The isolated porcine granulosa and thecal cells were grown on both sides of the amnion membrane, with granulosa cells in the inner chamber and thecal cells in the outer chamber. The concentrations of estradiol (E,), progesterone (P) and testosterone (T) in the culture media were mea-sured by radioimmunoassay.Results: The growth of both cells and their steroidogenic function were more active in amnion dual chamber system than in cellulose dual chamber system: (1) more T produced by thecal cells in the outer chamber, passing into inner chamber through the amnion membrane. T was used by granulosa cells as the substrate of aromatization, so that granulosa cells produced more E2 (up to 2 435 pmol/L); (2) the production of P (52. 5 μmol/L) and T (10. 2μmol/L) by granulosa cells cultured in the amnion mem-brane dual chamber system were also higher.Conclusions:The dual chamber system made of amnion mpmbrane showed better effect in studying steroidogenesis than with cellulose dual chamber system, and can be used as a model for studying paracrine regulatory interaction between granulosa and thecal cells in vitro.展开更多

Ovarian thecal metaplasia of the adrenal gland is a rare tumorlike ...Beckwith-Wiedemann syndrome(BWS) is an overgrowth syndrome associated with increased risk to develop malignancies including adrenocortical carcinoma. Ovarian thecal metaplasia of the adrenal gland is a rare tumorlike mesenchymal lesion in BWS patients that lacks detailed radiological description. We report a 17-yearold female patient with BWS, associated with bilateral Wilms tumor, hepatic hemangiomatosis, pancreatic neuroendocrine tumor, and a phyllodes tumor of the right breast. Surveillance abdominal ultrasound identified a right adrenal mass that was further characterized by computed tomography and magnetic resonance imaging. Radiologically, this mass displayed features that overlap with adrenocortical carcinoma and pheochromocytoma but after pathological examination this proved to be an ovarian thecal metaplasia of the adrenal gland. Adrenal masses in BWS should raise the suspicion for adrenocortical carcinoma though other adrenal tumors including ovarian thecal metaplasia can be seen in these patients.展开更多

Isolated porci...In order to investigate the mechanism of progestin and antiprogestin in the regulation of ovarian steroidogenesis, a dual chamber culture system was prepared with the amnion membrane of human placenta. Isolated porcine granulosa and thecal cells from 4~6 mm diameter follicles were grown on both sides of the amnion, respectively, and co cultured with or without LNG and RU486. After 48 h incubation, the mRNAs of FSH receptor (FSH R) and LH receptor (LH R) of both cells were observed by in situ hybridization. The results showed that granulosa cells expressed both FSH R mRNA and LH R mRNA, while thecal cells expressed LH R mRNA only. Under the stimulation of FSH, both LNG and RU486 increased FSH R mRNA expression of granulosa cells. Under the stimulation of LH, LNG enhanced LH R mRNA expression of thecal cells; while RU486 decreased its expression. When granulosa and thecal cells were exposed to FSH and LH both, the actions of LNG and RU486 in thecal cells showed the same result as that stimulated by LH alone. In granulosa cells LNG decreased LH R mRNA expression, while RU486 increased its expression. These data suggest that: (1) granulosa cells expressed FSH R mRNA significantly; (2) both the progestin and antiprogestin directly acted on the mRNA expression of gonadotropin receptors of ovarian cells, but effects were different; (3) the response of granulosa or thecal cells to the action of LNG and RU486 was not the same. A small armed dinoflagellate bloomed in the aquaculture ponds off the coast of Liaodong Bay, Bohai Sea of China, resulting in heavy mortalities of the cultured prawns (Penaeusjaponicus) and larvae of Chinese mitten ...A small armed dinoflagellate bloomed in the aquaculture ponds off the coast of Liaodong Bay, Bohai Sea of China, resulting in heavy mortalities of the cultured prawns (Penaeusjaponicus) and larvae of Chinese mitten handed crabs (Eriocheir sinensis). The bloom-forming species was successfully isolated, and cellular morphology of the specimen was consequently investigated through light, fluorescent and electron microscopy. The small ((14.4±1.6) μm in length) ellipsoid cells show typical Heterocapsa thecal plate arrangement (Po, cp, 5′, 3a, 7″, 6c, 5s, 5′″, 2″″). The episome is evidently bigger than the hyposome. One to three spherical pyrenoids are located above or beside the large elongated nucleus. The body scale is characterized by a triangle basal plate with one central upright and nine peripheral spines. Above all, Heterocapsa bohaiensis could be distinguished from other Heterocapsa species by the combination of the cell size, morphology, cellular structure and body scale. Sequence analyses of both ITS and LSU regions reveal the significant genetic divergence between H. bohaiensis and other established species in this genus, further supporting novelty of this species. Noticeably, different sample treatment methods resulted in morphological variation of the apical pore complex (APC) of H. bohaiensis, which needs to be taken into account in future study.展开更多
