With the development of high-coverage sequencing technology, researchers have started to notice that genomic repeat regions, previously neglected in search of disease culprits, are active contributors to multiple neurodegenerative diseases. In this review, we describe the association between repeat element variants and multiple degenerative diseases through genome-wide association studies and targeted sequencing. We discuss the identification of disease-relevant repeat element variants, further powered by the advancement of long-read sequencing technologies and their related tools, and summarize recent findings in the molecular mechanisms of repeat element variants in brain degeneration, such as those causing transcriptional silencing or RNA-mediated gain of toxic function. Furthermore, we describe how in silico predictions using innovative computational models, such as deep learning language models, could enhance and accelerate our understanding of the functional impact of repeat element variants. Finally, we discuss future directions to advance current findings for a better understanding of neurodegenerative diseases and the clinical applications of genomic repeat elements.展开更多

Some of these techniques using topography are used in several fields (civil engineering, geodesy, topography, mechanics, nuclear engineering, hydraulics, physics, etc.). These topometric techniques have undergone major changes as a result of technological advances, growing needs in the monitoring of movements or deformations, increased requirements and new challenges. The methodology adopted depends on the measuring instrument used, the parameters to be estimated and access to the area to be measured. There are two types of methods: destructive and non-destructive. In addition to the visual method, they can also be classified as mechanical, physico-chemical, dynamometric, electrophysical and geometric. The estimated parameter varies according to the methodology adopted. It can be defined by coordinates, distances, potential, electrical resistance, etc.展开更多

文摘Currently, the Al_(2)O_(3)content in the high-alumina slag systems within blast furnaces is generally limited to 16wt%–18.5wt%,making it challenging to overcome this limitation.Unlike most studies that concentrated on managing the MgO/Al_(2)O_(3)ratio or basicity, this paper explored the effect of equimolar substitution of MgO for CaO on the viscosity and structure of a high-alumina CaO-MgO-Al_(2)O_(3)-SiO_(2)slag system, providing theoretical guidance and data to facilitate the application of high-alumina ores. The results revealed that the viscosity first decreased and then increased with higher MgO substitution, reaching a minimum at 15mol%MgO concentration.Fourier transform infrared spectroscopy(FTIR)results found that the depths of the troughs representing[SiO_(4)]tetrahedra,[AlO_(4)]tetrahedra,and Si-O-Al bending became progressively deeper with increased MgO substitution.Deconvolution of the Raman spectra showed that the average number of bridging oxygens per Si atom and the X_(Q^(3))/X_(Q^(2))(X_(Q^(i))is the molar fraction of Q^(i) unit, and i is the number of bridging oxygens in a[SiO_(4)]tetrahedral unit)ratio increased from 2.30 and 1.02 to 2.52 and 2.14,respectively,indicating a progressive polymerization of the silicate structure.X-ray photoelectron spectroscopy(XPS)results highlighted that non-bridging oxygen content decreased from 77.97mol% to 63.41mol% with increasing MgO concentration, whereas bridging oxygen and free oxygen contents increased.Structural analysis demonstrated a gradual increase in the polymerization degree of the tetrahedral structure with the increase in MgO substitution. However, bond strength is another important factor affecting the slag viscosity. The occurrence of a viscosity minimum can be attributed to the complex evolution of bond strengths of non-bridging oxygens generated during depolymerization of the[SiO_(4)]and[AlO_(4)]tetrahedral structures by CaO and MgO.

With the development of high-coverage sequencing technology, researchers have started to notice that genomic repeat regions, previously neglected in search of disease culprits, are active contributors to multiple neurodegenerative diseases. 文摘γ-Secretase,called“the proteasome of the membrane,”is a membrane-embedded protease complex that cleaves 150+peptide substrates with central roles in biology and medicine, including amyloid precursor protein and the Notch family of cell-surface receptors.Mutations inγ-secretase and amyloid precursor protein lead to early-onset familial Alzheimer’s disease.γ-Secretase has thus served as a critical drug target for treating familial Alzheimer’s disease and the more common late-onset Alzheimer’s disease as well. However, critical gaps remain in understanding the mechanisms of processive proteolysis of substrates, the effects of familial Alzheimer’s disease mutations, and allosteric modulation of substrate cleavage byγ-secretase.In this review,we focus on recent studies of structural dynamic mechanisms ofγ-secretase.Different mechanisms, including the“Fit-Stay-Trim,”“Sliding-Unwinding,”and“Tilting-Unwinding,”have been proposed for substrate proteolysis of amyloid precursor protein byγ-secretase based on all-atom molecular dynamics simulations. While an incorrect registry of the Notch1 substrate was identified in the cryo-electron microscopy structure of Notch1-boundγ-secretase,molecular dynamics simulations on a resolved model of Notch1-boundγ-secretase that was reconstructed using the amyloid precursor protein-boundγ-secretase as a template successfully capturedγ-secretase activation for proper cleavages of both wildtype and mutant Notch, being consistent with biochemical experimental findings. The approach could be potentially applied to decipher the processing mechanisms of various substrates byγ-secretase.In addition, controversy over the effects of familial Alzheimer’s disease mutations, particularly the issue of whether they stabilize or destabilizeγ-secretase-substrate complexes,is discussed. Finally,an outlook is provided for future studies ofγ-secretase,including pathways of substrate binding and product release, effects of modulators on familial Alzheimer’s disease mutations of theγ-secretase-substrate complexes.Comprehensive understanding of the functional mechanisms ofγ-secretase will greatly facilitate the rational design of effective drug molecules for treating familial Alzheimer’s disease and perhaps Alzheimer’s disease in general.

文摘Full concentration gradient lithium-rich layered oxides are catching lots of interest as the next generation cathode for lithium-ion batteries due to their high discharge voltage, reduced voltage decay and enhanced rate performance, whereas the high lithium residues on its surface impairs the structure stability and long-term cycle performance.Herein,a facile multifunctional surface modification method is implemented to eliminate surface lithium residues of full concentration gradient lithium-rich layered oxides by a wet chemistry reaction with tetrabutyl titanate and the post-annealing process. It realizes not only a stable Li_(2)TiO_(3)coating layer with 3D diffusion channels for fast Li^(+)ions transfer, but also dopes partial Ti^(4+)ions into the sub-surface region of full concentration gradient lithium-rich layered oxides to further strengthen its crystal structure.Consequently, the modified full concentration gradient lithium-rich layered oxides exhibit improved structure stability, elevated thermal stability with decomposition temperature from 289.57℃to 321.72℃,and enhanced cycle performance(205.1 mAh g^(-1)after 150 cycles)with slowed voltage drop(1.67 mV per cycle). This work proposes a facile and integrated modification method to enhance the comprehensive performance of full concentration gradient lithium-rich layered oxides, which can facilitate its practical application for developing higher energy density lithium-ion batteries.

文摘As a promising anode material for magnesium ion rechargeable batteries, magnesium metavanadate(MgV_(2)O_(6))has attracted considerable research interest in recent years.A MgV_(2)O_(6)sample was synthesized via a facile solid-state reaction by multistep-firing stoichiometric mixtures of MgO and V2O5 powder under an air atmosphere. The solid-state phase transition fromα-MgV_(2)O_(6)toβ-MgV_(2)O_(6)occurred at 841 K and the enthalpy change was 4.37±0.04 kJ/mol.The endothermic effect at 1014 K and the enthalpy change was 26.54±0.26 kJ/mol,which is related to the incongruent melting ofβ-MgV_(2)O_(6).In situ XRD was performed to investigate phase transition of the as-prepared MgV_(2)O_(6)at high temperatures. The cell parameters obtained by Rietveld refinement indicated that it crystallizes in a monoclinic system with the C2/m space group, and the lattice parameters of a=9.280 A°,b=3.501 A°,c=6.731 A°,β=111.76°.The solid-state phase transition fromα-MgV_(2)O_(6)toβ-MgV_(2)O_(6)was further studied by thermal kinetics, indicating that this process is controlled first by a fibril-like mechanism and then by a spherulitic-type mechanism with an increasing heating rate.Additionally, the enthalpy change of MgV_(2)O_(6)at high temperatures was measured utilizing the drop calorimetry, heat capacity was calculated and given as:Cp=208.3+0.03583T-4809000T^(−2)(298-923 K)(J mol^(−1)K^(−1)),the high-temperature heat capacity can be used to calculate Gibbs free energy of MgV_(2)O_(6)at high temperatures.

文摘Ovalbumin(OVA)is the major allergenic protein that can induce T helper 2(Th2)-allergic reactions, for which current treatment options are inadequate. In this study,we developed a polymerized hypoallergenic OVA product via laccase/caffeic acid(Lac/CA)-catalyzed crosslinking in conjunction with galactomannan(Man). The formation of high molecular weight crosslinked polymers and the Ig G-binding were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)and Western blotting. The study indicated that Lac/CA-catalyzed crosslinking plus Man conjugation substantially altered secondary and tertiary structures of OVA along with the variation in surface hydrophobicity.Gastrointestinal digestion stability assay indicated that crosslinked OVA exhibited less resistance in simulated gastric fluid(SGF)and simulated intestinal fluid(SIF).Mouse model study indicated that Lac-Man/OVA ameliorated eosinophilic airway inflammatory response and efficiently downregulated the expression of Th2-related cytokines(interleukin(IL)-4,IL-5,and IL-13),and upregulated IFN-γand IL-10 expression.Stimulation of bone marrow-derived dendritic cells with Lac-Man/OVA suppressed the expression of phenotypic maturation markers(CD80 and CD86)and MHC class II molecules, and suppressed the expression levels of proinflammatory cytokines. The knowledge obtained in the present study offers an effective way to acquire a hypoallergenic OVA product that can have a therapeutic effect in alleviating OVA-induced allergic asthma.

文摘In this work, the structure, viscosity and ion-exchange process of Na_(2)O-MgO-Al_(2)O_(3)-SiO_(2) glasses with different Al_(2)O_(3)/SiO_(2) molar ratios were investigated. The results showed that, with increasing Al_(2)O_(3)/SiO_(2) ratio, the simple structural units Q_(1) and Q_(2) transformed into highly aggregated structural units Q_(3) and Q_(4),indicating the increase of polymerization degree of glass network.Meanwhile, the coefficient of thermal expansion decreased from 9.23×10^(-6)℃^(-1) to 8.88×10^(-6)℃^(-1).The characteristic temperatures such as melting, forming, softening and glass transition temperatures increased with the increase of Al_(2)O_(3)/SiO_(2) ratio, while the glasses working temperature range became narrow. The increasing Al_(2)O_(3)/SiO_(2) ratio and prolonging ion-exchange time enhanced the surface compressive stress(CS)and depth of stress layer(DOL).However, the increase of ion exchange temperature increased the DOL and decreased the CS affected by stress relaxation. There was a good linear relationship between stress relaxation and surface compressive stress.Chemical strengthening significantly improved the hardness of glasses, which reached the maximum value of(622.1±10)MPa for sample with Al_(2)O_(3)/SiO_(2) ratio of 0.27 after heat treated at 410℃for 2 h.
文摘Topometric auscultation is used to monitor the durability of structures, measure deformations linked to the structure of a structure or to the movement of the ground over a part of the globe, set up warning systems, etc. It first appeared as a visual method and rapidly evolved through the various techniques used. It can be defined by coordinates, distances, potential, electrical resistance, etc.

文摘Lithium-sulfur battery(LSB)has brought much attention and concern because of high theoretical specific capacity and energy density as one of main competitors for next-generation energy storage systems. The widely commercial application and development of LSB is mainly hindered by serious“shuttle effect”of lithium polysulfides(Li PSs),slow reaction kinetics, notorious lithium dendrites, etc. In various structures of LSB materials, array structured materials, possessing the composition of ordered micro units with the same or similar characteristics of each unit, present excellent application potential for various secondary cells due to some merits such as immobilization of active substances, high specific surface area, appropriate pore sizes, easy modification of functional material surface, accommodated huge volume change, enough facilitated transportation for electrons/lithium ions, and special functional groups strongly adsorbing Li PSs.Thus many novel array structured materials are applied to battery for tackling thorny problems mentioned above. In this review, recent progresses and developments on array structured materials applied in LSBs including preparation ways, collaborative structural designs based on array structures, and action mechanism analyses in improving electrochemical performance and safety are summarized.Meanwhile,we also have detailed discussion for array structured materials in LSBs and constructed the structure-function relationships between array structured materials and battery performances.Lastly, some directions and prospects about preparation ways, functional modifications, and practical applications of array structured materials in LSBs are generalized. We hope the review can attract more researchers'attention and bring more studying on array structured materials for other secondary batteries including LSB.

文摘On an agrosilvopastoral farm in central Italy where Maremmana cattle graze in Turkey oak forests,we evaluated the impact of different livestock densities on stand structure, tree diversity and natural regeneration in four types of grazed areas based on the grazing regime adopted:calf-grazed,high-intensity-grazed,low-intensity-grazed,ungrazed control. For each area,we set up three permanent circular plots(radius of 15 m)to survey the structural and dasometric characteristics of the overstorey, understorey, and regeneration layer. The results showed that grazing negatively affected the complexity of the forest structure and its potential to regenerate and maintain a high level of biodiversity. The differences in stand structure observed between the grazing areas were closely related to livestock density. The most sensitive components of the system were the understorey and the regeneration layers.Contrarily, the current grazing management did not affect the dominant tree structure or its composition. Our findings identified medium-term monitoring and regeneration management as the two significant aspects to consider when assessing sustainable livestock.New forests can be established by excluding graz-ing for about 20–25 years.

文摘Recent experimental advancements reported a chemical reaction between antimony and nitrogen under high temperature and high pressure, yielding crystalline antimony nitride(Sb_(3)N_(5))with an orthorhombic structure.Motivated by this statement,we calculate the stability, elastic properties, electronic properties and energy density of the Cmc2_(1) structure for pnictogen nitrides X_(3)N_(5)(X=P,As,Sb,and Bi)using first-principles calculations combined with particle swarm optimization algorithms.Calculations of formation enthalpies, elastic constants and phonon spectra show that P_(3)N_(5),As_(3)N_(5) and Sb_(3)N_(5) are thermodynamically, mechanically and kinetically stable at 35 GPa,whereas Bi_(3)N_(5) is mechanically and kinetically stable but thermodynamically unstable. The computed electronic density of states shows strong covalent bonding between the N atoms and the phosphorus group atoms in the four compounds, confirmed by the calculated electronic localization function. We also calculate the energy densities for Sb_(3)N_(5) and find it to be a potentially high-energy-density material.

文摘Cu^(+)-doped alkali borosilicate glasses with different Na_(2)O contents were prepared by the melting method, and the effects of different R values(R=Na_(2)O/B_(2)O_(3))on the structure, ion presence state and luminescence properties of Cu^(+)-doped alkali borosilicate glasses were investigated. The analysis by FT-IR and Raman spectroscopy shows that, with the increase of R value of the glass,the[BO_(3)]in the structure of Cu^(+)-doped alkali borosilicate glass transforms into[BO_(4)]and the number of non-bridging oxygen in the glass network appears to be slightly increased. The absorption spectra and EPR analysis reveal that the Cu^(+)content in the glass gradually decreases and the Cu^(2+)content gradually increases as the R value of the glass increases.XPS and PL tests further indicate that the transformation of the octahedral coordination structure of Cu^(+)to the octahedral coordination structure of Cu^(2+)and the cubic coordination structure of Cu^(+)occurs in the glass as the R value of the glass increases. This transformation can effectively reduce the concentration quenching phenomenon of Cu^(+)and improve the fluorescence luminescence intensity of the glass samples.Meanwhile, the samples were found to have luminescence tunability as well as good thermal stability.

文摘As a crucial component of terrestrial ecosystems, urban forests play a pivotal role in protecting urban biodiversity by providing suitable habitats for acoustic spaces.Previous studies note that vegetation structure is a key factor influencing bird sounds in urban forests;hence, adjusting the frequency composition may be a strategy for birds to avoid anthropogenic noise to mask their songs. However,it is unknown whether the response mechanisms of bird vocalizations to vegetation structure remain consistent despite being impacted by anthropogenic noise. It was hypothesized that anthropogenic noise in urban forests occupies the low-frequency space of bird songs, leading to a possible reshaping of the acoustic niches of forests, and the vegetation structure of urban forests is the critical factor that shapes the acoustic space for bird vocalization.Passive acoustic monitoring in various urban forests was used to monitor natural and anthropogenic noises, and sounds were classified into three acoustic scenes(bird sounds, human sounds, and bird-human sounds)to determine interconnections between bird sounds, anthropogenic noise, and vegetation structure.Anthropogenic noise altered the acoustic niche of urban forests by intruding into the low-frequency space used by birds, and vegetation structures related to volume(trunk volume and branch volume)and density(number of branches and leaf area index)significantly impact the diversity of bird sounds. Our findings indicate that the response to low and high frequency signals to vegetation structure is distinct.By clarifying this relationship, our results contribute to understanding of how vegetation structure influences bird sounds in urban forests impacted by anthropogenic noise.

文摘The calcium aluminosilicate-based glasses(CaO-Al_(2)O_(3)-SiO_(2),CAS)with different Fe_(2)O_(3)content(0.10wt%,0.50wt%,0.90wt%,and 1.30wt%)were prepared by traditional melt-quenching method. The glass network structure, thermal and mechanical properties, and crystallization behavior changes were investigated by nuclear magnetic resonance spectrometer,Fourier-transform infrared spectro-photometer,X-ray diffractometer, differential scanning calorimetry and field emission scanning electron microscope measurements. The change of Q^(n)in glass structures reveals the glass network connectivity decreases due to the increasing content of Fe_(2)O_(3)addition,resulting in the increasing of non-bridging number in glass structure. The glass densities slightly rise from 2.644 to 2.681 g/cm^(3),while Vickers’s hardness increases at first, from 6.469 to 6.901 GPa,then slightly drops to 6.745 GPa,with Fe_(2)O_(3)content increase. There is almost no thermal expansion coefficient change from different Fe_(2)O_(3)content.The glass transmittance in visible range gradually decreases with higher Fe_(2)O_(3)content,resulting from the strong absorption of Fe^(2+)and Fe^(3+)ions.The calculated activation energy from thermal analysis results first decreases from 282.70 to 231.18 kJ/mol,and then increases to 244.02 kJ/mol,with the Fe_(2)O_(3)content increasing from 0.10wt%to 1.30wt%.Meanwhile,the maximum Avrami constant of 2.33 means the CAS glasses exhibit two-dimensional crystallization. All of the CAS glass-ceramics samples contain main crystal phase of anorthite, the microstructure appears lamellar and columnar crystals.

文摘The Yellow Sea(YS)and East China Sea(ECS)are highly dynamic marginal seas of the northwestern Pacific Ocean. To gain an in-depth understanding of zooplankton community structure, zooplankton abundance, biovolume, and size structure in summer 2017 in the YS and ECS were assessed using ZooScan imaging analysis.Zooplankton abundance and biovolume ranged 2.94–1187.14 inds./m^(3)and 3.13–3438.51 mm^(3)/m^(3),respectively.Based on the biovolume data of the categorized size classes of 26 identified taxonomic groups, the zooplankton community was classified into five groups, and each group was coupled with distinctive oceanographic features.Under the influence of the Yellow Sea Cold Water Mass, the Yellow Sea offshore group featured the lowest bottom temperature(10.84±3.42℃)and the most abundant Calanoids(mainly in the 2–3 mm size class). In the Yellow Sea inshore group,Hydrozoans showed the largest biovolume and dominated in the 3–4-mm and>5-mm size classes. The East China Sea offshore group, which was affected by the Kuroshio Branch Current, featured high temperature and salinity, and the lowest bottom dissolved oxygen(2.58±0.5 mg/L).The lowest values of zooplankton abundance and biovolume in the East China Sea offshore group might be attributed to the bottom dissolved oxygen contents. The East China Sea inshore group, which was mainly influenced by the Zhejiang-Fujian Coastal Current and Changjiang Diluted Water, was characterized by high chlorophyll a and the largest biovolume of carnivorous Siphonophores(280.82±303.37 mm^(3)/m^(3)).The Changjiang River estuary offshore group showed the most abundant Cyclopoids, which might be associated with the less turbid water mass in this region.Seawater temperature was considered the most important factor in shaping the size compositions of Calanoids in different groups.

文摘In an effort to clarify the formation mechanism of LPSO structure in Mg-Y-Zn alloy, the chemical environment and structural ordering in liquid Mg-rich Mg-Y-Zn system are investigated with the aid of ab-initio molecular dynamics simulation. In liquid Mg-rich Mg-Y alloys, the strong Mg-Y interaction is determined, which promotes the formation of fivefold symmetric local structure. For Mg-Zn alloys, the weak Mg-Zn interaction results in the fivefold symmetry weakening in the liquid structure.Due to the coexistence of Y and Zn,the strong attractive interaction is introduced in liquid Mg-Y-Zn ternary alloy, and contributes to the clustering of Mg,Y,Zn launched from Zn.What is more, the distribution of local structures becomes closer to that in pure Mg compared with that in binary Mg-Y and Mg-Zn alloys. These results should relate to the origins of the Y/Zn segregation zone and close-packed stacking mode in LPSO structure, which provides a new insight into the formation mechanism of LPSO structure at atomic level.

文摘Indicating the tectonic features of the Hanshan-Wuwei basin can reconstruct the framework of the basins formed in Mesozoic and further understand the Mesozoic tectonic evolution of the South China Block.Studies on surface structure, regional stress field and deep geophysical characteristics of the Mesozoic Hanshan-Wuwei basin in Lower Yangtze region were carried out.NE-NNE trending folds and faults developed in the northern margin of the basins. The reconstruction of tectonic stress fields indicates four stress stages dominating the basins'evolution including NW-SE compression,N-S compression,NW-SE extension and NWW-SEE compression.2D seismic profiles reveal coexistence of thrust,strike-slip and normal faults in the basin.Combined with regional geological studies, the geodynamic processes for the formation of the Hanshan-Wuwei basin can be divided into five stages:1)During the Late Triassic,EW trending foreland basin was formed by N-S compression;2)From Mid-Jurassic to Late Jurassic, continuous compression strengthened the foreland deformation and formed thrust nappes. In this stage, the integrated foreland basin was compartmentalized or fragmented, and transferred to the broken foreland basin;3)NE-trending sinistral strike-slip movement at the beginning of the Early Cretaceous;4)Regional extension resulted in normal faults and rift basins developing in the Late Cretaceous;5)The NWW-SEE compression at the end of the Late Cretaceous caused NW sinistral strike-slip faults to form, which partly transformed the rift basin.
