Stem cell-like memory T(TSCM)cells possess stem cell properties including multipotency and self-renewal and are being recognized as emerging players in various human diseases.Advanced technologies such as multiparametric flowcytometry and single cell sequencing have enabled their identification and molecular characterization.In case of chronic viral diseases such as human immunodeficiency virus-1,CD4+T_(SCM) cells,serve as major reservoirs of the latent virus.However,during immune activation and functional exhaustion of effector T cells,these cells also possess the potential to replenish the pool of functional effector cells to curtail the infection.More recently,these cells are speculated to play important role in protective immunity following acute viral infections such as coronavirus disease 2019 and might be amenable for therapeutics by ex vivo expansion.Similarly,studies are also investigating their pathological role in driving autoimmune responses.However,there are several gaps in the understanding of the role of T_(SCM) cells in viral and autoimmune diseases to make them potential therapeutic targets.In this minireview,we have attempted an updated compilation of the dyadic role of these complex T_(SCM) cells during such human diseases along with their biology and transcriptional programs. 展开更多

Influencing program satisfaction for undergraduate students in STEM areas: The impact of interdisciplinary features, disciplinary connectedness, faculty support, and examination difficulty

Ensuring program satisfaction for undergraduate students in the areas of science,technology,engineering and mathematics(STEM)matters in student retention and education quality improvement.This study explores how four rarely examined variables support from faculty members,interdisciplinary features of STEM program courses,disciplinary connectedness of STEM program core courses,and examination difficulty impact Chinese STEM undergraduates’program satisfaction.With data from 619 Chinese STEM undergraduates,structural equation modeling shows that course satisfaction partially mediates the impact of support from faculty members on program satisfaction,while fully mediating that of interdisciplinary features of STEM program courses and disciplinary connectedness of STEM program core courses on program satisfaction.Examination difficulty exerts no significant impact on program satisfaction neither directly nor indirectly.Support from faculty members impact course satisfaction significantly stronger for junior and senior students than for freshmen and sophomores,while interdisciplinary features of STEM program courses impact course satisfaction stronger for freshmen and sophomores than for juniors and seniors.The study ends with practical implications for the higher education reform in relevant areas. 展开更多

BACKGROUND: S100 protein can promote axonal growth. Therefore, transplantation of induced bone marrow-derived mesenchymal stem cells (MSCs) that can secrete S100 may provide a beneficial microenvironment for neural regeneration. OBJECTIVE: To explore the changes in S100 expression during rat MSCs differentiation into Schwann ceils in vitro. DESIGN, TIME AND SETTING: This cytology experiment was performed at the Jiangsu Key Laboratory of Neuroregeneration, Nantong University in China, from January 2006 to May 2007. MATERIALS: The rabbit anti-S100 polyclonal antibody was purchased from Dako, Denmark; the mouse anti-rat S100 monoclonal antibody was purchased from Sigma, USA. METHODS: MSCs were cultured from adult Sprague-Dawley rat femur and tibia. Cell proliferation was determined by the MTT method and CD markers, and cell cycle was measured by flow cytometry. MSCs were induced to differentiate into SC cells. SC cells were stained for S100 protein, glial fibrillary acidic protein, and low-affinity nerve growth factor receptor. S100 protein and mRNA levels were evaluated by flow cytometry, Western blot, and reverse transcription-polymerase chain reaction. MAIN OUTCOME MEASURES: S100 protein and mRNA expression. RESULTS: MSCs exhibited high amplification potential over eight passages. Prior to induction, the majority of MSCs were at the G0/G1 phase of the cell cycle. After induction, MSCs displayed morphology changes similar to Schwann cells. Moreover, induction increased S100 mRNA levels. Immunofluorescence showed that MSCs expressed S100 protein, glial fibrillary acidic protein, and low-affinity nerve growth factor receptor at 7 days of induction. Induction also increased S100 protein levels compared with untreated MSCs. CONCLUSION: MSCs are capable of differentiating into Schwann cells-like cells under conditional induction in vitro, with increasing S100 mRNA and protein expression. 展开更多

Identification of precursor mi... MicroRNAs are an important subclass of non-coding RNAs (ncRNA), and serve as main players into RNA interference (RNAi). Mature microRNA derived from stem-loop structure called precursor. Identification of precursor microRNA (pre-miRNA) is essential step to target microRNA in whole genome. The present work proposed 25 novel local features for identifying stem- loop structure of pre-miRNAs, which captures characteristics on both the sequence and structure. Firstly, we pulled the stem of hairpins and aligned the bases in bulges and internal loops used ‘―’, and then counted 24 base-pairs (‘AA’, ‘AU’, …, ‘―G’, except ‘――’) in pulled stem (formalized by length of pulled stem) as features vector of Support Vector Machine (SVM). Performances of three classifiers with our features and different kernels trained on human data were all superior to Triplet-SVM-classifier’s in po- sitive and negative testing data sets. Moreover, we achieved higher prediction accuracy through combining 7 global sequence-structure. The result indicates validity of novel local features. 展开更多

目的探讨中线部位和脑室节细胞胶质瘤的临床病理及分子遗传学特点。方法收集11例中线部位和脑室节细胞胶质瘤患者的临床特征、免疫表型及分子检测结果,并复习总结相关文献。结果镜下观察节细胞胶质瘤由异常神经元和肿瘤性增生的胶质细... 目的探讨中线部位和脑室节细胞胶质瘤的临床病理及分子遗传学特点。方法收集11例中线部位和脑室节细胞胶质瘤患者的临床特征、免疫表型及分子检测结果,并复习总结相关文献。结果镜下观察节细胞胶质瘤由异常神经元和肿瘤性增生的胶质细胞构成,免疫表型NeuN、MAP2可标记肿瘤中的神经元成分,GFAP和Olig-2标记出了肿瘤性神经胶质细胞成分;分子遗传学常存在BRAFV600E点突变,但也可发现存在其他形式BRAF基因以及MAPK信号通路基因的改变。随访病例多数预后良好。结论发生在中线部位及脑室的节细胞胶质瘤较为罕见,对于发生于不典型部位的这种低级别神经上皮肿瘤,要将其纳入鉴别诊断中,可在形态学基础上,结合免疫组织化学以及分子检测结果进行综合分析。 展开更多

Smart Approaches to Efficient Text Mining for Categorizing Sexual Reproductive Health Short Messages into Key Themes

To promote behavioral change among adolescents in Zambia, the National HIV/AIDS/STI/TB Council, in collaboration with UNICEF, developed the Zambia U-Report platform. This platform provides young people with improved access to information on various Sexual Reproductive Health topics through Short Messaging Service (SMS) messages. Over the years, the platform has accumulated millions of incoming and outgoing messages, which need to be categorized into key thematic areas for better tracking of sexual reproductive health knowledge gaps among young people. The current manual categorization process of these text messages is inefficient and time-consuming and this study aims to automate the process for improved analysis using text-mining techniques. Firstly, the study investigates the current text message categorization process and identifies a list of categories adopted by counselors over time which are then used to build and train a categorization model. Secondly, the study presents a proof of concept tool that automates the categorization of U-report messages into key thematic areas using the developed categorization model. Finally, it compares the performance and effectiveness of the developed proof of concept tool against the manual system. The study used a dataset comprising 206,625 text messages. The current process would take roughly 2.82 years to categorise this dataset whereas the trained SVM model would require only 6.4 minutes while achieving an accuracy of 70.4% demonstrating that the automated method is significantly faster, more scalable, and consistent when compared to the current manual categorization. These advantages make the SVM model a more efficient and effective tool for categorizing large unstructured text datasets. These results and the proof-of-concept tool developed demonstrate the potential for enhancing the efficiency and accuracy of message categorization on the Zambia U-report platform and other similar text messages-based platforms. 展开更多

It has become general for surface waters being polluted by micro organic compounds. In order to know the current pollution situation and the properties of micro organic compounds in the Changjiang River, a test was performed on micro organic compounds in the water, bottom material and fish bodies which were sampled from major city river reaches of the Changjiang River. Based on the test result, researchers described and analyzed the sorts, concentration level and distribution features of micro organic compounds. A comprehensive evaluation was conducted by adopting the method of MEG (Multimedia Environmental Goals). The study indicated that ① the water body of major city river reaches of the Changjiang River has been generally polluted. In the test, totally 12 types with 308 kinds of organic compounds were detected. The main pollutants were paraffins, PAHs and lipids; and ② micro organic pollutant content in fish bodies was generally higher than that in bottom material which is in turn higher than that in water; and ③ pollution is relatively severe in the river reaches of mid-to-large comprehensive industrial cities with fairly great TAS (Total Ambient Severity) of public health and ecological system. 展开更多

Stem cell microterritories(niches),as a specialized part of the extracellular matrix(ECM),are considered an important target and tool for the development of new materials,medical implants,and devices.However,tissue bioengineering products that have stem cell niches of known size on the surface or in the bulk structure of artificial materials are practically unknown.This brief review attempts to draw attention to the problematic aspects of niches as specific parts of the ECM,such as their hierarchy and size for mesenchymal stromal/stem cells(MSCs).These parameters arise directly from numerous definitions of stem cell niches as specialized morphological microterritories found in various tissues.The authors of this review analyze the known information on the hierarchy of MSC microterritories by analogy with that of hematopoietic stem cells.Occasional reports on the size of artificial MSC niches compared to natural niche candidates are summarized.A consensus on a hierarchy and optimal range of niche sizes for MSCs and other stem cells is needed to accelerate the development of prototyping technologies and additive manufacturing in applications to precise tissue bioengineering and regenerative medicine. 展开更多

目的·分析弥漫大B细胞淋巴瘤(diffuse large B-cell lymphoma,DLBCL)患者临床特点和预后危险因素,评估自体造血干细胞移植(autologous stem cell transplantation,ASCT)及利妥昔单抗维持治疗对DLBCL患者预后的影响。方法·收... 目的·分析弥漫大B细胞淋巴瘤(diffuse large B-cell lymphoma,DLBCL)患者临床特点和预后危险因素,评估自体造血干细胞移植(autologous stem cell transplantation,ASCT)及利妥昔单抗维持治疗对DLBCL患者预后的影响。方法·收集上海交通大学医学院附属新华医院血液科2015年1月—2020年1月收治的160例经病理及免疫分型初次确诊的DLBCL患者的临床资料,分析影响患者疗效与预后的危险因素。分析复发/难治性DLBCL患者的临床特征,评估挽救性ASCT对患者总生存(overall survival,OS)的影响。对中期评估达到完全缓解(complete remission,CR)的高危患者,评估ASCT及利妥昔单抗维持治疗对其生存预后的影响。结果·初治年龄>60岁(P=0.005)、国际预后指数(International Prognostic Index,IPI)3~5分(P=0.032)、低白蛋白水平(P=0.001)及贫血(P=0.007)患者的近期疗效不佳。多因素分析结果显示:患者初治年龄>60岁(HR=2.788,95%CI 1.575~4.936,P=0.000),non-GCB亚型(HR=2.230,95%CI 1.150~4.324,P=0.018),乳酸脱氢酶水平升高(HR=2.064,95%CI 1.006~4.234,P=0.048),低白蛋白水平(HR=2.052,95%CI1.169~3.602,P=0.012)是影响患者无进展生存(progression-free survival,PFS)的独立危险因素;患者初治年龄>60岁(HR=2.269,95%CI 1.060~4.860,P=0.035),IPI评分3~5分(HR=2.557,95%CI 1.132~5.778,P=0.024)作为独立因素影响患者OS。对于复发/难治性DLBCL患者,挽救性ASCT能显著改善其预后,是患者死亡事件的保护性因素(P=0.030)。对于化学治疗后中期评估达到CR的高危患者,巩固性ASCT及利妥昔单抗维持治疗者至随访终点尚未出现死亡事件,但并未延长患者OS(P>0.05)。结论·挽救性ASCT能显著延长复发/难治性DLBCL患者OS,但巩固性ASCT及利妥昔单抗维持治疗并不能延长高危DLBCL患者OS。 展开更多

调控TGF-β1-PI3K/AKT轴对骨肉瘤细胞恶性增殖和干性表达的影响

目的探讨调控转化生长因子-β1(transforming growth factor-β1,TGF-β1)基因对骨肉瘤细胞的恶性增殖和干细胞样特性的影响以及可能的机制。方法本文采用试验对照研究。研究时间为2021年6月至12月。选取对数生长期的人骨肉瘤细胞,采用T... 目的探讨调控转化生长因子-β1(transforming growth factor-β1,TGF-β1)基因对骨肉瘤细胞的恶性增殖和干细胞样特性的影响以及可能的机制。方法本文采用试验对照研究。研究时间为2021年6月至12月。选取对数生长期的人骨肉瘤细胞,采用TGF-β1-siRNA及TGF-β1重组腺病毒分别下调、过表达TGF-β1基因;反转录聚合酶链反应(qRT-PCR)和蛋白免疫印迹(Western blotting)来检测TGF-β1-siRNA干扰效率;通过四甲基偶氮唑盐(methyl thiazolyl tetrazolium,MTT)法和平板克隆形成实验检测细胞的增殖特性;流式细胞术分选CD133阳性表达的肿瘤干细胞(cancer stem cells,CSCs),微球体形成实验检测细胞自我更新能力;Western blotting检测细胞凋亡和干细胞特性相关蛋白。组间比较采用t检验。结果与人正常成骨hFOB1.19细胞相比,TGF-β1在骨肉瘤细胞系均有不同程度的增高;其中U2OS细胞中上调程度最高(t=8.76,P<0.05),MG-63细胞中上调程度最少(t=4.33,P<0.05)。因此,U2OS细胞用于基因沉默实验,MG-63细胞用于基因过表达实验。qRT-PCR和Western blotting实验显示,si-TGF-β1#1的基因沉默效率最高(t=10.88,P<0.05),因此,后续实验采用si-TGF-β1#1。MTT法、流式细胞术结果显示,抑制TGF-β1表达后骨肉瘤细胞的细胞活力明显降低,细胞凋亡增加;而TGF-β1过表达则提高细胞活力,细胞凋亡减少。悬浮成球、CD133阳性细胞筛选实验结果显示,抑制TGF-β1表达后明显削弱骨肉瘤细胞的微球体形成能力(t=6.88,P<0.05)和CD133阳性细胞比例(t=9.37,P<0.05);反之,过表达TGF-β1会使骨肉瘤干细胞干性表达增强。同时,裸鼠体内实验亦证实Lv-TGF-β1可扩大肿瘤的体积和质量;反之si-TGF-β1预处理后瘤体的体积和质量有所减轻(t=9.27,P<0.05)。结论TGF-β1在骨肉瘤中发挥促癌基因角色,TGF-β1增强骨肉瘤细胞的恶性增殖和干细胞特性,其作用机制可能与磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/AKT)信号通路有关。 展开更多

尽管多数林木非同化器官树干、树枝内都存在绿色组织(Chlorenchyma),但是其生态学意义很少为人注意。综合前人研究结果,得出以下结论:(1)树枝的光合速率(暗呼吸与饱和光照下呼吸之差)的在0~10μmolm^-2s^-1之间,而大部分的... 尽管多数林木非同化器官树干、树枝内都存在绿色组织(Chlorenchyma),但是其生态学意义很少为人注意。综合前人研究结果,得出以下结论:(1)树枝的光合速率(暗呼吸与饱和光照下呼吸之差)的在0~10μmolm^-2s^-1之间,而大部分的结果认为光合速率在0.5~3.Oμmolm^-2s^-1之间。而且多数研究认为其是对自身呼吸的再固定,而不是对外界大气CO2的吸收,对再固定率(光合/呼吸比值)分析表明80%以上的研究结果认为树干光合作用能够固定40%~100%呼吸所释放的CO2。但其对个体乃至林分整体碳平衡的影响报道的较少。(2)不仅非同化器官光合作用直接影响其呼吸作用,冠层叶片光合产物对非同化器官的气体代谢也产生显著影响,但新形成光合产物和储藏碳水化合物是否存在功能上的差异不同学者意见不一。(3)尽管叶绿素含量多在80~450mgm^-2之间,低于相应叶片叶绿素含量340~620mgm^-2,但其叶绿素a/b平均值(2.5)显著低于叶片的平均值(3.6)(P〈0.0001),说明更加适应于阴生环境。(4)有关类胡萝卜素在非同化器官的功能,比较传统的观点认为其单位质量或者单位面积含量远低于叶片,是一种适应于低光照环境的表现,但最近研究表明其单位叶绿素含量远高于叶片,可能是在高浓度CO2条件下、叶绿体片层结构酸性化导致光合过程中光化学淬灭能力低下,需要以叶黄素循环为主导能量淬灭过程保证其功能正常。(5)在树皮光学特性方面,大部分光照被吸收,而有少部分被反射和透射。在透光率方面,70%左右的结果认为树皮的透光率在5%~15%之间,而超过85%的结果认为透光率在0%~20%之间。而且光照可以在木质化树干导管、纤维及管胞有效进行轴向导光,树干内部的光以红外和远红外光为主,其它低波长光的透射能力远低于红外光。(6)尽管已有研究表明某些草本植物茎具有C4特征,但对于多数、特别是木本植物未见报道,特别是对于茎内高浓度CO2、高红/蓝光比、低氧特征对光合机构的影响尚需要深入研究。 展开更多
