Ultrastructural pathological changes in the gut-associated lymphoid tissues of sacculus rotundus (SR) of rabbits infected with rabbit haemorrhagic disease virus (RHDV) were first observed. There were numerous holes at the luminal and basement membrane surfaces of the dome epithelium (DE), consistently accompanied by necrosis of lymphocytes and M-cells, and pronounced depletion of lymphocytes in the domes and follicles, decrease of DE complex with formation of pseudomembranous structure on the surface of the dome epithelium. A specific finding in lymphocytes and macrophages was that severe destruction detraction of the membrane of rough endoplasmic reticulum(RER) was accompanied by conspicious increase of solitary, ribo-some-like particles in the cytoplasm, with appearances of intranuclear particles and intranuclear inclusions. It was found that there were many round and dense virion-like particles, with 26 nm in diameter, in the nuclei and cytoplasm of lymphoctes, plasma cells, macrophages and fibroblasts, or in degenerated cells and cellular debris. At the same time, another round virion-like particles about 34 nm in diameter were also seen in the cytoplasm of some cells and interstitium. The results indicated that the appearances of the ribosome-like particles, virion-like particles and inclusion bodies were related to the replication and assembly of RHDV. The present observations suggested that DE of sacculus rotundus could be a open pathway and a transporting route for the entry of antigens into hosts. While the antigen is profoundly deleterious, DE may be as a closed portal or a barrier preventing the foreign antigenic materials from invading. 展开更多

The distribution of bursin in sacculus rotundus and gut-associated lymphoid tissues was investigated in rabbit by immuno-histochemical staining method with anti-bursin monoclonal antibody (McAb)2F9-4. The results indicated that the appearance of the bursin-containing cells coincided with the morphodifferentiation and growth process in rabbit. Bursin immunized cells not only distributed in dome epithelium and mucosal epithelium, but also in follicular germinal center, dome, T-cell area, goblet cell, inter-epithelial lymphocyte and cap-zone of sacculus rotundus. This founding suggested that bursincontaining cells in sacculus rotundus might play an important role in intestinal mucosal immunity. 展开更多

Phytoremediation potential of Cyperus rotundus for diesel-contaminated wetland

Oil spills may considerably damage sensitive coastal wetlands. The phytoremediation potential and restoration of a dominant coastal marsh plant, Cyperus rotundus, for diesel pollutant and its phytoremediation effectiveness were investigated in this open-air pot experiment. Cyperus rotundus was transplanted into soil contaminated with diesel at concentrations of 1 000, 5 000, 10 000, 15 000, 20 000 mg/kg. In order to better elucidate the biochemical and physiological responses to diesel pollutants, activity of the antioxidant enzymes peroxidase (POD), catalase (CAT) and ascorbic acid oxidase (AAO) were determined in the plant tissue after 50 d treatment at the levels mentioned above. The results showed that CAT and AAO of stem and leaf exhibited peak enzyme activities on 15 000 mg/kg soil and 10 000 mg/kg soil respectively, and declined at higher concentrations. Additionally, the increment of biomass and the content of soluble protein, as well as chlorophyll content were affected by diesel. The highest restoration effectiveness appeared at the level of 5 000 mg/kg. Collectively, Cyperus rotundus is a potential plant which can be used for restoring the diesel-contaminated soil. 展开更多

Chemical composition, antiproliferative, antioxidant and antibacterial activities of essential oils from aromatic plants growing in Sudan

Objective: To explore the potential of essential oil, as therapeutic molecule source, from olibanum of Boswellia papyrifera(Burseraceae), leafy stems of Cymbopogon schoenanthus(Poaceae) and Croton zambesicus(Euphorbiaceae) and rhizome of Cyperus rotundus(Cyperaceae) found in Sudan. Respective essential oil was evaluated for antiproliferative, antibacterial and antioxidant activity. Methods: Essential oils were extracted by hydrodistillation and then analysed by gas chromatography coupled to mass spectrometry(GCMS). Anti-proliferative activity was determined against human cell lines(MCF7 and MDAMB231, HT29 and HCT116) by the thiazolyl blue tetrazolium bromide(MTT) procedure. Antioxidant activity was evaluated by diphenyl 2 pycril hydrazil(DPPH) assay. Antibacterial activity was determined against two Gram-positive and two Gram-negative bacteria by microdilution method. Results: The essential oil from olibanum of Boswellia papyriferacontained mainly alcohol and ester derivatives(46.82%) while monoterpenes(69.84%) dominated in Corton zambesicus oil. Sesquiterpenes were the most highly represented classes of terpene derivatives in Cyperus schoenanthus(71.59%) and Cyperus rotundus(44.26%). Oil of Cymbopogon schoenanthus revealed the best anti-proliferative activity against HCT116 cell line with IC50 value at(19.1 ± 2.0) μg/m L. Oil of Croton zambesicus showed the best antioxidant activity [EC50(4.20 ± 0.19) mg/m L]. All oils showed good antibacterial activity against Escherichia coli, Bacillus subtilis and Staphylococcus aureus with minimum inhibitory concentration(MIC) value ranged from 16 to 250 μg/m L. Conclusions: The results suggest that the essential oils of these plants could be used as a source of natural anti-proliferative, antioxidant and antibacterial agents. 展开更多

Background: Human Cytomegalovirus(HCMV) infections can be found throughout the body, especially in epithelial tissue. Animal model was established by inoculation of HCMV(strain AD-169) or coinoculation with Hepatitis E virus(HEV) into the ligated sacculus rotundus and vermiform appendix in living rabbits. The specimens were collected from animals sacrificed 1 and a half hours after infection.Results: The virus was found to be capable of reproducing in these specimens through RT-PCR and Western-blot.Severe inflammation damage was found in HCMV-infected tissue. The viral protein could be detected in high amounts in the mucosal epithelium and lamina propria by immunohistochemistry and immunofluorescense.Moreover, there are strong positive signals in lymphocytes, macrophages, and lymphoid follicles. Quantitative statistics indicate that lymphocytes among epithlium cells increased significantly in viral infection groups.Conclusions: The results showed that HCMV or HEV + HCMV can efficiently infect in rabbits by vivo ligated intestine loop inoculation. The present study successfully developed an infective model in vivo rabbit ligated intestinal Loop for HCMV pathogenesis study. This rabbit model can be helpful for understanding modulation of the gut immune system with HCMV infection. 展开更多
