Soxllb regulates the migration and fate determination of Müller glia-derived progenitors during retina regeneration in zebrafish

The transcription factor Sox11 plays important roles in retinal neurogenesis during vertebrate eye development.However,its function in retina regeneration remains elusive.Here we report that Sox11 b,a zebrafish Sox11 homolog,regulates the migration and fate determination of Müller glia-derived progenitors(MGPCs)in an adult zebrafish model of mechanical retinal injury.Following a stab injury,the expression of Sox11 b was induced in proliferating MGPCs in the retina.Sox11 b knockdown did not affect MGPC formation at 4 days post-injury,although the nuclear morphology and subsequent radial migration of MGPCs were alte red.At 7 days post-injury,Sox11 b knockdown res ulted in an increased proportion of MGPCs in the inner retina and a decreased propo rtion of MGPCs in the outer nuclear layer,compared with controls.Furthermore,Sox11 b knockdown led to reduced photoreceptor regeneration,while it increased the numbe rs of newborn amacrines and retinal ganglion cells.Finally,quantitative polymerase chain reaction analysis revealed that Sox11 b regulated the expression of Notch signaling components in the retina,and Notch inhibition partially recapitulated the Sox11 b knockdown phenotype,indicating that Notch signaling functions downstream of Sox11 b.Our findings imply that Sox11 b plays key roles in MGPC migration and fate determination during retina regeneration in zebrafish,which may have critical im plications for future explorations of retinal repair in mammals.

Fate of fertilizer nitrogen and residual nitrogen in paddy soil in Northeast China

The relationship between the fate of nitrogen (N) fertilizer and the N application rate in paddy fields in Northeast China is unclear,as is the fate of residual N.To clarify these issues,paddy field and15N microplot experiments were carried out in 2017 and 2018,with N applications at five levels:0,75,105,135 and 165 kg N ha–1(N0,N75,N105,N135 and N165,respectively).15N-labeled urea was applied to the microplots in 2017,and the same amount of unlabeled urea was applied in 2018.Ammonia (NH3) volatilization,leaching,surface runoff,rice yield,the N contents and15N abundances of both plants and soil were analyzed.The results indicated a linear platform model for rice yield and the application rate of N fertilizer,and the optimal rate was 135 kg N ha–1.N uptake increased with an increasing N rate,and the recovery efficiency of applied N (REN) values of the difference subtraction method were 45.23 and 56.98%on average in 2017and 2018,respectively.The RENwas the highest at the N rate of 135 kg ha–1in 2017 and it was insignificantly affected by the N application rate in 2018,while the agronomic efficiency of applied N (AEN) and physiological efficiency of applied N (PEN) decreased significantly when excessive N was applied.N loss through NH3volatilization,leaching and surface runoff was low in the paddy fields in Northeast China.NH3volatilization accounted for 0.81 and 2.99%of the total N application in 2017 and 2018,respectively.On average,the leaching and surface runoff rates were 4.45% and less than 1.05%,respectively,but the apparent denitrification loss was approximately 42.63%.The residual N fertilizer in the soil layer (0–40 cm) was 18.37–31.81 kg N ha–1in 2017,and the residual rate was 19.28–24.50%.Residual15N from fertilizer in the soil increased significantly with increasing N fertilizer,which was mainly concentrated in the 0–10 cm soil layer,accounting for 58.45–83.54% of the total residual N,and decreased with increasing depth.While the ratio of residual N in the 0–10 cm soil layer to that in the 0–40 cm soil layer was decreased with increasing N application.Furthermore,of the residual N,approximately 5.4%was taken up on average in the following season and 50.2%was lost,but 44.4%remained in the soil.Hence,the amount of applied N fertilizer should be reduced appropriately due to the high residual N in paddy fields in Northeast China.The appropriate N fertilizer rate in the northern fields in China was determined to be 105–135 kg N ha–1in order to achieve a balance between rice yield and high N fertilizer uptake.

A virtual character is a design of a fictitious creature with distinctive characteristics created by people,and Disney virtual characters are those Intellectual Property(IP)images that appeared in Disneyland and Disney movies.This investigation aimed to explore why many younger females are keen to spend money on Disney virtual characters.This paper adopted the Marketing Mix Theory strategy(product,price,placement,and promotion(4P)),and the SWOT analysis method has been utilized.This paper investigated the relationship between the 4Ps and consumers’purchasing intentions,and it turned out that unique design and effective promotion in this Disney case would promote consumers’purchase intention,while the higher price and less accessible placements affected their purchase intentions.Thus,the high price and limited places somewhat inhibit customers’desire to buy;due to the attractiveness of the product itself and the promotion on the internet,the target consumers are still willing to consume.

A Comparative Analysis of the Origin and Formation Between Chinese Characters and English Alphabets

This research is based on the framework of social constructivism,utilizing the principle of Human Universals as a methodology to compare the similarities and differences between the ideation and formation methods of Chinese characters and English alphabets.Through comparative analysis of the ideation of English letters(pictogramme)and the origin of Chinese characters,known as the"Six Categories Theory,"we discover their alignment in terms of social,traditional,and cultural aspects.This suggests that different ethnic groups share common features in terms of life experience,learning cognitive development,and thinking habits.This study also finds that the origins of English letters and Chinese characters share similar linguistic features in their methods of constructing letters/characters,such as pictographic,ideographic,and semantic characteristics.Exploring these commonalities contributes to promoting learning and communication between Chinese and English characters.Additionally,by focusing on socio-cultural aspects,traditional customs,and cognitive learning,this study aims to break away from the traditional linguistic research approach that solely focuses on language differences.This provides a broader perspective and richer dimensions for Chinese and English language learning,facilitating the development of cross-linguistic and cross-cultural communication.

A Harmonic Approach to Handwriting Style Synthesis Using Deep Learning

The challenging task of handwriting style synthesis requires capturing the individuality and diversity of human handwriting.The majority of currently available methods use either a generative adversarial network(GAN)or a recurrent neural network(RNN)to generate new handwriting styles.This is why these techniques frequently fall short of producing diverse and realistic text pictures,particularly for terms that are not commonly used.To resolve that,this research proposes a novel deep learning model that consists of a style encoder and a text generator to synthesize different handwriting styles.This network excels in generating conditional text by extracting style vectors from a series of style images.The model performs admirably on a range of handwriting synthesis tasks,including the production of text that is out-of-vocabulary.It works more effectively than previous approaches by displaying lower values on key Generative Adversarial Network evaluation metrics,such Geometric Score(GS)(3.21×10^(-5))and Fréchet Inception Distance(FID)(8.75),as well as text recognition metrics,like Character Error Rate(CER)and Word Error Rate(WER).A thorough component analysis revealed the steady improvement in image production quality,highlighting the importance of specific handwriting styles.Applicable fields include digital forensics,creative writing,and document security.

Yes-associated protein 1(YAP1)is a downstream effector of the Hippo signaling pathway,and it is involved in tumorigenesis,tissue repair,growth,and development.In this review,the biological roles and the mechanisms of YAP1 in mediating stem cell fate decisions are discussed,including cell proliferation,differentiation,and apoptosis.In general,YAP1 promotes the proliferation and differentiation of stem cells,including embryonic stem cells and adult stem cells.It inhibits apoptosis by binding to the transcription factors,e.g.,transcriptional enhanced associate domain(TEAD),Smad,runt-related transcription factor 1/2,p73,p63,and Erb84,to maintain tissue homeostasis.The translocalization of YAP1 in cellular nuclei and the phosphorylation in the cytoplasm work as important and unusual events for the activation of YAP1.Moreover,YAP1 serves as the crosstalk for the Hippo pathway and other signaling pathways,including the Wnt and Notch pathways.It is highlighted in this review that YAP1 is an essential regulator for stem cells that have significant applications in regenerative medicine and reproductive medicine.

Identification of a Reliable DFT Calculation Method to Predict the Ground-and Excited-State Properties of Nickel(Ⅱ) Complexes with Porphyrin in Crude Oil

Metalloporphyrins have devastating effects on the deep processing and efficient utilization of petroleum resources. Various porphyrins are derived from porphin by substitution at the β, meso, or both positions. Herein, we focus on the interaction between nickel(Ⅱ) and the porphyrin skeleton. Five different density functionals were selected for determining a reliable approach to simulate porphyrin-nickel(Ⅱ) complexes. At D4h symmetry, the 10b1g, 15a1g, 4e1g and 9b2g were dominated by the 3d character of nickel. Compared with pure density functional, the hybrid functional gave larger highest occupied molecular orbital(HOMO) and lowest unoccupied molecular orbital(LUMO) gap, and the higher HOMO-LUMO gap also corresponded to a higher excitation energy. The characteristic Soret and Q bands modeled by timedependent density functional theory(TDDFT) calculations matched well with experimental results, and the absorption was discussed in detail with natural transition orbitals(NTO) analysis. Furthermore, the binding character of nickel(Ⅱ) with various substituted porphyrins were calculated at perdew-burke-ernzerhof(PBE) level, showed that the geometry and complexation behavior of porphyrin-nickel(Ⅱ) complexes could be significantly tuned by different substituents. The symmetry of the complex was reduced and the skeleton ring get distortion when introducing different substituents. Compared with ETIO porphyrin, the introduction of phenyl on the β position can expand the conjugated system and promote the covalent character of Ni-N bond.
