Mechanosensitive(MS) ion channels play an important role in various physiological processes.Although the determination of the structure of mechanosensitive channel of large conductance(MscL) makes the simulation s...Mechanosensitive(MS) ion channels play an important role in various physiological processes.Although the determination of the structure of mechanosensitive channel of large conductance(MscL) makes the simulation study possible,it has not so far been possible to directly simulate the gating mechanism of MscL in atomic detail.In this article,MscL has been studied via molecular dynamic(MD) simulations to gain a detailed description of the sensitivity to lateral tension and the gating pathway.MscL undergoes conformational rearrangement in sustaining lateral tension,and the open state is obtained when 2.0 MPa lateral tension is directly applied on the pure protein.During the opening process,Loop region responds to tension first,and the mechanical sensitivity is followed by S1 domain.Transmembrane(TM) bundle is the key position for channel opening,and the motion of TM1 helices finally realizes the significant expansion of the constricted gating pore.C-terminus domain presents expansion later during the TM opening.In our study,return of the whole protein to the initial closed state is achieved only in the early opening stage.During the relaxation from the open state,the TM helices are the most mobile domain,which is different from the opening process.展开更多
We use the carbon nanotube (CNT) as the material of the pH sensing layer of the separative structure for the extended gate H^+-ion sensitive field effect transistor (EGFET) device.The CNT paste was prepared with CNT p...We use the carbon nanotube (CNT) as the material of the pH sensing layer of the separative structure for the extended gate H^+-ion sensitive field effect transistor (EGFET) device.The CNT paste was prepared with CNT powder,Ag powder,silicagel,the di-n-butyl phthalate and the toluene solvents by appropriate ratio,then immobilized on the silicon substrate to form the carbon nanotube sensing layer.We measured theⅠ_(DS)-Ⅴ_G curves of the carbon nanotube separative structure EGFET device in the different pH buffer solutions by the Keithley 236Ⅰ-Ⅴmeasurement system.According to the experimental results,we can obtain the pH sensitivities of the carbon nanotube separative structure EGFET device,which is 62.54mV/pH from pH1 to pH13.展开更多
The slack-taut state of tether is a particular adverse circumstance, which may influence the normal operation state of tension leg platform (TLP). The dynamic responses of TLP with slack-taut tether are studied with...The slack-taut state of tether is a particular adverse circumstance, which may influence the normal operation state of tension leg platform (TLP). The dynamic responses of TLP with slack-taut tether are studied with consideration of several nonlinear factors introduced by large amplitude motions. The time histories of stresses of tethers of a typical TLP in slack- taut state are given. In addition, the sensitivities of slack to stiffness and mass are investigated by varying the stiffness of tether and mass of TLP. It is found that slack is sensitive to the mass of TLP. The critical curved surfaces ( over which indicates the slack) for the increase of mass are obtained.展开更多
