Mapping of QTLs for Dehiscence Length at Basal Part of Thecae in Rice Based on TD70/Kasalath RIL Population

A population of 240 recombinant inbred lines (RIL) was derived from a cross between Japonica rice cultivar TD70 and Indica cultivar Kasalath, and their linkage map including 141 SSR markers was used to locate the QTLs controlling the length of dehiscence at the basal part of thecae (LDBT). Then, the correlations between LDBT and other agronomic traits of rice were analyzed. Two QTLs con- trolling LDBT were mapped on chromosome 6 and 8, respectively, qLDBT6 was mapped between RM3 and RM3628 markers on chromosome 6 with a LOD value of 3.23, contribution rate of 6.18% and additive effect of 17.74%. qLDBT8 was mapped between RM1376-RM4085 markers on chromosome 8 with a LOD value 2.95, contribution rate of 8.00% and additive effect of 20.09%. This domain probably had a relationship with the sterility of rice. The LDBT of RILs had extremely signifi- cant positive correlations with flag leaf width, 1 000-grain weight, grain width, grain length and grain thickness, but no significant correlations with other agronomic traits, such as plant height, heading date, tiller number, flag leaf length, average and the maximum temperature during heading period or rice. 展开更多

Observation on human ovarian theca cell and granulosa cell interaction in polycystic ovary syndrome

Objective: To investigate the role of human theca cell(TC) and granulo sa cell(GC) interaction and insulin(INS) in steroidogenesis in normal ovarian cy cle and in patients with PCOS. Methods: Ovarian theca and granulosa cells from eleven normal wo men and eight PCOS patients were co-cultured on opposite side of collagen with or without INS. The concentrations of estradiol(E2), progesterone(P) and andro stenedione (A) in the culture medium were examined by ELISA method.Results: When co-cultured with GC, TC in PCOS group produced mo re A and less P than those of normal group. When co-cultured with theca cells, granulosa cells in PCOS group produced more E2 than those of normal group. Add ition of INS increased the difference significantly.Conclusions: The GC and TC interaction from the normal and PCOS ovaries is different. There is a high A and high E2 intraovary loop of PC OS leading to premature arrest of follicle growth and anovulation. Insulin may p lay an important regulatory role. 展开更多

Mammalian follicles are composed of oocytes,granulosa cells,and theca cells.Theca cells form in the secondary follicles,maintaining follicular structural integrity and secreting steroid hormones.Two main sources of theca cells exist:Wilms tumor 1 positive(Wt1+)cells native to the ovary and Gli1+mesenchymal cells migrated from the mesonephros.Normal folliculogenesis is a process where oocytes,granulosa cells,and theca cells constantly interact with and support each other through autocrine and paracrine mechanisms.The proliferation and differentiation of theca cells are regulated by oocyte-derived factors,including growth development factor 9 and bone morphogenetic protein 15,and granulosa cell-derived factors,including desert hedgehog,Indian hedgehog,kit ligand,insulin-like growth factor 1,as well as hormones such as insulin and growth hormones.Current research on the origin of theca cells is limited.Identifying the origin of theca cells will help us to systematically elaborate the mechanisms of follicular formation and development. 展开更多
