Trehalose, a non reducing disaccharide used as protective agent and additive in foodstuffs and pharmaceutical products, can be prepared by trehalose synthase(TreS). The present work aims to construct a robust recombinant B. subtilis to achieve the secretory expression of TreS. In this study, the treS gene from Pseudomonas putida ATCC47054 was amplified by PCR and further cloned and expressed in B. subtilis WB800 N using pHT01 as expression vector. For avoiding the use of inducer, promoter P_(srfA) was used to replace the promoter P_(grac) in pHT01 and verify the activity of recombinant trehalose synthase. The TreS activity assay was employed to evaluate the performance of recombinant B. subtilis W800 N under different phosphate concentrations, carbon sources, carbon source concentrations, nitrogen sources and pH. The results showed that the P_(srfA) promoter had a good regulation effect under pH 8.0 condition, and the enzyme activity reached 6 000 U/L. Using the PhoD as the secretory signal peptide, TreS was effectively secreted, and the extracellular enzyme activity reached 2 100 U/L, accounting for 35% of the total enzyme activity. By optimizing the medium and fermentation conditions, the extracellular enzyme activity reached 6 900 U/L in 5 L of fermentor, and the proportion reached 48%. The pHT01-P_(srfA)-PhoD-treS secretory recombinant B. subtilis constructed in this study has great potential in trehalose synthase production.展开更多

A theoretical model with ex plicit expressions is firstly developed to calculate the self-inductance of a single layer, and then numerical calcu lation of the mutual inductance between two layers is introduced. Our research aim is to investigate the buffet alleviation effect of static or vibrating bulges attached on the forebody surface of the model.Experiments and numerical simulations on a model consisting of a sharp-edged...Our research aim is to investigate the buffet alleviation effect of static or vibrating bulges attached on the forebody surface of the model.Experiments and numerical simulations on a model consisting of a sharp-edged,70°-leading edge sweep delta wing and twin swept back vertical tails were conducted.Models with different bulges were tested and computed at 10 and 20 m/s of free stream velocity at angles of attack ranging from 20°–50°.Dynamic strain gauge and multichannel data acquisition and analysis system were employed for the measurement of unsteady root strain on the vertical tails.Experimental and computational results show that both static and vibrating bulges behave effectively as a novel tool to alleviate tail buffet,and the alleviation effect depends largely on the vibrating frequency.Besides,one-sided bulge can only alleviate the buffeting response for the tail of the same side,and it has no obvious alleviation effect for the opposite tail.Results of the spectral analysis reveal that there are generally three peaks of spectral density for aircrafts of this configuration,and bulges used in this paper could alleviate tail buffeting,but the total lift and drag of the whole model show no obvious deviation compared to the base model and the dominant frequency of the vibration of the tails has not shifted.展开更多

The electrical-magnetic characteristics of a Switched Reluctance Motor(SRM)exhibit highly nonlinear relationshipwith respect to the rotor position and excitation current,which poses challenges for both precise static ...The electrical-magnetic characteristics of a Switched Reluctance Motor(SRM)exhibit highly nonlinear relationshipwith respect to the rotor position and excitation current,which poses challenges for both precise static measurements and exact calculation of these properties in real-time control.To guarantee that an in-lab test result can be used in the application,firstly a measurementmethod is proposed to characterize the SRM's electromagnetic properties such as the flux linkage,magnetic co-energy,phase inductance and electromagnetic torque on the basis of an installed SRM control circuitry and half-bridge power converter.By this means the characterization process is equivalent to the online observation in its results.Secondly,a theoreticalmodel is built to discriminate the physical meaning between the incremental inductance and the phase inductance,which is the origin of other relevant parameters.This helps to guide the correct utilization of the characterization result.Thirdly an in-situ cross-validation experimentation according to the magnetizing and demagnetizing status measurement verifies the feasibilities and accuracy of the proposed inductance measuring method,which avoid a dubious FEM-based comparison between the numerical calculation and experimental results.Cross-validation experiment shows that the proposed in-situ characterization scheme obtains an accurate full-range electromagnetic properties.The proposed methodology breaks the barrier between the in-lab measurement and on-line utilization of the SRM parameters,highlighting the merits that it completely includes the in-situ factors and replicates the operational scenario without the need of specifically designed instrumentation,which is especially suitable for rapid field characterization for high power motors.展开更多
