Tyrosine was c...Through the method of initial heat release rate, the kinetic property of tyrosine oxidation catalyzed by tyrosinase from Pseudomonas maltophilia was investigated using a LKB-2107 batch microcalorimeter. Tyrosine was catalyzed and oxidized into L-dopa, then into melanin catalyzed by tyrosinase. We found that the tyrosinase reaction obeyed the Michaelis-Menten kinetics, and at 298.15K and pH 7.0, the initial exothermic rate (W0 ) are in the range of 0.1567~0.5704 mJ·s-1, the maximum exothermic rate (Wmax) are in 0.4152 ~ 0.8143mol·L-1, and mean value of the Michaelis constant (Km) is 2.199±0.105104 mol·L-1.展开更多

T...The differential scanning calorimetric (DSC)curves of the mitochondria isolated from two varieties of sporophyte cytoplasmic male sterile and their fertile lines of Yie Bai and Ma Xie type rice have been determined. The curves show that the energy is released continuously as temperature rise to 70℃. Some thermodynamic and kinetic parameters of the energy release of the mitochondria have been obtained. The presented results showed that the mitochondria from cytoplasmic male sterile rice released more heat and they had higher energy barrier, less rate, and more complicated mechanism than that of their fertile lines in the energy release process.展开更多

The process of the reaction has been analyzed from changes in energy by using the transition state theory. Two methods for enhancing catalytic power of Laccase are proposed. The results shown that formation of an enzyme substrate complex is“anticatalytic”. In this paper, the effect of Na_2SeO_3 on Bacillussubtilis growth was investigated at 37℃ by microcalorimetry. The relationship between growth rateconstants (k) and concentration of Na_2SeO_3(c) shows a logarithmic normal distribution, and IC_(50)is 20.3 μg/mL. All these thermokinetic information is readily obtained by an LKB 2277-204 heatconduction microcalorimeter. The standard enthalpy of formation of 32Cr(His)+(aq) and 22M(His)+(aq) (M=Mn, Fe, Co, Ni and Cu) were calculated. Based on the thermodynamic and kinetic equations of the reactions, three thermody-namic parameters (the activation enthalpy, the activation entropy, the activation free energy), the rate constants, and three kinetic parameters (the apparent activation energy, the pre-exponential constant and the reaction order) are obtained. The solid complexes of CrCl3, MnCl2, FeCl2, CoCl2, NiCl2 and CuCl2 with histidine were prepared and identified as Cr(His)2Cl3H2O, Mn(His)2Cl24H2O, Fe(His)2Cl2H2O, Co(His)2Cl2H2O, Ni(His)2Cl2H2O and Cu(His)2Cl2H2O by chemical and elemental analyses. The bonding characteristics of these complexes were char-acterized by IR as well. The results showed that, with the atomic number increasing, three thermodynamic parame-ters, GD!, HD! and SD! of the complexation reaction of these metal chlorides with L-a-histidine in water present an analogy regularity.展开更多
Thermokinetics of the dismutation of superoxide anion (O ·- 2) catalyzed by superoxide dismutase (SOD), in which the decomposition of hydrogen peroxide catalyzed by catalase is utilized as a source of oxygen and ...Thermokinetics of the dismutation of superoxide anion (O ·- 2) catalyzed by superoxide dismutase (SOD), in which the decomposition of hydrogen peroxide catalyzed by catalase is utilized as a source of oxygen and the autoxidation of pyrogallol as a source of the substrate (O ·- 2), has been studied by microcalorimetry. The molar reaction enthalpies of the SOD reaction and the pyrogallol autoxidation have been measured as -160.1 and -218 kJ·mol -1, respectively, at 298.15 K and pH 7.0. A novel method for the determination of SOD activity, the microcalorimetric assay for SOD, has been proposed. The experimental results showed that SOD had no effect on both kinetic parameters and the mechanism for the pyrogallol autoxidation. This autoxidation followed the second_order reaction kinetics in the presence of limited oxygen (the first order with respect to both pyrogallol and O 2), and the second-order rate constants were determined at 298.15 K and pH 8.0 to be 1.25 and 1.30 L·mol -1·s -1 in the absence and presence of SOD, respectively. A possible mechanism for the autoxidation of pyrogallol inhibited by SOD was also suggested.展开更多

The reaction that arginase-catalyzed hydrolysis of L-arginine to L-ornithine and urea and the inhibition of this reaction by the product, L-ornithine, and exogenous L-lysine were studied at 37 ℃ in 40 mmolL-1 sodium barbiturate-HCl buffer solution (pH=9.4). Michealis con-stant Km for arginine and maximum velocity Vm of the reaction were determined to be 5.14 mmolL-1 and 1.13× 10-2 mmolL-1s-1, respectively. The product inhibition constant KP and inhibitory constant KI of L-lysine were de-termined to be 1.18 and 5.6 mmolL-1, respectively. All the results have better repeatability and self-consistency and are in agreement with literature values. This new method using more direct thermal information from the proc-ess would give more reliable kinetic information than the traditional initial rate method.展开更多

The solid complexes of CrCl3, MnCl2, FeCl2, CoCl2, NiCl2 and CuCl2 with histidine were prepared and identified as Cr(His)2Cl3H2O, Mn(His)2Cl24H2O, Fe(His)2Cl2H2O, Co(His)2Cl2H2O, Ni(His)2Cl2H2O and Cu(His)2Cl2H2O by chemical and elemental analyses. The bonding characteristics of these complexes were char-acterized by IR as well. The results showed that, with the atomic number increasing, three thermodynamic parame-ters, GD!, HD! and SD! of the complexation reaction of these metal chlorides with L-a-histidine in water present an analogy regularity.
文摘Thermokinetics of the dismutation of superoxide anion (O ·- 2) catalyzed by superoxide dismutase (SOD), in which the decomposition of hydrogen peroxide catalyzed by catalase is utilized as a source of oxygen and the autoxidation of pyrogallol as a source of the substrate (O ·- 2), has been studied by microcalorimetry. The molar reaction enthalpies of the SOD reaction and the pyrogallol autoxidation have been measured as -160.1 and -218 kJ·mol -1, respectively, at 298.15 K and pH 7.0. A novel method for the determination of SOD activity, the microcalorimetric assay for SOD, has been proposed. The experimental results showed that SOD had no effect on both kinetic parameters and the mechanism for the pyrogallol autoxidation. This autoxidation followed the second_order reaction kinetics in the presence of limited oxygen (the first order with respect to both pyrogallol and O 2), and the second-order rate constants were determined at 298.15 K and pH 8.0 to be 1.25 and 1.30 L·mol -1·s -1 in the absence and presence of SOD, respectively. A possible mechanism for the autoxidation of pyrogallol inhibited by SOD was also suggested.

文摘A double-parameter theoretical model for the conduction calorimeter, in which the delay of sensing has been considered, is suggested, and a simple method for the determination of the parameters of the model is proposed. The experimental results show that on the basis of this model, the phenomenon of thermogram maximum in thermogram reconstruction can be eliminated, and it can be applied to the thermokinetic investigations of faster thermal change processes.

文摘A new thermokinetic reduced extent method for studying of the reversible competitive inhibition of single sub-strate enzyme-catalyzed reactions was proposed in this paper. The reaction that arginase-catalyzed hydrolysis of L-arginine to L-ornithine and urea and the inhibition of this reaction by the product, L-ornithine, and exogenous L-lysine were studied at 37 ℃ in 40 mmolL-1 sodium barbiturate-HCl buffer solution (pH=9.4). Michealis con-stant Km for arginine and maximum velocity Vm of the reaction were determined to be 5.14 mmolL-1 and 1.13× 10-2 mmolL-1s-1, respectively. The product inhibition constant KP and inhibitory constant KI of L-lysine were de-termined to be 1.18 and 5.6 mmolL-1, respectively. All the results have better repeatability and self-consistency and are in agreement with literature values. This new method using more direct thermal information from the proc-ess would give more reliable kinetic information than the traditional initial rate method.
