Although Nudix hydrolases are involved in plant detoxification processes in response to abiotic and biotic stresses, the biological functions of Nudix hydrolases remain largely unclear in grapevine. In the present study, a total of 25 putative grapevine Nudix hydrolases(VvNUDXs) were identified by bioinformatics analysis and classified into eight subfamilies based to their preferred substrates. Both tandem and segmental duplications were responsible for the evolution and expansion of the NUDX gene family in grapevine. To investigate the regulatory roles of VvNUDX genes during growth and development, as well as in response to abiotic and biotic stresses in grapevine, the expression patterns were revealed in publicly available microarray data. The spatial and temporal expression patterns of the VvNUDX genes indicated that they might play important roles in multiple developmental processes. Transcriptome and qRT-PCR analyses showed that ten VvNUDX genes were specifically expressed in grapevine berries, suggesting potential roles in grapevine berry development. Expression and phylogenetic analyses demonstrated that VvNUDX1 and VvNUDX3 might be involved in terpenoid biosynthesis in grapevine. Furthermore, most VvNUDX genes active toward the ADP-ribose/NADH showed different patterns in response to various abiotic and biotic stresses, such as salinity and drought, as well as different types of biotic treatments, such as Erysiphe necator,Bois Noir phytoplasma and leaf-roll-associated virus-3(GLRaV-3). These results indicated that VvNUDX genes were associated with plant detoxification processes in response to abiotic and biotic stresses, and regulate the disease immunity and resistance pathways. The information obtained here may provide good opportunities to explore the physiological functions of VvNUDX genes in berry development and stress response networks in grapevine.展开更多

However, species may exhibit stress response variation depending on their specific environmental and selection pressures. Blind cavefish dwell in cave environments, which differ markedly in stressors and resource availability compared to surface aquatic environments. However, whether blind cavefish exhibit differences in stress response as an adaptation to their cave environments remains unclear. Here, we investigated differences in stress response in six closely related Triplophysa species, including three blind cavefish (T. longibarbata, T. jiarongensis, and T. rosa) and three normal-sighted river fish (T. nasobarbatula, T. dongsaiensis, and T. bleekeri). Results showed that blind cavefish exhibited a range of distinct behavioral responses compared to sighted river fish, including greater levels of activity, shorter duration of freezing, absence of erratic movements or thrashing behavior, and opposite behavioral trends over time. Furthermore, the cavefish species demonstrated attenuated increases in metabolic rate in response to stressors related to novel environments. Cave-dwelling T. rosa also exhibited lower basal hypothalamic-pituitary-inter-renal (HPI) axis-related gene expression levels and stress hormone concentrations compared to river-dwelling T. bleekeri. These results suggest that blind cavefish may have lost their behavioral stress response, potentially mediated by a reduction in basal activity of the HPI axis, thus enabling the conservation of energy by reducing unnecessary expenditure in energy-limited caves.展开更多

As a representative of the wine bacterium with remarkable adaptability to the very harsh physicochemical conditions of wine, many studies were carded out for its applied interest and focused mainly on its stress response mechanisms of O. oeni. on both physiological and molecular levels. In this review, three main stress response mechanisms in O. oeni during culturing process were addressed. Of them, various solute transporters and secondary metabolic energy-generating systems were utilized to control the intracellular environment and the energetic status of O. oeni. The changes in cell membrane fatty acid composition profiles and synthesis of stress proteins, especially small heat shock proteins were required for active cell response to maintain membrane integrity and function under stress conditions. The study on stress response of O. oeni played an important role on culture bacteria selection, making inoculation culture and construction of other engineering bacteria.展开更多

We hypothesized that edaravone, a low molecular weight, lipophilic free radical scavenger, would reduce OGSD/R-induced apoptosis of spinal cord astrocytes. To test this, we established primary cultures of rat astrocytes, and exposed them to 8 hours/6 hours of OGSD/R with or without edaravone(0.1, 1, 10, 100 μM) treatment. We found that 100 μM of edaravone significantly suppressed astrocyte apoptosis and inhibited the release of reactive oxygen species. It also inhibited the activation of caspase-12 and caspase-3, and reduced the expression of homologous CCAAT/enhancer binding protein, phosphorylated(p)-PERK, p-eIF2α, and ATF4. These results point to a new use of an established drug in the prevention of OGSD/R-mediated spinal cord astrocyte apoptosis via the integrated stress response.展开更多

Physiological mechanisms that underlie traits for salt tolerance plantlets have been used to test some wheat species and to identify sources of salty tolerance. Stress conditions were founded by means of exposure of the plantlets in NaCI water solution, 1.68% that demonstrably differentiates the samples by growth parameters. Under laboratory conditions the ability of seedlings to bear a significant shortage of water, their ability to accumulate biomass, changing the parameters of linear growth processes and changes in water content in leaves and roots of seedlings under stress was determined. Interest is also the rate of growth of root fibrils in saline conditions. These experiments allowed to provide a comprehensive diagnosis of studied forms of wheat and make an initial opinion on their salt tolerance. Different features of abiotic stress responses of some wheat species at the early stages of development were shown that testifies necessi~ of great number of approaches at selection of genetic material for improvement of modern wheat eultivars. This work is the first phase of research for practical breeding.展开更多

The model extends the Quemeda model to describe the viscosity function with a structural parameter λ which varies according to differ- ent kinetic orders of particle aggregation and segregation. The transient stress response is obtained by solving an instantaneous Maxwell model with an assumed shear modulus function G of the same form as the viscosity function η. Accuracy of the proposed model is verified experimentally with the results obtained for two oil (creosote)/water emulsions. The model that gives the best fit of experimental data appears to be the one with kinetic orders n = m = 2.展开更多

Cells can initiate either protective ...Cellular stress responses are powerful mechanisms that prevent and cope with the accumulation of macromolecular damage in the cells and also boost host defenses against pathogens. Cells can initiate either protective or destructive stress responses depending, to a large extent, on the nature and duration of the stressing stimulus as well as the cell type. The productive replication of a virus within a given cell places inordinate stress on the metabolism machinery of the host and, to assure the continuity of its replication, many viruses have developed ways to modulate the cell stress responses. Poxviruses are among the viruses that have evolved a large number of strategies to manipulate host stress responses in order to control cell fate and enhance their replicative success. Remarkably, nearly every step of the stress responses that is mounted during infection can be targeted by virally encoded functions. The fine-tuned interactions between poxviruses and the host stress responses has aided virologists to understand specific aspects of viral replication; has helped cell biologists to evaluate the role of stress signaling in the uninfected cell; and has tipped immunologists on how these signals contribute to alert the cells against pathogen invasionand boost subsequent immune responses. This review discusses the diverse strategies that poxviruses use to subvert host cell stress responses.展开更多

文摘The homeodomain-leucine zipper(HD-Zip)family has been shown to perform amultitude of functions during plant development and stress responses;however, the familymembers and functions have not been identified in apple(Malus×domestica). In this study,83 HD-Zips(MdHDZs)were identified in the apple genome. They were assembled into four subgroups according to the classification in Arabidopsis, where MdHDZs in the same subgroup had similar gene structures and conserved protein motifs.Putative cis-element analysis of MdHDZs promoter regions uncovered numerous elements related to the response of stress and plant hormones. In addition, twelve transcripts of the MdHDZs showed different expression patterns under salt, drought, low temperature and ABA stresses by quantitative reverse transcription-PCR(qRT-PCR)assay.To further explore the function of MdHDZs in apple,MdHDZ3 was selected to verify its function under salt, low temperature and ABA stresses;and genetic transformation was used to obtain MdHDZ3 transgenic apple calli. The results demonstrated that MdHDZ3 increased sensitivity to salt, low temperature and abscisic acid in apple calli, suggesting that MdHDZ3 plays an important role in response to stresses.Subcellular localization and three-dimensional structural analysis revealed that MdHDZ3 was a nuclear-localized protein.Taken together, these findings provide potential information for further identification of HD-Zip proteins in apple.

文摘BACKGROUND Radical gastrectomy(RG)is commonly used in the treatment of patients with gastric cancer(GC),but this procedure may lead to stress responses, postoperative cognitive dysfunction, and blood coagulation abnormalities in patients.AIM To investigate the influences of dexmedetomidine(DEX)on stress responses and postoperative cognitive and coagulation functions in patients undergoing RG under general anesthesia(GA).METHODS One hundred and two patients undergoing RG for GC under GA from February 2020 to February 2022 were retrospectively reviewed.Of these,50 patients had received conventional anesthesia intervention[control group(CG)]and 52 patients had received DEX in addition to routine anesthesia intervention[observation group(OG)].Inflammatory factor(IFs;tumor necrosis factor-α,TNF-α;interleukin-6,IL-6),stress responses(cortisol,Cor;adrenocorticotropic hormone,ACTH),cognitive function(CF;Mini-Mental State Examination,MMSE),neurological function(neuron-specific enolase,NSE;S100 calciumbinding protein B,S100B),and coagulation function(prothrombin time,PT;thromboxane B2,TXB2;fibrinogen,FIB)were compared between the two groups before surgery(T0),as well as at 6 h(T1)and 24 h(T2)after surgery.RESULTS Compared with T0,TNF-α,IL-6,Cor,ACTH,NSE,S100B,PT,TXB2,and FIB showed a significant increase in both groups at T1 and T2,but with even lower levels in OG vs CG.Both groups showed a significant reduction in the MMSE score at T1 and T2 compared with T0,but the MMSE score was notably higher in OG compared with CG.CONCLUSION In addition to a potent inhibitory effect on postoperative IFs and stress responses in GC patients undergoing RG under GA,DEX may also alleviate the coagulation dysfunction and improve the postoperative CF of these patients.

文摘Nudix hydrolases are widely distributed across all classes of organisms and provide the potential capacity to hydrolyze a wide range of organic pyrophosphates. The information obtained here may provide good opportunities to explore the physiological functions of VvNUDX genes in berry development and stress response networks in grapevine.

文摘The stress response is essential for animal self-defense and survival. Cave-dwelling T. rosa also exhibited lower basal hypothalamic-pituitary-inter-renal (HPI) axis-related gene expression levels and stress hormone concentrations compared to river-dwelling T. bleekeri. These results suggest that blind cavefish may have lost their behavioral stress response, potentially mediated by a reduction in basal activity of the HPI axis, thus enabling the conservation of energy by reducing unnecessary expenditure in energy-limited caves.

文摘To investigate the stress response characteristics and shear stress transfer mechanism of BFRP(basalt fiber reinforced plastics)anchors under rainfall conditions and to explore the reinforcement effect of BFRP anchors,a comparative indoor physical model test was conducted in this study using loess mudstone slope as a typical case, and multi-attribute response data, such as slope displacement,BFRP anchor strain and axial force, were obtained.Based on the variation law of slope displacement,it can be concluded that the deformation displacement of the slope on the side reinforced by BFRP anchors is smaller than that of slopes reinforced with steel anchors;the bearing capacity of BFRP anchor is given priority in time, and the anchoring effect of BFRP anchor at the top of the slope is given priority, and the axial force value of anchor is characterized by a surface amplification effect in space;the axial force is higher around the anchor head and tends to decay as a negative exponential function as the anchor extends in the direction of the interior of the slope;the shear stress of BFRP anchor and anchorage body are unevenly distributed along the axial direction, with the maximum value occurring in the free section near the slope surface and decreases as the BFRP anchor extends towards the interior of the slope. These results can provide a theoretical basis for the optimal design of BFRP anchors.

文摘Nutrient and stress factor 1(Nsf1),a transcription factor containing the classical Cys2-His2(C_(2)H_(2))zinc finger motif,is expressed under non-fermentable carbon conditions and in response to salt stress in Saccharomyces cerevisiae. However, the role of Nsf1 in filamentous fungi is not well understood. In this study, the orthologue of Nsf1 was investigated in Fusarium graminearum(named FgNsf1),a causal agent of Fusarium head blight(FHB).The functions of FgNsf1 were evaluated by constructing a FgNSF1 deletion mutant, designated asΔFgNsf1,and its functional complementation mutantΔFgNsf1-C.Gene deletion experiments showed that the mycelial growth rate, asexual and sexual reproduction ofΔFgNsf1 were significantly reduced, but the pigment production ofΔFgNsf1 was remarkably increased compared with the PH-1 andΔFgNsf1-C.In addition, the tolerance ofΔFgNsf1 to osmotic pressures, cell wall-damaging agents and oxidative stress increased significantly.Sensitivity tests to different fungicides revealed thatΔFgNsf1 exhibited increased sensitivity to carbendazim(MBC)and tebuconazole, and enhanced tolerance to fludioxonil and iprodione than PH-1 andΔFgNsf1-C.The virulence ofΔFgNsf1 to wheat coleoptiles and flowering wheat heads were dramatically decreased, which was consistent with the decrease in the yield of deoxynivalenol(DON).All of these defects were restored by target gene complementation. These results indicated that FgNsf1 plays a crucial role in vegetative growth, asexual and sexual reproduction, stress responses, fungicide sensitivity, and full virulence in F.graminearum.

文摘Oenoccoccus oeni is an alcohol-tolerant, acidophilic lactic acid bacterium with its ability to perform malolactic fermentation in wine, which is of fundamental importance in oenology. The study on stress response of O. oeni played an important role on culture bacteria selection, making inoculation culture and construction of other engineering bacteria.

文摘We previously found that oxygen-glucose-serum deprivation/restoration(OGSD/R) induces apoptosis of spinal cord astrocytes, possibly via caspase-12 and the integrated stress response, which involves protein kinase R-like endoplasmic reticulum kinase(PERK), eukaryotic initiation factor 2-alpha(eIF2α) and activating transcription factor 4(ATF4). These results point to a new use of an established drug in the prevention of OGSD/R-mediated spinal cord astrocyte apoptosis via the integrated stress response.
文摘BACKGROUND Radical surgery is a common treatment for patients with gastric cancer;however,it can lead to postoperative complications and intestinal barrier dysfunction.Ultrasound-guided quadratus lumborum block is often used for postoperative analgesia, but its effects on stress response and intestinal barrier function are not well understood.AIM To investigate the effects of an ultrasound-guided quadratus lumborum block on stress response and intestinal barrier function in patients undergoing radical surgery for gastric cancer.METHODS A total of 100 patients undergoing radical surgery for gastric cancer were randomly categorized into observation and control groups.Plasma adrenaline and cortisol levels, intestinal mucosal barrier indexes, and complication rates were compared between the two groups before, during, and 1 day after surgery.RESULTS The observation group had significantly lower plasma adrenaline and cortisol levels during surgery and at 1 day postoperatively than that of the control group(P<0.05).Additionally,intestinal barrier indexes(endotoxin and D-dimer)at 1 day postoperatively were significantly lower in the observation group than in the control group(P<0.05).CONCLUSION Ultrasound-guided quadratus lumborum block could reduce stress response, protect intestinal barrier function, and decrease the incidence of complications in patients undergoing radical surgery for gastric cancer. This technique has the potential for clinical applications.
文摘The aim of this work was to study the reactions of wheat (T. aestivum L.) and its wild congeners under salt stress. This work is the first phase of research for practical breeding.
文摘The symmetrical push-pull fatigue tests at strain amplitude of 2×10~3 with different slip orientation of Al single crystals, were carried out at room temperature in air.The peak stresses of various cycles were measured and the stress-strain hysteresis loops for selected cy- cles were recorded. The energy losses.friction stresses, back stresses and shape
文摘The variation of cyclic stress, internal friction and ultrasonic attenuation during cyclic deformation and relations among them have been investigated with different slip orientation Al single crystals. The results indicate that the value of cyclic stress σ,internal friction Q^(-1)and ultrasonic attenuation △α depend obviously on the slip orientation. There are large differences in above three parameters for different slip orientation Al crystals. In early stage of fatigue life,σ and △α increase and Q^(-1)decreases with cycles N,and △α reached maxi- mum before σ,while Q^(-1)and σ get the valley and the peak, respectively,at same cycles.
文摘A phenomenological model for dispersed systems which exhibit complex theological behaviour such as shear and time-dependent viscosity, yield stress, and elasticity is proposed. The model that gives the best fit of experimental data appears to be the one with kinetic orders n = m = 2.

文摘Plant male reproduction is a fine-tuned developmental process that is susceptible to stressful environments and influences crop grain yields.Phytohormone signaling functions in control of plant normal growth and development as well as in response to external stresses, but the interaction or crosstalk among phytohormone signaling, stress response, and male reproduction in plants remains poorly understood.Cross-species comparison among 514 stress-response transcriptomic libraries revealed that ms33-6038,a genic male sterile mutant deficient in the Zm Ms33/Zm GPAT6 gene, displayed an excessive drought stress-like transcriptional reprogramming in anthers triggered mainly by disturbed jasmonic acid(JA)homeostasis. An increased level of JA appeared in Zm Ms33-deficient anthers at both meiotic and postmeiotic stages and activated genes involved in JA biosynthesis and signaling as well as genes functioning in JA-mediated drought response.Excessive accumulation of JA elevated expression level of a gene encoding a WRKY transcription factor that activated the Zm Ms33 promoter. These findings reveal a feedback loop of Zm Ms33-JA-WRKY-Zm Ms33 in controlling male sterility and JA-mediated stress response in maize, shedding light on the crosstalk of stress response and male sterility mediated by phytohormone homeostasis and signaling.

文摘Although tocopherols are essential for rice development, the molecular details by which their absence affects development remain to be determined. To study how tocopherols function during rice development,we performed a transcriptome deep sequencing(RNA-seq)analysis of the rice cultivar Nipponbare(Nip)and the tocopherol-deficient mutant small grain and dwarf 1-2(sgd1-2).We identified 563 differentially expressed genes that were enriched in Gene Ontology categories associated with metabolism, stress, cellular responses, and transcriptional regulation. We determined that the total fatty acid composition of Nip and sgd1-2 was comparable, although cell membrane penetrability in sgd1-2 was significantly higher than in Nip under optimal growth conditions, indicating that tocopherol deficiency induces cell membrane damage. The expression levels of dehydration-responsive element binding 1(DREB1)genes and free proline content in sgd1-2 were also higher than those in Nip.We also showed that the DELLA protein SLENDER RICE1(SLR1)accumulated in sgd1-2,resulting in significant changes in the global transcriptome. Our study confirms that the lack of tocopherol accumulation in rice induced ectopic stress responses and limited growth by enhancing SLR1 abundance through increasing SLR1 transcript levels. These results provide new insights into tocopherol during rice development.
文摘In 1945,Porter et al.published an electon microscopy study of cultured chick fibroblasts in which they observed:'a granular background and details of a darker lacelike reticulum which in places appears to be made up of chains of"vesicles"'(Porter et al.,1945).This constituted the first published observation of the endoplasmic reticulum(ER)and, while it was not evident at that time, this cytoplasmic system of interconnecting membrane-lined channels, comprising vesicles, tubules and cisternae, has numerous important functions.
文摘Cellular stress responses are powerful mechanisms that prevent and cope with the accumulation of macromolecular damage in the cells and also boost host defenses against pathogens. This review discusses the diverse strategies that poxviruses use to subvert host cell stress responses.
