Climate change strongly affected the structure and functions of natural ecosystems,e.g.the vegetation productivity decreased in the Northeast permafrost region due to the higher temperature and less precipitation,whereas in the Tibetan Plateau,the vegetation productivity increased,owing to the improved thermal resource.Climate change led to reduced precipitation in North and Northeast China and thus the reduced surface runoff.The public needs for energy were changed because of climate change,e.g.the shorter heating period in winter.Climate change profoundly influenced human health,pathophoresis and major projects by increasing extreme events,including frequency and magnitude,and causing more serious water shortage.Under the background of climate change,although the improved thermal resources can be helpful for extending the crop growth period,more extreme events may resulted in more instability in agricultural productivity.Not only did climate change indirectly affect the secondary and tertiary industries through the impacts on agriculture and natural resources,but also climate change mitigation measures,such as carbon tax,tariff and trading,had extensive and profound influences on the socioeconomic system.Further analysis indicated that the impact of climate change presented significant regional differences.The impact had its pros and cons,while the advantages outweighed the disadvantages.Based on the above analysis on the impacts of climate change,we put forward suggestions on coping with climate change.First,scientifically dealing with climate change will need to seek advantages while avoiding the disadvantages of climate change in order to achieve the orderly adaptation to climate change,which is characterized with"Overall best,long-term benefit."Second,quantitative adaptation should be given more attention,e.g.proposing operational schemes and predictable goals and using uncertainty analysis on adaptation measures.Third,more active coping strategy should be adopted to enhance China's future comprehensive competitiveness.The strategies include but are not limited to gradually adjusting the industrial structure,intensifying the research and development(R&D)of emission reduction technology and actively responding to the influence of carbon tax,tariff and trading on socioeconomic development in China. 展开更多

Colorectal cancer incidence and mortality in patients younger than 50 years are increasing, but screening before the age of 50 is not offered in Europe. Advancedstage diagnosis and mortality from colorectal cancer before 50 years of age are increasing. This is not a detection-bias effect;it is a real issue affecting the entire population. Three independent computational models indicate that screening from 45 years of age would yield a better balance of benefits and risks than the current start at 50 years of age. Experimental data support these predictions in a sex- and race-independent manner. Earlier screening is seemingly affordable, with minimal impediments to providing younger adults with colonoscopy. Indeed, the American Cancer Society has already started to recommend screening from 45 years of age in the United States. Implementing early screening is a societal and public health problem. The three independent computational models that suggested earlier screening were criticized for assuming perfect compliance. Guidelines and recommendations should be derived from well-collected and reproducible data, and not from mathematical predictions. In the era of personalized medicine, screening decisions might not be based solely on age, and sophisticated prediction software may better guide screening. Moreover, early screening might divert resources away from older individuals with greater biological risks. Finally, it is still unknown whether early colorectal cancer is part of a continuum of disease or a biologically distinct disease and, as such, it might not benefit from screening at all. The increase in early-onset colorectal cancer incidence and mortality demonstrates an obligation to take actions. Earlier screening would save lives, and starting at the age of 45 years may be a robust screening option. 展开更多

As a main pro-inflammatory factor, the up-regulation of TNF-α in RA patients plays a pivotal role in RA pathogenesis. The process of TNF production starts with the binding of a ligand( which is commonly a microbial product) to a cell surface Toll receptor, which stimulates a signal transduction pathway that activates NF - κB transcription factors. Activated NF - κB enters nuclei and induces the transcription of genes associated with inflammation, including those coding for TNF. The production of TNF and other inflammatory cytokines serves to recruit other inflammatory cells, which in turn release cytokines and subsequently amplify the immune response. TNF exerts its effect via binding of TNFRland TNFR2 receptors. Many experimental and clinical study have proved the effectiveness of TNF - αa ctivity blockade. Recent therapeutic interventions, including TNF - α and IL - 1 inhibitors, strongly support the importance of cytokines in RA. The mechanism of anti - TNF - α action in vivo is complex and probably includes suppression of other pro - inflammatory cytokines, decreased synovial cellular infiltration, interference with osteoclast activation, and decreased angiogenisis. Studies using anti -TNF agents clearly show that they can slow or prevent the progression of bone and cartilage damage in RA. This activity probably involves suppression of osteoclasts in joint lesions. Etanereept, Infliximab and adalimumab are the three main biologic agents that were fo-cused on clinically at present. 展开更多

扶正化瘀方调控代谢抑制巨噬细胞促炎表型极化的机制研究

目的利用label-free蛋白质组学技术探究扶正化瘀方(FZHY)抑制巨噬细胞向促炎表型(M1)极化的分子机制。方法脂多糖(LPS)和γ干扰素(IFN-γ)诱导小鼠巨噬细胞RAW264.7向M1极化。细胞分为空白对照组、M1组和FZHY低、高剂量组(100、200 mg/... 目的利用label-free蛋白质组学技术探究扶正化瘀方(FZHY)抑制巨噬细胞向促炎表型(M1)极化的分子机制。方法脂多糖(LPS)和γ干扰素(IFN-γ)诱导小鼠巨噬细胞RAW264.7向M1极化。细胞分为空白对照组、M1组和FZHY低、高剂量组(100、200 mg/L),通过非标记定量蛋白质组学遴选差异蛋白(DEPs)及进行生物信息学分析,并通过实验验证差异蛋白及其信号通路。结果受LPS和IFN-γ诱导后,巨噬细胞向M1极化,与空白对照组比较,共有223个DEPs,其中120个上调,103个下调。经FZHY处理后,与M1组比较,共有103个DEPs,其中80个上调,23个下调。以上各组DEPs的交集为28个,其中脂肪分化相关蛋白2(Plin2)、溶酶体酸脂肪酶A(Lipa)、含犰狳重复序列蛋白6(Armc6)等25个DEPs在M1组中水平降低,在FZHY高剂量组中显著上调;IFN-γ诱导的GTP酶1(Iigp1)、三结构域蛋白质30a(Trim30a)和RAB11家族相互作用蛋白1(Rab11fip1)共3个蛋白在M1组中水平提高,在FZHY高剂量组中显著下调。差异蛋白采用基因本体(GO)数据库分析,发现其生物学功能包括定位修复等、细胞组成涉及细胞质部分等、分子功能包括催化活性等。采用京都基因与基因组百科全书(KEGG)数据库分析显示差异蛋白富集于代谢通路、溶酶体以及谷胱甘肽代谢等。与空白对照组比较,M1组的炎症表面标志物白细胞分化抗原40(CD40)、白细胞分化抗原86(CD86)和诱导型一氧化氮合酶(iNOS)及差异蛋白Iigp1的表达显著提高,而差异蛋白Plin2的表达降低。FZHY处理后可逆转上述炎症诱导的蛋白表达。M1巨噬细胞代谢以糖酵解为主,α-酮戊二酸脱氢酶(AKGDH)和谷氨酰胺连接酶(GLUL)在M1组中的表达降低,伴随着糖酵解速率提高,FZHY的处理可显著提高AKGDH和GLUL的基因表达并降低糖酵解速率。结论FZHY方可通过调控代谢中的关键代谢酶AKGDH、GLUL和差异蛋白Plin2、Iigp1及炎症标记物CD40、CD86、iNOS的表达,下调糖酵解速率而抑制巨噬细胞向M1的极化。 展开更多

个体亲环境行为对酒店的可持续发展具有重要意义,但已有研究缺乏对酒店特定空间情境的关注。绿色实践是酒店构建绿色空间情境的重要途径。文章以长沙市4家金叶级绿色饭店为研究案例,基于刺激-机体-反应理论,采用质性研究与结构方程模型... 个体亲环境行为对酒店的可持续发展具有重要意义,但已有研究缺乏对酒店特定空间情境的关注。绿色实践是酒店构建绿色空间情境的重要途径。文章以长沙市4家金叶级绿色饭店为研究案例,基于刺激-机体-反应理论,采用质性研究与结构方程模型相结合的方法,探讨了酒店绿色实践对个体亲环境行为的影响机制。质性研究结果表明,酒店绿色实践主要包括四大维度,分别为环境设计、绿色产品、绿色宣传和绿色管理。结构方程模型结果表明:(1)无论顾客组还是员工组,环境设计、绿色产品、绿色宣传和绿色管理对个体亲环境行为具有显著正向影响;(2)地方依恋在绿色实践与亲环境行为之间具有中介效应,由于绿色产品对员工地方依恋无显著影响,导致员工组在此维度上的中介效应不显著,而其他中介效应均显著;(3)在绿色管理维度,地方依恋的中介效应存在显著的组间差异。该研究强调了酒店空间情境的作用,有利于丰富酒店内亲环境行为的研究,并对酒店的绿色运营管理具有一定的启示。 展开更多

Many neurological disorders such Parkinson’s disease, Alzheimer’s disease, amyotrophic lateral sclerosis, Huntington’s disease, cerebral ischemia, and acute spinal cord injury (ASCI), are closely related to autophagy.
