Boulder spacing in mountain rivers and near-wake flow zones within the boulder array is very useful for fish habitat and growth of aquatic organisms.The present study aims to investigate how the boulder array and spac...Boulder spacing in mountain rivers and near-wake flow zones within the boulder array is very useful for fish habitat and growth of aquatic organisms.The present study aims to investigate how the boulder array and spacing influence the near-bed flow structures in a gravel-bed stream.Boulders are staggered over a gravel-bed stream with three different inter-boulder spacing namely(a)large(b)medium and(c)small spacing.An acoustic Doppler velocimeter was used for flow measurements in a rectangular channel and the results were compared with those acquired from numerical simulation.The time-averaged velocity profiles at the near-wake flow zones of boulders experience maximum flow retardation which is an outcome of the boulder-induced form roughness.The ratio of velocity differences associated to form and skin roughness and its positive magnitude reveals the dominance of form roughness closest to the boulders.Form roughness computed is 1.75 to 2 times higher than the skin roughness at the near-wake flow region.In particular,the collective immobile boulders placed at different inter-boulder spacings developed high and low bed shear stresses closest to the boulders.The low bed shear stresses characterised by a secondary peak developed at the trough location of the boulders is attributed to the skin shear stress.Further,the spatial averaging of time-averaged flow quantities gives additional impetus to present an improved illustration of fluid shear stresses.The formation of form-induced shear stress is estimated to be 17%to 23%of doubleaveraged Reynolds shear stress and partially compensates for the damping of time-averaged Reynolds shear stress in the interfacial sub-layer.The quadrant analysis of spatial velocity fluctuations depicts that the form-induced shear stresses are dominant in the interfacial sub-layer and have no significance above the gravel-bed surface.展开更多

We present a 42-year-old female, with multiple sclerosis, whose neurological symptoms and respiratory decline were finally explained by the diagnosis of West Nile-encephalitis. During her admission, the isolated peaked T-waves indicated the underlying stress-induced cardiomyopathy. The absence of all other causes of hyperacute T-waves, their subsequent resolution with the resolution of infection and improvement in wall motion abnormalities, further supported the association. This case highlights the importance of considering hyperacute T-waves in an approach towards the diagnosis of WNV-encephalitis related atypical variant of stress-induced cardiomyopathy.展开更多
Evaluating the physical mechanisms that link hydraulic fracturing(HF) operations to induced earthquakes and the anticipated form of the resulting events is significant in informing subsurface fluid injection operation...Evaluating the physical mechanisms that link hydraulic fracturing(HF) operations to induced earthquakes and the anticipated form of the resulting events is significant in informing subsurface fluid injection operations. Current understanding supports the overriding role of the effective stress magnitude in triggering earthquakes, while the impact of change rate of effective stress has not been systematically addressed. In this work, a modified critical stiffness was brought up to investigate the likelihood, impact,and mitigation of induced seismicity during and after hydraulic fracturing by developing a poroelastic model based on rate-and-state fraction law and linear stability analysis. In the new criterion, the change rate of effective stress was considered a key variable to explore the evolution of this criterion and hence the likelihood of instability slip of fault. A coupled fluid flow-deformation model was used to represent the entire hydraulic fracturing process in COMSOL Multiphysics. The possibility of triggering an earthquake throughout the entire hydraulic fracturing process, from fracturing to cessation, was investigated considering different fault locations, orientations, and positions along the fault. The competition between the effects of the magnitude and change rate of effective stress was notable at each fracturing stage. The effective stress magnitude is a significant controlling factor during fracturing events, with the change rate dominating when fracturing is suddenly started or stopped. Instability dominates when the magnitude of the effective stress increases(constant injection at each fracturing stage) and the change rate of effective stress decreases(the injection process is suddenly stopped). Fracturing with a high injection rate, a fault adjacent to the hydraulic fracturing location and the position of the junction between the reservoir and fault are important to reduce the Coulomb failure stress(CFS) and enhance the critical stiffness as the significant disturbance of stresses at these positions in the coupled process. Therefore,notable attention should be given to the injection rate during fracturing, fault position, and position along faults as important considerations to help reduce the potential for induced seismicity. Our model was verified and confirmed using the case of the Longmaxi Formation in the Sichuan Basin, China, in which the reported microseismic data were correlated with high critical stiffness values. This work supplies new thoughts of the seismic risk associated with HF engineering.展开更多
DI-3-n-butylphthalide is used to treat mild and moderate acute ischemic stroke.However,the precise underlying mechanism requires further investigation.In this study,we investigated the molecular mechanism of DI-3-n-bu...DI-3-n-butylphthalide is used to treat mild and moderate acute ischemic stroke.However,the precise underlying mechanism requires further investigation.In this study,we investigated the molecular mechanism of DI-3-n-butylphthalide action by various means.We used hydrogen peroxide to induce injury to PC12cells and RAW264.7 cells to mimic neuronal oxidative stress injury in stroke in vitro and examined the effects of DI-3-n-butylphthalide.We found that DI-3-nbutylphthalide pretreatment markedly inhibited the reduction in viability and reactive oxygen species production in PC12 cells caused by hydrogen peroxide and inhibited cell apoptosis.Furthermore,DI-3-n-butylphthalide pretreatment inhibited the expression of the pro-apoptotic genes Bax and Bnip3.DI-3-nbutylphthalide also promoted ubiquitination and degradation of hypoxia inducible factor 1α,the key transcription factor that regulates Bax and Bnip3 genes.These findings suggest that DI-3-n-butylphthalide exhibits a neuroprotective effect on stroke by promoting hypoxia inducible factor-1α ubiquitination and degradation and inhibiting cell apoptosis.展开更多
Unconventional reservoirs usually contain many weak surfaces such as faults,laminae and natural fractures,and effective activation and utilization of these weak surfaces in reservoirs can significantly improve the ext...Unconventional reservoirs usually contain many weak surfaces such as faults,laminae and natural fractures,and effective activation and utilization of these weak surfaces in reservoirs can significantly improve the extraction effect.In hydraulic fracturing,when the artificial fracture approaches the natural fracture,the natural fracture would be influenced by both the original in-situ stress field and the hydraulic fracturing-induced stress field.In this paper,the hydraulic fracturing-induced stress field is calculated based on the relative position of hydraulic fracture and natural fracture,the original in-situ stress,the net pressure inside the hydraulic fracture and the pore pressure of the formation.Furthermore,the stability model of the natural fracture is established by combining the Mohr-Coulomb rupture criterion,and extensive parametric studies are conducted to explore the impact of each parameter on the stability of the natural fracture.The validity of the proposed model is verified by comparing with the reservoir characteristics and fracturing process of the X-well 150e155 formation in the Songliao Basin.It is found that the stress field induced by the hydraulic fracture inhibits the activation of the natural fracture after the artificial fracture crossed the natural fracture.Therefore,for similar reservoirs as X-well 150e155,it is suggested to connect natural fractures with hydraulic fractures first and then activate natural fractures which can effectively utilize the natural fractures and form a complex fracture network.展开更多

At present, great attention has been placed on oxidative stress. However,little is known about fighting oxidative stress. In the current study, we evaluated antioxidant-induced cochlear damage by applying several different additional antioxidants. To determine whether excessive antioxidants can cause damage to cochlear cells, we treated cochlear explants with 50 m M M40403, a superoxide dismutase mimetic, 50 m M coenzyme Q-10, a vitamin-like antioxidant, or 50 m M d-methionine, an essential amino acid and the important antioxidant glutathione for 48 h. Control cochlear explants without the antioxidant treatment maintained their normal structures after incubation in the standard serum-free medium for 48 h, indicating the maintenance of the inherent oxidative and antioxidant balance in these cochlear explants. In contrast, M40403 and coenzyme Q-10-treated cochlear explants displayed significant hair cell damage together with slight damage to the auditory nerve fibers.Moreover, d-methiodine-treated explants exhibited severe damage to the surface structure of hair cells and the complete loss of the spiral ganglion neurons and their peripheral fibers. These results indicate that excessive antioxidants are detrimental to cochlear cells, suggesting that inappropriate dosages of antioxidant treatments can interrupt the balance of the inherent oxidative and antioxidant capacity in the cell.展开更多

The simulation results show that the SILC is formed by trap-assisted tunnelling (TAT) process which is dominated by oxide traps induced by high field stresses. Their energy levels obtained by this work are approximately 1.9eV from the oxide conduction band, and the traps are believed to be the oxygen-related donor-like defects induced by high field stresses. There is abundant evidence that the ROCK pathway participates in the pathogenesis of retinal endothelial injury and proliferative epiretinal membrane traction. In this study, we investigated the effect of the ROCK pathway inhibitor Y-27632 on retinal Müller cells subjected to hypoxia or oxidative stress. Müller cells were subjected to hypoxia or oxidative stress by exposure to CoCl2 or H2O2. After a 24-hour treatment with Y-27632, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay was used to assess the survival of Müller cells. Hoechst 33258 was used to detect apoptosis, while 2′,7′-dichlorodihydrofluorescein diacetate was used to measure reactive oxygen species generation. A transwell chamber system was used to examine the migration ability of Müller cells. Western blot assay was used to detect the expression levels of α-smooth muscle actin, glutamine synthetase and vimentin. After treatment with Y-27632, Müller cells subjected to hypoxia or oxidative stress exhibited a morphology similar to control cells. Y-27632 reduced apoptosis, α-smooth muscle actin expression and reactive oxygen species generation under oxidative stress, and it reduced cell migration under hypoxia. Y-27632 also upregulated glutamine synthetase expression under hypoxia but did not impact vimentin expression. These findings suggest that Y-27632 protects Müller cells against cellular injury caused by oxidative stress and hypoxia by inhibiting the ROCK pathway.展开更多

In this paper, the charac...The entry at Zhangcun coal mine in Lu'an coal mining area in Shanxi Province suffered from severe mining-induced stresses with the heading face driven oppositely to an adjacent working face. In this paper, the characteristics of deformation and failure of the entry were investigated in terms of the tempo-spatial relations between heading and working faces through field study and numerical modeling. The three-dimensional (3D) finite difference models were built to investigate stresses, displacements and damages in the surrounding rocks of the entry and the working face. The field study includes selection of reinforcing methods and materials, design parameters, and determination of cable prestress. The monitoring data of entry deformation and stress along the cables during every stage were presented. The state of the reinforced entry was evaluated based on the monitoring data. The results demonstrate that before the heading face of the entry crosses the adjacent working face, the influence of advanced abutment pressure caused by adjacent working face upon the entry is not significant. After they cross each other, however, the lateral abutment pressure will have an evident impact on the entry. The displacement rate of the entry will be greatly increased and reaches a certain value within a certain distance between the heading face and the working face. Then, it will increase again with the presence of secondary mining-induced pressure on the entry when the present working face advances. The fully-grouted cable with short length, high strength and high prestress is an effective way to reinforce the entry suffering from severe mining-induced stresses, which greatly reduces the displacement and failure possibility of the entry. Finally, the principles and recommendations for reinforcing design of entries suffering from severe mining-induced stresses were proposed according to field study, numerical modeling and experiences from other coal mines. Problems encountered in field study and suggestions for reinforcement were also discussed.展开更多

Met...Objective To investigate the anti-inflammatory, antioxidant and anti-arthritic effects of Centello asiatica methanolfraction (CAME) on collagen-induced arthritis (ClA), an animal model of rheumatoid arthritis. Methods Arthritis was induced in female wistar rats by immunization with porcine type II collagen. The CIA rats were treated orally with CaME (50, 150, and 250 mg/kg/day) for 15 d (beginning on day 21 of the experimental period). The clinical, histological, biochemical, and immunological parameters were assessed. Results CaME treatment (150 and 250 mg/kg) significantly attenuated the severity of CIA and reduced the synovial inflammation, cartilage erosion, and bone erosion as evident from both histological and radiographic data. The escalated plasma levels of pro-inflammatory cytokines TNF-α, IL-1β, IL-6, and IL-12 alongwith nitric oxide in CIA rats decreased significantly on CaME treatment. The serum levels of type-Ⅱ collagen antibody were significantly lower in rats of CaME (150 and 250 mg/kg) treated group than those in the arthritic group. Furthermore, by inhibiting the above mediators, CaME also contributed towards the reversal of the disturbed antioxidant levels and peroxidative damage. Conclusion Our results clearly indicate that oral administration of CaME suppresses joint inflammation, cytokine expression as well as antioxidant imbalance, thereby contributing to an amelioration of arthritis severity in CIA rats.展开更多

Through theoretical analysis and experimental observations, it was found that the stress-induced martensitic transformation process of single crystal Cu-based SMA under uniaxial tension condition consisted of three periods: nucleation, mixed nucleation and growth, and merging due to growth. During the nucleation, the stress dropped rapidly and the number of interfaces increased very fast while the phase fraction increased slowly. In the second period, both the stress and the interface number changed slightly but the phase fraction increased dramatically. Finally, the stress and the phase fraction changed slowly while the number of interfaces decreased quickly. Moreover, it was found that the transformation could be of multi-stage: sharp stress drops at several strains and correspondingly, the nucleation and growth process occurred quasi-independently in several parts of the sample.展开更多

Restraint water-immersion stress(RWIS) can cause serious gastrointestinal dysfunction and has been widely used to study the pathogenesis of SGU to identify medications that can cure the disease. The mediodorsal thalamic nucleus(MD) is the centre integrating visceral and physical activity and contributes to SGU induced by RWIS. Hence, the role of the MD during RWIS needs to be studied.AIM To screen for differentially expressed proteins in the MD of the RWIS rats to further elucidate molecular mechanisms of SGU.METHODS Male Wistar rats were selected randomly and divided into two groups, namely, a control group and an RWIS group. Gastric mucosal lesions of the sacrificed rats were measured using the erosion index and the proteomic profiles of the MD were generated through isobaric tags for relative and absolute quantitation(iTRAQ) coupled with two-dimensional liquid chromatography and tandem mass spectrometry. Additionally, iTRAQ results were verified by Western blot analysis.RESULTS A total of 2853 proteins were identified, and these included 65 dysregulated(31 upregulated and 34 downregulated) proteins(fold change ratio ≥ 1.2). Gene Ontology(GO) analysis showed that most of the upregulated proteins are primarily related to cell division, whereas most of the downregulated proteins are related to neuron morphogenesis and neurotransmitter regulation. Ingenuity Pathway Analysis revealed that the dysregulated proteins are mainly involved in the neurological disease signalling pathways. Furthermore, our results indicated that glycogen synthase kinase-3 beta might be related to the central mechanismthrough which RWIS gives rise to SGU.CONCLUSION Quantitative proteomic analysis elucidated the molecular targets associated with the production of SGU and provides insights into the role of the MD. The underlying molecular mechanisms need to be further dissected.展开更多

Methods Seventy women with PIH and seventy women with uncomplicated normotensive pregnancy (UNP) whose age, nutritional conditions, levels of hemoglobin and albumin were all matched, were enrolled in a randomized controlled trial. Their plasma concentrations of nitric oxide (NO), vitamin C (VC), vitamin E (VE), and β-carotene (β-CAR) as well as their erythrocyte malondialdehyde (MDA), and activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) were determined by spcctrophotometry. Results Compared with average values of the above experimental parameters in the women with UNP, the average value of erythrocyte MDA in the women with PIH significantly increased (P〈0.0001), while the average values of plasma NO, VC, VE, and β-CAR as well as those of erythrocyte SOD, CAT, and GPX in the women with PIH significantly decreased (P〈0.0005-0.0001). The findings from partial correlation analysis (controlling for age) for 70 women with PIH showed that with elevated systolic blood pressure (SBP) and diastolic blood pressure (DBP), MDA value gradually increased (P〈0.001), and NO, VC, VE, β-CAR, SOD, CAT, and GPX values gradually decreased (P〈0.02-0.001). The findings from reliability analysis for NO, VC, VE, β-CAR, SOD, CAT, GPX, and MDA values used to reflect increased oxidative stress and potential free radical damage in women with PIH showed that the reliability coefficients (alpha, 8 items) = 0.7062, P〈 0.0001, and the standardized item alpha = 0.9116, P〈 0.0001. Conclusion The findings in the present research suggest that pregnancy-induced hypertension can increase oxidative stress and potential free radical damage in women with pregnancy-induced hypertension.展开更多

文摘Boulder spacing in mountain rivers and near-wake flow zones within the boulder array is very useful for fish habitat and growth of aquatic organisms.The present study aims to investigate how the boulder array and spacing influence the near-bed flow structures in a gravel-bed stream.Boulders are staggered over a gravel-bed stream with three different inter-boulder spacing namely(a)large(b)medium and(c)small spacing.An acoustic Doppler velocimeter was used for flow measurements in a rectangular channel and the results were compared with those acquired from numerical simulation.The time-averaged velocity profiles at the near-wake flow zones of boulders experience maximum flow retardation which is an outcome of the boulder-induced form roughness.The ratio of velocity differences associated to form and skin roughness and its positive magnitude reveals the dominance of form roughness closest to the boulders.Form roughness computed is 1.75 to 2 times higher than the skin roughness at the near-wake flow region.In particular,the collective immobile boulders placed at different inter-boulder spacings developed high and low bed shear stresses closest to the boulders.The low bed shear stresses characterised by a secondary peak developed at the trough location of the boulders is attributed to the skin shear stress.Further,the spatial averaging of time-averaged flow quantities gives additional impetus to present an improved illustration of fluid shear stresses.The formation of form-induced shear stress is estimated to be 17%to 23%of doubleaveraged Reynolds shear stress and partially compensates for the damping of time-averaged Reynolds shear stress in the interfacial sub-layer.The quadrant analysis of spatial velocity fluctuations depicts that the form-induced shear stresses are dominant in the interfacial sub-layer and have no significance above the gravel-bed surface.
文摘Several cardiac outcomes have been reported with West Nile-encephalitis;however, the underlying pathophysiology remains complex. We present a 42-year-old female, with multiple sclerosis, whose neurological symptoms and respiratory decline were finally explained by the diagnosis of West Nile-encephalitis. During her admission, the isolated peaked T-waves indicated the underlying stress-induced cardiomyopathy. The absence of all other causes of hyperacute T-waves, their subsequent resolution with the resolution of infection and improvement in wall motion abnormalities, further supported the association. This case highlights the importance of considering hyperacute T-waves in an approach towards the diagnosis of WNV-encephalitis related atypical variant of stress-induced cardiomyopathy.

文摘Evaluating the physical mechanisms that link hydraulic fracturing(HF) operations to induced earthquakes and the anticipated form of the resulting events is significant in informing subsurface fluid injection operations. Current understanding supports the overriding role of the effective stress magnitude in triggering earthquakes, while the impact of change rate of effective stress has not been systematically addressed. In this work, a modified critical stiffness was brought up to investigate the likelihood, impact,and mitigation of induced seismicity during and after hydraulic fracturing by developing a poroelastic model based on rate-and-state fraction law and linear stability analysis. In the new criterion, the change rate of effective stress was considered a key variable to explore the evolution of this criterion and hence the likelihood of instability slip of fault. A coupled fluid flow-deformation model was used to represent the entire hydraulic fracturing process in COMSOL Multiphysics. The possibility of triggering an earthquake throughout the entire hydraulic fracturing process, from fracturing to cessation, was investigated considering different fault locations, orientations, and positions along the fault. The competition between the effects of the magnitude and change rate of effective stress was notable at each fracturing stage. The effective stress magnitude is a significant controlling factor during fracturing events, with the change rate dominating when fracturing is suddenly started or stopped. Instability dominates when the magnitude of the effective stress increases(constant injection at each fracturing stage) and the change rate of effective stress decreases(the injection process is suddenly stopped). Fracturing with a high injection rate, a fault adjacent to the hydraulic fracturing location and the position of the junction between the reservoir and fault are important to reduce the Coulomb failure stress(CFS) and enhance the critical stiffness as the significant disturbance of stresses at these positions in the coupled process. Therefore,notable attention should be given to the injection rate during fracturing, fault position, and position along faults as important considerations to help reduce the potential for induced seismicity. Our model was verified and confirmed using the case of the Longmaxi Formation in the Sichuan Basin, China, in which the reported microseismic data were correlated with high critical stiffness values. This work supplies new thoughts of the seismic risk associated with HF engineering.
文摘DI-3-n-butylphthalide is used to treat mild and moderate acute ischemic stroke.However,the precise underlying mechanism requires further investigation.In this study,we investigated the molecular mechanism of DI-3-n-butylphthalide action by various means.We used hydrogen peroxide to induce injury to PC12cells and RAW264.7 cells to mimic neuronal oxidative stress injury in stroke in vitro and examined the effects of DI-3-n-butylphthalide.We found that DI-3-nbutylphthalide pretreatment markedly inhibited the reduction in viability and reactive oxygen species production in PC12 cells caused by hydrogen peroxide and inhibited cell apoptosis.Furthermore,DI-3-n-butylphthalide pretreatment inhibited the expression of the pro-apoptotic genes Bax and Bnip3.DI-3-nbutylphthalide also promoted ubiquitination and degradation of hypoxia inducible factor 1α,the key transcription factor that regulates Bax and Bnip3 genes.These findings suggest that DI-3-n-butylphthalide exhibits a neuroprotective effect on stroke by promoting hypoxia inducible factor-1α ubiquitination and degradation and inhibiting cell apoptosis.

文摘Unconventional reservoirs usually contain many weak surfaces such as faults,laminae and natural fractures,and effective activation and utilization of these weak surfaces in reservoirs can significantly improve the extraction effect.In hydraulic fracturing,when the artificial fracture approaches the natural fracture,the natural fracture would be influenced by both the original in-situ stress field and the hydraulic fracturing-induced stress field.In this paper,the hydraulic fracturing-induced stress field is calculated based on the relative position of hydraulic fracture and natural fracture,the original in-situ stress,the net pressure inside the hydraulic fracture and the pore pressure of the formation.Furthermore,the stability model of the natural fracture is established by combining the Mohr-Coulomb rupture criterion,and extensive parametric studies are conducted to explore the impact of each parameter on the stability of the natural fracture.The validity of the proposed model is verified by comparing with the reservoir characteristics and fracturing process of the X-well 150e155 formation in the Songliao Basin.It is found that the stress field induced by the hydraulic fracture inhibits the activation of the natural fracture after the artificial fracture crossed the natural fracture.Therefore,for similar reservoirs as X-well 150e155,it is suggested to connect natural fractures with hydraulic fractures first and then activate natural fractures which can effectively utilize the natural fractures and form a complex fracture network.

文摘The imbalance of reactive oxygen species and antioxidants is considered to be an important factor in the cellular injury of the inner ear. At present, great attention has been placed on oxidative stress. However,little is known about fighting oxidative stress. In the current study, we evaluated antioxidant-induced cochlear damage by applying several different additional antioxidants. To determine whether excessive antioxidants can cause damage to cochlear cells, we treated cochlear explants with 50 m M M40403, a superoxide dismutase mimetic, 50 m M coenzyme Q-10, a vitamin-like antioxidant, or 50 m M d-methionine, an essential amino acid and the important antioxidant glutathione for 48 h. Control cochlear explants without the antioxidant treatment maintained their normal structures after incubation in the standard serum-free medium for 48 h, indicating the maintenance of the inherent oxidative and antioxidant balance in these cochlear explants. In contrast, M40403 and coenzyme Q-10-treated cochlear explants displayed significant hair cell damage together with slight damage to the auditory nerve fibers.Moreover, d-methiodine-treated explants exhibited severe damage to the surface structure of hair cells and the complete loss of the spiral ganglion neurons and their peripheral fibers. These results indicate that excessive antioxidants are detrimental to cochlear cells, suggesting that inappropriate dosages of antioxidant treatments can interrupt the balance of the inherent oxidative and antioxidant capacity in the cell.
文摘The conduction mechanism of stress induced leakage current (SILC) through 2nm gate oxide is studied over a gate voltage range between 1.7V and stress voltage under constant voltage stress (CVS). The simulation results show that the SILC is formed by trap-assisted tunnelling (TAT) process which is dominated by oxide traps induced by high field stresses. Their energy levels obtained by this work are approximately 1.9eV from the oxide conduction band, and the traps are believed to be the oxygen-related donor-like defects induced by high field stresses. The dependence of the trap density on stress time and oxide electric field is also investigated.

文摘Rho kinase (ROCK) was the first downstream Rho effector found to mediate RhoA-induced actin cytoskeletal changes through effects on myosin light chain phosphorylation. There is abundant evidence that the ROCK pathway participates in the pathogenesis of retinal endothelial injury and proliferative epiretinal membrane traction. In this study, we investigated the effect of the ROCK pathway inhibitor Y-27632 on retinal Müller cells subjected to hypoxia or oxidative stress. Müller cells were subjected to hypoxia or oxidative stress by exposure to CoCl2 or H2O2. After a 24-hour treatment with Y-27632, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay was used to assess the survival of Müller cells. Hoechst 33258 was used to detect apoptosis, while 2′,7′-dichlorodihydrofluorescein diacetate was used to measure reactive oxygen species generation. A transwell chamber system was used to examine the migration ability of Müller cells. Western blot assay was used to detect the expression levels of α-smooth muscle actin, glutamine synthetase and vimentin. After treatment with Y-27632, Müller cells subjected to hypoxia or oxidative stress exhibited a morphology similar to control cells. Y-27632 reduced apoptosis, α-smooth muscle actin expression and reactive oxygen species generation under oxidative stress, and it reduced cell migration under hypoxia. Y-27632 also upregulated glutamine synthetase expression under hypoxia but did not impact vimentin expression. These findings suggest that Y-27632 protects Müller cells against cellular injury caused by oxidative stress and hypoxia by inhibiting the ROCK pathway.

文摘The entry at Zhangcun coal mine in Lu'an coal mining area in Shanxi Province suffered from severe mining-induced stresses with the heading face driven oppositely to an adjacent working face. In this paper, the characteristics of deformation and failure of the entry were investigated in terms of the tempo-spatial relations between heading and working faces through field study and numerical modeling. The three-dimensional (3D) finite difference models were built to investigate stresses, displacements and damages in the surrounding rocks of the entry and the working face. The field study includes selection of reinforcing methods and materials, design parameters, and determination of cable prestress. The monitoring data of entry deformation and stress along the cables during every stage were presented. The state of the reinforced entry was evaluated based on the monitoring data. The results demonstrate that before the heading face of the entry crosses the adjacent working face, the influence of advanced abutment pressure caused by adjacent working face upon the entry is not significant. After they cross each other, however, the lateral abutment pressure will have an evident impact on the entry. The displacement rate of the entry will be greatly increased and reaches a certain value within a certain distance between the heading face and the working face. Then, it will increase again with the presence of secondary mining-induced pressure on the entry when the present working face advances. The fully-grouted cable with short length, high strength and high prestress is an effective way to reinforce the entry suffering from severe mining-induced stresses, which greatly reduces the displacement and failure possibility of the entry. Finally, the principles and recommendations for reinforcing design of entries suffering from severe mining-induced stresses were proposed according to field study, numerical modeling and experiences from other coal mines. Problems encountered in field study and suggestions for reinforcement were also discussed.

文摘Objective To investigate the anti-inflammatory, antioxidant and anti-arthritic effects of Centello asiatica methanolfraction (CAME) on collagen-induced arthritis (ClA), an animal model of rheumatoid arthritis. Methods Arthritis was induced in female wistar rats by immunization with porcine type II collagen. The CIA rats were treated orally with CaME (50, 150, and 250 mg/kg/day) for 15 d (beginning on day 21 of the experimental period). The clinical, histological, biochemical, and immunological parameters were assessed. Results CaME treatment (150 and 250 mg/kg) significantly attenuated the severity of CIA and reduced the synovial inflammation, cartilage erosion, and bone erosion as evident from both histological and radiographic data. The escalated plasma levels of pro-inflammatory cytokines TNF-α, IL-1β, IL-6, and IL-12 alongwith nitric oxide in CIA rats decreased significantly on CaME treatment. The serum levels of type-Ⅱ collagen antibody were significantly lower in rats of CaME (150 and 250 mg/kg) treated group than those in the arthritic group. Furthermore, by inhibiting the above mediators, CaME also contributed towards the reversal of the disturbed antioxidant levels and peroxidative damage. Conclusion Our results clearly indicate that oral administration of CaME suppresses joint inflammation, cytokine expression as well as antioxidant imbalance, thereby contributing to an amelioration of arthritis severity in CIA rats.

文摘The mechanical behavior of shape memory alloys (SMAs) is closely related to the formation and evolution of its microstructures. Through theoretical analysis and experimental observations, it was found that the stress-induced martensitic transformation process of single crystal Cu-based SMA under uniaxial tension condition consisted of three periods: nucleation, mixed nucleation and growth, and merging due to growth. During the nucleation, the stress dropped rapidly and the number of interfaces increased very fast while the phase fraction increased slowly. In the second period, both the stress and the interface number changed slightly but the phase fraction increased dramatically. Finally, the stress and the phase fraction changed slowly while the number of interfaces decreased quickly. Moreover, it was found that the transformation could be of multi-stage: sharp stress drops at several strains and correspondingly, the nucleation and growth process occurred quasi-independently in several parts of the sample.

文摘In this study, the spatial distributions of stress and fracture fields for three typical underground coal mining layouts, Le, non-pillar mining (NM), top-coal caving mining (TCM) and protective coal-seam mining (PCM), are modeled using discrete element software UDEC, The numerical results show that different mining layouts can lead to different mining-induced stress fields, resulting in diverse fracture fields, For the PCM, the mining influenced area in front of the mining faces is the largest, and the stress concentration factor in front of the mining faces is the lowest, The spatial shapes of the mining-induced fracture fields under NM, TCM and PCM differ, and they are characterized by trapezoidal, triangular and tower shapes, respectively, The fractal dimensions of mining-induced fractures of the three mining layouts decrease in the order of PCM, TCM and NM, It is also shown that the PCM can result in a better gas control effect in coal mines with high outburst potential, The numerical results are expected to provide a basis for understanding of mining-induced gas seepage fields and provide a reference for high- efficiency coal mining,

文摘BACKGROUND Stress-induced gastric ulcer(SGU) is one of the most common visceral complications after trauma. Restraint water-immersion stress(RWIS) can cause serious gastrointestinal dysfunction and has been widely used to study the pathogenesis of SGU to identify medications that can cure the disease. The mediodorsal thalamic nucleus(MD) is the centre integrating visceral and physical activity and contributes to SGU induced by RWIS. Hence, the role of the MD during RWIS needs to be studied.AIM To screen for differentially expressed proteins in the MD of the RWIS rats to further elucidate molecular mechanisms of SGU.METHODS Male Wistar rats were selected randomly and divided into two groups, namely, a control group and an RWIS group. Gastric mucosal lesions of the sacrificed rats were measured using the erosion index and the proteomic profiles of the MD were generated through isobaric tags for relative and absolute quantitation(iTRAQ) coupled with two-dimensional liquid chromatography and tandem mass spectrometry. Additionally, iTRAQ results were verified by Western blot analysis.RESULTS A total of 2853 proteins were identified, and these included 65 dysregulated(31 upregulated and 34 downregulated) proteins(fold change ratio ≥ 1.2). Gene Ontology(GO) analysis showed that most of the upregulated proteins are primarily related to cell division, whereas most of the downregulated proteins are related to neuron morphogenesis and neurotransmitter regulation. Ingenuity Pathway Analysis revealed that the dysregulated proteins are mainly involved in the neurological disease signalling pathways. Furthermore, our results indicated that glycogen synthase kinase-3 beta might be related to the central mechanismthrough which RWIS gives rise to SGU.CONCLUSION Quantitative proteomic analysis elucidated the molecular targets associated with the production of SGU and provides insights into the role of the MD. The underlying molecular mechanisms need to be further dissected.
文摘Objective To investigate whether pregnancy-induced hypertension (PIH) may increase oxidative stress in women with PIH, and to explore the mechanisms by which PIH may increase oxidative stress and potential free radical damage. Methods Seventy women with PIH and seventy women with uncomplicated normotensive pregnancy (UNP) whose age, nutritional conditions, levels of hemoglobin and albumin were all matched, were enrolled in a randomized controlled trial. Their plasma concentrations of nitric oxide (NO), vitamin C (VC), vitamin E (VE), and β-carotene (β-CAR) as well as their erythrocyte malondialdehyde (MDA), and activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) were determined by spcctrophotometry. Results Compared with average values of the above experimental parameters in the women with UNP, the average value of erythrocyte MDA in the women with PIH significantly increased (P〈0.0001), while the average values of plasma NO, VC, VE, and β-CAR as well as those of erythrocyte SOD, CAT, and GPX in the women with PIH significantly decreased (P〈0.0005-0.0001). The findings from partial correlation analysis (controlling for age) for 70 women with PIH showed that with elevated systolic blood pressure (SBP) and diastolic blood pressure (DBP), MDA value gradually increased (P〈0.001), and NO, VC, VE, β-CAR, SOD, CAT, and GPX values gradually decreased (P〈0.02-0.001). The findings from reliability analysis for NO, VC, VE, β-CAR, SOD, CAT, GPX, and MDA values used to reflect increased oxidative stress and potential free radical damage in women with PIH showed that the reliability coefficients (alpha, 8 items) = 0.7062, P〈 0.0001, and the standardized item alpha = 0.9116, P〈 0.0001. Conclusion The findings in the present research suggest that pregnancy-induced hypertension can increase oxidative stress and potential free radical damage in women with pregnancy-induced hypertension.

文摘The passive film-induced stress and the susceptibility to SCC of 7050 aluminum alloy in 3.5%sodium chloride solution at various pH values were investigated by slow strain rate testing(SSRT) and flowing stress differential method.The results showed that the passive film-induced stress and the susceptibility to SCC decreased with increasing pH values when pH≤7,while they increased with increasing pH values when pH7.However,the corrosion type was interpreted as exfoliation corrosion when pH=l and 14,and there was no film formed on the surface of the specimens.The whole variation plots of film-induced stress and the SCC susceptibility with pH values were both presented as a valley shape.The symbol and amount of the film-induced stress were related to the compositions of the passive film,which were analyzed using X-ray photoelectron spectroscopy(XPS).

文摘When ocean waves propagate over the sea floor,dynamic wave pressures and bottom shear stresses exert on the surface of seabed.The bottom shear stresses provide a horizontal loading in the wave-seabed interaction system,while dynamic wave pressures provide a vertical loading in the system.However,the bottom shear stresses have been ignored in most previous studies in the past.In this study,the effects of the bottom shear stresses on the dynamic response in a seabed of finite thickness under wave loading will be examined,based on Biot's dynamic poro-elastic theory.In the model,an "u-p" approximation will be adopted instead of quasi-static model that have been used in most previous studies.Numerical results indicate that the bottom shear stresses has certain influences on the wave-induced seabed dynamic response.Furthermore,wave and soil characteristics have considerable influences on the relative difference of seabed response between the previous model(without shear stresses) and the present model(with shear stresses).As shown in the parametric study,the relative differences between two models could up to 10% of p0,depending on the amplitude of bottom shear stresses.
文摘Stacked chip scale package(SCSP) attracts more and more attentions in advanced packages application with light weight,thin and small size,high reliability,low power and high storage capability.However,more and more physical and electrical issues being caused by package-induced stress in SCSP were reported recently.The effect of structural factors,including die thickness,die attach film thickness,die attach film type,and spacer size on package induced stress,was investigated.Analyses were given based on simulation results and provide important suggestion for package design.

文摘The susceptibility to stress corrosion cracking (SCC) of brass in an ammoniasolution with various pH values or under various applied potentials was measured at slow strain ratetests. The additive stress in the same solution was measured using two methods. The resultsindicate that the variation of the susceptibility to SCC with pH value or with potential is in anexcellent agreement with the corrosion (passive film or dezincification layer)-induced stress. WhenpH >= 7, the corrosion-induced tensile stress and the susceptibility to SCC have maximum values andhardly change with increasing the pH value. However, when pH <7, both the corrosion-induced tensilestress and the susceptibility to SCC reduce rapidly with decreasing the pH value. Both thecorrosion-induced tensile stress and the susceptibility to SCC have maximum values at theopen-circuit potential, decrease slightly under the anodic polarization, and reduce gradually tozero under the cathodic polarization.
