As a data protection legislation with the dual objectives of market regulation and government supervision,the EU General Data Protection Regulation(GDPR)is designed to promote the cultivation of a market for data ele⁃ments and to regulate the data processing subjects'behaviour.The civil legal norms to participate in the data market regulation are introduced,and a right-protected mode with the obligation norms of data controllers as the core is started by GDPR.However,from the perspective of GDPR's global enforcement effectiveness,as an export-orientated global standard and protection framework,the power regulation model of the EU's single digital market is facing the in⁃novative incentives of data market expansion and the tension of human rights protection.A more modest and moderate data governance regulation model has been opened by a new round of the EU digital market regulation.The legal sce⁃narios transition and digital platforms regulation more fit the regulatory demand and development goals of global digital economy development.The EU's determination to enhance the global digital market share and competitiveness is dem⁃onstrated.For China,the impact and security risks of the new round should be evaluated carefully,drawing on the ex⁃perience of the technical standpoint and institutional framework and regulatory rules of the EU digital market regula⁃tion,providing the regulatory models and Chinese solutions to respond to the EU digital market regulation,and enhanc⁃ing Chinese institutional discourse and rule-making leadership in the global digital economy. 展开更多

Attachment Styles and Traumatic Responses: Exploring the Impact of Parental Interaction on Child Development and Coping Mechanisms

This article explores the intricate relationship between attachment styles formed during early childhood and the subsequent responses to traumatic events, particularly the death of a parent. Drawing on the theoretical framework of attachment theory and incorporating contemporary research, the paper discusses how parental interactions shape the neural circuitry of infants and children, influencing their ability to form secure or insecure attachments. These attachment styles, in turn, play a critical role in determining the child’s coping mechanisms when faced with trauma. This paper focuses on trying to understand how attachment theory is connected to the reaction to trauma with a highlight on the four major styles of attachments which are secure, anxious, avoidant, and disorganized to mention but a few, and how they influence stress and adversity in children. Attachment theory holds that human beings’ ability to form affectional bonds in infancy determines their patterns of relatedness across the life cycle. The type of attachment that is secure usually supports healthy adaptation and good coping mechanisms regardless of the trauma in the childhood of the child. While secure attachment mostly facilitates favorable trauma-related outcomes, anxious or avoidant attachment can exacerbate or alter the responses. The caregiving system that is avoidant attachment has implications of autonomous self-functioning which has features of suppression of the emotional response and poor search for emotional support during stress. From the principles of developmental psychology and trauma theory, the paper also focuses on the major significance of the child’s early caregivers’ interactions that define the resilience and vulnerability factor. This knowledge is therefore critical in designing specific interventions based on the improvement of coping behaviors and emotional regulatory systems of children who have been exposed to trauma. Finally, we have the synthesis of new knowledge about the role of secure attachment relationships as its fundamental element in shaping adaptive traumatization and psychological development. The article also delves into the physiological processes involved in emotional regulation and the role of cortisol in disrupting attachment. Finally, the implications of these findings for therapeutic interventions and the challenges of addressing prolonged grief and traumatic responses in clinical settings are considered. 展开更多

Foxtail millet(Setaria italica L.),a member of the Paniceae family,is a temperate and tropical grass species that is widely cultivated on the Eurasian continent.It is Chinese in origin and possesses a small genome,short growth cycle,and strong natural abiotic stress resistance.Elucidating the mechanism of millet tolerance to salt stress is becoming increasingly important with increasing soil salinization limiting crop productivity.The responses and mechanisms of tolerance to salt stress from other model plants such as Arabidopsis and rice,were compared with those from foxtail millet to summarize current research on responses to salt stress.Numerous processes are involved in these processes,including physiological reactions,sensing,signaling,and control at the transcriptional,post-transcriptional,and epigenetic levels.To increase crop productivity and agricultural sustainability,a variety of technologies can be used to investigate how salt tolerance is mediated by physiological and molecular processes in foxtail millet. 展开更多

Plant N starvation response is closely associated with the N signaling components that involve transduction of the low-N cues. In this study, we functionally characterized Ta ARR1, a cytokinin(CK) response regulator gene in Triticum aestivum, in mediating the N starvation adaptation in plants. Ta ARR1 harbors two conserved domains specified by plant ARR family members;subcellular localization analysis indicated its target onto nucleus after endoplasmic reticulum assortment. Ta ARR1 displayed modified expression upon the N starvation stressor, showing upregulated expression in roots and leaves over a 27-h N starvation treatment and whose induced transcripts were gradually recovered along with progression of the N recovery treatment. The tobacco lines overexpressing Ta ARR1 displayed improved low-N stress tolerance, displaying enlarged phenotype, increased biomass and N accumulation, and enhanced glutamine synthetase(GS) activities compared with wild type(WT) following the N starvation treatment. Expression analysis on genes encoding the nitrate transporter(NRT) and GS proteins in Nicotiana tabacum revealed that Nt NRT2.2 and Nt GS3 are upregulated in expression in the N-deprived transgenic lines, whose expression patterns were contrasted to other above family genes that were unaltered on transcripts between the transgenic lines and WT. Transgene analysis validated the function of Nt NRT2.2 and Nt GS3 in regulating N accumulation, GS activity, growth traits, and N use efficiency in plants. These results suggested the internal connection between the Ta ARR1-mediated N starvation tolerance and the modified transcription of distinct N acquisitionand assimilation-associated genes. Our investigation together indicates that Ta ARR1 is essential in plant N starvation adaptation due to the gene function in transcriptionally regulating distinct NRT and GS genes that affect plant N uptake and assimilation under the N starvation condition. 展开更多

Regulators are important components in pneumatic system, and their flow-rate characteristics are the key parameters for designers. According to the correlatively international standard and national standard of China, which describe the flow-rate characteristics measurement method of pneumatic regulators, the pressure and the flow are measured point by point, and then the flow-rate characteristics curve is plotted point to point. This method has some disadvantages, such as equipment complexity, much air consumption, and low efficiency. To settle the problems presented above, this paper puts forward a new high efficient and energy saving flow-rate characteristics measurement method of regulators, which is based on the pressure response when charging and discharging to an isothermal tank without any flow meters. The measurement principle, the system and the steps are introduced. And the tracking differentiator is used for the data processing of the pressure difference. Two typical kinds of regulators were experimentally investigated, and their flow-rate characteristics curves were obtained with the new and the conventional method, respectively. Comparatively, it's proved that this new method is feasible because it is not only able to meet the demand of the measurement precision, but also to save energy and improve efficiency. Compared to the conventional method, the new method takes only about 1/10 amount of time and consumes about only 1/30 amount of air. Hopefully it will be able to serve as an international standard of flow-rate characteristics measurement method of regulators. 展开更多

BACKGROUND Pancreatic ductal cancer(PDAC)has high malignancy and poor prognosis.Long noncoding RNAs(lncRNAs)are associated with high levels of malignancy,including PDAC.However,the biological and clinical significance of negative regulator of antiviral response(NRAV)in PDAC is unclear.AIM To study the regulatory role of lncRNA NRAV in PDAC.METHODS GEPIA analyzed lncRNA NRAV and miRNA(miR-299-3p)expression levels in PDAC tissues and measured them in PDAC cells by quantitative measurements in real time.The specific role of NRAV and miR-299-3p in cell proliferation and transfer potential was evaluated by cell formation analysis,Cell Counting Kit-8 and Transwell analysis.The relationship between NRAV and miR-299-3p was studied by predictive bioinformatics,RNA immunoassay,and fluorescence enzyme analysis.In vivo experiments included transplantation of simulated tumor cells under naked mice.RESULTS The expression level of lncRNA NRAV was higher in both tumor tissues and cell lines of PDAC and was negatively associated with the clinical survival of PDAC patients.Functionally,overexpression of NRAV promoted cell proliferation and metastasis of PDAC cells,while knockdown of NRAV reversed these effects.Finally,NRAV was performed as a molecular sponge of miR-299-3p.Moreover,overexpression of miR-299-3p could reverse the promoting effects of NRAV on cell proliferation and metastasis of PDAC cells.CONCLUSION NRAV facilitates progression of PDAC as a molecular sponge of miR-299-3p and may be a potential molecular marker for diagnosis and treatment of PDAC. 展开更多

Objective:To study the effects of silent information regulator of transcription 2 (SIRT2) on inflammatory response and bone destruction in cartilage tissue of osteoarthritis.Methods: A total of 200 patients who underwent knee replacement due to knee osteoarthritis in Kashgar Prefecture First People's Hospital between September 2014 and September 2017 were selected as the osteoarthritis (OA) group of the research, and 80 patients who underwent knee replacement or meniscus operation due to trauma in Kashgar Prefecture First People's Hospital during the same period were selected as the control group. Articular cartilage tissue was collected after surgery to measure the expression of SIRT2 and bone destruction-related apoptosis molecules as well as the levels of inflammatory response molecules and bone destruction-related collagen metabolism molecules.Results: SIRT2 and Bcl-2 mRNA expression as well as SOX9 and Col-II levels in articular cartilage tissue of OA group were significantly lower than those of control group whereas TNF-α, bFGF, NO, IP-10, CCL2, PAR-2,β-catenin, OPN and MMP13 levels as well as Fas, GRP78, ATF6 and Caspase-3 mRNA expression were significantly higher than those of control group;SIRT2 mRNA expression in articular cartilage tissue of OA group was positively correlated with Bcl-2 mRNA expression as well as SOX9 and Col-II levels, and negatively correlated with TNF-α, bFGF, NO, IP-10, CCL2, PAR-2,β-catenin, OPN and MMP13 levels as well as Fas, GRP78, ATF6 and Caspase-3 mRNA expression.Conclusion: The lowly expressed SIRT2 in cartilage of osteoarthritis can aggravate inflammatory response and bone destruction. 展开更多

Global warming threatens food security.Rice(Oryza sativa L.),a vital food crop,is vulnerable to heat stress,especially at the reproductive stage.Here we summarize putative mechanisms of high-temperature perception(via RNA secondary structure,the phyB gene,and phase separation)and response(membrane fluidity,heat shock factors,heat shock proteins,and ROS(reactive oxygen species)scavenging)in plants.We describe how rice responds to heat stress at different cell-component levels(membrane,endoplasmic reticulum,chloroplasts,and mitochondria)and functional levels(denatured protein elimination,ROS scavenging,stabilization of DNA and RNA,translation,and metabolic flux changes).We list temperature-sensitive genetic male sterility loci available for use in rice hybrid breeding and explain the regulatory mechanisms associated with some of them.Breeding thermotolerant rice species without yield penalties via natural alleles mining and transgenic editing should be the focus of future work. 展开更多

With the increase in the power receiving proportion and an insufficient peak regulation capacity of the local units, the receiving-end power grid struggles to achieve peak regulation in valley time. To solve this problem while considering the potential of the large-scale charge load of electric vehicles(EVs), an aggregator-based demand response(DR) mechanism for EVs that are participating in the peak regulation in valley time is proposed in this study. In this aggregator-based DR mechanism, the profits for the power grid’s operation and the participation willingness of the EV owners are considered. Based on the characteristics of the EV charging process and the day-ahead unit generation scheduling, a rolling unit commitment model with the DR is established to maximize the social welfare. In addition, to improve the efficiency of the optimization problem solving process and to achieve communication between the independent system operator(ISO) and the aggregators, the clustering algorithm is utilized to extract typical EV charging patterns. Finally, the feasibility and benefits of the aggregator-based DR mechanism for saving the costs and reducing the peak-valley difference of the receiving-end power grid are verified through case studies. 展开更多

Atherosclerosis or fibrosis and cirrhosis undergo chronic inflammation associated with the adhesion between neutrophils and endothelial cells(ECs)that is mediated by their respective cellular adhesive molecules on stiffened blood vessel wall or extracellular matrix(ECM)under shear flow[1-3].However,the mechanical dependence of calcium flux and trail formation in neutrophils remains unclear yet in these processes.First,the effect of substrate stiffness through ECs on neutrophil calcium spike was quantified when the individual neutrophils adhered to EC monolayer pre-placed onto stiffness-varied polyacrylamide(PA)substrate(5 or 34.88 kPa)or glass surface.Our data indicated that E-/P-selectins and ICAM-1s on HUVECs and b2-integrins,PSGL-1s,and CD44s on neutrophils were all involved in mediating neutrophil calcium spike in a stiffness-dependent manner,in which the increase of substrate stiffness enhanced the calcium intensity and spike number.Such stiffness-dependent calcium response is associated with selectin-induced b2-integrin activation through Syk/Src signaling pathway and the F-actin/myosin II function.Moreover,tension-activated calcium ion channels displayed critical roles in initiating stiffness-dependent calcium spike [4].Second,the trail formation of neutrophils to ECs monolayer pre-placed onto the same PA substrate were also tested under shear flow.Live fluorescence imaging showed that neutrophils are able to form long membrane tethers during migration and subsequently leave behind membranous long-lasting trails under shear,which are enriched in LFA-1,Mac-1,and CD44.Moreover,the formation of the trails was inhibited by blocking LFA-1s and Mac-1s,suggesting an important role forβ2-integrins in the trial formation.The recruitment of monocytes was inhibited when pre-blocking ICAM-1s on flowing monocytes,indicating that the neutrophil’s trails employβ2-integrin-ICAM-1 binding to recruit the monocytes.Intriguingly,both the length and the area of the trails increase with increasing substrate stiffness,resulting in the enhanced monocyte recruitment.Inhibition of actin binding protein Arp2/3 impairs the trail formation and dramatically decreases the neutrophil-dependent monocyte recruitment.These data provide an insight into understanding how stiffening of vascular wall could regulate the calcium flux of adhered neutrophils and thus the immune responses in atherosclerosis.They also imply that local mechanical microenvironment is remodeled with the migration of neutrophils,leaving the trails presented to induce and regulate monocyte recruitment.All the results are meaningful in elucidating the occurrence and development of atherosclerosis or fibrosis from the viewpoint of mechanotransduction and also for the potential intervention of cardiovascular disease progress. 展开更多

In the present review we summarize the known roles of extracellular signal-regulated kinase, cAMP response element-binding protein and brain-derived neurotrophic factor in the pathogenesis of depression and in the mechanism of action of antidepressant medicines. The extracellular signal-regulated kinase/cAMP response element-binding protein/brain-derived neurotrophic factor pathway has potential to be used as a biological index to help diagnose depression, and as such it is considered as an important new target in the treatment of depression. 展开更多

Monitoring expression at the transcriptional level is the first essential step for the functional analysis of plant genes. Genes-encoding proteins directly involved in early response to elicitor constitute only a small fraction of all the genes affected by elicitor. Transcriptional responses to various elicitors have been extensively studied in different plants including Nicotiana and Arabidopsis thaliana; however, corresponding data aren't available for non-heading Chinese cabbage. To address this problem, we describe a suppression subtractive library-based approach to isolate the plant's ESTs up-regulated in the early induction/execution of the HR induced by elicitor PB90 from Phytophthora boehmeriae. According to their putative identification in BLAST searches against the three genome databases, 70 up-regulated genes were classified into 9 parts: some aspect of primary 'metabolism' or 'energy' production; 'protein synthesis' or 'protein fate'; cellular communication/signal transduction mechanism; cell fates including Beclin, SPT1, and SPT2; HLA-B and AGO1 which participate in transcription; cellular transport and hypothetical proteins or proteins for which a function has yet to be determined. Seven selected genes such as Beclin, thioredoxin, HLA-B, MAP3K, SPT1, SPT2, and AGO1 were up-regulated induced by PB90, suggesting that the genes may play an important role in PB90-triggered HR. 展开更多

Granular power management in a power-efficient system on a chip(SoC)requires multiple integrated voltage regulators with a small area,process scalability,and low supply voltage.Conventional on-chip analog low-dropout regulators(ALDOs)can hardly meet these requirements,while digital LDOs(DLDOs)are good alternatives.However,the conventional DLDO,with synchronous control,has inherently slow transient response limited by the power-speed trade-off.Meanwhile,it has a poor power supply rejection(PSR),because the fully turned-on power switches in DLDO are vulnerable to power supply ripples.In this comparative study on DLDOs,first,we compare the pros and cons between ALDO and DLDO in general.Then,we summarize the recent DLDO advanced techniques for fast transient response and PSR enhancement.Finally,we discuss the design trends and possible directions of DLDO. 展开更多

Pancreatic adenocarcinoma is one of the most lethal cancers with rising incidence.Despite progress in its treatment,with the introduction of more effective chemotherapy regimens in the last decade,prognosis of metastatic disease remains inferior to other cancers with long term survival being the exception.Molecular characterization of pancreatic cancer has elucidated the landscape of the disease and has revealed common lesions that contribute to pancreatic carcinogenesis.Regulation of proteostasis is critical in cancers due to increased protein turnover required to support the intense metabolism of cancer cells.The proteasome is an integral part of this regulation and is regulated,in its turn,by key transcription factors,which induce transcription of proteasome structural units.These include FOXO family transcription factors,NFE2L2,hHSF1 and hHSF2,and NF-Y.Networks that encompass proteasome regulators and transduction pathways dysregulated in pancreatic cancer such as the KRAS/BRAF/MAPK and the Transforming growth factor beta/SMAD pathway contribute to pancreatic cancer progression.This review discusses the proteasome and its transcription factors within the pancreatic cancer cellular microenvironment.We also consider the role of stemness in carcinogenesis and the use of proteasome inhibitors as therapeutic agents. 展开更多

Soil salinity severely limits crop yields and quality.Plants have evolved several strategies to mitigate the adverse effects of salinity,including redistribution and compartmentalization of toxic ions using ion-specific transporters.However,the mechanisms underlying the regulation of these ion transporters have not been fully elucidated.Loss-of-function mutants of OsHKT2;1,which is involved in sodium uptake,exhibit strong salt stress-resistant phenotypes.In this study,OsHKT2;1 was identified as a transcriptional target of the type-B response regulator OsRR22.Loss-of-function osrr22 mutants showed resilience to salt stress,and OsRR22-overexpression plants were sensitive to salt stress.OsRR22 was found to activate the expression of OsHKT2;1 by directly binding to the promoter region of OsHKT2;1 via a consensus cis-element of type-B response regulators.Moreover,rice DELLA protein OsSLR1 directly interacted with OsRR22 and functioned as a transcriptional co-activator.This study has uncovered a novel transcriptional regulatory mechanism by which a type-B response regulator controls sodium transport under salinity stress. 展开更多

Stress adaptation is fundamental for health, and the hypothalamic-pituitary-adrenal axis (HPA) is one of its main mechanisms. Considerable data indicate that arginine vasopressin (AVP) related disturbances of stress adaptation can occur with aging. However, most studies of such kind have been performed on rodents, give contradictory results and fail to consider individual characteristics of the animals. The purpose of this study was to investigate individual HPA responsiveness to acute stress and its vasopressinergic regulation in old female rhesus monkeys that differ in their behavioral responses to stress. Animals with depression-like or anxiety-like behavior (DAB) responded with higher plasma levels of ACTH and AVP, lower levels of corticosteroids and higher cortisol/DHEAS molar ratios to restraint stress and to insulin-induced hypoglycemia compared with animals with healthy adaptive behavior. AVP and ACTH dynamics were closely correlated in most animals. AVP treatment produced differences in HPA responses similar to those produced by the stressors. The ACTH response to hypoglycemic stress in the DAB animal with highest HPA responsiveness was dramatically reduced by prior administration of a V1b receptor antagonist. These results suggest that the dysfunctions of HPA observed in old animals with DAB are caused by increased tone of the vasopressinergic system in regulation of HPA stress reactivity. 展开更多

Any observer of current economic development in the world and in the Europe should ask about the causes of the current crisis situation. The authors of this article perceive current situation in the European Union and many of its member states associated with long-term developmental tendencies still less respecting the objective differences in various economic situation of the member states. Establishment and further functioning of monetary union in Europe is gradually becoming a burden for many countries, which prevents full utilization of instruments of monetary and fiscal policy by daily, and in many cases qualitative differences in the problems of economies of individual member states. The fulfillment of these tendencies in particular in the Euro Area monetary policy is confronted with the dominant position of member states in fiscal policy and in the mutual differences among member states. As considerations of differences in economic level and in member states fiscal policies, both of them are based on official figures, some of them are part of the state. In addition, EU institutions face to face to crisis they are increasingly resorting to deepening statist centralization and enter more often to the processes which are usually domains of business subjects, at most of nation states. All these trends lead to a deepening asymmetry between the decision-making processes and responsibilities for economic development and they could lead to even larger failure of national economies or at European or global level. Since the accession of the Czech Republic to the European Union in 2004 our department has dealing with issues connecting to the economic management of insurance companies and in relation to the ongoing integration process research interest is extended also to other segments of financial markets. 展开更多

The relevance of constipation to the development and progression of colorectal cancer(CRC)is currently a controversial issue.Studies have shown that changes in the composition of the gut microbiota,a condition known as ecological imbalance,are correlated with an increasing number of common human diseases,including CRC and constipation.CRC is the second leading cause of cancerrelated deaths worldwide,and constipation has been receiving widespread attention as a risk factor for CRC.Early colonoscopy screening of constipated patients,with regular follow-ups and timely intervention,can help detect early intestinal lesions and reduce the risks of developing colorectal polyps and CRC.As an important regulator of the intestinal microenvironment,the gut microbiota plays a critical role in the onset and progression of CRC.An increasing amount of evidence supports the thought that gut microbial composition and function are key determinants of CRC development and progression,with alterations inducing changes in the expression of host genes,metabolic regulation,and local and systemic immunological responses.Furthermore,constipation greatly affects the composition of the gut microbiota,which in turn influences the susceptibility to intestinal diseases such as CRC.However,the crosstalk between the gut microbiota,constipation,and CRC is still unclear. 展开更多
