Fast,simple,efficient Agrobacterium rhizogenes-mediated transformation system to non-heading Chinese cabbage with transgenic roots

Non-heading Chinese cabbage, a variety of Brassica campestris, is an important vegetable crop in the Yangtze River Basin of China. However,the immaturity of its stable transformation system and its low transformation efficiency limit gene function research on non-heading Chinese cabbage. Agrobacterium rhizogenes-mediated(ARM) transgenic technology is a rapid and effective transformation method that has not yet been established for non-heading Chinese cabbage plants. Here, we optimized conventional ARM approaches(one-step and two-step transformation methods) suitable for living non-heading Chinese cabbage plants in nonsterile environments. Transgenic roots in composite non-heading Chinese cabbage plants were identified using phenotypic detection, fluorescence observation, and PCR analysis. The transformation efficiency of a two-step method on four five-day-old non-heading Chinese cabbage seedlings(Suzhouqing, Huangmeigui, Wuyueman, and Sijiu Caixin) was 43.33%-51.09%, whereas using the stout hypocotyl resulted in a transformation efficiency of 54.88% for the 30-day-old Sijiu Caixin.The one-step method outperformed the two-step method;the transformation efficiency of different varieties was above 60%, and both methods can be used to obtain transgenic roots for functional studies within one month. Finally, optimized ARM transformation methods can easily,quickly, and effectively produce composite non-heading Chinese cabbage plants with transgenic roots, providing a reliable foundation for gene function research and non-heading Chinese cabbage genetic improvement breeding.

Critique of TBLT in the Chinese Context

Task-based language teaching(TBLT) is an influential approach which prevails and thrives all over the world and is still favored by a number of teachers and researchers for its unparalleled advantages that overshadow the traditional ones. However, it still remains a question whether the foreign approach could be imported to China without modification and localized to adapt to Chinese educational background. This paper, aiming at answering this question, investigates the strengths and weaknesses of TBLT in the Chinese context at length.

Actinidia chinensis Planch.root extract downregulates the Wnt/β-catenin pathway to treat gastric cancer:a mechanism study based on network pharmacology

Background:Actinidia chinensis Planch.roots(AcRoots)have been applied as an anti-inflammatory and antitumor drug in the treatment of gastric cancer(GC).However,their mechanisms against GC cells remain unclear.To investigate the anticancer effect of AcRoots in GC and the possible underlying mechanism by using network pharmacology.Methods:Differentially expressed genes between gastric precancerous lesions and cancer were analyzed in Gene Expression Omnibus datasets,and these genes were overlapped with potential targets of AcRoots.Potential targets and pathways for AcRoots treatment of GC predicted by network pharmacology.Furthermore,we used the GC cell line HGC27 to explore the molecular mechanisms in the context of hub genes in apoptosis,invasion,metastasis,and epithelial to mesenchymal transition-promoting factors.Molecular docking between hub targets and active drug components was also performed.Results:Network pharmacological analysis suggested that the potential mechanism was related to the Wnt pathway and predicted nine hub genes.In in vitro studies,AcRoots significantly decreased HGC27 cell viability and promoted apoptosis by upregulating caspase3 and downregulating Bcl2.Moreover,it suppressed invasion and metastasis as well as the expression of epithelial to mesenchymal transition-related factors.In addition,AcRoots affected the phosphorylation level of GSK3β(Ser9)in the Wnt pathway to promote the degradation ofβ-catenin,resulting in the downregulation of the downstream target genes c-myc,cyclin D1 and snail.All the experimental results were consistent with the network pharmacology results.Conclusion:This study combined network pharmacology with in vitro experiments to provide valid evidence for the clinical promotion of AcRoots.

Effects of IBA on the rooting of branch cuttings of Chinese jujube (Zizyphus jujuba Mill.) and changes to nutrients and endogenous hormones

‘Zhongqiusucui’ jujube secondary shoots were treated with 3-indolebutyric acid(IBA) at three concentrations, 500, 1000 and 1500 mg/L. Results show that IBA could significantly enhance rooting and root characteristics of cuttings and were best with IBA at 1500 mg/L. In the rooting process, the formation of adventitious roots was related to the consumption and accumulation of nutrients(soluble sugars and proteins) and the changes in endogenous hormones in phloem, leaf tips and leaf bases. The rooting of cuttings had a positive correlation with the consumption of soluble sugars during the period of callus formation and with the accumulation of soluble sugars during adventitious root formation and growth. Rooting was positively related to the breakdown of soluble proteins in the phloem when the callus formed, and had a positive correlation with its accumulation during adventitious root formation and growth. Leaf tips and leaf bases showed a reverse trend in changes of soluble protein. However, together with the phloem, leaf tips and leaf bases regulated and controlled the formation and development of adventitious roots. The main activities of soluble proteins exist in the leaf tips as this was the main source of soluble proteins. The relation between rooting and IAA(indole-3-acetic acid) content in phloem was positive and thus a high concentration of IAA could benefit the induction and formation of adventitious roots. However, rooting was negatively related with ABA(abscisic acid) and GA(gibberellic acid) and a high concentration of both could inhibit the induction and formation of adventitious roots. Rooting had a positive correlation with phloem IAA/ABA ratios, and higher ratios could improve rooting. Low concentrations of ZR(zeatin riboside) triggered the induction of adventitious roots, while higher concentrations promoted root growth.Endogenous hormones in leaf tips and bases had an impact on rooting. The activities of endogenous hormones mainly existed in leaf tips because they play a major role in the production and consumption of IAA and its ABA content increased during rooting. The ZR in leaf tips influenced the rooting of cuttings, especially in the callus formation and rooting stage. Leaf tips were the main source of GA.

A Study On the Structural Dimension of Middle Class Relational Identification in Chinese Context

Through in-depth interviews,this study makes an exploratory study on the dimension of the middle class relational identification in Chinese context by using the grounded theory.It is mainly composed of 4 dimensions:relational orientation,relational adaptation,relational evaluation and relational development.According to the four dimension model of middle class relational identification,the middle class relational identification in China is the cognitive and behavioral orientation that in the action of achieving a certain purpose,because of the perception of consistence with each other in role cognition and personal cognition,the middle class is willing to unite and trust,respect and understand and coordinate the conflict to adapt to this relationship,and then to form a positive evaluation of the relationship and a sense of belonging and commitment to this relationship.The construction of the structural dimension of middle class relational identification in China is conducive to explaining the endemic phenomena of Chinese middle class.

A Survey of English Majors'Chinese Culture Learning and Teaching Strategies in the Context of Culture Going Out

At present,there is a serious phenomenon of"Chinese cultural aphasia"in English teaching for English majors in Colleges and universities,and Chinese cultural knowledge has not received due attention in foreign language teaching.Therefore,in the context of culture going out,the author conducts a research on the current situation of Chinese culture learning and teaching strategies of English majors.Relevant research shows that solving these problems can be solved by rationally dealing with the relationship between Chinese culture and Western culture,enriching the cultural reserves of college English teachers,creating a Chinese cultural learning environment and improving students'intercultural communication skills.In addition,the Chinese cultural literacy of English teachers should be promoted to explore the teaching methods of diversified Chinese cultural resources.

Teaching Grammar in a Communicative Framework in the Chinese EFL Context

Communicative language teaching(CLT) has been introduced to the English-as-a-foreign-language(EFL) context in China for four decades.One of the most significant remaining issues is related to whether this approach to grammar teaching is more effective and efficient than traditional ones.Based on an overview of trends in approaches to grammar instruction,discussion on both advantages of teaching grammar communicatively and the challenges posed by the Chinese EFL context are provided.It is proposed that grammar instruction should be integrated into a communicative framework and this would be the optimal approach to grammar teaching on condition that some constraints in the EFL context in China are dealt with carefully.Teaching implications conclude this paper.

Exploration of the feasibility for MOOC to be developed for EFL learning in the Chinese Context

MOOC is an abbreviation of Massive Open Online Course.Whether MOOC can be developed for EFL learning in the Chinese context is discussed regarding both the advantages and disadvantages.MOOC provides open access to resources of high quality,systematic set of social interactions and assessment,but still needs to enlarge structured EFL learning courses and examine their effectiveness(Wu&Zhou,2014).The lack of needs analysis and existence of language obstacles are the defects of MOOC for EFL learning in China.

Developing College Students' English Speaking Skills in Chinese Context

As one of the four important communication skills,speaking has long been neglected in the language teaching,while oral English teaching has always been the weak points in college English education.Since China began to reform and open the door to the outside world,oral English becomes more and more important.However,the traditional oral English education failed to meet the demand of social development,for many graduates couldn't communicate in English smoothly.This is due to the inappropriateness of traditional oral English education.The thesis is to discuss the present condition of Chinese students in oral classroom and sets up a new teaching model.In this part the new model is discussed in detail from the perspective of students,teaching materials,teacher,and teaching method.

Reduction in nerve root compression by the nucleus pulposus after Feng's Spinal Manipulation

Ninety-four patients with lumbar intervertebral disc herniation were enrolled in this study. Of these, 48 were treated with Feng＇s Spinal Manipulation, hot fomentation, and bed rest （treatment group）. The remaining 46 patients were treated with hot fomentation and bed rest only （control group）. After 3 weeks of treatment, clinical parameters including the angle of straight-leg raising, visual analogue scale pain score, and Japanese Orthopaedic Association score for low back pain were improved. The treatment group had significantly better improvement in scores than the control group. Magnetic resonance myelography three-dimensional reconstruction imaging of the vertebral canal demonstrated that filling of the compressed nerve root sleeve with cerebrospinal fluid increased significantly in the treatment group. The diameter of the nerve root sleeve was significantly larger in the treatment group than in the control group. However, the sagittal diameter index of the herniated nucleus pulposus and the angle between the nerve root sleeve and the thecal sac did not change significantly in either the treatment or control groups. The effectiveness of Feng＇s Spinal Manipulation for the treatment of symptoms associated with lumbar intervertebral disc herniation may be attributable to the relief of nerve root compression, without affecting the herniated nucleus pulposus or changing the morphology or position of the nerve root.

Response of density-related fine root production to soil and leaf traits in coniferous and broad-leaved plantations in the semiarid loess hilly region of China

Fine roots are the most active and functional component of root systems and play a significant role in the acquisition of soil resources. Density is an important structural factor in forest plantations but information on changes in fine roots along a density gradient is limited. In this study, plantations of black locust (Robinia pseudoacacia L.) and Chinese pine (Pinus tabuliformis Carr.) with four density classes were analyzed for the influence of soil and leaf traits on fine root growth. Fine root biomass increased with stand density. High fine root biomass was achieved through increases in the fine root production and turnover rate in the high-density black locust plantations and through an increase in fine root production in the pine plantations. In the high-density Chinese pine stand, there was a high fine root turnover which, coupled with high fine root production, contributed to a high fine root biomass. Overall, fine root production and turnover rate were closely related to soil volumetric water content in both kinds of plantations, while fine root biomass, especially the component of necromass, was related to soil nutrient status, which refers to phosphorous content in black locust plantations and nitrogen content in Chinese pine plantations. There was a close linkage between leaf area index and fine root dynamics in the black locust plantations but not in the pine plantations.

Intercultural Trust in Global Contexts:Synthesizing a Western Nomological Approach with a Chinese Systems Approach

Intercultural trust in global contexts plays a central role in helping people from different cultures to communicate comfortably,which is essential for cooperation.Attempting to construct a framework that might foster international cooperation,and thus be helpful for coping with global emergencies,we relate a Western nomological approach to an Eastern systems approach to analyse intercultural trust in global contexts.Considering cultural impacts on intercultural trust and the nomological framework of cultural differences,we propose an intercultural trust model to interpret how cultural differences influence trust.A qualitative study of Chinese-Irish interactions was conducted to interpret this model.We organized 10 seminars on intercultural trust,and interviewed 16 people to further explore the respondents'deeper feelings and experiences about intercultural trust in global contexts.Through this study,we have identified factors impacting on intercultural trust,and found that intercultural trust can be developed and improved in various ways.To llustrate these ways,we have provided tactics and methods for building intercultural trust in global contexts.Implications are highlighted for organizations to avoid cultural clashes and relevant political or economic risks.

Chinese word segmentation with local and global context representation learning

A local and global context representation learning model for Chinese characters is designed and a Chinese word segmentation method based on character representations is proposed in this paper.First,the proposed Chinese character learning model uses the semantics of local context and global context to learn the representation of Chinese characters.Then,Chinese word segmentation model is built by a neural network,while the segmentation model is trained with the character representations as its input features.Finally,experimental results show that Chinese character representations can effectively learn the semantic information.Characters with similar semantics cluster together in the visualize space.Moreover,the proposed Chinese word segmentation model also achieves a pretty good improvement on precision,recall and f-measure.

A New Word Detection Method for Chinese Based on Local Context Information

Finding out out-of-vocabulary words is an urgent and difficult task in Chinese words segmentation. To avoid the defect causing by offline training in the traditional method,the paper proposes an improved prediction by partical match (PPM) segmenting algorithm for Chinese words based on extracting local context information,which adds the context information of the testing text into the local PPM statistical model so as to guide the detection of new words. The algorithm focuses on the process of online segmentation and new word detection which achieves a good effect in the close or opening test,and outperforms some well-known Chinese segmentation system to a certain extent.
