Archived samples are irreplaceable and therefore it is valuable to develop analytical techniques that require minimal sample vo-lume. This work describes the measurement of trace elements Mg, Cu, Fe, Zn and ultratrace elements Cd, Co, Mn, Pb in limited quantity (150 μL) human serum or plasma samples. Samples were digested using a hotblock and analyzed using inductively coupled plasma mass spectrometry (ICP-MS). The analytical method was evaluated using a quadrupole (Q) and sector field high resolution (SF) instrument to analyze trace elements in Seronorm? quality control serum material. The method was used to analyze 1888 blinded human plasma samples which were archived for the National Cancer Institute from the Nutrition Intervention Trial in Linxian China. The inductively coupled plasma method was capable of accurately analyzed limited quantity samples of human serum and plasma for the trace elements Mg, Cu, Fe Zn and the ultra trace elements Co, Mn and Pb. The concentration of Cd in human plasma samples was below the level of detection for 75% of the samples analyzed.展开更多

It is proved in recent studies that human umbilical cord blood mononuclear cells induced and cultured in vitro by recombinant human granulocyte-macrophage colony stimulating factor (rhG-MCSF) and recombinant human interleukin-4 (rhlL-4) can generate a great many dendritic cells and promote the lethal effect of T cells on human neuroblastoma, but it is unclear that whether the lethal effect is associated with the most proper concentration of dendritic cells. OBJEETIVE: To investigate the lethal effect of human umbilical cord blood mononuclear cells induced in vitro by cytokines differentiating into dendritic cells on human neuroblastoma, and its best concentration range. DESIGN : Open experiment SEI-FING: Department of Pediatrics, the Medical School Hospital of Qingdao University MATERIALS : The study was carried out in the Shandong Provincial Key Laboratory (Laboratory for the Department of Pediatrics of the Medical School Hospital of Qingdao University) during September 2005 to May 2006. Human umbilical cord blood samples were taken from the healthy newborn infants of full-term normal delivery during October to November 2005 in the Medical School Hospital of Qingdao University, and were voluntarily donated by the puerperas. Main instruments: type 3111 CO2 incubator (Forma Scientific, USA), type 550 ELISA Reader (Bio-Rad, USA). Main reagents: neuroblastoma cell line SK-N-SH (Shanghai Institute of Life Science, Chinese Academy of Sciences), RPMI-1640 culture fluid and fetal bovine serum (Hyclone), rhlL-4 (Promega, USA), rhG-MCSF (Harbin Pharmaceutic Group Bioengineering Co.Ltd), rat anti-human CDla monoclonal antibody and FITC-labeled rabbit anti-rat IgG (Xiehe Stem cell Gene Engineering Co.Ltd). METHODS: ① Human umbilical cord blood mononuclear cells obtained with attachment methods differentiated into human umbilical cord blood dendritic cells, presenting typical morphology of dendritic cells after in vitro induction by rhG-MCSF and rhlL-4. ② Different concentrations of dendritic cells[ dendritic cells: neuroblastoma cells=20:1,50:1,100:1 (2×10^8 L^-1,5×10^8 L^-1,1×10^9 L^-1)], 1×10^9 L^-1 T cells and 1×10^7 L^-1 neuroblastoma cells were added in the experimental group. 1 ×10^9 L^-1 T cells and 1 ×10^7 L^-1 neuroblastoma cells were added in the control group. ③ Main surface marker CDla molecules of dendritic cells were detected with indirect immunofluorescence, and the percent rate of dendritic cells was counted with ultraviolet light and expressed as the expression rate of CD1a^+ cells. ④Single effector cells and target cells were respectively set in the experimental group and control group to obtain the lethal effect. The lethal effect of dendritic cells on neuroblastoma cells was indirectly evaluated by detecting cellular survival with MTT assay. The lethal effect(%)= (1-A experimentat well-A effector cell /A target cell well)×100%.⑤The expenmental data were presented as Mean ±SD, and paired t test was used. MAIN OUTCOME MEASURES: ① Morphological characters of dendritic cells in the process of induction and differentiation. ②CD1a^+ cellular expression rate. ③Lethal effect of dendntic cells on neuroblastoma cells. RESULTS: ①Morphological characters of dendritic cells in the process of induction and differentiation: On the 15^th day after human umbilical cord blood mononuclear cells were induced by rhG-MCSF and rhlL-4, typical morphology of dendritic cells could be seen under an inverted microscope. ②Expression rate of CD1a^+ cells was (43.12±5.83)%. ③Lethal effect of dendritic cells on neuroblastoma cells: Lethal effect of dendritic cells stimulated T cells in each experimental group ( dendritic cells: neuroblastoma cells=100:1,50:1, 20:1 respectively) on neuroblastoma cells was significantly higher than that in control group[(31.00 ±4.41 )%, (30.92±5.27)%,(33.57±5.35)%,(26.23±5.20)%, t=3.51,2.98,4.24, P〈 0.01 ); But the lethal effect of dendntic cells on neuroblastoma was significantly lower when their ratio was 100:1 and 50:1 in comparison with 20:1 (t=2.01,2.36, P 〈 0.05), and no significant difference in lethal effect existed between the ratio at 100:1 and 50:1 (t=0.06,P 〉 0.05). CONCLUSION: Dendritic cells differentiated from human umbilical cord blood mononuclear cells after in vitro induction of cytokines can promote the lethal effect of T cells on neuroblastoma cells. The lethal effect is associated with the concentration of dendritic cells within some range.展开更多

Present study was designed to find out the association of socio-economic, clinical and heavy metals such as chromium (Cr), lead (Pb) copper (Cu), zinc (Zn), iron (Fe), manganese (Mn) selenium (Se) and arsenic (As) as the principle environmental factors that appear to impact schizophrenic condition in Pakistani population. In this study we have tried to expose some factors such as drug abuse, marital status, education, monthly income that could be related to the disease. These parameters were not investigated before in Pakistani subjects. A self-made questionnaire was developed to collect and record the history of patient’s social and economical status. The level of transition metals in the whole blood was also measured by using Inductive couple plasma optical emission spectroscopy (ICP-OES). Present study has found higher levels of Cr, Pb, Zn, Se, As and Cu and lower levels of Fe and Mn in the blood of newly diagnosed (ND) patients when compared to the controls. While comparing ND with the patients who were on medication (Old Diagnosed) we, found lower level of these metals except for copper in ND. However no significant differences were observed between any trace metal levels between the studied groups. The survey concludes that economic status, marital status and illegal drugs are significantly associated with schizophrenia. Pakistani men who use cannabis are significantly higher in numbers when compared to women in acquiring the psychiatric symptoms.展开更多
Rationale:The current literature has a surprising controversy regarding the use of low-dose human chorionic gonadotropin(hCG)for luteal support as an explanation for the development of ovarian hyperstimulation syndrom...Rationale:The current literature has a surprising controversy regarding the use of low-dose human chorionic gonadotropin(hCG)for luteal support as an explanation for the development of ovarian hyperstimulation syndrome,and this is because of the gap in the listing of the predisposing factors that put women at an increased risk of ovarian hyperstimulation syndrome.Patient concerns:A case of 25-year-old woman presented with abdominal pain,distention,dyspnea,and nausea with a 6.5 kg increase in weight from baseline.Ultrasonographic examination showed bilaterally enlarged multicystic ovaries after gonadotropin-releasing hormone(GnRH)agonist triggering and cycle segmentation with no hCG rescue administration.Diagnosis:Moderate/severe ovarian hyperstimulation syndrome.Interventions:The woman was admitted to the hospital for medical management of moderate/severe ovarian hyperstimulation syndrome,and pain management was advanced to patient-controlled anesthesia with the start of low molecular weight heparin.On day 2,albumin therapy followed by a furosemide chase was started due to an increase in abdominal girth.On day 1,Cabergoline was maintained,and on day 2 the GnRH antagonist Cetrorelix was started.Outcomes:The woman’s clinical condition improved,and a clinical pregnancy was eventually achieved during the first cryo-warmed blastocyst cycle.Lessons:Ovarian hyperstimulation syndrome can still happen even after the use of GnRH agonist and avoidance of hCG support.Segmentation of in vitro fertilization with complete avoidance of hCG for luteal support remains the best approach.展开更多
AIM: To report 3 of 120 patients on the German MALT lymphoma trial with H. pylori associated gastric MALT lymphoma who developed early gastric cancer 4 and 5 years, after complete lymphoma remission following cure of ...AIM: To report 3 of 120 patients on the German MALT lymphoma trial with H. pylori associated gastric MALT lymphoma who developed early gastric cancer 4 and 5 years, after complete lymphoma remission following cure of H. pylori infection. PATIENTS AND RESULTS: Three patients (two men, 74 and 70 years; one women, 77 years) with H. pylori-associated low-grade MALT lymphoma achieved complete lymphoma remission after being cured. Surveillance endoscopies were performed twice a year in accordance to the protocol. Four years after complete lymphoma remission in two patients, and after 5 years in the other, early gastric adenocarcinoma of the mucosa-type, type IIa and type IIc, respectively, was detected, which were completely removed by endoscopic mucosa resection. In one patient, the gastric cancer was diagnosed at the same location as the previous MALT lymphoma, in the other patients it was detected at different sites of the stomach distant from location of the previous MALT lymphoma. The patients were H. pylori negative during the whole follow-up time. CONCLUSION: These findings strengthen the importance of regular Long-term follow-up endoscopies in patients with complete remission of gastric MALT lymphoma after cure of H. pylori infection. Furthermore, gastric adenocarcinoma may develop despite eradication of H. pylori.展开更多

In this study, an effective noncontact and nonattached technique that is based on electrostatic induction current generated during walking motion is proposed for the detection and assessment of human physical activity...In this study, an effective noncontact and nonattached technique that is based on electrostatic induction current generated during walking motion is proposed for the detection and assessment of human physical activity. In addition, a theoretical model is proposed for the electrostatic induction current generated owing to variation in the electric potential of the human body. The proposed electrostatic induction current model is compared with the theoretical model, and the proposed model is shown to effectively explain the behavior of the electrostatic induction current waveform. The normal walking motions of daily living are recorded with a portable sensor located in a regular house. The obtained results show that detailed information of physical activity such as a gait cycle can be estimated using our proposed technique. Additionally, the walking signal was measured when the subject walked with the ankle and knee fastened to a splint with bandages to simulate a limp. Therefore, the proposed technique, which is based on the detection of signal generated during walking, can be successfully employed to assess human physical activity.展开更多

Since the reliable earthquake precursor signals described by Ada and the characteristics of Alfvén waves match quite well, this paper proposes a hypothesis on how earthquakes are triggered based on the Alfvén (Q G) torsional wave model of Gillette et al. When the plume of the upper mantle column intrudes into the magma and lithosphere of the soft flow layer during the exchange of hot and cold molten material masses deep inside the Earth’s interior during ascent and descent, it is possible to form body and surface plasma sheets under certain conditions to form Alfven nonlinear isolated waves, and Alfven waves often perturb the geomagnetic field, releasing huge heat and kinetic energy thus triggering earthquakes. To explain the complex phenomenon of how Ada senses Alvfen waves and how to locate epicenters, we venture to speculate that special magnetosensory cells in a few human bodies can sense earthquake precursors and attempt to hypothesize an algorithm that analyzes how the human biological nervous system encodes and decodes earthquake precursors and explains how human magnetosensory cells can solve complex problems such as predicting earthquake magnitude and locating epicenters.展开更多

文摘AIM:To analyze bone inductive capacity of the compound material of decalcified bone matrix combined with rhBMP 2 and impregnated with bone cement.METHODS:To assess the experimental study, histological and Masson’s methods were used.RESULTS:The effects of compound material on the induction of bone formation were investigated in NIH mouse models.It was observed that in the with rhBMP 2 group, mesenchymal cells gathered in the implanted material at the 7th day postoperation,chondrogenesis were found at 14 to 21 days after implantation,new bone formation were observed at about 21 to 28 days after surgery and the DBM particles were absorbed by the new generated tissues gradually.CONCLUSIONS:The compound material of DBM combined with rhBMP 2 and impregnated with bone cement could induce the proliferation and migration of mesenchymal tissues that could be differentiated into cartilage and formed new bone finally.The new bone could absorb DBM particles gradually.The compound material had fair capacity of bone induction.
文摘Exposure to toxic chemicals appears to be one of the major factors in the onset of Schizophrenia. Present study was designed to find out the association of socio-economic, clinical and heavy metals such as chromium (Cr), lead (Pb) copper (Cu), zinc (Zn), iron (Fe), manganese (Mn) selenium (Se) and arsenic (As) as the principle environmental factors that appear to impact schizophrenic condition in Pakistani population. In this study we have tried to expose some factors such as drug abuse, marital status, education, monthly income that could be related to the disease. These parameters were not investigated before in Pakistani subjects. A self-made questionnaire was developed to collect and record the history of patient’s social and economical status. The level of transition metals in the whole blood was also measured by using Inductive couple plasma optical emission spectroscopy (ICP-OES). Present study has found higher levels of Cr, Pb, Zn, Se, As and Cu and lower levels of Fe and Mn in the blood of newly diagnosed (ND) patients when compared to the controls. While comparing ND with the patients who were on medication (Old Diagnosed) we, found lower level of these metals except for copper in ND. However no significant differences were observed between any trace metal levels between the studied groups. The survey concludes that economic status, marital status and illegal drugs are significantly associated with schizophrenia. Pakistani men who use cannabis are significantly higher in numbers when compared to women in acquiring the psychiatric symptoms.
文摘Rationale:The current literature has a surprising controversy regarding the use of low-dose human chorionic gonadotropin(hCG)for luteal support as an explanation for the development of ovarian hyperstimulation syndrome,and this is because of the gap in the listing of the predisposing factors that put women at an increased risk of ovarian hyperstimulation syndrome.Patient concerns:A case of 25-year-old woman presented with abdominal pain,distention,dyspnea,and nausea with a 6.5 kg increase in weight from baseline.Ultrasonographic examination showed bilaterally enlarged multicystic ovaries after gonadotropin-releasing hormone(GnRH)agonist triggering and cycle segmentation with no hCG rescue administration.Diagnosis:Moderate/severe ovarian hyperstimulation syndrome.Interventions:The woman was admitted to the hospital for medical management of moderate/severe ovarian hyperstimulation syndrome,and pain management was advanced to patient-controlled anesthesia with the start of low molecular weight heparin.On day 2,albumin therapy followed by a furosemide chase was started due to an increase in abdominal girth.On day 1,Cabergoline was maintained,and on day 2 the GnRH antagonist Cetrorelix was started.Outcomes:The woman’s clinical condition improved,and a clinical pregnancy was eventually achieved during the first cryo-warmed blastocyst cycle.Lessons:Ovarian hyperstimulation syndrome can still happen even after the use of GnRH agonist and avoidance of hCG support.Segmentation of in vitro fertilization with complete avoidance of hCG for luteal support remains the best approach.
文摘AIM: To report 3 of 120 patients on the German MALT lymphoma trial with H. pylori associated gastric MALT lymphoma who developed early gastric cancer 4 and 5 years, after complete lymphoma remission following cure of H. pylori infection. PATIENTS AND RESULTS: Three patients (two men, 74 and 70 years; one women, 77 years) with H. pylori-associated low-grade MALT lymphoma achieved complete lymphoma remission after being cured. Surveillance endoscopies were performed twice a year in accordance to the protocol. Four years after complete lymphoma remission in two patients, and after 5 years in the other, early gastric adenocarcinoma of the mucosa-type, type IIa and type IIc, respectively, was detected, which were completely removed by endoscopic mucosa resection. In one patient, the gastric cancer was diagnosed at the same location as the previous MALT lymphoma, in the other patients it was detected at different sites of the stomach distant from location of the previous MALT lymphoma. The patients were H. pylori negative during the whole follow-up time. CONCLUSION: These findings strengthen the importance of regular Long-term follow-up endoscopies in patients with complete remission of gastric MALT lymphoma after cure of H. pylori infection. Furthermore, gastric adenocarcinoma may develop despite eradication of H. pylori.

文摘In this study, an effective noncontact and nonattached technique that is based on electrostatic induction current generated during walking motion is proposed for the detection and assessment of human physical activity. In addition, a theoretical model is proposed for the electrostatic induction current generated owing to variation in the electric potential of the human body. The proposed electrostatic induction current model is compared with the theoretical model, and the proposed model is shown to effectively explain the behavior of the electrostatic induction current waveform. The normal walking motions of daily living are recorded with a portable sensor located in a regular house. The obtained results show that detailed information of physical activity such as a gait cycle can be estimated using our proposed technique. Additionally, the walking signal was measured when the subject walked with the ankle and knee fastened to a splint with bandages to simulate a limp. Therefore, the proposed technique, which is based on the detection of signal generated during walking, can be successfully employed to assess human physical activity.
文摘We statistically validate the 2011-2022 earthquake prediction records of Ada, the sixth finalist of the 2nd China AETA in 2021, who made 147 earthquake predictions (including 60% of magnitude 5.5 earthquakes) with a prediction accuracy higher than 70% and a confidence level of 95% over a 12-year period. Since the reliable earthquake precursor signals described by Ada and the characteristics of Alfvén waves match quite well, this paper proposes a hypothesis on how earthquakes are triggered based on the Alfvén (Q G) torsional wave model of Gillette et al. When the plume of the upper mantle column intrudes into the magma and lithosphere of the soft flow layer during the exchange of hot and cold molten material masses deep inside the Earth’s interior during ascent and descent, it is possible to form body and surface plasma sheets under certain conditions to form Alfven nonlinear isolated waves, and Alfven waves often perturb the geomagnetic field, releasing huge heat and kinetic energy thus triggering earthquakes. To explain the complex phenomenon of how Ada senses Alvfen waves and how to locate epicenters, we venture to speculate that special magnetosensory cells in a few human bodies can sense earthquake precursors and attempt to hypothesize an algorithm that analyzes how the human biological nervous system encodes and decodes earthquake precursors and explains how human magnetosensory cells can solve complex problems such as predicting earthquake magnitude and locating epicenters.
