γ-Secretase,called“the proteasome of the membrane,”is a membrane-embedded protease complex that cleaves 150+peptide substrates with central roles in biology and medicine,including amyloid precursor protein and the ...γ-Secretase,called“the proteasome of the membrane,”is a membrane-embedded protease complex that cleaves 150+peptide substrates with central roles in biology and medicine,including amyloid precursor protein and the Notch family of cell-surface receptors.Mutations inγ-secretase and amyloid precursor protein lead to early-onset familial Alzheimer’s disease.γ-Secretase has thus served as a critical drug target for treating familial Alzheimer’s disease and the more common late-onset Alzheimer’s disease as well.However,critical gaps remain in understanding the mechanisms of processive proteolysis of substrates,the effects of familial Alzheimer’s disease mutations,and allosteric modulation of substrate cleavage byγ-secretase.In this review,we focus on recent studies of structural dynamic mechanisms ofγ-secretase.Different mechanisms,including the“Fit-Stay-Trim,”“Sliding-Unwinding,”and“Tilting-Unwinding,”have been proposed for substrate proteolysis of amyloid precursor protein byγ-secretase based on all-atom molecular dynamics simulations.While an incorrect registry of the Notch1 substrate was identified in the cryo-electron microscopy structure of Notch1-boundγ-secretase,molecular dynamics simulations on a resolved model of Notch1-boundγ-secretase that was reconstructed using the amyloid precursor protein-boundγ-secretase as a template successfully capturedγ-secretase activation for proper cleavages of both wildtype and mutant Notch,being consistent with biochemical experimental findings.The approach could be potentially applied to decipher the processing mechanisms of various substrates byγ-secretase.In addition,controversy over the effects of familial Alzheimer’s disease mutations,particularly the issue of whether they stabilize or destabilizeγ-secretase-substrate complexes,is discussed.Finally,an outlook is provided for future studies ofγ-secretase,including pathways of substrate binding and product release,effects of modulators on familial Alzheimer’s disease mutations of theγ-secretase-substrate complexes.Comprehensive understanding of the functional mechanisms ofγ-secretase will greatly facilitate the rational design of effective drug molecules for treating familial Alzheimer’s disease and perhaps Alzheimer’s disease in general.展开更多
The correlation coefficients of random variables of mechanical structures are generally chosen with experience or even ignored,which cannot actually reflect the effects of parameter uncertainties on reliability.To dis...The correlation coefficients of random variables of mechanical structures are generally chosen with experience or even ignored,which cannot actually reflect the effects of parameter uncertainties on reliability.To discuss the selection problem of the correlation coefficients from the reliability-based sensitivity point of view,the theory principle of the problem is established based on the results of the reliability sensitivity,and the criterion of correlation among random variables is shown.The values of the correlation coefficients are obtained according to the proposed principle and the reliability sensitivity problem is discussed.Numerical studies have shown the following results:(1) If the sensitivity value of correlation coefficient ρ is less than(at what magnitude 0.000 01),then the correlation could be ignored,which could simplify the procedure without introducing additional error.(2) However,as the difference between ρs,that is the most sensitive to the reliability,and ρR,that is with the smallest reliability,is less than 0.001,ρs is suggested to model the dependency of random variables.This could ensure the robust quality of system without the loss of safety requirement.(3) In the case of |Eabs|ρ0.001 and also |Erel|ρ0.001,ρR should be employed to quantify the correlation among random variables in order to ensure the accuracy of reliability analysis.Application of the proposed approach could provide a practical routine for mechanical design and manufactory to study the reliability and reliability-based sensitivity of basic design variables in mechanical reliability analysis and design.展开更多

Based on the first-order upwind and second-order central type of finite volume (UFV and CFV) scheme, upwind and central type of perturbation finite volume (UPFV and CPFV) schemes of the Navier-Stokes equations were de...Based on the first-order upwind and second-order central type of finite volume (UFV and CFV) scheme, upwind and central type of perturbation finite volume (UPFV and CPFV) schemes of the Navier-Stokes equations were developed. In PFV method, the mass fluxes of across the cell faces of the control volume (CV) were expanded into power series of the grid spacing and the coefficients of the power series were determined by means of the conservation equation itself. The UPFV and CPFV scheme respectively uses the same nodes and expressions as those of the normal first-order upwind and second-order central scheme, which is apt to programming. The results of numerical experiments about the flow in a lid-driven cavity and the problem of transport of a scalar quantity in a known velocity field show that compared to the first-order UFV and second-order CFV schemes, upwind PFV scheme is higher accuracy and resolution, especially better robustness. The numerical computation to flow in a lid-driven cavity shows that the under-relaxation factor can be arbitrarily selected ranging from (0.3) to (0.8) and convergence perform excellent with Reynolds number variation from 10~2 to 10~4.展开更多

The measurement matrix is, however, often perturbed in a practical...It is understood that the sparse signal recovery with a standard compressive sensing(CS) strategy requires the measurement matrix known as a priori. The measurement matrix is, however, often perturbed in a practical application.In order to handle such a case, an optimization problem by exploiting the sparsity characteristics of both the perturbations and signals is formulated. An algorithm named as the sparse perturbation signal recovery algorithm(SPSRA) is then proposed to solve the formulated optimization problem. The analytical results show that our SPSRA can simultaneously recover the signal and perturbation vectors by an alternative iteration way, while the convergence of the SPSRA is also analytically given and guaranteed. Moreover, the support patterns of the sparse signal and structured perturbation shown are the same and can be exploited to improve the estimation accuracy and reduce the computation complexity of the algorithm. The numerical simulation results verify the effectiveness of analytical ones.展开更多

BACKGROUND The main clinical manifestation of Alzheimer’s disease(AD)is memory loss,which can be accompanied by neuropsychiatric symptoms at different stages of the disease.Amygdala is closely related to emotion and ...BACKGROUND The main clinical manifestation of Alzheimer’s disease(AD)is memory loss,which can be accompanied by neuropsychiatric symptoms at different stages of the disease.Amygdala is closely related to emotion and memory.AIM To evaluate the diagnostic value of amygdala on structural magnetic resonance imaging(sMRI)for AD.METHODS In this study,22 patients with AD and 26 controls were enrolled.Their amygdala volumes were measured by sMRI and analyzed using an automatic analysis software.RESULTS The bilateral amygdala volumes of AD patients were significantly lower than those of the controls and were positively correlated with the hippocampal volumes.Receiver operating characteristic curve analyses showed that the sensitivity of the left and right amygdala volumes in diagnosing AD was 80.8%and 88.5%,respectively.Subgroup analyses showed that amygdala atrophy was more serious in AD patients with neuropsychiatric symptoms,which mainly included irritability(22.73%),sleep difficulties(22.73%),apathy(18.18%),and hallucination(13.64%).CONCLUSION Amygdala volumes measured by sMRI can be used to diagnose AD,and amygdala atrophy is more serious in patients with neuropsychiatric symptoms.展开更多
Components with strong adsorption capacity for cholates from buckwheat proteins were screened, separated and purified by several methods, and the effects of ultra-high-pressure(UHP) on the structure and function of bu...Components with strong adsorption capacity for cholates from buckwheat proteins were screened, separated and purified by several methods, and the effects of ultra-high-pressure(UHP) on the structure and function of buckwheat 13S globulin(BW13SG) were studied. Samples were treated by UHP at different p H(3.0 and 7.0) value(s) and at 100–500 MPa for 10–30 min. The results showed that the tertiary structure of BW13SG was partially denatured and aggregated. The decrease in the unordered structure indicated that UHP resulted in a looser secondary structure of BW13SG. UHP treatment also increased solubility, emulsion activity and stability, foaming capacity and stability. The samples treated at 500 MPa, p H 3.0 for 30 min had the most enhanced functionality. Moreover, under this condition, the sodium cholate and sodium deoxycholate adsorption capacities of BW13SG were both higher than 98% and the adsorption capacity of sodium taurocholate, which can be difficult to adsorb, was higher than 60%.展开更多

Although some progresses have been made, there is no evident study on applying multifractal analysis to evaluate the white matter structural changes on MRI for Alzheimer's disease(AD) research. In this paper, to explore multifractal analysis of white matter structural changes on 3D MRI volumes between normal aging and early AD, we not only extend the traditional box-counting multifractal analysis(BCMA) into the 3D case, but also propose a modified integer ratio based BCMA(IRBCMA) algorithm to compensate for the rigid division rule in BCMA. We verify multifractal characteristics in 3D white matter MRI volumes. In addition to the previously well studied multifractal feature,△α, we also demonstrated △ f as an alternative and effective multifractal feature to distinguish NC from AD subjects.Both △α and △ f are found to have strong positive correlation with the clinical MMSE scores with statistical significance.Moreover, the proposed IRBCMA can be an alternative and more accurate algorithm for 3D volume analysis. Our findings highlight the potential usefulness of multifractal analysis, which may contribute to clarify some aspects of the etiology of AD through detection of structural changes in white matter.展开更多

The hydro-elastic structure is equal to the non-proportional damping system. In this paper a perturbation analysis method is put forward. The structure motion equation is strictly solved mathematically, and the non-proportional damping problem is transformed into a series of proportional damping ones in the superposition form. The paper also presents the calculation formula of the dynamic response of the structure being subjected to harmonic and arbitrary load. The convergence of the proposed method is also studied in this paper, and the corresponding convergence conditions are given. Finally, the proposed method is used to analyze the displacement response of a real offshore platform. The calculation results show that this method has the characteristics of high accuracy and fast convergence.展开更多
Neurological disorders are a diverse group of conditions that affect the nervous system and include neurodegenerative diseases(Alzheimer’s disease,multiple sclerosis,Parkinson’s disease,Huntington’s disease),cerebr...Neurological disorders are a diverse group of conditions that affect the nervous system and include neurodegenerative diseases(Alzheimer’s disease,multiple sclerosis,Parkinson’s disease,Huntington’s disease),cerebrovascular conditions(stroke),and neurodevelopmental disorders(autism spectrum disorder).Although they affect millions of individuals around the world,only a limited number of effective treatment options are available today.Since most neurological disorders express mitochondria-related metabolic perturbations,metformin,a biguanide type II antidiabetic drug,has attracted a lot of attention to be repurposed to treat neurological disorders by correcting their perturbed energy metabolism.However,controversial research emerges regarding the beneficial/detrimental effects of metformin on these neurological disorders.Given that most neurological disorders have complex etiology in their pathophysiology and are influenced by various risk factors such as aging,lifestyle,genetics,and environment,it is important to identify perturbed molecular functions that can be targeted by metformin in these neurological disorders.These molecules can then be used as biomarkers to stratify subpopulations of patients who show distinct molecular/pathological properties and can respond to metformin treatment,ultimately developing targeted therapy.In this review,we will discuss mitochondria-related metabolic perturbations and impaired molecular pathways in these neurological disorders and how these can be used as biomarkers to guide metformin-responsive treatment for the targeted therapy to treat neurological disorders.展开更多

In addition to the previously well studied multifractal feature,△α, we also demonstrated △ f as an alternative and effective multifractal feature to distinguish NC from AD subjects.Both △α and △ f are found to have strong positive correlation with the clinical MMSE scores with statistical significance.Moreover, the proposed IRBCMA can be an alternative and more accurate algorithm for 3D volume analysis. Our findings highlight the potential usefulness of multifractal analysis, which may contribute to clarify some aspects of the etiology of AD through detection of structural changes in white matter.
文摘The principle aim of this paper is to explore the existence of periodic solution of n-Species Gilpin-Ayala competition system with impulsive perturbations. Sufficient and realistic conditions are obtained by using Mawhin's continuation theorem of the coincidence degree. Further, some numerical simulations show that our model can occur in many forms of complexities including periodic oscillation and chaotic strange attractor.
文摘With conditions of approximately actual background wind field in typhoons and a barotropic nondivergent model in a cylindrical coordinate, the problem of instability of the perturbation in typhoons isdiscussed. The results show that the warm core structure of typhoons and the activity of fanly strongcold air in the periphery are advantageous to the perturbation development in typhoons,and the strongerthe typhoons, i. e. the lower the pressure, the caster the perturbation in typhoons would develop.
文摘Theoretically speaking, it is impossible to make the differential equation of motion uncoupled for the natural modes of a system in consideration of the attached water. The hydro-elastic structure is equal to the non-proportional damping system. In this paper a perturbation analysis method is put forward. The structure motion equation is strictly solved mathematically, and the non-proportional damping problem is transformed into a series of proportional damping ones in the superposition form. The paper also presents the calculation formula of the dynamic response of the structure being subjected to harmonic and arbitrary load. The convergence of the proposed method is also studied in this paper, and the corresponding convergence conditions are given. Finally, the proposed method is used to analyze the displacement response of a real offshore platform. The calculation results show that this method has the characteristics of high accuracy and fast convergence.
文摘Neurological disorders are a diverse group of conditions that affect the nervous system and include neurodegenerative diseases(Alzheimer’s disease,multiple sclerosis,Parkinson’s disease,Huntington’s disease),cerebrovascular conditions(stroke),and neurodevelopmental disorders(autism spectrum disorder).Although they affect millions of individuals around the world,only a limited number of effective treatment options are available today.Since most neurological disorders express mitochondria-related metabolic perturbations,metformin,a biguanide type II antidiabetic drug,has attracted a lot of attention to be repurposed to treat neurological disorders by correcting their perturbed energy metabolism.However,controversial research emerges regarding the beneficial/detrimental effects of metformin on these neurological disorders.Given that most neurological disorders have complex etiology in their pathophysiology and are influenced by various risk factors such as aging,lifestyle,genetics,and environment,it is important to identify perturbed molecular functions that can be targeted by metformin in these neurological disorders.These molecules can then be used as biomarkers to stratify subpopulations of patients who show distinct molecular/pathological properties and can respond to metformin treatment,ultimately developing targeted therapy.In this review,we will discuss mitochondria-related metabolic perturbations and impaired molecular pathways in these neurological disorders and how these can be used as biomarkers to guide metformin-responsive treatment for the targeted therapy to treat neurological disorders.
