BACKGROUND Diabetic intracerebral hemorrhage(ICH)is a serious complication of diabetes.The role and mechanism of bone marrow mesenchymal stem cell(BMSC)-derived exosomes(BMSC-exo)in neuroinflammation post-ICH in patients with diabetes are unknown.In this study,we investigated the regulation of BMSC-exo on hyperglycemia-induced neuroinflammation.AIM To study the mechanism of BMSC-exo on nerve function damage after diabetes complicated with cerebral hemorrhage.METHODS BMSC-exo were isolated from mouse BMSC media.This was followed by transfection with microRNA-129-5p(miR-129-5p).BMSC-exo or miR-129-5poverexpressing BMSC-exo were intravitreally injected into a diabetes mouse model with ICH for in vivo analyses and were cocultured with high glucoseaffected BV2 cells for in vitro analyses.The dual luciferase test and RNA immunoprecipitation test verified the targeted binding relationship between miR-129-5p and high-mobility group box 1(HMGB1).Quantitative polymerase chain reaction,western blotting,and enzyme-linked immunosorbent assay were conducted to assess the levels of some inflammation factors,such as HMGB1,interleukin 6,interleukin 1β,toll-like receptor 4,and tumor necrosis factorα.Brain water content,neural function deficit score,and Evans blue were used to measure the neural function of mice.RESULTS Our findings indicated that BMSC-exo can promote neuroinflammation and functional recovery.MicroRNA chip analysis of BMSC-exo identified miR-129-5p as the specific microRNA with a protective role in neuroinflammation.Overexpression of miR-129-5p in BMSC-exo reduced the inflammatory response and neurological impairment in comorbid diabetes and ICH cases.Furthermore,we found that miR-129-5p had a targeted binding relationship with HMGB1 mRNA.CONCLUSION We demonstrated that BMSC-exo can reduce the inflammatory response after ICH with diabetes,thereby improving the neurological function of the brain. 展开更多

This study examined the impact of workplace-based sports participation on the mental status of Japanese workers. It involved a single-arm intervention comprising participation in a company’s volleyball team. Data on mental status were collected using a simplified version of the Brief Job Stress Questionnaire. Overall, 11 individuals participated, 9 (81.8%) of whom were women;the mean participant age was 38.2 years. Because the intervention was conducted during busy periods, the median (25th and 75th percentiles) score for qualitative job overload significantly increased from 3.0 (2.0, 4.0) to 3.0 (3.0, 4.3) (p = 0.03). Nonetheless, the fatigue score significantly decreased from 3.0 (3.0, 4.3) to 3.0 (2.0, 4.0) (p = 0.03). These results indicate that participation in workplace-based sports can prevent the deterioration of mental status during busy periods. 展开更多

The major concentration of these unions was in Bratislava and Eastern Slovakia. Concerning their specialized field, they can be divided into tourism-practising, physical and sport ones However, the union activities were strongly influenced by political parties. Objective Innate lymphoid cells(ILCs)are a class of newly discovered immunocytes.Group 1 ILCs(ILC1s)are identified in the decidua of humans and mice.High mobility group box 1(HMGB1)is predicted to be one of the target genes of miR-142-3p,which is closely related to pregnancy-related diseases.Furthermore,miR-142-3p and HMGB1 are involved in regulating the NF-κB signaling pathway.This study aimed to examine the regulatory effect of miR-142-3p on ILC1s and the underlying mechanism involving HMGB1 and the NF-κB signaling pathway.Methods Mouse models of normal pregnancy and abortion were constructed,and the alterations of ILC1s,miR-142-3p,ILC1 transcription factor(T-bet),and pro-inflammatory cytokines of ILC1s(TNF-α,IFN-γand IL-2)were detected in mice from different groups.The targeting regulation of HMGB1 by miR-142-3p in ILC1s,and the expression of HMGB1 in normal pregnant mice and abortive mice were investigated.In addition,the regulatory effects of miR-142-3p and HMGB1 on ILC1s were detected in vitro by CCK-8,Annexin-V/PI,ELISA,and RT-PCR,respectively.Furthermore,changes of the NF-κB signaling pathway in ILC1s were examined in the different groups.For the in vivo studies,miR-142-3p-Agomir was injected in the uterus of abortive mice to evaluate the abortion rate and alterations of ILC1s at the maternal-fetal interface,and further detect the expression of HMGB1,pro-inflammatory cytokines,and the NF-κB signaling pathway.Results The number of ILC1s was significantly increased,the level of HMGB1 was significantly upregulated,and that of miR-142-3p was considerably downregulated in the abortive mice as compared with the normal pregnant mice(all P<0.05).In addition,miR-142-3p was found to drastically inhibit the activation of the NF-κB signaling pathway(P<0.05).The number of ILC1s and the levels of pro-inflammatory cytokines were significantly downregulated and the activation of the NF-κB signaling pathway was inhibited in the miR-142-3p Agomir group(all P<0.05).Conclusion miR-142-3p can regulate ILC1s by targeting HMGB1 via the NF-κB signaling pathway,and attenuate the inflammation at the maternal-fetal interface in abortive mice. 展开更多

[Objective] The aim of the study is to clone and analyze the gene encoding 14-3-3 protein from banana. [Method] Combined with PCR amplification, RACE (rapid amplification of cDNA ends) technique was employed to clone 14-3-3 gene from banana; then the amplified sequence was sequenced and homologically analyzed. [Result] A new cDNA homologous with 14-3-3 protein genes were obtained by RT-PCR and RACE ( rapid amplification of cDNA ends ) approaches. The full length of this cDNA was 866 bp encoding 197 amino acids. Alignment of deduced amino acid sequence with those from other plants revealed that the cDNA shared high homology with 14-3-3 protein genes from other plants, and was designated as Musa acuminata 14-3-3 gene (Ma-14-3-3d). Phylogenetic analysis reveals that Ma-14-3-3d has closer genetic relationship with those from monocotyledon species than those from other species. [Conclusion] Ma-14-3-3d belongs to the same lineage of 14-3-3 from monocotyledon. 展开更多

目的探讨胃癌组织中高迁移率族蛋白A2(HMGA2)及基质金属蛋白酶-9(MMP-9)表达与肿瘤侵袭和转移能力的关系及预后意义。方法采用免疫组化法(SP法)检测93例胃癌组织、30例正常胃黏膜组织中HMGA2、MMP-9的表达;收集患者临床病历资料,并进行... 目的探讨胃癌组织中高迁移率族蛋白A2(HMGA2)及基质金属蛋白酶-9(MMP-9)表达与肿瘤侵袭和转移能力的关系及预后意义。方法采用免疫组化法(SP法)检测93例胃癌组织、30例正常胃黏膜组织中HMGA2、MMP-9的表达;收集患者临床病历资料,并进行随访。结果 HGMA2蛋白在胃癌组织中阳性表达率分别为明显高于正常胃黏膜(P<0.01);MMP-9蛋白在胃癌组织中阳性表达率明显高于正常胃黏膜(P<0.01)。HMGA2与MMP-9在胃癌组织中的蛋白阳性表达均与胃癌组织的肿瘤浸润深度、肿瘤分化程度、淋巴结转移、TNM分期有关(P<0.05);与患者的性别、年龄、肿瘤直径无关(P>0.05)。相关性分析发现,HMGA2与MMP-9在胃癌组织中的表达情况呈正相关(r=0.317,P<0.01)。经Kaplan-Meier生存分析显示,HMGA2、MMP-9蛋白表达阳性患者的生存率均低于表达阴性患者(P<0.01)。结论 HMGA2、MMP-9与胃癌浸润和转移有关,两者表达具有相互协同作用,对胃癌的侵袭和转移起重要的促进作用。HMGA2、MMP-9是影响预后的危险因素,可能成为胃癌治疗的新靶点。 展开更多

目的探讨丙泊酚的肺保护作用及其可能作用机制。方法采用尾静脉注射内毒素建立急性肺损伤模型,丙泊酚和锌原卟啉同样经过尾静脉给药。将Wistar大鼠按随机数字表法分为实验对照组(C组)、脂多糖组(L组)、脂多糖+丙泊酚组(LP组)。C组尾静... 目的探讨丙泊酚的肺保护作用及其可能作用机制。方法采用尾静脉注射内毒素建立急性肺损伤模型,丙泊酚和锌原卟啉同样经过尾静脉给药。将Wistar大鼠按随机数字表法分为实验对照组(C组)、脂多糖组(L组)、脂多糖+丙泊酚组(LP组)。C组尾静脉注射生理盐水;L组尾静脉注射LPS 7.5mg·kg-1;LP组尾静脉注射LPS 7.5mg·kg-1,同时静脉注射丙泊酚30mg·kg-1。给药前及开始后第3、6和12h测定动脉血氧分压(PaO2),实验结束后观察肺湿/干重(W/D)比值、肺组织病理学检查并进行肺损伤轻重程度评分,同时观察各组大鼠肺组织高迁移率族蛋白1(HMGB-1)含量。结果实验前各组PaO2无明显差异,注射LPS后L组PaO2持续下降,和C组相比其PaO2显著降低(P<0.05)。实验结束后L组和C组相比其W/D比值、肺组织病理学检查评分、HO-1及HMGB-1含量显著增加(P<0.05)。而LP组和L组相比其PaO2显著改善,而W/D比值、肺组织病理学检查评分及HMGB-1含量显著降低(P<0.05)。结论丙泊酚具有肺保护作用,且该作用可能与其抑制HMGB-1过度表达作用相关。 展开更多

Learning activities interactions between small groups is a key step in understanding team sports videos.Recent research focusing on team sports videos can be strictly regarded from the perspective of the audience rather than the athlete.For team sports videos such as volleyball and basketball videos,there are plenty of intra-team and inter-team relations.In this paper,a new task named Group Scene Graph Generation is introduced to better understand intra-team relations and inter-team relations in sports videos.To tackle this problem,a novel Hierarchical Relation Network is proposed.After all players in a video are finely divided into two teams,the feature of the two teams’activities and interactions will be enhanced by Graph Convolutional Networks,which are finally recognized to generate Group Scene Graph.For evaluation,built on Volleyball dataset with additional 9660 team activity labels,a Volleyball+dataset is proposed.A baseline is set for better comparison and our experimental results demonstrate the effectiveness of our method.Moreover,the idea of our method can be directly utilized in another video-based task,Group Activity Recognition.Experiments show the priority of our method and display the link between the two tasks.Finally,from the athlete’s view,we elaborately present an interpretation that shows how to utilize Group Scene Graph to analyze teams’activities and provide professional gaming suggestions. 展开更多

针对中国女运动员在奥运会的优势现象,从项群角度对第27-31届夏季奥运会我国女子冠军比赛成绩比较分析,发现这连续5届奥运会中不同项群的女运动员运动寿命有所不同,技能主导项群的运动员运动寿命长于体能主导项群运动员,且技能主导项群... 针对中国女运动员在奥运会的优势现象,从项群角度对第27-31届夏季奥运会我国女子冠军比赛成绩比较分析,发现这连续5届奥运会中不同项群的女运动员运动寿命有所不同,技能主导项群的运动员运动寿命长于体能主导项群运动员,且技能主导项群中隔网对抗类运动员运动寿命最长。而在体能主导项群中,速度性项目运动员的运动寿命长于耐力性和快速力量性项目运动员的运动寿命。同时也发现技能主导项群是我国女运动员夺金的主要项群,体能主导项群是我国女运动员夺金的弱势项群;技能主导项群运动员的运动寿命与运动成绩有可能存在正比的关系。 展开更多
