Oleuropein alleviates sepsis-induced acute lung injury via the AMPK/Nrf-2/HO-1 signaling

Objective:To explore the effect of oleuropein on sepsis-induced acute lung injury(ALI)in vitro and in vivo and investigate the underlying mechanism.Methods:In an lipopolysaccharide(LPS)-mediated cell model of sepsis-induced ALI and a cecal ligation and puncture-induced mouse model of septic ALI,CCK-8 assay and flow cytometry analysis were used to detect cell activity and apoptosis.ELISA and relevant assay kits were used to measure the levels of inflammatory cytokines and oxidative stress,respectively.Western blot was applied to determine the expression of apoptosis-and AMP-activated protein kinase(AMPK)/nuclear factor erythroid 2-related factor-2(Nrf-2)/heme oxygenase-1(HO-1)signaling-associated proteins.JC-1 staining,adenosine triphosphate(ATP)assay kit,and MitoSOX Red assays were performed to detect mitochondrial membrane potential,ATP content,and mitochondrial ROS formation,respectively.Moreover,lung injury was evaluated by measuring lung morphological alternations,lung wet-to-dry ratio,myeloperoxidase content,and total protein concentration.Results:Oleuropein reduced inflammatory reaction,oxidative damage,and apoptosis,and ameliorated mitochondrial dysfunction in LPS-exposed BEAS-2B cells and mice with septic ALI.Besides,oleuropein activated the AMPK/Nrf-2/HO-1 signaling pathway.However,these effects of oleuropein were abrogated by an AMPK inhibitor compound C.Conclusions:Oleuropein can protect against sepsis-induced ALI in vitro and in vivo by activating the AMPK/Nrf-2/HO-1 signaling,which might be a potential therapeutic agent for the treatment of sepsis-induced ALI. 展开更多

Investigation of the Thermal Decomposition Behavior of Oleuropein with Many Pharmacological Activities from Olive by Thermogravimetry

Due to the existence of poly-hydroxyl structures,the temperature may have an effect on the thermal stability of oleuropein for its applications.In the current study,the thermal decomposition process and kinetics behavior of oleuropein from the olive resource were researched by thermogravimetric theoretical analysis methods and non-isothermal kinetics simulation.The results of thermogravimetry analysis showed the whole thermal decomposi-tion process of oleuropein involved two stages,with 21.22%of residue.It was also revealed that high heating rates of more than 20 K min^(-1) led to significant thermal hysteresis and inhibited the whole thermal decomposition behavior of oleuropein.Moreover,an investigation of the thermal decomposition kinetics indicated that the non-isothermal decomposition behavior followed a D3 model during thefirst stage(three-dimensional diffusion,Jander equation)and a D1 model in the second stage(one-dimensional diffusion).For thefirst and second ther-mal decomposition stages,the Kissinger,Friedman,Flynn-Wall-Ozawa,and Coats–Redfern four methods were applied to determine the activation energy(E=143.72 and 247.01 kJ mol^(-1))and Arrhenius preexponential factor(ln A=26.34 and 42.45 min^(-1)),respectively.Therefore,the study will provide good theoretical guidance for ther-mal stability and thermal transformation application of oleuropein.It will be suitable for low-temperature appli-cations in the cosmetic,food supplement and pharmaceutical industries. 展开更多

An ultrahigh performance liquid chromatography-triple quadrupole tandem mass spectrometry(UPLC-MS/MS)was established to quickly and accurately determine the content of oleuropein in cosmetics.The samples were extracted with methanol-aqueous solution,and the mobile phase with methanol-formic acid solution(0.1 mol/L)=40∶60 was separated by Agilent ZORBAX Eclipse Plus C18(2.1 mm×50 mm×1.8μm-Micron)column temperature 30℃,flow rate 0.3 mL/min.The MS end was detected by electrospray negative mode ionization(ESI-)and multiple reaction monitoring(MRM)mode.The results show a good linear relationship in the range of 0.002~5 mg/L,with a correlation coefficient R2 of 0.999,5.Method recovery range from 84.2%~107.6%and the relative standard deviation RSD is 5.8%.The detection time is 5 min,the detection limit is 0.000,6 mg/L,and the limit of quantification is 0.002 mg/L.This method has the advantages of convenient operation,low quantification limit,high precision and good repeatability,and is suitable for measuring the content of oleuropein in many kinds of cosmetics. 展开更多

橄榄苦苷酶水解产物具有良好的生物活性,为了促进纤维素酶水解橄榄苦苷,本研究通过天然低共熔溶剂(natural deep eutectic solvent,NADES)对纤维素酶稳定性的促进作用,形成NADES-纤维素酶溶剂系统促进橄榄苦苷的酶水解。以橄榄苦苷酶水... 橄榄苦苷酶水解产物具有良好的生物活性,为了促进纤维素酶水解橄榄苦苷,本研究通过天然低共熔溶剂(natural deep eutectic solvent,NADES)对纤维素酶稳定性的促进作用,形成NADES-纤维素酶溶剂系统促进橄榄苦苷的酶水解。以橄榄苦苷酶水解率为指标,考察温度、pH和DES种类及浓度对纤维素酶水解橄榄苦苷的影响。结果表明:在50℃,pH=5时水解3 h,以10%(V/V)DES-5(甜菜碱∶1,4-丁二醇=1∶2,n/n)为溶剂系统的橄榄苦苷酶水解率是缓冲液的1.7倍;证明以氯化胆碱或甜菜碱为氢键受体(hydrogen bond acceptor,HBA),多元醇为氢键供体(hydrogen bond donor,HBD)的NADES对增强纤维素酶稳定性、促进橄榄苦苷酶水解具有良好的效果,为高效制备橄榄苦苷酶水解产物提供了科学依据。 展开更多

阴离子树脂分离橄榄苦苷的过程及性能研究

目前橄榄苦苷的分离、纯化研究主要集中在大孔吸附树脂,但鲜有阴离子树脂分离、纯化橄榄苦苷的报道。为了研究阴离子树脂(LX-68M)吸附、分离过程及性能,采用吸附动力学及等温吸附模型来研究阴离子树脂静态吸附、解吸橄榄苦苷的行为。结... 目前橄榄苦苷的分离、纯化研究主要集中在大孔吸附树脂,但鲜有阴离子树脂分离、纯化橄榄苦苷的报道。为了研究阴离子树脂(LX-68M)吸附、分离过程及性能,采用吸附动力学及等温吸附模型来研究阴离子树脂静态吸附、解吸橄榄苦苷的行为。结果表明阴离子树脂对橄榄苦苷的吸附符合准二级动力学模型(R^(2)=0.9934),Freundlich等温吸附模型(R^(2)=0.9642)。进一步通过动态实验优化阴离子树脂纯化橄榄苦苷工艺,最佳操作工艺为:上样浓度为0.25 g/mL,解吸液为45%乙醇,吸附、解吸速率均为2.0 BV/h。该条件下橄榄苦苷的吸附率、解吸率分别为86.86%、87.08%,纯度为52.33%。此外,阴离子树脂重复使用4次后吸附率为54.9%,经再生处理后,吸附率恢复到93.51%,表明其具有一定的工业化前景。阴离子树脂稳定性好、分离条件温和,适合用于橄榄苦苷及其他相似天然产物的分离纯化。 展开更多

Olive fruits are seriously deteriorated by pre and postharvest damage due to the attack of insects, such as Bactrocera olaea, which strongly alters the quality of olives. Defence response in olive fruits injured both by pathogens and by mechanical damages has been associated with the enzyme β-glucosidase, which specifically hydrolyses oleuropein, producing highly reactive aldehyde molecules. In situ detection of ^-glucosidase activity in olive fruit tissues following injury, which simulates Bactrocera oleae punctures, is reported. The assay was performed in two cultivars showing different degrees of susceptibilities to fly infestation. In both cultivars, the histochemical assay for β-glucosidase showed that within 20 min after the injury, a strong ^-glucosidase activity could be observed in the damaged tissues. Thereafter a progressive enzyme inactivation occurred starting from tissues around the boundary of the injury with decrease of the enzyme activity and stopped after 3 h. Whereas the mass of active cells reached a distance of (300±50) μm from the edge of the injury. Biochemical analyses showed that in extracts of the injured fruit, β-glucosidase activity rapidly increased within 20 min from injury, thereafter decreasing and reaching values comparable with those in intact fruits. Following puncture, the oleuropein contents did not change significantly in the high susceptibility cultivar, whereas it rapidly decreased in the cultivar showing low susceptibility. The results strongly suggest that olive fruits susceptible towards fly infestation could be related to the ability of the oleuropein-degrading-β-glucosidase to produce the highly reactive molecules in the damaged tissues. As a consequence of puncture, high level of peroxidase activity was detected. This feature also suggested that this enzyme could play a key role in the defence response against insect injuries. 展开更多

Olive leaves extract compounds, such as Oleuropein, have been used in many therapeutic and preventive cases. The present study is designed to evaluate the effect of Oleuropein as antioxidant on the heart histological and oxidative features in diabetic rats. Diabetes was induced by alloxan at 180 mg/kg b.w by intra-peritoneal method once. Four weeks Oleuropein rich olive leaves administration restore significantly the heart histological alteration established by diabetes and enhanced the cardiac antioxidant potential. 展开更多

Results showed that higher concentration was obtained from olive leaves dried at room temperature (10.0 mg per gram of dry olive leaves), compared to those dried at 50&deg;C (1.7 mg/g), and those that collected dry from the tree (2.5 mg/g). Oleuropein content of dried olive leaves was compared with leaves that chopped and extracted fresh, and results show that fresh leaves showed the lowest oleuropein content (<0.1 mg/g) denoting that drying of leaves is required for high oleuropein recovery. 展开更多

目的研究橄榄苦苷(oleuropein,OP)对高脂饮食(high-fatdiet,HFD)联合链脲佐菌素(streptozotocin,STZ)诱导的糖尿病模型小鼠的降血糖作用。方法使用高脂饮食(high-fat diet)联合STZ建立高血糖小鼠模型,将高血糖小鼠分为模型组(HFD/STZ)... 目的研究橄榄苦苷(oleuropein,OP)对高脂饮食(high-fatdiet,HFD)联合链脲佐菌素(streptozotocin,STZ)诱导的糖尿病模型小鼠的降血糖作用。方法使用高脂饮食(high-fat diet)联合STZ建立高血糖小鼠模型,将高血糖小鼠分为模型组(HFD/STZ)及橄榄苦苷组(HFD/STZ+OP),将正常小鼠作为对照组(chow)。对于橄榄苦苷组小鼠,以200 mg/kg体重的剂量灌胃橄榄苦苷11 w。测定3组小鼠的空腹血糖、葡萄糖耐受性及肝脏、胰脏和附睾脂肪的质量和系数,用H&E染色的方法检测OP对糖尿病小鼠胰岛损伤的影响。结果橄榄苦苷能够显著降低糖尿病小鼠的空腹血糖(337.5vs.477.0mg/dL);橄榄苦苷能促进胰岛素的早期分泌,在一定程度上改善糖尿病小鼠的葡萄糖不耐受;橄榄苦苷增加了HFD/STZ诱导的糖尿病小鼠的胰腺胰岛面积。结论橄榄苦苷具有良好的改善HFD/STZ诱导的2型糖尿病的作用。 展开更多

Antioxidants are used to minimize oxidation process. There is a growing interest in the substitution of synthetic food antioxidants by natural ones from vegetable sources. In meat industry, sodium erythorbate is antioxidant that is usually used and is an example of chemical antioxidant. Effect of olive leaf extract rich in olenropein on the quality of frozen hamburger was investigated. The objective of this study was to evaluate the usage of oleuropein from olive leaf extract as natural antioxidant in frozen hamburger stored at -12 ℃ compared with sodium erythorbate. Results suggested that olive leaf extracts might be useful to the meat industry as an efficient alternative to synthetic antioxidants by retarding oxidation of hamburger compared with sodium erythorbate 0.5% of olenropein and 0.5% of sodium erythorbate are the best concentrations to be used in frozen hamburger. 展开更多

为优选可抑制秦艽次生代谢产物含量变化的条件,该文采用3因素4水平正交试验设计方法,共设计16组处理,研究洛伐他汀(MVA途径抑制剂)、膦胺霉素(MEP途径抑制剂)和取样天数对秦艽中马钱苷酸、獐牙菜苷、獐牙菜苦苷、龙胆苦苷4种主要环烯醚... 为优选可抑制秦艽次生代谢产物含量变化的条件,该文采用3因素4水平正交试验设计方法,共设计16组处理,研究洛伐他汀(MVA途径抑制剂)、膦胺霉素(MEP途径抑制剂)和取样天数对秦艽中马钱苷酸、獐牙菜苷、獐牙菜苦苷、龙胆苦苷4种主要环烯醚萜类化合物含量的影响。结果表明:(1)4种环烯醚萜类化合物含量变化受取样天数影响最大,其次为膦胺霉素浓度,次之为洛伐他汀浓度。(2)以最佳抑制条件处理后,马钱苷酸、獐芽菜苦苷、龙胆苦苷和獐芽菜苷含量分别下降了69%、36%、33%和4%。基于正交法优选的抑制条件,对4种化合物均可抑制。综上所述,可确定调控秦艽次生代谢产物含量变化的最佳抑制条件为膦胺霉素400μmol·L^(-1),洛伐他汀50μmol·L^(-1),取样天数6 d,该条件为进一步研究MEP和MVA途径在环烯醚萜类化合物代谢合成中的调控机制奠定基础。 展开更多

该研究探讨素馨花水提物(WE)及其化学成分在3T3-L1前脂肪细胞上的降脂作用。通过将3T3-L1细胞诱导为成熟脂肪细胞,油红O染色定量和检测甘油三酯(TG)含量判断素馨花样品对脂滴的抑制作用;液质联用仪分析WE中化学成分;MTT检测素馨花及其... 该研究探讨素馨花水提物(WE)及其化学成分在3T3-L1前脂肪细胞上的降脂作用。通过将3T3-L1细胞诱导为成熟脂肪细胞,油红O染色定量和检测甘油三酯(TG)含量判断素馨花样品对脂滴的抑制作用;液质联用仪分析WE中化学成分;MTT检测素馨花及其成分对3T3-L1细胞增殖的影响;Western Blot检测AMPK、C/EBPα、FAS、ACC、CPT1、Bax、Bcl-2等蛋白表达。结果发现,WE能剂量依赖性抑制脂滴形成,其中高浓度WE(500μg/mL)能降低中性脂质含量23.59%,降低TG含量30.20%,在脂肪积累早期作用最显著,并证明其活性成分主要是橄榄苦苷,含量为13.77%。WE及橄榄苦苷能激活AMPK(123.19%和115.98%)和其下游靶点ACC(451.06%和1050.0%)的磷酸化、下调其下游靶点FAS(81.72%和60.50%)的蛋白表达,减少脂肪形成,提高CPT1(164.84%和292.19%)蛋白的表达,加快脂肪酸氧化;抑制C/EBPα(68.97%和34.57%)的表达,影响3T3-L1细胞分化;上调Bax(154.60%和139.37%)、下调Bcl-2(41.10%和45.62%)蛋白的表达,诱导脂肪细胞凋亡。结果提示,WE能在3T3-L1细胞分化过程早期抑制细胞生长、分化和脂肪合成并促进脂肪酸氧化从而有效抑制脂质积累,机制上可能通过调控AMPK和Bax/Bcl-2通路。 展开更多
