Many studies have revealed that gut microbes modulate host metabolism.In this study,we characterized the therapeutic effects of a novel gut commensal Luoshenia tenuis against host metabolic disorders.First,by in silic...Many studies have revealed that gut microbes modulate host metabolism.In this study,we characterized the therapeutic effects of a novel gut commensal Luoshenia tenuis against host metabolic disorders.First,by in silico analysis,we demonstrated that the L.tenuis was prevalent in the gut microbiomes of healthy humans but were depleted specifically in obesity cohorts.Further in vitro cultivation revealed that L.tenuis produced short chain fatty acids that were verified to modulate host metabolism and some other volatile metabolites to benefit hosts by anti-inflammation and anti-tumor.Second,gavage of the L.tenuis significantly decreased the body weight gain and food intake of high-fat diet-feeding C57BL/6J mice,which was in parallel with the changed expression level of genes related to satiety and feeding behavior.We then performed the gavage trial using diet induced obese mice,and it revealed that the administration of L.tenuis alleviated significantly the abnormal glucose and lipid metabolisms and reduced the inflammatory response.In summary,this study revealed a previously-unknown human gut commensal microbe that benefited host metabolism,and set the stage for the development of novel next-generation probiotic applicable for treatment of obesity and related metabolic disorders.展开更多

Diuron is one of the most frequently applied herbicides in sugarcane farming in southern Japan,and Australia.In addition,it is used as a booster substance in copper-based antifouling paints.Due to these various uses,D...Diuron is one of the most frequently applied herbicides in sugarcane farming in southern Japan,and Australia.In addition,it is used as a booster substance in copper-based antifouling paints.Due to these various uses,Diuron is released into the marine environment;however,little information is available on gene expression in corals and their symbiotic algae exposed to Diuron.We investigated the efects of Diuron on stress-responsive gene expression in the hermatypic coral Acropora tenuis and its symbiotic dinofagellates.After seven days of exposure to 1µg/L and 10µg/L Diuron,no signifcant changes in the body colour of corals were observed.However,quantitative reverse transcription-polymerase chain reaction analyses revealed that the expression levels of stress-responsive genes,such as heat shock protein 90(HSP90),HSP70,and calreticulin(CALR),were signifcantly downregulated in corals exposed to 10µg/L of Diuron for seven days.Moreover,aquaglyceroporin was signifcantly downregulated in corals exposed to environmentally relevant concentrations of 1µg/L Diuron.In contrast,no such efects were observed on the expression levels of other stress-responsive genes,such as oxidative stress-responsive proteins,methionine adenosyltransferase,and green/red fuorescent proteins.Diuron exposure had no signifcant efect on the expression levels of HSP90,HSP70,or HSP40 in the symbiotic dinofagellates.These results suggest that stress-responsive genes,such as HSPs,respond diferently to Diuron in corals and their symbiotic dinofagellates and that A.tenuis HSPs and CALRs may be useful molecular biomarkers for predicting stress responses induced by the herbicide Diuron.展开更多
Objective:To investigate the antioxidant and cytotoxic activity of the fruits of Calamus tenuis Roxb.Methods:The preliminary phytochemical group tests were done,which revealed the presence of alkaloid,tannin,flavonoid...Objective:To investigate the antioxidant and cytotoxic activity of the fruits of Calamus tenuis Roxb.Methods:The preliminary phytochemical group tests were done,which revealed the presence of alkaloid,tannin,flavonoid and steroid.The dried fruit was extracted in soxhlet apparatus using petroleum ether,ethyl acetate and methanol.Antioxidant potential of each extract was evaluated using total phenol content,total flavonoid content,cupric reducing antioxidant capacity,1,1-diphenyl-2-picrylhydrazyl radical scavenging activity,and total antioxidant capacity determinations.Results:The extracts were found to possess moderate to high amounts of phenolic and flavonoid contents.In cupric reducing antioxidant capacity assay the extracts showed moderate reducing power which increases with concentration.Scavenging of 1,1-diphenyl-2-picrylhydrazyl radical was found to rise with concentration with lowest IC_(50)value for methanol extract,which was confirmed by total antioxidant activity test that shows highest(95 mg/g of extract)in ascorbic acid equivalent for methanol extract.In Brine shrimp lethality bioassay the methanol and petroleum ether extracts were found to be toxic to Brine shrimp nauplii,with LC_(50)of 25.53μg/mL and 28.07μg/mL respectively while the LC_(50)of the reference vincristine sulphate was 1.32μg/mL.Ethyl acetate extract was found to be moderately cytotoxic showing LC_(50)of 47.79μg/mL.Conclusions:The results of the present study suggest that the fruits of Calamus tenuis Roxb possess antioxidant and cytotoxic potential.Moreover,phytochemical screening reveals the presence of alkaloid,tannin,flavonoid and steroid,which may be responsible for the observed bioactivities.展开更多

文摘长身高原鳅(Triplophysa tenuis)对维持木扎提河的水生态系统物质和能量流动具有重要作用。文章基于脂肪酸生物标记法和碳、氮稳定同位素(δ^(13)C、δ^(15)N)技术研究了长身高原鳅的食性和营养生态位特征。结果显示,其肌肉中共检测出22种脂肪酸,其中有8种饱和脂肪酸、6种单不饱和脂肪酸、8种多不饱和脂肪酸;由特征脂肪酸组成情况推测,长身高原鳅对浮游动物、硅藻、陆地植物或喜摄食硅藻的鱼虾均有摄食,表现为杂食性。长身高原鳅的δ^(13)C和δ^(15)N分别介于-27.09‰~-20.98‰和5.71‰~8.45‰,营养级介于2.68~3.48。雌雄样本间的δ^(13)C、δ^(15)N和营养级均不存在显著性差异(P>0.05);雄性样本核心生态位(Standard ellipse area,SEAc)和总生态位(Total area of convex hull,TA)均高于雌性。δ^(13)C与体长间表现为极显著正相关性(P<0.01),δ^(15)N和营养级与体长间不具有显著相关性。70~80 mm体长组与90 mm以上体长组SEAc面积不存在重叠且在聚类分析中被分为不同组。综上,长身高原鳅的食性为杂食性且食物组成随体长变化而不同,作为营养级偏高的捕食者,长身高原鳅能延长食物链长度,增加食物网复杂性,有利于维持水域生态系统的稳定性。
