Locoweed is a poisonous plant wildly distributed in most areas of the world,which causes livestock poisoning or death with serious economic loss.The Astragalus strictus belongs to a species of Iocoweed.It is mainly distributed in Tibet of China and is a serious hazard to the local livestock industry.The objective of this study is extracting and purifying condition of Swainsonine from Astragalus strictus with the cellulase extraction method.An optimum extracting technology of SW from Astragalus strictus was investigated through the orthogonal experiment under the cellulose assistance conditions,and then the content of swainsonine was analyzed with gas chromatography (GC) method.The optimized extraction conditions were as follow:The crushed mesh is 40; solid-liquid ratio is 1∶40 (g/ml); enzyme dosage is 3.5%; enzymatic time is 3 h.Under the above conditions,the extraction percentage of the swainsonine was 0.003 941%.The conditions for extracting swainsonine with cellulase extraction method are mild,it is easy to utilize industrial production.It is benefit for producing swainsonine from Astragalus strictus. 展开更多

利用透射电镜观察长竹蛏Solen strictus精子发生和精子的超微结构,揭示了从精原细胞发育为精子过程中超微结构的变化。其过程主要包括:生精细胞体积逐级减小,细胞核由椭圆形转变为圆形,染色质以颗粒状形式浓缩,内质网和高尔基体分泌的... 利用透射电镜观察长竹蛏Solen strictus精子发生和精子的超微结构,揭示了从精原细胞发育为精子过程中超微结构的变化。其过程主要包括:生精细胞体积逐级减小,细胞核由椭圆形转变为圆形,染色质以颗粒状形式浓缩,内质网和高尔基体分泌的前顶体颗粒融合形成前顶体囊、前顶体囊最终形成顶体,线粒体逐渐融合与发达,以及中心体移动及鞭毛形成。精细胞的分化可分为4个时期。成熟精子属原生型,全长约38—43μm,由头部、中段和尾部3部分组成。头部顶体较短,约为核径的五分之一,呈倒置的碗状,轴向纵切面为马鞍形,顶体物质为分布均匀的高电子密度物质;亚顶体腔几乎伸达顶体的顶端,内含均匀的较低电子密度物质。细胞核呈圆球形,无核前窝,但有一浅的核后窝。中段由4—5个椭圆形线粒体和2个相互垂直的中心粒组成。尾部为典型的“9+2”型结构。 展开更多

Total concentration of heavy metals(Cd, Cr, Cu, Fe, Pb, Ni, Mn and Zn) was estimated in the redeveloping soil of mine spoil under 5 yr old plantations of four woody species namely: Albizia lebbeck, Albizia procera, Tectona grandis and Dendrocalamus strictus. The data recorded in the present study were compared with other unplanted coal mine spoil colliery, which was around to the study site and adjoining area of dry tropical forest. Among all the heavy metals, the maximum concentration was found for Fe and minimum for Cd. However, among all four species, total concentrations of these heavy metals were recorded maximally in the plantation plots of T. grandis except for Fe, while minimally in A. lebbeck except for Zn, whereas, the maximum concentration of Fe and Zn was in the plantation plots of D. strictus and A. procera. Statistical analysis revealed significant differences due to species for all the heavy metals except Cu. Among four species, A. lebbeck, A. procera and D. strictus showed more efficient for reducing heavy metal concentrations whereas T. grandis was not more effective to reduce heavy metal concentrations in redeveloping soil of mine spoil. 展开更多

Micropropagation of Three Bamboo Species from Nodal Explants of Mature Culms

A micropropagation technique is developed for the multiplication of Dendrocalamus strictus (D. strictus), Dendrocalamus asper (19. asper) and Bambusa bambos (B. bambos) through shoot proliferation. Nodal explants obtained from field gown clumps were used to initiate cultures. Shoots were induced on Murashige and Skoog's (MS) medium supplemented with 5 mg L^-1 6-benzylamino purine (BAP). Rapid shoot multiplication was obtained on MS medium containing 3 mg Lt BAP in D. asper, B. bambos and 2 mg L^-1 BAP in D. strictus. In vitro multiplied shoots showed best root induction on half strength MS supplemented with 1 mg L^-1 indole-3 butyric acid (IBA) and 0.5 mg L^-1 1-naphthalene acetic acid (NAA) in D. asper. Pre-rooting conditioning followed by culturing on half strength MS supplemented with 1 mg L^-1 IBA and 2 mg L^-1 IBA showed maximum root induction in D. strictus and B. bambos, respectively. Further root proliferation was obtained on hormone free medium. The micropropagated plantlets were acclimatized and successfully transferred on soil in green house. 展开更多
