In order to seek suitable protein extraction method, three different deep eutectic solvents(DESs)(including choline chlorideglycerol, choline chloride-oxalic acid and choline chloride-urea) were developed for extracting protein from SSM and compared with alkaline. Result indicated that alkaline could effectively extract 56.9% protein from SSM and its protein content was 73.1%, higher than DES at 31.0%-41.4% and 64.3%-67.5%, respectively. However, compared to alkali, DES led to a product with less β-sheet, more β-turn, more essential amino acids, higher total amino acid content, especially choline chloride-urea which extracted protein showing an integrated and similar protein weight distribution compared to SSM. Also, this protein extracted chloride-urea showed a highest digestibility in vitro(by pepsin)(54.2%). These results indicated that choline chloride-urea extraction is better than alkaline extraction for SSM.展开更多

The berry was extracted by ethanol-water solution. MTT assay and growth rate method were applied to determine antibacterial ability in vitro of the extract solution. The antibacterial and antifungal assay indicated that the extract exhibited different degree of inhibitory activity. The result showed that the extract at a concentration of 1 250 mg/L had the maximum inhibitory effect against Escherichia coli, Bacillus subtilis, and Staphylococcus aureus. Inhibitory effect against B. subtilis was much larger than that against the other two bacterial species. MIC values(minimum inhibitory concentration) of the extract on three bacterial species were 5 000, 1 250, and 1 000 mg/L respectively. IC_(50)(50% inhibiting concentration) evaluation of Trichoderma viride, Rhizopus stolonifer, Aspergillus niger, Penicillium expansum, and Vintage Red indicated that the berry had the strongest inhibitory effect against P. expansum(5 520 mg/L), while it had the weakest inhibitory effect against R. stolonifer(18 870 mg/L). The results proved that seabuckthorn berry had highly effective and comprehensive antimicrobial function.展开更多

The aim of this demarcation is to avoid a random introduction of seabuckthorn. In each of the five zones, objectives should be set and suitable seabuckthorn species, subspecies and varieties should be planted according to site conditions, seed sources and methods of tree breeding. The cultivation centers, bases, stations, or units should be established and successful models of seedling and planting methods should be encouraged. The principle of matching trees with suitable site conditions and adjusting measures to local conditions should be practiced. From a strategic viewpoint of solving ecological and economic problems of seabuckthorn development in the three northern areas, every seabuckthorn center must have its own germplasm nursery, standard plantation for popularizing, excellent seed and seedling nurseries and sufficient afforestation areas for demonstration and propaganda purposes. These measures would improve the ecological environment and promote economic and social development in the three northern areas of China.展开更多

Although logging is prohibited, habitat destruction has increased because of regular road construction, repairing and broadening activities. In addition, Hippophae resources are continuously being harvested by lopping (both partial and complete) for fuelwood, fodder and fruits in higher Himalayan region. This paper presents a detailed analysis of relationship between density, demographic structure, and harvesting of H. salicifolia growing pockets in the five major valleys (Gangotri, Yamunotri, Niti, Mana and Bhyundhar) of Uttarakhand in Central Himalaya, India. A total of 12o quadrats were laid randomly to study population structure, regeneration, sex ratio and lopping using quadrats of lOO m2 (a4 in each valley) in Hippophae growing patches. Our study shows that the density, size distribution, and regeneration of Hippophae vary considerably among the major valleys. Trees in the Yamunotri valley have the highest density of large trees but the lowest density of seedlings. In contrast, there are few large trees but many seedlings in the Mana valley. The number and size of lopped trees also varied among the valleys. Lopping was greatest in Bhyundhar (11.4%) and Yamunotri (19.7%) and least in Niti (3.9%). The size of lopped trees differed substantially as well. In Bhyundhar, the largest trees were taken while saplings were taken in Yamunotri. Our study revealed that unsustainable harvesting from plants for fuel, fencing and fruits along with road broadening activities in Central Himalaya are the main cause ofhabitat destruction. Our research highlights the urgent need for in-situ and ex-situ conservation of Hippophae salicifolia so that it's potential can be harnessed sustainably by rural hill societies for their socio-economic development.展开更多

Six different branded SBT oils purchased from e-commerce suppliers(Amazon) and in-house prepared SBT oil was analyzed by qNMR and fatty acyl composition of TAGs determined by using NMR. In-house oil was also analysed by GC–MS and FTIR spectroscopy. Results: The qNMR results showed that the oil contained 80.3% of triacylglycerol(TAG). The SBT oil TAGs comprised of linolenate 6.6%, palmitoleate/oleate 65.4%, and total saturated fatty acyl chain including palmitate 28% as determined by qNMR. GC–MS analysis revealed that the major acyl functionalities present in the TAG were palmitoleic acid 36.5%, oleic acid 12.9%, palmitic acid 21.2%, and linoleic acid 18%. Of the six commercial samples analyzed, samples from only one supplier(SW) were fruit oil;All others were the seed oils or mix of fruit oil and seed oil. The labels for samples except for the SW did not indicate whether it was fruit oil or seed oil. Conclusion: The results suggest that SBT oil should be analyzed by combination of GC–MS, FTIR and qNMR for factual content of free fatty acid or TAGs, which are chemically different in nature and affect the quality of oil. GC–MS showed the content of omega free fatty acids after hydrolysis, while qNMR and FTIR showed the content of TAGs. The major acyl functionalities found in SBT fruit oil TAGs are palmitoleate/palmitate/oleate, while linoleate and linonelate make up a minor fraction. Furthermore, analysis of commercial samples showed discrepancies between label claims and actual content.展开更多

Furthermore, analysis of commercial samples showed discrepancies between label claims and actual content.

文摘Pisha sandstone is a special kind of soft rock in the semiarid region along the up and middle reaches of Yellow River and Loess Plateau and is severely eroded by wind, rainfall and gravity force.Pisha sandstone region is called 'the most severe water loss and soil erosion in the world' and referred to as the 'Earth Cancer' by local residents and experts both at home and abroad. The plan for the control of Pisha sandstone started in the 1950s.Since then a series of projects have been implemented, both engineering and vegetation measures have been developed and widely used to protect Pisha sandstone from erosion.Since seabuckthom was introduced as a viable approach for soil erosion protection and its implementation resulted in a breakthrough to control Loess Plateau,it was considered as an important and efficient measure. Then the seabuckthom flexible dam was proposed for coarse sediment retention and fine sediment deposition. And then a new comprehensive control model using consolidation materials combined with vegetation was proposed and promising results were achieved both in laboratory and field demonstrations. In the future, the complex erosion mechanism and ecological carrying capacity should he studied further and more efficient and practical comprehensive measures shall be developed. Moreover, monitoring systems shall be used to predict and detect the changes in the Pisha sandstone slope and structure.

文摘Objective:To investigate the effect of seabuckthorn berries extract(SBE)on pulmonary vascular hyperpermeability in the mice model of acute lung injury(ALI)induced by lipopolysaccharide(LPS).Methods:Sixty Kunming mice were allocated into 6 groups by a random number table, including control,LPS,dexamethasone(Dex,1 mg/kg),and 120,240 and 480 mg/kg SBE groups,10 mice in each group.Except the control group, mice were pre-treated with Dex and SBE,respectively, for 7 days before LPS was intraperitoneally injected to induce ALl.Pulmonary vascular hyperpermeabilitywas evaluated by histopathologic observation and transvascular leakagedetermination.Tumor necrosis factor alpha(TNF-α)and interleukin-6(IL-6)levels in serum were measured using enzyme-linked immunosorbent assay. The expression of nuclear factor-kappa B(NF-k B)p65 in lung cells was determined by immunofluorescence analysis. The contents of cytoplasmic inhibitorof nuclear factor-kBkinase(IKK)and nuclearp65,as wellasdownstreamproteins of E-selectin(CD62E)and intercellular adhesion molecule-1(ICAM-1),were determined using Western blot analysis. Results:Histopathological observation confirmed SBEtreatment alleviatedmorphological lesion induced by LPS.Compared with the LPS group,480 mg/kg SBE significantly decreased the water content of lung,Evans blue accumulation in lung tissue, and protein concentration and neutrophils count in bronchoalveolar lavage fluid(P<o.01);moreover,480 mg/kg SBE significantly suppressed release of TNF-αand IL-6,and down-regulated expressions of IKK,nuclear p65,ICAM-1 and CD62E(P<0.01).Conclusion:SBE maintained alveolar-capillary barrier integrity under endotoxin challenge in mice by suppressing the key factors in the pathogenesis of ALl.

文摘Objective:To compare the proximate composition, mineral contents, antimicrobial, phytochemical and Fourier transforms infrared(FTIR)spectroscopy analysis of bark, stem and seed of Hippophae rhamnoides. Methods:Proximate composition was determined according to the described methods.Mineral analysis was carried out by atomic absorption spectroscopy and flame photometer.Antimicrobial activities were evaluated according to the agar well diffusion method.Phytochemical qualitative analysis was carried out according to the described methods and functional groups were determined by FTIR Prestige-21 Shimadzu Japan. Results:The proximate analysis showed high content of protein and fiber in stem and bark.High content of Na(900 mg/L)and K(670 mg/L)was found in bark powder, while in seed, high contents of Ca(800 mg/L),Mg(725 mg/L),Fe(250 mg/L)Zn(90 mg/L)and Mn(65 mg/L)were found compared to stem and bark.Phenols, flavonoids and tannins showed high contents in stem and bark of all extracts. The bark aqueous extract showed high zone of inhibition against Staphylococcus aureus(21 mm)and Escherichia coli(20 mm),while methanol extract of stem showed high zone of inhibition(14 mm and 13 mm)against Enterococcus faecalis and Escherichia coli respectively. The aqueous extract of bark documented high zone of inhibition against Aspergillus niger(21 mm)and Aspergillus parasiticus(20 mm).FTIR spectra revealed the presence of OH,C-O and C=O functional groups. Conclusions:The study concludes that bark, stem and seed extracts will be useful guideline for the new syntheses of feed, food supplements and herb drugs with various combination, which can be used for the treatment of many diseases at global level especially in tropical regions as well as the male nutrition problems in these areas.
文摘Objective:To evaluate the antitumor and anti-proliferative activities of methanol, aqueous, acetone, ethyl acetate, ethanol, chloroform and n-hexane extracts of Hippophae rhamnoides leaves. Methods:Antitumor activities were evaluated by using the antitumor potato disc assay by using inoculums(Agrobacterium tumefaciens)with three different concentrations of test samples(10,100 and 1000 mg/L).Anti-proliferative activity was evaluated by the given method of methyl thiazolyl tetrazolium assay. The concentrations of the extract ranging from 0.039 to 10 mg/mL were tested against HeLa cells. Results:Highest tumors inhibition activity(60.9%and 55.8%)was shown by methanol and ethanol extracts, with EC_(50) values of 424.41 and 434.61 mg/L respectively. At 10 mg/mL,The highest cell inhibition 75.61%was observed in methanol extract and the lowest 36.59%were calculated in n-hexane extract. The difference in tumor and cell inhibition(%)may be due to the different concentration of active compounds responsible for antitumor and anti-proliferative activities. All extracts have considerable level of tumor and cell inhibitiory effect in a dose dependent manner. Conclusions:Our finding showed that Hippophae rhamnoides leaves are a potent natural source of antitumor and antiproliferative agent.
