Methods The effects of ST, DON and AFG1 on apoptosis of HPBLs were studied with cell culture, flow cytometric (FCM) DNA analysis and DNA agarose gel electrophoresis. Results DNA agarose gel electrophoresis results showed the characteristic 'ladder' pattern of apoptosis in HPBLs treated with ST, DON and AFG1. Flow cytometric DNA analysis revealed that typical subdiploid peaks of apoptosis in DNA histogram could be seen in all groups treated with the three mycotoxins. Significant time-effect and dose-effect relationships were found between the apoptosis rates and treatment time as well as concentrations of the three mycotoxins. Conclusion ST, DON and AFG1 can induce apoptosis of HPBLs in vitro and may have some negative effects on human immunosystem.展开更多
Fourteen strains of Aspergillus versicolor and 2 strains of A. nidulans were screened for sterigmatocystin (ST) production on a semi-synthetic solid substrate by high performence liquid chromatography (HPLC) analysis....Fourteen strains of Aspergillus versicolor and 2 strains of A. nidulans were screened for sterigmatocystin (ST) production on a semi-synthetic solid substrate by high performence liquid chromatography (HPLC) analysis. Two strains of A. versicolor producing ST at 550.5 mg.kg-1 substrate and 1160.8 mg·kg-1 substrate were selected to inoculate 4 kg solid ST-producing media. After 30 days stationary incubation at 28 ℃ in the dark, 2271.6 mg of pale-yellow needle-shaped crystals were isolated and purified from the culture with a procedure applying column chromatography and recrystallization method. The crystal was proved to be sterigmatocystin by spectroanalysis and some physico-chemical analysis. The purity of the final material obtained were more than 99.9% as shown by HPLC and TLC detection. With this procedure, ST was obtained at about one tenth of its commercial cost展开更多

Mice fed with Aspergillus-inoculated corn flour developed adenocarcinoma of the lung in 15 of 35 mice (42.9%) and atypical hyperplasia of the glandular stomach in 13 of 35 mice (37.4%). Sterigmatocystin was identified by high performance liquid chromato-graphy (HPLC) and fluorescence spectrophotometry in the extract of Aspergillus-inoculated corn flour. The results suggest that the mycotoxin Sterigmatocystin may play a potential role in carcinogenesis in human.展开更多
