Cold atmospheric plasma(CAP)driven by pulsed high-voltage power has specific advantages in various fields,such as the growth promotion of edible fungi in plasma agriculture.Compared with other strains,Ganoderma lucidu...Cold atmospheric plasma(CAP)driven by pulsed high-voltage power has specific advantages in various fields,such as the growth promotion of edible fungi in plasma agriculture.Compared with other strains,Ganoderma lucidum has the significant advantage of high medicinal value,but the shortcomings of low yield,long growth cycle,and an uneven market quality.In this study,Ganoderma lucidum was treated with a CAP powered by a homemade pulsed highvoltage power supply.Three groups of Ganoderma lucidum were treated under different conditions.The stalk lengths and pileus areas of Ganoderma lucidum were recorded to evaluate growth status.Results showed that treatment frequency affected growth status considerably.Ganoderma lucidum was treated with 40 s plasma once a week and showed an improved growth status with a 33.63%increase in average stalk length and a 28%increase in the number of individuals whose pileus areas was greater than the average.Meanwhile,the growth speed was accelerated.However,treatment at an excessive frequency would damage and slow down the growth of Ganoderma lucidum.展开更多

The response of phytoplankton and its satellite bacteria to various concentrations(0.01%-10%v/v)of ethanol is studied.To elucidate the effect of ethanol,single-strains of phytoplankton(SSP)culture,pure strains of sate...The response of phytoplankton and its satellite bacteria to various concentrations(0.01%-10%v/v)of ethanol is studied.To elucidate the effect of ethanol,single-strains of phytoplankton(SSP)culture,pure strains of satellite bacteria isolated from nonaxenic SSP cultures,and Escherichia coli were screened.Results indicate that ethanol could promote the growth and photo synthetic efficiency(F_(v)/F_(m))of S SP at 0.01%and the growth of satellite bacteria at 0.01%-1%.Nevertheless,ethanol inhibited the growth and F_(v)/F_(m)of SSP at 0.1%-1%,and killed bacteria and SSP at 10% concentration.Further investigation on a satellite bacterium(Mameliella alba)revealed that ethanol promotes growth by serving as a growth stimulant rather than a metabolic carbon source.The 16 S rRNA gene amplicon indicated that all nonaxenic S SP cultures harbor distinct satellite bacteria communities where the SSP culture of Skeletonema costatum,Phaeodactylum tricornutum,and Dunaliella bardawil were dominated by bacteria genera of Marivita(~80%),Dinoroseobacter(~47%),and Halomonas(~87%),respectively,indicating that every SSP cultures have their own distinct satellite bacterial community.The bacteria family Rhodobacteraceae was dominant in the two marine diatoms,whereas Halomonadaceae was dominant in the saline green microalga.Compared to their respective controls,the supply of 0.5% ethanol to SSP cultures promoted the growth of the satellite bacteria but did not cause a significant difference in species composition of satellite bacteria.Therefore,a low concentration of ethanol can promote the growth of bacteria in a non-selective way.This study enriched our knowledge about the effect of ethanol on aquatic microbes and provided a baseline for basic and applied biotechnological re search in the aquatic environment in the future.展开更多
<strong>Objective:</strong> To explore the level of soluble growth-stimulated expression gene 2 (sST2) in patients with atrial fibrillation (AF) and its relationship with atrial remodeling. <strong>M...<strong>Objective:</strong> To explore the level of soluble growth-stimulated expression gene 2 (sST2) in patients with atrial fibrillation (AF) and its relationship with atrial remodeling. <strong>Methods:</strong> From December 2018 to July 2019, 86 patients were enrolled in the cardiovascular department of Jingzhou Central Hospital in Hubei Province. AF group was 60 patients with AF diagnosed according to the diagnostic criteria of electrophysiology and pacing branch of Chinese Medical Association, while the control group was 26 normal ECG patients with sinus rhythm examined by ECG and/or DCG. Clinical data, NT proBNP and birth rate of all patients were collected chemical indicators. All the patients in the groups were examined by ECG and color Doppler echocardiography. The serum level of ST2 was detected by enzyme-linked immunosorbent assay (ELISA), and the indexes related to atrial remodeling in echocardiography, left atrial diameter (LAD), were collected, compared and analyzed. <strong>Results:</strong> Serum sST2 levels and LAD and RAD values in the AF group were higher than those in the control group (P < 0.05 or P < 0.01). Serum sST2 levels were higher in the AF group than in the paroxysmal atrial fibrillation group (P = 0.031). 1) The sST2 level was positively correlated with LAD and RAD measured by cardiac ultrasound (r = 0.388, P = 0.029). <strong>Conclusion:</strong> Serum sST2 levels have early recognition value in patients with high-risk atrial fibrillation, which contributes to the stratification of risk of atrial fibrillation and the prediction of disease activity. Elevated serum sST2 levels may be associated with atrial remodeling. sST2 may be an objective biomarker that predicts the risk of emergency admission in patients with atrial fibrillation. Elevated sST2 levels may be associated with atrial fibrillation progression.展开更多
Aim:The effects of hepatocyte growth factor(HGF)on a non-invasive prostate cancer cell line(CAHPV-10),expressing cMET were studied,to mimic the possible effects neo-adjuvant androgen deprivation therapy may have in pr...Aim:The effects of hepatocyte growth factor(HGF)on a non-invasive prostate cancer cell line(CAHPV-10),expressing cMET were studied,to mimic the possible effects neo-adjuvant androgen deprivation therapy may have in promoting tumour progression.Methods:Prostate epithelial cells and prostate cancer cells derived from cancer metastatic sites were analysed using cell culture assays,immunofluorescence,quantitative real-time polymerase chain reaction and western blotting,with or without HGF stimulation.Results:HGF significantly enhanced cell proliferation and induced cell scattering and invasion in CAHPV-10 cells compared to untreated controls.Active adenosine diphosphate-ribosylation factor 6(ARF6)was found to be present in all metastatic prostate cancer cells,with levels highest in the most aggressive cell line,PC-3.Following stimulation with HGF,active ARF6 expression was substantially elevated in CAHPV-10 cells.Conclusion:These findings provide further molecular insight into the progression of prostate cancer and highlights potential issues for early prostate cancer therapeutic strategies.展开更多

Diadenosine polyphosphates have been identified as modulators of the cardiovascular system....The proliferation of vascular smooth muscle cells (VSMC) plays a role in the maintenance of hypertension and atherosclerosis. Diadenosine polyphosphates have been identified as modulators of the cardiovascular system. We detected the existence of diadenosine polyphosphates in the heart and studied their growth stimulating effect on VSMC. Porcine cardiac tissue was deproteinated with perchloric acid and concentrated with a preparative reversed phase C18 column. Then the extract was separated with a size exclusion column, an anion exchange column, and an affinity column. It was then chromatographed with an anion exchange HPLC and a reversed phase HPLC and a purified substance was obtained. Med. Klinik I, Nephrol. Labor, Univ. Klinik Marien hospital, University of Bochum, Herne, Germany (Luo JK, Wang H, Schlüter H, JankowskifJ, Potthoff W, Tepel M and Zidek W) Using the BrdU method, this substance was shown to have a growth stimulating effect that is dose-dependent on VSMC. Matrix assisted laser desorption/ionization mass spectrometry (MALDI MS) analysis showed that the molecular mass of the substance was 757.0, the same as diadenosine triphosphate (AP 3A). UV spectrum indicated the maximum absorption at 259 nm, similar to that of adenosine. Postsource decay (PSD) MALDI MS showed that the substance contained adenine, adenosine, AMP, ADP, and ATP, respectively. Enzymatic cleavage revealed that phosphates in this molecule connected adenosine in 5' position. It was determined from the above data that the substance was Ap 3A. In this study the existence of Ap 3A was demonstrated in the heart. It has a growth stimulating effect on VSMC.展开更多
The cytochrome P450 enzyme CYP-sb21 from the rare actinomycete Sebekia benihana is capable of hydroxylating the immunosuppressive drug molecule cyclosporine A(CsA)primarily at the 4th N-methyl leucine(MeLeu4),giving r...The cytochrome P450 enzyme CYP-sb21 from the rare actinomycete Sebekia benihana is capable of hydroxylating the immunosuppressive drug molecule cyclosporine A(CsA)primarily at the 4th N-methyl leucine(MeLeu4),giving rise toγ-hydroxy-N-methyl-L-Leu4-CsA(CsA-4-OH).This oxidative modification of CsA leads to dramatically reduced immunosuppressive activity while retaining the hair growth-promoting side-effect,thus demonstrating great application potential in both pharmaceutical and cosmetic industries.However,this P450 enzyme also hydroxylates CsA at the unwanted position of the 9th N-methyl leucine(MeLeu9),indicating that the regioselectivity needs to be improved for the development of CsA-4-OH into a commercial hair growth stimulator.Herein,we report the crystal structure of CYP-sb21 in its substrate-free form at 1.85Å.Together with sequence and 3D structure comparisons,Autodock-based substrate docking,molecular dynamics(MD)simulation,and site-directed mutagenesis,we identified a number of key residues including R294,E264,and M179 that can improve catalytic efficiency or change the regioselectivity of CYP-sb21 towards CsA,setting the stage for better enzymatic preparation of CsA-4-OH.This study also provides new insights into the substrate recognition and binding mechanism of P450 enzymes that accommodate bulky substrates.展开更多
