assist in efficient development in the future. 展开更多

目的 基于RhoA/ROCK信号通路探讨L-3-正丁苯酞(L-3-n-butylphthalide, NBP)对实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis, EAE)小鼠的疗效及潜在机制。方法 32只雌性C57BL/6小鼠随机分为4组:对照组、EAE组、... 目的 基于RhoA/ROCK信号通路探讨L-3-正丁苯酞(L-3-n-butylphthalide, NBP)对实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis, EAE)小鼠的疗效及潜在机制。方法 32只雌性C57BL/6小鼠随机分为4组:对照组、EAE组、NBP低剂量组(L-NBP)和NBP高剂量组(H-NBP),每组8只。使用髓鞘少突胶质细胞糖蛋白(myelin oligodendrocyte glycoprotein 35-55, MOG35-55)作为抗原乳剂免疫诱导C57BL/6小鼠建立EAE模型。EAE模型制作成功后,L-NBP组和H-NBP组分别以3.25,6.5 mg/(kg·d)NBP腹腔注射,连续28 d。免疫当日起,每7 d记录各组小鼠的体质量,每天进行神经功能障碍评分;对脊髓组织进行HE染色和劳克坚牢蓝(Luxol fast blue, LFB)染色,观察病理改变;使用酶联免疫吸附试验法(ELISA)检测外周血白细胞介素-10(IL-10)、IL-1β、IL-6和IL-18含量;real time-PCR检测脊髓组织炎症相关因子肿瘤坏死因子(TNF)-α、TNF受体1(TNFR1)、IL-1β的表达;Western blot检测RhoA、ROCKⅠ及ROCKⅡ蛋白表达情况。结果 与对照组相比,EAE组小鼠体质量下降,神经功能障碍评分、组织病理学评分升高,IL-10含量降低,IL-18、IL-6和IL-1β含量增加,TNF-α、TNFR1及IL-1β mRNA表达升高,RhoA、ROCKⅠ及ROCKⅡ蛋白表达增高,差异均有统计学意义(P<0.05)。与EAE组相比,L-NBP和H-NBP组小鼠体质量增加,发病潜伏期延长,高峰期延迟,神经功能障碍评分、组织病理学评分降低,IL-10含量增高,IL-1β、IL-6和IL-18含量降低,TNF-α、TNFR1及IL-1β mRNA表达降低,RhoA、ROCKⅠ和ROCKⅡ蛋白表达降低,差异均有统计学意义(P<0.05)。与L-NBP组相比,H-NBP组小鼠体质量增加,发病潜伏期延长,高峰期延迟,神经功能障碍评分、组织病理学评分降低,IL-10含量增高,IL-1β、IL-6和IL-18含量降低,TNF-α、TNFR1和IL-1β mRNA表达降低,RhoA和ROCKⅡ蛋白表达降低,差异均有统计学意义(P<0.05)。结论 NBP能够减轻EAE小鼠外周及中枢炎症反应,进而改善EAE的神经障碍症状,其作用机制可能是通过抑制RhoA/ROCK信号通路来实现的。 展开更多

目的探讨丹参酚酸B盐(Sal B)对大鼠肝星状细胞(HSC)中内皮素-1(ET-1)激活的RhoA/ROCK信号通路的影响。方法采用肝脏原位灌流酶消化、Nycodenz密度梯度离心法分离大鼠HSC。免疫蛋白印迹法检测肌球蛋白磷酸酶靶亚基1(MYPT1)磷酸化。特异... 目的探讨丹参酚酸B盐(Sal B)对大鼠肝星状细胞(HSC)中内皮素-1(ET-1)激活的RhoA/ROCK信号通路的影响。方法采用肝脏原位灌流酶消化、Nycodenz密度梯度离心法分离大鼠HSC。免疫蛋白印迹法检测肌球蛋白磷酸酶靶亚基1(MYPT1)磷酸化。特异性抗体沉淀ROCK,进行体外磷酸化反应,以磷酸化底物Thr696-MYPT1(654-880)的含量反映ROCK活性。GST下拉实验检测RhoA活性。结果在大鼠HSC中,ET-1刺激后,RhoA和ROCK Ⅱ活性显著增加,ROCK I活性无明显变化,MYPT1 Thr696和Thr850磷酸化水平均显著增加。ET-1刺激1 min和10 min时,RhoA活性分别是基础状态下的1.95倍(P<0.05)和5.84倍(P<0.01)。ET-1刺激2.5 min和15 min时,ROCK Ⅱ活性分别是基础状态下的3.49倍和4.83倍,均P<0.01,差异有统计学意义。ET-1刺激2.5 min时,MYPT1 Thr696磷酸化水平是基础状态下的3.86倍;刺激15 min时,Thr696磷酸化达到高峰,是基础状态下的5.17倍。Thr850磷酸化亦在ET-1刺激15 min达到高峰,是基础状态下的3.33倍,均P<0.01,差异有统计学意义。在ET-1刺激前给予10-5 mol/L Sal B预处理,则使ET-1诱导的RhoA和ROCK Ⅱ活性分别下降66.84%和76.79%,ET-1诱导的MYPT1 Thr696磷酸化下降80.09%,对Thr850磷酸化水平无影响。结论 Sal B能显著抑制大鼠HSC中ET-1诱导的RhoA和ROCK Ⅱ活化,抑制MYPT1Thr696磷酸化。 展开更多

目的观察透骨消痛胶囊对TNF-α诱导体外培养大鼠关节软骨细胞Rho/Rock信号通路的影响,探讨其防治骨性关节炎的作用机制。方法采用4周龄SD大鼠膝关节软骨建立软骨细胞体外培养体系,选取第3代软骨细胞进行实验,20 ng/mL的TNF-α诱导体外... 目的观察透骨消痛胶囊对TNF-α诱导体外培养大鼠关节软骨细胞Rho/Rock信号通路的影响,探讨其防治骨性关节炎的作用机制。方法采用4周龄SD大鼠膝关节软骨建立软骨细胞体外培养体系,选取第3代软骨细胞进行实验,20 ng/mL的TNF-α诱导体外培养软骨细胞的凋亡,诱导成功后,给予透骨消痛胶囊(500、100、20μg/mL)孵育24 h后,用流式细胞仪检测软骨细胞Annexin-Ⅴ观察细胞凋亡情况,采用RT-PCR和Western Blot测定各组软骨细胞RhoA、Rock1和PKN等mRNA和蛋白表达。结果 TNF-α组细胞凋亡明显增多,透骨消痛胶囊各组能减少软骨细胞的凋亡,TNF-α组的RhoA、Rock1、PKN、MKK3、ERK6、Cbfα1等mRNA和蛋白表达较空白组上升,而透骨消痛胶囊各组对RhoA/Rock信号传导通路关键信号分子RhoA、Rock1、PKN及Cbfα1等的mRNA和蛋白的表达有抑制,差异均具有统计学意义(P<0.05)。结论透骨消痛胶囊可减轻TNF-α所致的软骨细胞凋亡,其机制可能与抑制软骨细胞的RhoA/Rock信号通路有关。 展开更多

背景:Lingo-1在神经元轴突再生、髓鞘形成等过程中具有重要作用,但其是否参与对神经干细胞分化的调控以及具体机制尚无定论。目的:观察干预神经干细胞Lingo-1表达水平对其分化方向的影响,并探索其可能机制。方法:从新生大鼠脑组织分离... 背景:Lingo-1在神经元轴突再生、髓鞘形成等过程中具有重要作用,但其是否参与对神经干细胞分化的调控以及具体机制尚无定论。目的:观察干预神经干细胞Lingo-1表达水平对其分化方向的影响,并探索其可能机制。方法:从新生大鼠脑组织分离培养神经干细胞,经神经干细胞标志物检测鉴定后分别使用Lingo-1 shRNA慢病毒载体及阴性对照病毒转染神经干细胞,再用成神经元诱导分化培养基培养7 d,随后运用免疫荧光、免疫RhoA印迹技术检测神经元相关标志物表达,并检测、ROCK1蛋白表达水平。结果与结论:(1)从新生大鼠脑组织所分离培养的神经干细胞呈浮球状生长,高表达神经干细胞特异性标志物Nestin、Sox2与Pax6,符合神经干细胞特征;(2)慢病毒载体转染可在mRNA及蛋白质水平实现对神经干细胞Lingo-1的干扰,经诱导培养7 d后Lingo-1敲低的神经干细胞分化为神经元的比例显著增加,且伴随着RhoA、ROCK1蛋白表达水平的下降;(3)结果表明,Lingo-1参与对神经干细胞分化方向的调控,抑制Lingo-1可促进其向神经元方向分化,该过程可能与调控Rho-ROCK通路有关。 展开更多

The importance of confinement stresses has been recognized in the literature by many researchers, in particular, its influence on strength and on the angle of fracture, but still there is not a clear description for the influence of confining stress on the crack propagation mechanism of rocks. This paper presents a numerical pro- cedure for the analysis of crack propagation in rock-like ma- terials under compressive biaxial loads. Several numerical simulations of biaxial tests on the rock specimen have been carried out by a bonded particle model (BPM) and the influ- ence of confinement on the mechanism of crack propagation from a single flaw in rock specimens is studied. For this purpose, several biaxial compressive tests on rectangular spec- imens under different confinement stresses were modeled in (2 dimensional particle flow code) PFC2D. The results show that wing cracks initiate perpendicular to the flaw and trend toward the direction of major stress, however, when the lat- eral stresses increase, this initiation angle gets wider. Also it is concluded that in addition to the material type, the initiation direction of the secondary cracks depends on confine- ment stresses, too. Kerogen content and kerogen porosity play a significant role in elastic properties of organic-rich shales. We construct a rock physics model for organic-rich shales to quantify the effect of kerogen content and kerogen porosity using the Kuster and Toks6z theory and the selfconsistent approximation method. Rock physics modeling results show that with the increase of kerogen content and kerogen-related porosity, the velocity and density of shales decrease, and the effect of kerogen porosity becomes more obvious only for higher kerogen content. We also find that the Poisson's ratio of the shale is not sensitive to kerogen porosity for the case of gas saturation. Finally, for the seismic reflection responses of an organic-rich shale layer, forward modeling results indicate the fifth type AVO re- sponses which correspond to a negative intercept and a positive gradient. The absolute values of intercept and gradient increase with kerogen content and kerogen porosity, and present predictable variations associated with velocities and density. 展开更多

β-1,3-半乳糖-O-糖基-糖蛋白β-1,6-N-乙酰氨基葡萄糖转移酶(β-1,3-galactosyl-O-glycosyl-glycoproteinβ-1,6-N-acetylglucosaminyltransferase,GCNT3)是黏液蛋白质生物合成中不可缺少的一类N-乙酰葡萄糖转移酶。越来越多的证据表明... β-1,3-半乳糖-O-糖基-糖蛋白β-1,6-N-乙酰氨基葡萄糖转移酶(β-1,3-galactosyl-O-glycosyl-glycoproteinβ-1,6-N-acetylglucosaminyltransferase,GCNT3)是黏液蛋白质生物合成中不可缺少的一类N-乙酰葡萄糖转移酶。越来越多的证据表明,GCNT3的异常表达与肿瘤的侵袭以及病人的生存率有关,然而相关的研究仍较少报道。本研究旨在揭示GCNT3在调控肝细胞癌进程中的潜在机制。本研究首先利用高通量基因表达数据库(Gene Expression Omnibus,GEO)和癌症基因组图谱(Cancer Genome Atlas databases,TCGA)数据库分析肝癌和正常组织中GCNT3的mRNA表达水平,结果表明,肝癌组织中GCNT3的mRNA表达水平高于正常组织(P≤0.001)。同时利用免疫组化、Western印迹进一步分析了肝癌组织、癌旁组织、正常组织中GCNT3蛋白的表达,发现有30%肝癌组织GCNT3表达水平高于癌旁组织和正常组织。随后,在肝癌细胞系HCCLM3和Huh7细胞中,采用CCK-8、平板克隆、划痕实验、Transwell实验分析GCNT3对肝癌细胞增殖、迁移和侵袭能力的影响,结果显示,敲低GCNT3可抑制肝癌细胞的增殖、迁移和侵袭,而过表达GCNT3发挥相反作用。细胞周期和Western印迹结果显示,GCNT3调控G_(0/)G_1期关键蛋白质的表达来促进肝癌细胞周期G_1/S的转换。进一步探究发现,GCNT3可能通过激活PI3K/AKT/mTOR信号通路促进细胞增殖;以及GCNT3上调RhoA/ROCK/Cofilin通路关键蛋白质的表达来促进F-肌动蛋白(F-actin)的形成,从而参与细胞的迁移和侵袭。总之,本研究通过对GCNT3在肝癌细胞中的功能分析,初步证明,GCNT3与肝癌细胞增殖、迁移和侵袭中的功能有关,证实了GCNT3在肝癌临床治疗中的潜在价值,为肝癌治疗和诊断提供了新思路。 展开更多

目的建立SD大鼠后囊膜混浊(posterior capsular opacification,PCO)动物模型,检测Rho/Rock通路中的关键因子Rho A、Rho激酶Ⅰ(Rho associated coiledcoil forming protein kinase-Ⅰ,RockⅠ)和肌球蛋白轻链(phosphorylated myosin light... 目的建立SD大鼠后囊膜混浊(posterior capsular opacification,PCO)动物模型,检测Rho/Rock通路中的关键因子Rho A、Rho激酶Ⅰ(Rho associated coiledcoil forming protein kinase-Ⅰ,RockⅠ)和肌球蛋白轻链(phosphorylated myosin light chain,p-MLC)在PCO中的表达。探讨Rho/Rock通路在PCO形成过程中的作用。方法选用健康SD大鼠30只,随机分为对照组(0d)和实验组(3 d、7 d、14 d、28 d),每个时间点各6只。所有大鼠(右眼)均行晶状体囊外摘出术,显微镜下观察不同时间点术眼前节的炎症反应及PCO发生情况。ELISA法检测不同时间点房水中TGF-β的表达。处死大鼠后摘除眼球行组织病理学检查,免疫组织化学和Western Blot法检测Rho A、RockⅠ、p-MLC在晶状体后囊上的表达。结果随着术后观察时间的推移,PCO混浊程度逐渐增加,HE染色显示晶状体上皮细胞不同程度的增生、移行。对照组术后0 d、3 d、7 d、14 d、28 d房水中TGF-β的表达量分别为(37.28±1.31)μg·L-1、(40.29±3.17)μg·L-1、(42.12±3.45)μg·L-1、(60.42±4.75)μg·L-1、(68.69±8.06)μg·L-1,实验组各时间点与对照组相比,差异有统计学意义(F=137.158,P<0.05);不同时间点两两相比,差异均有统计学意义(均为P<0.05)。术后0 d、3 d、7 d、14 d和28 d晶状体后囊膜上Rho A蛋白相对表达量(Rho A/GAPDH)分别为1.00±0.00、1.49±0.01、1.57±0.03、1.77±0.02和1.97±0.02,实验组各时间点与对照组相比,差异有统计学意义(F=94.546,P<0.05);不同时间点两两相比,差异均有统计学意义(均为P<0.05)。RockⅠ蛋白的表达量(RockⅠ/GAPDH)分别为1.00±0.00、0.35±0.12、1.42±0.14、1.78±0.09和1.95±0.14,实验组各时间点与对照组相比,差异有统计学意义(F=30.942,P<0.05);不同时间点两两相比,差异均有统计学意义(均为P<0.05)。p-MLC蛋白的相对表达量(p-MLC/GAPDH)分别为1.00±0.00、1.44±0.08、1.47±0.13、1.63±0.10和2.12±0.20,实验组各时间点与对照组相比,差异有统计学意义(F=34.313,P<0.05);不同时间点两两相比,差异均有统计学意义(均为P<0.05)。结论 Rho A、RockⅠ、p-MLC在PCO形成过程中表达逐渐增多,Rho/Rock信号转导通路可能与PCO的发生、发展有关。 展开更多

Hydrocarbon-producing lacustrine basins are widely developed in the world, and China has a large number of lacustrine basins that have developed since the early Permian. The organic-rich shale-dominated heterogeneous source rock intervals in Chinese lacustrine basins generally contain frequent thin interbeds of stratigraphically associated sandstone, siltstone, marl, dolomite, and limestone. The concept of ‘‘Hybrid Plays' ' as put forth in this article recognizes this pattern of alternating organicrich shale and organic-lean interbeds and existence of mixed unconventional and conventional plays. Hybrid Plays in lacustrine source rock intervals present a unique closed petroleum system hosting continuous hydrocarbons.The interbedded organic-lean siliciclastic and/or carbonateplays are efficiently charged with hydrocarbons via short migration pathways from the adjacent organic-rich shale that is often also a self-sourced play. We assert ‘‘Hybrid Plays' ' provide the most realistic exploration model for targeting multiple-stacked and genetically related very tight shale, tight and conventional plays together in the entire source rock interval rather than individual plays only. The Hybrid Play model has been proven and works for a wide variety of lacustrine rift, sag and foreland basins in China. 展开更多
