Association between semen collection time and semen parameters:an observational study

The process of semen collection plays a key role in the quality of semen specimens.However,the association between semen collection time and semen quality is still unclear.In this study,ejaculates by masturbation from 746 subfertile men or healthy men who underwent semen analysis were examined.The median(interquartile range)semen collection time for all participants was 7.0(5.0-11.0)min,and the median time taken for semen collection was lower in healthy men than that in subfertile men(6.0 min vs 7.0 min).An increase in the time required to produce semen samples was associated with poorer semen quality.Among those undergoing assisted reproductive technology(ART),the miscarriage rate was positively correlated with the semen collection time.After adjusting for confounders,the highest quartile(Q4)of collection time was negatively associated with semen volume and sperm concentration.A longer time to produce semen samples(Q3 and Q4)was negatively correlated with progressive and total sperm motility.In addition,there was a significant negative linear association between the semen collection time and the sperm morphology.Higher risks of asthenozoospermia(adjusted odds ratio[OR]=2.06,95%confidence interval[CI]:1.31-3.25,P=0.002)and teratozoospermia(adjusted OR=1.98,95%CI:1.10-3.55,P=0.02)were observed in Q3 than those in Q1.Our results indicate that a higher risk of abnormal semen parameter values was associated with an increase in time for semen collection,which may be related to male fertility through its association with semen quality. 展开更多

Intracytoplasmicsperm injection (ICSI) provides an effective manipulation for bypassing some severe etiologies of male fertility. However, some men may encounter difficulty in semen collection by masturbation on demand at the time of undergoing assisted reproductive technology (ART). If a patient is unable to provide semen by masturbation, he has to undergo testicular sperm extraction (TESE) to collect spermatozoa for ICSI, which is expensive and invasive. Background:Collecting and studying live sperm is central to many important fields of biology.Yet,a simple method to collect live sperm is lacking in wild seabird species.Here,we describe a non?invasive method to collect viable sperm samples based on a simple massage technique applied to male Black?legged Kittiwakes(Rissa tridactyla).Methods:We studied a colony breeding at Kongsfjorden,Svalbard and successfully obtained sperm samples from 32 males.With a subset of samples(n = 12 males),we compared the suitability of several extenders(0.9% NaCl,PBS,Earle's balance salt solution,Dulbecco's modified Eagle medium) in maintaining sperm alive long enough for analyses.With another 18 ejaculates,we conducted computer assisted sperm analyses using the CASA plugin for ImageJ.We provide details about the settings to be used for such analyses.Lastly,droplets from 20 ejaculates were smeared on glass slides and preserved with formalin to characterize sperm morphology in terms of total sperm length,sperm head length,midpiece length and flagellum length,and percentage of abnormal sperm.Results:With this method and under field conditions,we were able to obtain sufficient amounts of live sperm to assess traits related to sperm quality(e.g.sperm morphology,percentage of motile sperm,sperm velocity).We found that two extenders,Earle's balanced salt solution and Dulbecco modified Eagle's medium,yielded similarly good results.Additionally,we investigated whether specific behaviours were associated with successful sperm collection and whether sperm collection success depended on how long before laying sperm collection was attempted.Finally,we provide mean values for sperm morphology,sperm swimming ability and percentage of motile sperm,which may prove useful for future comparative analyses,and we report high levels of sperm abnormality and within?ejaculate variation in sperm morphology.Conclusions:We discuss the high percentage of abnormal sperm and high within?ejaculate variation in sperm morphology in light of sperm competition theory and conclude that these figures are likely due to relaxed post?cop?ulatory sexual selection,kittiwakes being strictly monogamous.Finally,we suggest that this method could be applied to other seabird species sharing similar ecology. 展开更多

Sperm competition has been studied in numerous species as a representative example of postcopulatory sexual selection,where sampling sperm from male is the most basic and important step.Sperm collection can be tricky in birds,however,because unlike mammals,the genitals of birds are generally latent in the cloacal region and their characteristics vary among species.Various methods to collect sperm from different birds have been tested,such as cloacal massage,feces collection,and electro-stimulation,but their applicability varies depending on species.In this study,we introduced the urodeum stimulation method(UroS method)to collect sperm from Cuculus cuckoos,such as the Common Cuckoo(C.canorus).These species are expected to have interesting patterns of pair bonding and sperm competition because of their unique breeding strategy called brood parasitism;however,it remains unexplored.In this study,we described the application of our new method to expel semen from male common cuckoos,measured the volume of semen collected,checked the presence of sperm in the semen sample,and finally estimated its success rate among 82 males.Samples were successfully collected from 76 cuckoos(approximately 93%)and the colors and volumes of the samples were very diverse.Sperm was present in 43 of these samples(57%),showing a sperm observation rate approximately twice as high as that of the conventional cloacal massage method.We believe that this novel method will contribute to a better understanding of postcopulatory sexual selection in avian brood parasites and facilitate the process of sperm collection and artificial insemination in other medium-sized birds. 展开更多

Oral supplementation of selenium improves post-thaw sperm quality in Saanen bucks

Objective:To determine the effect of oral selenium supplementation and semen collection methods on various post thaw semen quality parameters in Saanen bucks.Methods:Sixteen healthy bucks were divided into two equal groups(n=8 each).The treatment group received selenium at 10-day intervals for three months.Sperm kinematic parameters,morphological parameters,mitochondrial membrane potential,plasma membrane functionality,and sperm DNA integrity were evaluated weekly pre and post-cryopreservation.Results:The mean percentages of the morphological abnormalities were significantly lower in the selenium-supplemented samples when semen was collected by using artificial vagina method(P<0.05).Proximal droplet defects were significantly lower in the selenium supplementation group when semen was collected by electro-ejaculation(P<0.05).Post-thaw sperm parameters such as total motility and progressive motility were significantly higher when semen was obtained by artificial vagina in the selenium-supplemented bucks compared to the electro-ejaculation and the control groups(P<0.05).The sperm kinematic parameters such as curvilinear velocity,average path velocity,and amplitude of lateral head displacement were significantly higher when semen was collected by artificial vagina in the selenium-treated bucks(P<0.05).The percentages of sperm with intact and functional plasma membrane and functional mitochondria were significantly higher in the selenium-supplemented samples collected with artificial vagina compared to the electro-ejaculation method and the control groups(P<0.05).In vitro fertilizing potential was significantly higher in the selenium-supplemented samples collected with artificial vagina compared to the electro-ejaculation method and the control groups,respectively(P<0.05).Conclusions:Oral supplementation of selenium and artificial vagina semen collection improve post thaw sperm quality parameters of Saanen buck. 展开更多
