文摘Iron-sulfur clusters(ISC)are essential cofactors for proteins involved in various biological processes, such as electron transport, biosynthetic reactions,DNA repair, and gene expression regulation.ISC assembly protein IscA1(or MagR)is found within the mitochondria of most eukaryotes.Magnetoreceptor(MagR)is a highly conserved A-type iron and iron-sulfur cluster-binding protein, characterized by two distinct types of iron-sulfur clusters,[2Fe-2S]and[3Fe-4S],each conferring unique magnetic properties.MagR forms a rod-like polymer structure in complex with photoreceptive cryptochrome(Cry)and serves as a putative magnetoreceptor for retrieving geomagnetic information in animal navigation.Although the N-terminal sequences of MagR vary among species, their specific function remains unknown. In the present study,we found that the N-terminal sequences of pigeon MagR,previously thought to serve as a mitochondrial targeting signal(MTS),were not cleaved following mitochondrial entry but instead modulated the efficiency with which iron-sulfur clusters and irons are bound. Moreover, the N-terminal region of MagR was required for the formation of a stable MagR/Cry complex. Thus, the N-terminal sequences in pigeon MagR fulfil more important functional roles than just mitochondrial targeting. These results further extend our understanding of the function of MagR and provide new insights into the origin of magnetoreception from an evolutionary perspective.

文摘The recycling of spent batteries has become increasingly important owing to their wide applications, abundant raw material supply, and sustainable development.Compared with the degraded cathode, spent anode graphite often has a relatively intact structure with few defects after long cycling.Yet,most spent graphite is simply burned or discarded due to its limited value and inferior performance on using conventional recycling methods that are complex, have low efficiency, and fail in performance restoration.Herein,we propose a fast, efficient, and“intelligent”strategy to regenerate and upcycle spent graphite based on defect‐driven targeted remediation.Using Sn as a nanoscale healant,we used rapid heating(~50 ms)to enable dynamic Sn droplets to automatically nucleate around the surface defects on the graphite upon cooling owing to strong binding to the defects(~5.84 eV/atom),thus simultaneously achieving Sn dispersion and graphite remediation. As a result, the regenerated graphite showed enhanced capacity and cycle stability(458.9 mAh g^(−1) at 0.2 A g^(−1) after 100 cycles),superior to those of commercial graphite.Benefiting from the self‐adaption of Sn dispersion, spent graphite with different degrees of defects can be regenerated to similar structures and performance.EverBatt analysis indicates that targeted regeneration and upcycling have significantly lower energy consumption(~99%reduction)and near‐zero CO_(2) emission, and yield much higher profit than hydrometallurgy, which opens a new avenue for direct upcycling of spend graphite in an efficient, green, and profitable manner for sustainable battery manufacture.

文摘The majority of the projectiles used in the hypersonic penetration study are solid flat-nosed cylindrical projectiles with a diameter of less than 20 mm.This study aims to fill the gap in the experimental and analytical study of the evolution of the nose shape of larger hollow projectiles under hypersonic penetration. In the hypersonic penetration test, eight ogive-nose AerMet100 steel projectiles with a diameter of 40 mm were launched to hit concrete targets with impact velocities that ranged from 1351 to 1877 m/s.Severe erosion of the projectiles was observed during high-speed penetration of heterogeneous targets, and apparent localized mushrooming occurred in the front nose of recovered projectiles.By examining the damage to projectiles,a linear relationship was found between the relative length reduction rate and the initial kinetic energy of projectiles in different penetration tests.Furthermore, microscopic analysis revealed the forming mechanism of the localized mushrooming phenomenon for eroding penetration,i.e.,material spall erosion abrasion mechanism, material flow and redistribution abrasion mechanism and localized radial upsetting deformation mechanism. Finally,a model of highspeed penetration that included erosion was established on the basis of a model of the evolution of the projectile nose that considers radial upsetting;the model was validated by test data from the literature and the present study.Depending upon the impact velocity,v0,the projectile nose may behave as undistorted, radially distorted or hemispherical.Due to the effects of abrasion of the projectile and enhancement of radial upsetting on the duration and amplitude of the secondary rising segment in the pulse shape of projectile deceleration, the predicted DOP had an upper limit.

文摘Based on the organic geochemical data and the molecular and stable carbon isotopic compositions of natural gas of the Lower Permian Fengcheng Formation in the western Central Depression of Junggar Basin, combined with sedimentary environment analysis and hydrocarbon-generating simulation, the gas-generating potential of the Fengcheng source rock is evaluated, the distribution of large-scale effective source kitchen is described, the genetic types of natural gas are clarified, and four types of favorable exploration targets are selected. The results show that:(1)The Fengcheng Formation is a set of oil-prone source rocks, and the retained liquid hydrocarbon is conducive to late cracking into gas, with characteristics of high gas-generating potential and late accumulation;(2)The maximum thickness of Fengcheng source rock reaches 900 m.The source rock has entered the main gas-generating stage in Penyijingxi and Shawan sags, and the area with gas-generating intensity greater than 20×10^(8) m^(3)/km^(2) is approximately 6500 km^(2).(3)Around the western Central Depression, highly mature oil-type gas with light carbon isotope composition was identified to be derived from the Fengcheng source rocks mainly, while the rest was coal-derived gas from the Carboniferous source rock;(4)Four types of favorable exploration targets with exploration potential were developed in the western Central Depression which are structural traps neighboring to the source, stratigraphic traps neighboring to the source,shale-gas type within the source, and structural traps within the source.Great attention should be paid to these targets.
文摘This paper presents an investigation on the effect of JPEG compression on the similarity between the target image and the background, where the similarity is further used to determine the degree of clutter in the image.Four new clutter metrics based on image quality assessment are introduced, among which the Haar wavelet-based perceptual similarity index, known as HaarPSI,provides the best target acquisition prediction results. It is shown that the similarity between the target and the background at the boundary between visually lossless and visually lossy compression does not change significantly compared to the case when an uncompressed image is used.In future work, through subjective tests,it is necessary to check whether this presence of compression at the threshold of just noticeable differences will affect the human target acquisition performance.Similarity values are compared with the results of subjective tests of the well-known target Search_2 database, where the degree of agreement between objective and subjective scores, measured through linear correlation, reached a value of 90%.

文摘Ribosomopathies encompass a spectrum of disorders arising from impaired ribosome biogenesis and reduced functionality.Mutation or dysexpression of the genes that disturb any finely regulated steps of ribosome biogenesis can result in different types of ribosomopathies in clinic, collectively known as ribosomopathy genes.Emerging data suggest that ribosomopathy patients exhibit a significantly heightened susceptibility to cancer.Abnormal ribosome biogenesis and dysregulation of some ribosomopathy genes have also been found to be intimately associated with cancer development. The correlation between ribosome biogenesis or ribosomopathy and the development of malignancies has been well established. This work aims to review the recent advances in the research of ribosomopathy genes among human cancers and meanwhile,to excavate the potential role of these genes, which have not or rarely been reported in cancer,in the disease development across cancers. We plan to establish a theoretical framework between the ribosomopathy gene and cancer development,to further facilitate the potential of these genes as diagnostic biomarker as well as pharmaceutical targets for cancer treatment.

文摘Endometriosis is a common chronic gynecological disease with endometrial cell implantation outside the uterus.Angiogenesis is a major pathophysiology in endometriosis. Our previous studies have demonstrated that the prodrug of epigallocatechin gallate(ProEGCG)exhibits superior anti-endometriotic and anti-angiogenic effects compared to epigallocatechin gallate(EGCG).However, their direct binding targets and underlying mechanisms for the differential effects remain unknown. In this study,we demonstrated that oral ProEGCG can be effective in preventing and treating endometriosis.Additionally,1D and 2D Proteome Integral Solubility Alteration assay-based chemical proteomics identified metadherin(MTDH)and PX domain containing serine/threonine kinase-like(PXK)as novel binding targets of EGCG and ProEGCG,respectively.Computational simulation and BioLayer interferometry were used to confirm their binding affinity. Our results showed that MTDH-EGCG inhibited protein kinase B(Akt)-mediated angiogenesis, while PXK-ProEGCG inhibited epidermal growth factor(EGF)-mediated angiogenesis via the EGF/hypoxia-inducible factor(HIF-1a)/vascular endothelial growth factor(VEGF)pathway. In vitro and in vivo knockdown assays and microvascular network imaging further confirmed the involvement of these signaling pathways. Moreover, our study demonstrated that ProEGCG has superior therapeutic effects than EGCG by targeting distinct signal transduction pathways and may act as a novel antiangiogenic therapy for endometriosis.

文摘Astrocytes are the most abundant glial cells in the central nervous system;they participate in crucial biological processes, maintain brain structure, and regulate nervous system function.Exosomes are cell-derived extracellular vesicles containing various bioactive molecules including proteins, peptides, nucleotides, and lipids secreted from their cellular sources.Increasing evidence shows that exosomes participate in a communication network in the nervous system,in which astrocyte-derived exosomes play important roles. In this review,we have summarized the effects of exosomes targeting astrocytes and the astrocyte-derived exosomes targeting other cell types in the central nervous system. We also discuss the potential research directions of the exosome-based communication network in the nervous system. The exosome-based intercellular communication focused on astrocytes is of great significance to the biological and/or pathological processes in different conditions in the brain.New strategies may be developed for the diagnosis and treatment of neurological disorders by focusing on astrocytes as the central cells and utilizing exosomes as communication mediators.

文摘Objective Our previous studies established that microRNA(miR)-451 from human umbilical cord mesenchymal stem cell-derived exosomes(hUC-MSC-Exos)alleviates acute lung injury(ALI).This study aims to elucidate the mechanisms by which miR-451 in hUC-MSC-Exos reduces ALI by modulating macrophage autophagy. Methods Exosomes were isolated from hUC-MSCs.Severe burn-induced ALI rat models were treated with hUC-MSC-Exos carrying the miR-451 inhibitor.Hematoxylin-eosin staining evaluated inflammatory injury.Enzyme-linked immunosorbnent assay measured lipopolysaccharide(LPS),tumor necrosis factor-α,and interleukin-1βlevels.qRT-PCR detected miR-451 and tuberous sclerosis complex 1(TSC1)expressions.The regulatory role of miR-451 on TSC1 was determined using a dual-luciferase reporter system.Western blotting determined TSC1 and proteins related to the mammalian target of rapamycin(mTOR)pathway and autophagy.Immunofluorescence analysis was conducted to examine exosomes phagocytosis in alveolar macrophages and autophagy level. Results hUC-MSC-Exos with miR-451 inhibitor reduced burn-induced ALI and promoted macrophage autophagy.MiR-451 could be transferred from hUC-MSCs to alveolar macrophages via exosomes and directly targeted TSC1.Inhibiting miR-451 in hUC-MSC-Exos elevated TSC1 expression and inactivated the mTOR pathway in alveolar macrophages.Silencing TSC1 activated mTOR signaling and inhibited autophagy, while TSC1 knockdown reversed the autophagy from the miR-451 inhibitor-induced.Conclusion miR-451 from hUC-MSC exosomes improves ALI by suppressing alveolar macrophage autophagy through modulation of the TSC1/mTOR pathway, providing a potential therapeutic strategy for ALI.

文摘BACKGROUND Monopolar spindle-binding protein 3B(MOB3B)functions as a signal transducer and altered MOB3B expression is associated with the development of human cancers.AIM To investigate the role of MOB3B in colorectal cancer(CRC).METHODS This study collected 102 CRC tissue samples for immunohistochemical detection of MOB3B expression for association with CRC prognosis. After overexpression and knockdown of MOB3B expression were induced in CRC cell lines, changes in cell viability, migration, invasion, and gene expression were assayed.Tumor cell autophagy was detected using transmission electron microscopy, while nude mouse xenograft experiments were performed to confirm the in-vitro results.RESULTS MOB3B expression was reduced in CRC vs normal tissues and loss of MOB3B expression was associated with poor CRC prognosis.Overexpression of MOB3B protein in vitro attenuated the cell viability as well as the migration and invasion capacities of CRC cells, whereas knockdown of MOB3B expression had the opposite effects in CRC cells. At the molecular level,microtubule-associated protein light chain 3 II/I expression was elevated, whereas the expression of matrix metalloproteinase(MMP)2,MMP9,sequestosome 1,and phosphorylated mechanistic target of rapamycin kinase(mTOR)was downregulated in MOB3B-overexpressing RKO cells. In contrast, the opposite results were observed in tumor cells with MOB3B knockdown. The nude mouse data confirmed these in-vitro findings,i.e.,MOB3B expression suppressed CRC cell xenograft growth, whereas knockdown of MOB3B expression promoted the growth of CRC cell xenografts.CONCLUSION Loss of MOB3B expression promotes CRC development and malignant behaviors, suggesting a potential tumor suppressive role of MOB3B in CRC by inhibition of mTOR/autophagy signaling.
文摘Aptamers are a type of single-chain oligonucleotide that can combine with a specific target.Due to their simple preparation, easy modification, stable structure and reusability, aptamers have been widely applied as biochemical sensors for medicine, food safety and environmental monitoring. However, there is little research on aptamer-target binding mechanisms, which limits their application and development.Computational simulation has gained much attention for revealing aptamer-target binding mechanisms at the atomic level. This work summarizes the main simulation methods used in the mechanistic analysis of aptamer-target complexes, the characteristics of binding between aptamers and different targets(metal ions, small organic molecules, biomacromolecules, cells, bacteria and viruses),the types of aptamer-target interactions and the factors influencing their strength. It provides a reference for further use of simulations in understanding aptamer-target binding mechanisms.

文摘Multiple myeloma(MM)is the second most prevalent hematological malignancy.Current MM treatment strategies are hampered by systemic toxicity and suboptimal therapeutic efficacy. This study addressed these limitations through the development of a potent MM-targeting chemotherapy strategy, which capitalized on the high binding affinity of alendronate for hydroxyapatite in the bone matrix and the homologous targeting of myeloma cell membranes, termed T-PB@M.The results from our investigations highlight the considerable bone affinity of T-PB@M,both in vitro and in vivo.Additionally, this material demonstrated a capability for drug release triggered by low pH conditions.Moreover,T-PB@M induced the generation of reactive oxygen species and triggered cell apoptosis through the poly(ADP-ribose)polymerase 1(PARP1)-Caspase-3-B-cell lymphoma-2(Bcl-2)pathway in MM cells.Notably,T-PB@M preferentially targeted bone-involved sites, thereby circumventing systemic toxic side effects and leading to prolonged survival of MM orthotopic mice. Therefore, this designed target-MM nanocarrier presents a promising and potentially effective platform for the precise treatment of MM.

文摘The Medium-Resolution Spectral Imager-Ⅱ(MERSI-Ⅱ)instrument aboard China’s Fengyun-3D satellite shares similarities with NASA’s Moderate Resolution Imaging Spectroradiometer(MODIS)sensor, enabling the retrieval of global aerosol optical depth(AOD).However,no officially released operational MERSI-Ⅱ aerosol products currently exist over the ocean. This study focuses on adapting the MODIS dark target(DT)ocean algorithm to the MERSI-Ⅱ sensor.A retrieval test is conducted on the 2019 MERSI-Ⅱ data over the global ocean, and the retrieved AODs are validated against ground-based measurements from the automatic Aerosol Robotic Network(AERONET)and the shipborne Maritime Aerosol Network(MAN).The operational MODIS DT aerosol products are also used for comparison purposes. The results show that MERSI-Ⅱ AOD granule retrievals are in good agreement with MODIS products, boasting high correlation coefficients(R)of up to 0.96 and consistent spatial distribution trends.Furthermore, the MERSI-Ⅱ retrievals perform well in comparison to AERONET and MAN measurements, with high R-values(>0.86).However,the low-value retrievals from MERSI-Ⅱ tend to be slightly overestimated compared to MODIS,despite both AODs displaying a positive bias.Notably, the monthly gridded AODs over the high latitudes of the northern and southern hemispheres suggest that MERSI-Ⅱ exhibits greater stability in space and time, effectively reducing unrealistically high-value noise in the MODIS products. These results illustrate that the MERSI-Ⅱ retrievals meet specific accuracy requirements by maintaining the algorithmic framework and most of the algorithmic assumptions, providing a crucial data supplement for aerosol studies and climate change.

文摘This paper studies the target controllability of multilayer complex networked systems,in which the nodes are highdimensional linear time invariant(LTI)dynamical systems, and the network topology is directed and weighted. The influence of inter-layer couplings on the target controllability of multi-layer networks is discussed. It is found that even if there exists a layer which is not target controllable, the entire multi-layer network can still be target controllable due to the inter-layer couplings. For the multi-layer networks with general structure,a necessary and sufficient condition for target controllability is given by establishing the relationship between uncontrollable subspace and output matrix.By the derived condition,it can be found that the system may be target controllable even if it is not state controllable. On this basis, two corollaries are derived, which clarify the relationship between target controllability, state controllability and output controllability. For the multi-layer networks where the inter-layer couplings are directed chains and directed stars, sufficient conditions for target controllability of networked systems are given, respectively. These conditions are easier to verify than the classic criterion.

文摘Towards higher impact velocities, ballistic events are increasingly determined by the material temperatures.Related effects might range from moderate thermal softening to full phase transition. In particular,it is of great interest to quantify the conditions for incipient or full melting of metals during impact interactions, which result in a transition from still strength-affected to hydrodynamic material behavior. In this work,we investigate to which extent the respective melting thresholds are also dependent on the initial, and generally elevated, temperatures of projectiles and targets before impact. This is studied through the application of a model developed recently by the authors to characterize the transition regime between high-velocity and hypervelocity impact, for which the melting thresholds of materials were used as the defining quantities. The obtained results are expected to be of general interest for ballistic application cases where projectiles or targets are preheated.Such conditions might result, for example, from aerodynamic forces acting onto a projectile during atmospheric flight, explosive shapedcharge-jet formation or armor exposure to environmental conditions. The performed analyses also broaden the scientific understanding of the relevance of temperature in penetration events, generally known since the 1960s,but often not considered thoroughly in impact studies.
