BACKGROUND The self-assembly of solid organs from stem cells has the potential to greatly expand the applicability of regenerative medicine.Stem cells can self-organise into microsized organ units,partially modelling ...BACKGROUND The self-assembly of solid organs from stem cells has the potential to greatly expand the applicability of regenerative medicine.Stem cells can self-organise into microsized organ units,partially modelling tissue function and regeneration.Dental pulp organoids have been used to recapitulate the processes of tooth development and related diseases.However,the lack of vasculature limits the utility of dental pulp organoids.AIM To improve survival and aid in recovery after stem cell transplantation,we demonstrated the three-dimensional(3D)self-assembly of adult stem cell-human dental pulp stem cells(hDPSCs)and endothelial cells(ECs)into a novel type of spheroid-shaped dental pulp organoid in vitro under hypoxia and conditioned medium(CM).METHODS During culture,primary hDPSCs were induced to differentiate into ECs by exposing them to a hypoxic environment and CM.The hypoxic pretreated hDPSCs were then mixed with ECs at specific ratios and conditioned in a 3D environment to produce prevascularized dental pulp organoids.The biological characteristics of the organoids were analysed,and the regulatory pathways associated with angiogenesis were studied.RESULTS The combination of these two agents resulted in prevascularized human dental pulp organoids(Vorganoids)that more closely resembled dental pulp tissue in terms of morphology and function.Single-cell RNA sequencing of dental pulp tissue and RNA sequencing of Vorganoids were integrated to analyse key regulatory pathways associated with angiogenesis.The biomarkers forkhead box protein O1 and fibroblast growth factor 2 were identified to be involved in the regulation of Vorganoids.CONCLUSION In this innovative study,we effectively established an in vitro model of Vorganoids and used it to elucidate new mechanisms of angiogenesis during regeneration,facilitating the development of clinical treatment strategies.展开更多

Numerous studies have shown that cell replacement therapy can replenish lost cells and rebuild neural circuitry in animal models of Parkinson’s disease.Transplantation of midbrain dopaminergic progenitor cells is a p...Numerous studies have shown that cell replacement therapy can replenish lost cells and rebuild neural circuitry in animal models of Parkinson’s disease.Transplantation of midbrain dopaminergic progenitor cells is a promising treatment for Parkinson’s disease.However,transplanted cells can be injured by mechanical damage during handling and by changes in the transplantation niche.Here,we developed a one-step biomanufacturing platform that uses small-aperture gelatin microcarriers to produce beads carrying midbrain dopaminergic progenitor cells.These beads allow midbrain dopaminergic progenitor cell differentiation and cryopreservation without digestion,effectively maintaining axonal integrity in vitro.Importantly,midbrain dopaminergic progenitor cell bead grafts showed increased survival and only mild immunoreactivity in vivo compared with suspended midbrain dopaminergic progenitor cell grafts.Overall,our findings show that these midbrain dopaminergic progenitor cell beads enhance the effectiveness of neuronal cell transplantation.展开更多
Due to long-term human activity interference,the Hainan Tropical Rainforest National Park(HTRNP)of China has experienced ecological problems such as habitat fragmentation and biodiversity loss,and with the expanding s...Due to long-term human activity interference,the Hainan Tropical Rainforest National Park(HTRNP)of China has experienced ecological problems such as habitat fragmentation and biodiversity loss,and with the expanding scope and intensity of human activity impact,the regional ecological security is facing serious challenges.A scientific assessment of the interrelationship between human activity intensity and habitat quality in the HTRNP is a prerequisite for achieving effective management of ecological disturbances caused by human activities and can also provide scientific strategies for the sustainable development of the region.Based on the land use change data in 2000,2010,and 2020,the spatial and temporal variations and the relationship between habitat quality(HQ)and human activity intensity(HAI)in the HTRNP were explored using the integrated valuation of ecosystem services and trade-offs(InVEST)model.System dynamics and land use simulation models were also combined to conduct multi-scenario simulations of their relationships.The results showed that during 2000–2020,the habitat quality of the HTRNP improved,the intensity of human activities decreased each year,and there was a negative correlation between the two.Second,the system dynamic model could be well coupled with the land use simulation model by combining socio-economic and natural factors.The simulation scenarios of the coupling model showed that the harmonious development(HD)scenario is effective in curbing the increasing trend of human activity intensity and decreasing trend of habitat quality,with a weaker trade-off between the two compared with the baseline development(BD)and investment priority oriented(IPO)scenarios.To maintain the authenticity and integrity of the HTRNP,effective measures such as ecological corridor construction,ecological restoration,and the implementation of ecological compensation policies need to be strengthened.展开更多

Methods: Sperm chromosomes of 14 subjects (5 healthy controls and 9 HB patients, including 1 acute hepatitis B, 2 chronic active hepatitis B, 4 chronic persistent hepatitis B, 2 HBsAg chronic carriers with no clinical symptoms) were prepared using interspecific in vitro fertilization between zona-free hamster oocytes and human spermatozoa. Fluorescence in situ hybridization (FISH) to sperm chromosome spreads was carried out with biotin-labeled full length HBV DNA probe to detect the specific HBV DNA sequences in the sperm chromosomes. Results: Specific fluorescent signal spots for HBV DNA were seen in sperm chromosomes of one patient with chronic persistent hepatitis B. In 9(9/42) sperm chromosome complements containing fluorescent signal spots, one presented 5 obvious FISH spots and the others 2 to 4 signals. The fluorescence intensity showed significant difference among the signal spots. The distribution of signal sites among chromosomes seems to be random. Conclusion: HBV could integrate into human sperm chromosomes. Results suggest that the possibility of vertical transmission of HBV via the germ line to the next generation is present.展开更多

Methods Ten infertile men with immotile sperm were enrolled into this study (group A). The membrane integrity in the head and tail regions of individual spermatozoon of immotile sperm was examined by using the combined hypo-osmotic swelling-eosin Y exclusion test (HOS-EY test). Sperm morphology was observed by light, scanning and transmission electron microscopy. Ten semen samples from normospermic donors were used as the control (group B). Results The percentage of sperm with intact both head and tail membranes in group A was significantly lower than that in group B (P〈0.01), whereas the value of sperm with defective head membrane but intact tail membrane in group A was significantly higher than that in group B (P〈0.01). Abnormal sperm morphology in group A had a high incidence, and immotile sperm with viability and normal morphology could be observed in some cases. Most sperm had multiple ultrastructural defects. Conclussion Some immotile sperm had intact tail membrane but defective head membrane. Immotile sperm with viability and normal morphology could exist in some cases though abnormal sperm were in a great proportion. Carefully evaluating immotile sperm membrane integrity and morphology should benefit the treatment of patients with immotile sperm.展开更多

A common administrative framework of healthcare involves focus upon costs, quality and patient satisfaction (The Triple Aim). Many industries which support healthcare and healthcare administrators do not have firsthand knowledge of the complexities in delivering care. As a result, the experience and human factors of providing care are often overlooked at high level decision-making unless incorporated into the healthcare delivery framework, proposed as the fourth aim of The Quadruple Aim framework. Research is pointing to consequent negative effects on quality, safety, joy, meaning and sustainability of healthcare practice. High acute occupational stress and chronic occupational stress can cause direct and indirect effects on safety and quality of care. The biological, psychological and social consequences of burnout from excessive acute and chronic occupational stress are more of a threat to healthcare than commonly acknowledged. Patient safety, quality of care and clinician well-being are inextricably linked. This report will describe the process of transition from The Triple Aim to The Quadruple Aim administrative framework of healthcare delivery at the University of Rochester Medical Center. Developing the fourth aim of improving the experience of providing care, had high acceptability and aligned with other health system goals of optimization of safety, quality, and performance by applying a human factors/ergonomic (HFE) framework that considers human capabilities and human limitations. The goal of HFE is to fit the healthcare system to the human instead of the human to the healthcare system. Concepts include removal of extraneous cognitive load, using clinician neural resource (brain power) optimally for highest order decision making in patient care. As far as regions concerned, relative sea-level rising is more important than theoretical sea-level rising. The relative sea-level rising has resulted in series of environment disasters in coastal area of China, such as coastal erosion, seawater invasion, and floods in coastal lowlands, etc. The flux of sediments transported by rivers into sea is the main material source of marine pollution. The human activities are controlling factors of variations in river sediment loads. Under the influence of high strength development in the upstream region, the results are that the reduction of seagoing sediments in main rivers of China, and damage of coastal material balance. Land-based pollution is the main source of marine pollution. With rapid economic development in China s eastern coastal, pollutants flux into sea increase gradually. The results are that the environmental quality and seawater quality of marine have being deteriorated and pollution is serious increasingly. To meet the land demand of industrialization and urbanization, the sea reclamation has become an effective means to alleviate the contradiction between human and land in coastal area of China. This leads to large-scale reduction of coastal wetland area, and the heavy losses of ecological service value of coastal wetland. To face the environmental changes in coastal area of China and solve the contradiction among population, resources, environment, development, it is required to set up the system concept of man-land-sea relationship, so as to achieve sustainable development. Objective:This paper aims to study the correlativity between the number of pulmonary tuberculosis (PTB) cases and seasonal meteorological factors in Beijing.Methods:Based on theory of Human-Environmental Inter Relatio...Objective:This paper aims to study the correlativity between the number of pulmonary tuberculosis (PTB) cases and seasonal meteorological factors in Beijing.Methods:Based on theory of Human-Environmental Inter Relation in Huangdi's Internal Classics,we adopted monthly cases of PTB in Beijing from 2004 to 2011,and established a Seasonal Autoregressive Integrated Moving Average (SARIMA) model.Using the cross-correlation function (CCF),we then analyzed the correlation between meteorological factors and number of infected patients.The related meteorological factors were subsequently integrated,to establish a Seasonal Autoregressive Integrated Moving Average with explanatory variables (SARIMAX) model,which was used to estimate and verify the number of PTB cases in 2012.Results:In this study,a SARIMA(0,1,1) (0,1,1)12 model was established;CCF analysis was used to reveal the correlativity between PTB and precipitation with 1 lag,relative humidity with 1 lag.Then,integrated with relative humidity with 1 lag (β =2.405,95% confidence interval:0.433-4.377),the SARIMAX prediction model was proved to be an accurate approach for predicting local situations of PTB occurrence.Conclusions:The occurrence of PTB is correlated with seasonal meteorological factors.Combining these factors,an exact prediction model can be established,to estimate of the number of PTB infected patients.展开更多

The prevalence of human papilloma virus (HPV)-16 in patients with cervical cancer,the physical status of HPV-16 in patients with cervical lesions,and the role of HPV-16 integration in cervi-cal carcinogenesis were inv...The prevalence of human papilloma virus (HPV)-16 in patients with cervical cancer,the physical status of HPV-16 in patients with cervical lesions,and the role of HPV-16 integration in cervi-cal carcinogenesis were investigated.HPV genotyping was performed by using PCR approach with the primer GP5+/GP6+ and type-specific primer on biopsy specimens taken operatively from 198 women.Multiple PCR was done to detect physical status of HPV-16 in a series of cervical liquid-based cytology samples and biopsy specimens obtained from different cervical lesions with HPV-16 infection,includ-ing 112 specimens with cervical cancer,151 specimens with CINⅠ,246 specimens with CINⅡ and 120 specimens with CINⅢ.The results showed that there were 112 cervical cancer samples (56.57% of total cervical cancer patients) with HPV-16 infection.The frequency of HPV-16 pure integration was 65.18% (73/112),56.57% (47/120),23.58% (58/246) and 7.95% (12/151) in cervical cancer,CINⅢ,CINⅡand CINⅠ patients respectively.In situ hybridization was performed on some paraffin-embedded sections of CINⅡ,CINⅢ and cervical cancer to verify the physical status of HPV-16 infection.Sig-nificant difference was observed between cervical cancer and CINⅠ,CINⅡ,CINⅢ in the frequency of HPV-16 integration (P<0.01).It is suggested that HPV-16 is the most prevalent type and is associated with cervical cancer.In the case of HPV-16 infection there are close associations between the severity of cervical lesions and the frequency of HPV-16 integration.The application of testing HPV genotyping and physical status based on detection of HC-Ⅱ HPV DNA would be in favor of predicting the progno-sis of cervical precancerosis and enhancing the screening accuracy of cervical cancer.展开更多
AIM:To detect human papillomavirus(HPV) DNA in esophageal carcinoma(EC) 109 cells and investigate the relationship between HPV and EC.METHODS:Genomic DNA and total RNA from EC109 cells were isolated.HPV DNA was detect...AIM:To detect human papillomavirus(HPV) DNA in esophageal carcinoma(EC) 109 cells and investigate the relationship between HPV and EC.METHODS:Genomic DNA and total RNA from EC109 cells were isolated.HPV DNA was detected by polymerase chain reaction(PCR) with the general primer sets of My09/11 and GP5 +/6 + for the HPV L1 gene and type-specific primer sets for HPV18 E6 and HPV18 E6-E7.Reverse transcription(RT) of mRNA isolated from EC109 cells was performed to produce a cDNA.And then a PCR-based protocol for the amplification of papillomavirus oncogene transcripts was used to analyze HPV18 DNA and integrated transcripts of HPV18 in the chromosomes of EC109 cells.The final nested PCR products were cloned into a pMD-18T vector and sequenced to analyze the chromosomal location of HPV integration.RESULTS:HPV18 DNA was detected in EC109 cells by PCR using the general primer sets of My09/11 and GP5 +/6 + for HPV L1 and the type-specif ic primer sets for HPV18 E6 and E6-E7 to generate products of 450 bp,150 bp,335 bp and 944 bp,respectively.Approximately 600 bp of integrated HPV18-specific transcript was identified.The final nested PCR product of integrated HPV18 DNA was cloned into a pMD-18T vector and sequenced to analyze the chromosomal location of HPV integration.Sequence alignment showed that the HPV18 sequence from EC109 cells was identical to that of the encoded early protein E7-E1 of the standard HPV18 strain X05015,and another partial gene sequence was identical to a partial sequence of human chromosome 8.CONCLUSION:Integration of the HPV genome into the host cell chromosome suggests that persistent HPV infection is vital for malignant cell transformation and carcinogenesis.展开更多

The amount of rhGLP-I analogue fusion protein in transformant SG2 reached ca. 0.84 mg per gram of packed cells when SG2 was grown for 24 h in the YPD medium with a inoculum and medium ratio of 1:1. We believe these differences in optical properties should help differential diagnosis of human colon tissues by using optical methods.METHODS: In vitro optical properties were investigated for four kinds of tissues: normal human colon mucosa/submucosa and muscle layer/chorion, and adenomatous human colon mucosa/submucosa and muscle layer/chorion. Tissue samples were taken from 13 human colons (13 adenomatous, 13 normal). From the normal human colons a total of 26 tissue samples, with a mean thickness of 0.40 mm, were used (13 from mucosa/submucosa and 13 from muscle layer/chorion), and from the adenomatous human bladders a total of 26 tissue samples, with a mean thickness of 0.40 mm, were used (13 from mucosa/submucosa and 13 from muscle layer/chorion). The measurements were performed using a double-integratingsphere setup and the optical properties were assessed from these measurements using the adding-doubling method that was considered reliable.RESULTS: The results of measurement showed that there were significant differences in the absorption coefficients and scattering coefficients between normal and adenomatous human colon mucosa/submucosa at the same wavelength,and there were also significant differences in the two optical parameters between both colon muscle layer/chorion at the same wavelength. And there were large differences in the anisotropy factors between both colon mucosa/submucosa at the same wavelength, there were also large differences in the anisotropy factors between both colon muscle layer/chorion at the same wavelength.There were large differences in the value ranges of the absorption coefficients, scattering coefficients and anisotropy factors between both colon mucosa/submucosa,and there were also large differences in these value ranges between both colon muscle layer/chorion. There are the same orders of magnitude in the absorption coefficients for four kinds of colon tissues. The scattering coefficients of these tissues exceed the absorption coefficients by at least two orders of magnitude.CONCLUSION: There were large differences in the three optical parameters between normal and adenomatous human colon mucosa/submucosa at the same laser wavelength, and there were also large differences in these parameters between both colon muscle layer/chorion at the same laser wavelength. Large differences in optical parameters indicate that there were large differences in compositions and structures between both colon mucosa/submucosa, and between both colon muscle layer/chorion.Optical parameters for four kinds of colon tissues are wavelength dependent, and these differences would be useful and helpful in clinical applications of laser and tumors photodynamic therapy (PDT).展开更多

This study evaluates policy integration of human resource development (HRD) in agricultural sector within the context of multi-level governance (i.e., interdependence of administrative levels in decision making) and suggests means to enhance policy integration in Bangladesh. Conducting a critical review of major policy instruments, key informant interview and two case studies, this study concluded that HRD issue has fairly included and observed consistency (between the aim of HRD and policy) in all policies and strategies;majority of policy instruments have significantly addressed the significance of HRD and added plans for its development;reporting, monitoring and evaluation of policies and strategies are rather weak and there is no adequate flow of financial arrangement that is largely determined by the availability of government project. Based on the findings, the following means should be useful for improving HRD policy integration such as providing a handsome amount of development budget at the sub-district agricultural office, formulating coherent strategies to build effective cooperation, coordination and participation among stakeholders, initiating special capacity building programmes for rural women, young farmers and smallholders and mainstreaming HRD issue in governmental planning and programmes, and accordingly crafting policies and implementation.展开更多
Robot’s living space Human-robot relationship Robot humanization Hybrid intelligence Human-robot integrated society Robots,as the creation of humans,became an irreplaceable component in human society.With the advance...Robot’s living space Human-robot relationship Robot humanization Hybrid intelligence Human-robot integrated society Robots,as the creation of humans,became an irreplaceable component in human society.With the advancement of technologies,robots have become more and more intelligent and have been widely used in many fields,such as disease diagnosis,customer services,healthcare for the older people,and so on.As robots made our lives much more convenient than ever before,they also brought many potential risks and challenges in technology,security,and ethic.To better understand the development of robots,we proposed a concept of a robot’s living space and analyzed the role of robots in our society.In this paper,we focus on setting a theoretical framework of the robot’s living space to further understand the human-robot relationship.The research in this paper contains three central aspects.First,we interpret the concept of the robot’s living space and the functions of each space.Second,we analyze and summarize the relative technologies which support robots living well in each space.Finally,we provide advice and improvement measures based on a discussion of potential problems caused by the developments of robots.With the trend of robots humanization and human-robot society integration,we should seriously consider how to collaborate with intelligent robots to achieve hybrid intelligence.To build a harmonious human-robot integrated society,studying the robot’s living space and its relationship with humans is the prerequisite and roadmap.展开更多

Some programs have rigorous policy based programs in which potential adopters are screened in or out based on their responses to a series of qualifying questions. Some other organizations use a conversation based approach with-out policies around such things as income, vaccination status of animals in the home and landlord approval. Those organizations that use the policy-based approach do so with the premise that the animal will be loved and cared for better by those that meet their criteria. Policy based adoptions can be arduous and can decrease adoptions, as, for example, those living in apartments that are unable to prove their landlord accepts pets are turned away. We hypothesized that meeting or not meeting policy based criteria would have no impact on the care or bond of the adopter with the pet. This study examined the quality of care and attachment in two groups of adopters, a group that adopted while policy based adoptions were in place and a group that adopted when policies were eliminated. There were no substantial differences between the two groups. This important finding indicates that those that adopt through conversation based adoptions (policy-free) provide similar high quality care and are just as likely to be highly bonded to their pet as those that adopt through policy based adoptions.展开更多

文摘BACKGROUND The self-assembly of solid organs from stem cells has the potential to greatly expand the applicability of regenerative medicine.Stem cells can self-organise into microsized organ units,partially modelling tissue function and regeneration.Dental pulp organoids have been used to recapitulate the processes of tooth development and related diseases.However,the lack of vasculature limits the utility of dental pulp organoids.AIM To improve survival and aid in recovery after stem cell transplantation,we demonstrated the three-dimensional(3D)self-assembly of adult stem cell-human dental pulp stem cells(hDPSCs)and endothelial cells(ECs)into a novel type of spheroid-shaped dental pulp organoid in vitro under hypoxia and conditioned medium(CM).METHODS During culture,primary hDPSCs were induced to differentiate into ECs by exposing them to a hypoxic environment and CM.The hypoxic pretreated hDPSCs were then mixed with ECs at specific ratios and conditioned in a 3D environment to produce prevascularized dental pulp organoids.The biological characteristics of the organoids were analysed,and the regulatory pathways associated with angiogenesis were studied.RESULTS The combination of these two agents resulted in prevascularized human dental pulp organoids(Vorganoids)that more closely resembled dental pulp tissue in terms of morphology and function.Single-cell RNA sequencing of dental pulp tissue and RNA sequencing of Vorganoids were integrated to analyse key regulatory pathways associated with angiogenesis.The biomarkers forkhead box protein O1 and fibroblast growth factor 2 were identified to be involved in the regulation of Vorganoids.CONCLUSION In this innovative study,we effectively established an in vitro model of Vorganoids and used it to elucidate new mechanisms of angiogenesis during regeneration,facilitating the development of clinical treatment strategies.

文摘Efficient and flexible interactions require precisely converting human intentions into computer-recognizable signals,which is critical to the breakthrough development of metaverse.Interactive electronics face common dilemmas,which realize highprecision and stable touch detection but are rigid,bulky,and thick or achieve high flexibility to wear but lose precision.Here,we construct highly bending-insensitive,unpixelated,and waterproof epidermal interfaces(BUW epidermal interfaces)and demonstrate their interactive applications of conformal human–machine integration.The BUW epidermal interface based on the addressable electrical contact structure exhibits high-precision and stable touch detection,high flexibility,rapid response time,excellent stability,and versatile“cut-and-paste”character.Regardless of whether being flat or bent,the BUW epidermal interface can be conformally attached to the human skin for real-time,comfortable,and unrestrained interactions.This research provides promising insight into the functional composite and structural design strategies for developing epidermal electronics,which offers a new technology route and may further broaden human–machine interactions toward metaverse.

文摘Numerous studies have shown that cell replacement therapy can replenish lost cells and rebuild neural circuitry in animal models of Parkinson’s disease.Transplantation of midbrain dopaminergic progenitor cells is a promising treatment for Parkinson’s disease.However,transplanted cells can be injured by mechanical damage during handling and by changes in the transplantation niche.Here,we developed a one-step biomanufacturing platform that uses small-aperture gelatin microcarriers to produce beads carrying midbrain dopaminergic progenitor cells.These beads allow midbrain dopaminergic progenitor cell differentiation and cryopreservation without digestion,effectively maintaining axonal integrity in vitro.Importantly,midbrain dopaminergic progenitor cell bead grafts showed increased survival and only mild immunoreactivity in vivo compared with suspended midbrain dopaminergic progenitor cell grafts.Overall,our findings show that these midbrain dopaminergic progenitor cell beads enhance the effectiveness of neuronal cell transplantation.

文摘Due to long-term human activity interference,the Hainan Tropical Rainforest National Park(HTRNP)of China has experienced ecological problems such as habitat fragmentation and biodiversity loss,and with the expanding scope and intensity of human activity impact,the regional ecological security is facing serious challenges.A scientific assessment of the interrelationship between human activity intensity and habitat quality in the HTRNP is a prerequisite for achieving effective management of ecological disturbances caused by human activities and can also provide scientific strategies for the sustainable development of the region.Based on the land use change data in 2000,2010,and 2020,the spatial and temporal variations and the relationship between habitat quality(HQ)and human activity intensity(HAI)in the HTRNP were explored using the integrated valuation of ecosystem services and trade-offs(InVEST)model.System dynamics and land use simulation models were also combined to conduct multi-scenario simulations of their relationships.The results showed that during 2000–2020,the habitat quality of the HTRNP improved,the intensity of human activities decreased each year,and there was a negative correlation between the two.Second,the system dynamic model could be well coupled with the land use simulation model by combining socio-economic and natural factors.The simulation scenarios of the coupling model showed that the harmonious development(HD)scenario is effective in curbing the increasing trend of human activity intensity and decreasing trend of habitat quality,with a weaker trade-off between the two compared with the baseline development(BD)and investment priority oriented(IPO)scenarios.To maintain the authenticity and integrity of the HTRNP,effective measures such as ecological corridor construction,ecological restoration,and the implementation of ecological compensation policies need to be strengthened.
文摘Aim: To study the integration of hepatitis B virus (HBV) DNA into sperm chromosomes in hepatitis B patients and the features of its integration. Methods: Sperm chromosomes of 14 subjects (5 healthy controls and 9 HB patients, including 1 acute hepatitis B, 2 chronic active hepatitis B, 4 chronic persistent hepatitis B, 2 HBsAg chronic carriers with no clinical symptoms) were prepared using interspecific in vitro fertilization between zona-free hamster oocytes and human spermatozoa. Fluorescence in situ hybridization (FISH) to sperm chromosome spreads was carried out with biotin-labeled full length HBV DNA probe to detect the specific HBV DNA sequences in the sperm chromosomes. Results: Specific fluorescent signal spots for HBV DNA were seen in sperm chromosomes of one patient with chronic persistent hepatitis B. In 9(9/42) sperm chromosome complements containing fluorescent signal spots, one presented 5 obvious FISH spots and the others 2 to 4 signals. The fluorescence intensity showed significant difference among the signal spots. The distribution of signal sites among chromosomes seems to be random. Conclusion: HBV could integrate into human sperm chromosomes. Results suggest that the possibility of vertical transmission of HBV via the germ line to the next generation is present.

文摘Objective To determine the membrane integrity in the head and tail regions of individual spermatozoon, and observe sperm morphology for samples with totally immotile sperm. Methods Ten infertile men with immotile sperm were enrolled into this study (group A). The membrane integrity in the head and tail regions of individual spermatozoon of immotile sperm was examined by using the combined hypo-osmotic swelling-eosin Y exclusion test (HOS-EY test). Sperm morphology was observed by light, scanning and transmission electron microscopy. Ten semen samples from normospermic donors were used as the control (group B). Results The percentage of sperm with intact both head and tail membranes in group A was significantly lower than that in group B (P〈0.01), whereas the value of sperm with defective head membrane but intact tail membrane in group A was significantly higher than that in group B (P〈0.01). Abnormal sperm morphology in group A had a high incidence, and immotile sperm with viability and normal morphology could be observed in some cases. Most sperm had multiple ultrastructural defects. Conclussion Some immotile sperm had intact tail membrane but defective head membrane. Immotile sperm with viability and normal morphology could exist in some cases though abnormal sperm were in a great proportion. Carefully evaluating immotile sperm membrane integrity and morphology should benefit the treatment of patients with immotile sperm.
文摘Human factors in the delivery of service are considered in many occupations of high impact on others such as airline industry and nuclear power industry, but not sufficiently in healthcare delivery. A common administrative framework of healthcare involves focus upon costs, quality and patient satisfaction (The Triple Aim). Many industries which support healthcare and healthcare administrators do not have firsthand knowledge of the complexities in delivering care. As a result, the experience and human factors of providing care are often overlooked at high level decision-making unless incorporated into the healthcare delivery framework, proposed as the fourth aim of The Quadruple Aim framework. Research is pointing to consequent negative effects on quality, safety, joy, meaning and sustainability of healthcare practice. High acute occupational stress and chronic occupational stress can cause direct and indirect effects on safety and quality of care. The biological, psychological and social consequences of burnout from excessive acute and chronic occupational stress are more of a threat to healthcare than commonly acknowledged. Patient safety, quality of care and clinician well-being are inextricably linked. This report will describe the process of transition from The Triple Aim to The Quadruple Aim administrative framework of healthcare delivery at the University of Rochester Medical Center. Developing the fourth aim of improving the experience of providing care, had high acceptability and aligned with other health system goals of optimization of safety, quality, and performance by applying a human factors/ergonomic (HFE) framework that considers human capabilities and human limitations. The goal of HFE is to fit the healthcare system to the human instead of the human to the healthcare system. Concepts include removal of extraneous cognitive load, using clinician neural resource (brain power) optimally for highest order decision making in patient care. An integrative model of patient safety and clinician wellbeing is a product of this effort.

文摘Coastal zone is the complex area with intense sea-land interaction. There are various types, complex environments and developed economy in coastal area of China. Under the influence of global change and human activities, the ecosystem of coastal zone is vulnerable and has sensitive response to environment changes. The environmental problems of sea-level rising have attached close attention by countries all over the world. As far as regions concerned, relative sea-level rising is more important than theoretical sea-level rising. The relative sea-level rising has resulted in series of environment disasters in coastal area of China, such as coastal erosion, seawater invasion, and floods in coastal lowlands, etc. The flux of sediments transported by rivers into sea is the main material source of marine pollution. The human activities are controlling factors of variations in river sediment loads. Under the influence of high strength development in the upstream region, the results are that the reduction of seagoing sediments in main rivers of China, and damage of coastal material balance. Land-based pollution is the main source of marine pollution. With rapid economic development in China s eastern coastal, pollutants flux into sea increase gradually. The results are that the environmental quality and seawater quality of marine have being deteriorated and pollution is serious increasingly. To meet the land demand of industrialization and urbanization, the sea reclamation has become an effective means to alleviate the contradiction between human and land in coastal area of China. This leads to large-scale reduction of coastal wetland area, and the heavy losses of ecological service value of coastal wetland. To face the environmental changes in coastal area of China and solve the contradiction among population, resources, environment, development, it is required to set up the system concept of man-land-sea relationship, so as to achieve sustainable development. The feasible measures include improving coastal management system, strengthening integrated basin management, attaching importance to construction of the coastal environment engineering, and raising the environmental monitoring and early warning level.

文摘Objective:This paper aims to study the correlativity between the number of pulmonary tuberculosis (PTB) cases and seasonal meteorological factors in Beijing.Methods:Based on theory of Human-Environmental Inter Relation in Huangdi's Internal Classics,we adopted monthly cases of PTB in Beijing from 2004 to 2011,and established a Seasonal Autoregressive Integrated Moving Average (SARIMA) model.Using the cross-correlation function (CCF),we then analyzed the correlation between meteorological factors and number of infected patients.The related meteorological factors were subsequently integrated,to establish a Seasonal Autoregressive Integrated Moving Average with explanatory variables (SARIMAX) model,which was used to estimate and verify the number of PTB cases in 2012.Results:In this study,a SARIMA(0,1,1) (0,1,1)12 model was established;CCF analysis was used to reveal the correlativity between PTB and precipitation with 1 lag,relative humidity with 1 lag.Then,integrated with relative humidity with 1 lag (β =2.405,95% confidence interval:0.433-4.377),the SARIMAX prediction model was proved to be an accurate approach for predicting local situations of PTB occurrence.Conclusions:The occurrence of PTB is correlated with seasonal meteorological factors.Combining these factors,an exact prediction model can be established,to estimate of the number of PTB infected patients.

However,for a long time,their social integration situation in inflow places is not ideal. Academic circle has done a lot of researches,but no effective strategy is so far put forward. Through analysis of domestic and foreign researches,it is found that social capital and human capital have an important influence on social integration of new generation migrant workers. This paper takes this as starting point,combines characteristics of new generation migrant workers,and overviews theories and empirical researches of domestic and foreign famous scholars,to explore the relationship between social capital and human capital and social integration of new generation migrant workers.
文摘People in educational circles believe that humanism has an incomparable role and status in education and its ideology has a certain positive effect both in past education and in contemporary education. Moreover, the core of people-oriented thinking in humanism can be integrated with the quality education thought advocated by contemporary China. Therefore, in this context, the paper analyzes English education should focus on the integration of humanism. Firstly, the paper gives a brief overview of the meaning of humanism, and then summarizes the role of humanism in the full integration of English education. Finally, it analyzes in detail how to effectively integrate humanism into English education. 文摘The prevalence of human papilloma virus (HPV)-16 in patients with cervical cancer,the physical status of HPV-16 in patients with cervical lesions,and the role of HPV-16 integration in cervi-cal carcinogenesis were investigated.HPV genotyping was performed by using PCR approach with the primer GP5+/GP6+ and type-specific primer on biopsy specimens taken operatively from 198 women.Multiple PCR was done to detect physical status of HPV-16 in a series of cervical liquid-based cytology samples and biopsy specimens obtained from different cervical lesions with HPV-16 infection,includ-ing 112 specimens with cervical cancer,151 specimens with CINⅠ,246 specimens with CINⅡ and 120 specimens with CINⅢ.The results showed that there were 112 cervical cancer samples (56.57% of total cervical cancer patients) with HPV-16 infection.The frequency of HPV-16 pure integration was 65.18% (73/112),56.57% (47/120),23.58% (58/246) and 7.95% (12/151) in cervical cancer,CINⅢ,CINⅡand CINⅠ patients respectively.In situ hybridization was performed on some paraffin-embedded sections of CINⅡ,CINⅢ and cervical cancer to verify the physical status of HPV-16 infection.Sig-nificant difference was observed between cervical cancer and CINⅠ,CINⅡ,CINⅢ in the frequency of HPV-16 integration (P<0.01).It is suggested that HPV-16 is the most prevalent type and is associated with cervical cancer.In the case of HPV-16 infection there are close associations between the severity of cervical lesions and the frequency of HPV-16 integration.The application of testing HPV genotyping and physical status based on detection of HC-Ⅱ HPV DNA would be in favor of predicting the progno-sis of cervical precancerosis and enhancing the screening accuracy of cervical cancer.

文摘AIM:To detect human papillomavirus(HPV) DNA in esophageal carcinoma(EC) 109 cells and investigate the relationship between HPV and EC.METHODS:Genomic DNA and total RNA from EC109 cells were isolated.HPV DNA was detected by polymerase chain reaction(PCR) with the general primer sets of My09/11 and GP5 +/6 + for the HPV L1 gene and type-specific primer sets for HPV18 E6 and HPV18 E6-E7.Reverse transcription(RT) of mRNA isolated from EC109 cells was performed to produce a cDNA.And then a PCR-based protocol for the amplification of papillomavirus oncogene transcripts was used to analyze HPV18 DNA and integrated transcripts of HPV18 in the chromosomes of EC109 cells.The final nested PCR products were cloned into a pMD-18T vector and sequenced to analyze the chromosomal location of HPV integration.RESULTS:HPV18 DNA was detected in EC109 cells by PCR using the general primer sets of My09/11 and GP5 +/6 + for HPV L1 and the type-specif ic primer sets for HPV18 E6 and E6-E7 to generate products of 450 bp,150 bp,335 bp and 944 bp,respectively.Approximately 600 bp of integrated HPV18-specific transcript was identified.The final nested PCR product of integrated HPV18 DNA was cloned into a pMD-18T vector and sequenced to analyze the chromosomal location of HPV integration.Sequence alignment showed that the HPV18 sequence from EC109 cells was identical to that of the encoded early protein E7-E1 of the standard HPV18 strain X05015,and another partial gene sequence was identical to a partial sequence of human chromosome 8.CONCLUSION:Integration of the HPV genome into the host cell chromosome suggests that persistent HPV infection is vital for malignant cell transformation and carcinogenesis.
文摘The recombinant Saccharomyces cerevisiae strain stably expressing recombinant human glucagon-like peptide-l(rhGLP-1) analogue, as a potential oral drug delivery system for diabetes type II treatment, was successfully constructed by the homologous recombination between chromosomal DNA and yeast and integrating vector pNK-GLP containing yeast ribosomal DNA fragments. The amount of rhGLP-I analogue fusion protein in transformant SG2 reached ca. 0.84 mg per gram of packed cells when SG2 was grown for 24 h in the YPD medium with a inoculum and medium ratio of 1:1. Oral administration of 5 g lyophilized SG2/kg to hyperglycemic rats decreased serum glucose from (24.8±1.40) to (21.2±1.36) mmol/L.

文摘AIM: The purpose of the present study is to compare the optical properties of normal human colon mucosa/submucosa and muscle layer/chorion, and adenomatous human colon mucosa/submucosa and muscle layer/chorion in vitro at 476.5, 488, 496.5, 514.5 and 532 nm. We believe these differences in optical properties should help differential diagnosis of human colon tissues by using optical methods.METHODS: In vitro optical properties were investigated for four kinds of tissues: normal human colon mucosa/submucosa and muscle layer/chorion, and adenomatous human colon mucosa/submucosa and muscle layer/chorion. Tissue samples were taken from 13 human colons (13 adenomatous, 13 normal). From the normal human colons a total of 26 tissue samples, with a mean thickness of 0.40 mm, were used (13 from mucosa/submucosa and 13 from muscle layer/chorion), and from the adenomatous human bladders a total of 26 tissue samples, with a mean thickness of 0.40 mm, were used (13 from mucosa/submucosa and 13 from muscle layer/chorion). The measurements were performed using a double-integratingsphere setup and the optical properties were assessed from these measurements using the adding-doubling method that was considered reliable.RESULTS: The results of measurement showed that there were significant differences in the absorption coefficients and scattering coefficients between normal and adenomatous human colon mucosa/submucosa at the same wavelength,and there were also significant differences in the two optical parameters between both colon muscle layer/chorion at the same wavelength. And there were large differences in the anisotropy factors between both colon mucosa/submucosa at the same wavelength, there were also large differences in the anisotropy factors between both colon muscle layer/chorion at the same wavelength.There were large differences in the value ranges of the absorption coefficients, scattering coefficients and anisotropy factors between both colon mucosa/submucosa,and there were also large differences in these value ranges between both colon muscle layer/chorion. There are the same orders of magnitude in the absorption coefficients for four kinds of colon tissues. The scattering coefficients of these tissues exceed the absorption coefficients by at least two orders of magnitude.CONCLUSION: There were large differences in the three optical parameters between normal and adenomatous human colon mucosa/submucosa at the same laser wavelength, and there were also large differences in these parameters between both colon muscle layer/chorion at the same laser wavelength. Large differences in optical parameters indicate that there were large differences in compositions and structures between both colon mucosa/submucosa, and between both colon muscle layer/chorion.Optical parameters for four kinds of colon tissues are wavelength dependent, and these differences would be useful and helpful in clinical applications of laser and tumors photodynamic therapy (PDT).

文摘The great challenges of sustainable development highlight an urgent need to systematically understand the mech-anisms linking humans and nature.Resources and Environmental Sciences are a broad and practical discipline focused on coupled human and natural systems.They aim to study the formation and evolution of resources in the earth system,the drivers of various environmental problems,processes and relationships between resources and the environment,particularly under the combined impacts of natural conditions and human activities.The major resources and environmental problems drive the discipline development;international science programmes guide the direction of the discipline;interdisciplinary and transdisciplinary integration promotes new branches of the discipline;and technological progress results in a research paradigm shift.Facing the critical research re-quirements of strengthening trans-and interdisciplinarity,breaking through the key technology,targeting major environmental and disaster issues,and supporting sustainable development,nine critical scientific issues should be focused on climate change impact and adaptation,petroleum and mineral resources,water cycle and water re-sources,soil and land resources,ecosystems,remote sensing and geographic information science,environmental science and technology,disaster risk,and global and regional sustainable development.Suggestions to enhancing funding systems,improve talent cultivation,develop scientific platforms,and strength international cooperation are provided in this study to support scientific policymaking.The promotion of Resources and Environmental Sci-ences enables a more comprehensive and in-depth understanding of economic development and environmental changes relevant to assure a more sustainable global development.
文摘Human resource development assumes as a golden key for a knowledge-based economy and competitiveness. This study evaluates policy integration of human resource development (HRD) in agricultural sector within the context of multi-level governance (i.e., interdependence of administrative levels in decision making) and suggests means to enhance policy integration in Bangladesh. Conducting a critical review of major policy instruments, key informant interview and two case studies, this study concluded that HRD issue has fairly included and observed consistency (between the aim of HRD and policy) in all policies and strategies;majority of policy instruments have significantly addressed the significance of HRD and added plans for its development;reporting, monitoring and evaluation of policies and strategies are rather weak and there is no adequate flow of financial arrangement that is largely determined by the availability of government project. Based on the findings, the following means should be useful for improving HRD policy integration such as providing a handsome amount of development budget at the sub-district agricultural office, formulating coherent strategies to build effective cooperation, coordination and participation among stakeholders, initiating special capacity building programmes for rural women, young farmers and smallholders and mainstreaming HRD issue in governmental planning and programmes, and accordingly crafting policies and implementation.

文摘Robot’s living space Human-robot relationship Robot humanization Hybrid intelligence Human-robot integrated society Robots,as the creation of humans,became an irreplaceable component in human society.With the advancement of technologies,robots have become more and more intelligent and have been widely used in many fields,such as disease diagnosis,customer services,healthcare for the older people,and so on.As robots made our lives much more convenient than ever before,they also brought many potential risks and challenges in technology,security,and ethic.To better understand the development of robots,we proposed a concept of a robot’s living space and analyzed the role of robots in our society.In this paper,we focus on setting a theoretical framework of the robot’s living space to further understand the human-robot relationship.The research in this paper contains three central aspects.First,we interpret the concept of the robot’s living space and the functions of each space.Second,we analyze and summarize the relative technologies which support robots living well in each space.Finally,we provide advice and improvement measures based on a discussion of potential problems caused by the developments of robots.With the trend of robots humanization and human-robot society integration,we should seriously consider how to collaborate with intelligent robots to achieve hybrid intelligence.To build a harmonious human-robot integrated society,studying the robot’s living space and its relationship with humans is the prerequisite and roadmap.
文摘Animal shelters’ adoption processes vary across the US. Some programs have rigorous policy based programs in which potential adopters are screened in or out based on their responses to a series of qualifying questions. Some other organizations use a conversation based approach with-out policies around such things as income, vaccination status of animals in the home and landlord approval. Those organizations that use the policy-based approach do so with the premise that the animal will be loved and cared for better by those that meet their criteria. Policy based adoptions can be arduous and can decrease adoptions, as, for example, those living in apartments that are unable to prove their landlord accepts pets are turned away. We hypothesized that meeting or not meeting policy based criteria would have no impact on the care or bond of the adopter with the pet. This study examined the quality of care and attachment in two groups of adopters, a group that adopted while policy based adoptions were in place and a group that adopted when policies were eliminated. There were no substantial differences between the two groups. This important finding indicates that those that adopt through conversation based adoptions (policy-free) provide similar high quality care and are just as likely to be highly bonded to their pet as those that adopt through policy based adoptions.
