Background:Knee osteoarthritis(KOA)characterized by degeneration of knee cartilage and subsequent bone hyperplasia is a prevalent joint condition primarily affecting aging adults.The pathophysiology of KOA remains poorly understood,as it involves complex mechanisms that result in the same outcome.Consequently,researchers are interested in studying KOA and require appropriate animal models for basic research.Chinese herbal compounds,which consist of multiple herbs with diverse pharmacological properties,possess characteristics such as multicomponent,multipathway,and multitarget effects.The potential benefits in the treatment of KOA continue to attract attention.Purpose:This study aims to provide a comprehensive overview of the advantages,limitations,and specific considerations in selecting different species and methods for KOA animal models.This will help researchers make informed decisions when choosing an animal model.Methods:Online academic databases(e.g.,PubMed,Google Scholar,Web of Science,and CNKI)were searched using the search terms“knee osteoarthritis,”“animal models,”“traditional Chinese medicine,”and their combinations,primarily including KOA studies published from 2010 to 2023.Results:Based on literature retrieval,this review provides a comprehensive overview of the methods of establishing KOA animal models;introduces the current status of advantages and disadvantages of various animal models,including mice,rats,rabbits,dogs,and sheep/goats;and presents the current status of methods used to establish KOA animal models.Conclusion:This study provides a review of the animal models used in recent KOA research,discusses the common modeling methods,and emphasizes the role of traditional Chinese medicine compounds in the treatment of KOA. 展开更多

Background:As of 2023,coronavirus disease 2019(COVID-19)is still spreading globally.Therefore,we aim to integrate non-critical COVID-19 high-frequency and high-targeting Chinese medicines to provide a reference for clinical prescriptions to improve COVID-19-related symptoms.Materials and methods:The information on non-critical COVID-19 high-frequency Chinese medicines in the diagnosis and treatment of COVID-19 was obtained by the TCM inheritance support platform.Using network pharmacology and molecular docking technology,high-targeting Chinese medicines with good docking activity with COVID-19 receptors angiotensin-converting enzyme-II(ACE2),3CLpro and tyrosine-protein kinase receptor UFO(AXL)were obtained.A new prescription for non-critical COVID-19 was established by integrating high-frequency and high-targeting Chinese medicines.Rats with acute lung injury induced by lipopolysaccharide were used as the experimental model.The histopathological changes in the lungs of rats in each group were observed by hematoxylin-eosin staining.The lung coefficient of rats was measured.The levels of IL-6,TNF-α,and IL-1βin serum were detected by enzyme-linked immunosorbent assay.The mRNA and protein levels of ACE2 and AXL in lung tissue were detected by real-time quantitative polymerase chain reaction and western blot.Results:Through data mining,it was found that there were 39 high-frequency traditional Chinese medicines for non-critical COVID-19 in the diagnosis and treatment guidelines.According to network pharmacology and molecular docking,30 highly targeted traditional Chinese drugs for COVID-19 were found.The new prescriptions for non-critical COVID-19 were comprehensively obtained,including Glycyrrhizae Radix,Ephedra Herba,Amygdalus Communis Vas,Gypsum Fibrosum,Descurainiae Semen,Atractylodes Lancea,Scutellariae Radix,Amomum Tsao-Ko Crevostet,Forsythiae Fructus,Pogostemon cablin,Magnolia Officinalis.Compared with the LPS-induced lung injury model group,the medium dose of the new prescription group had significantly alleviated pathological changes in lung tissue,decreased lung coefficient,decreased contents of IL-6,TNF-αand IL-1β,and increased mRNA and protein expression of ACE2 and AXL(P<0.05).Conclusion:Based on data mining,network pharmacology and molecular docking technology,the new prescription for non-critical COVID-19 established by this method has an anti-inflammatory effect on rats with acute lung injury induced by lipopolysaccharide and can provide a reference for clinicians to alleviate the symptoms related to non-critical COVID-19. 展开更多

OBJECTIVE In recent years,the frequency of hyperuricemia has gradually risen along with the improvement of living standards,the irregular of diet and overfeeding greasy and surfeit flavor.However,hyperuricemia is a disorder of purine metabolism,and is strongly associated with insulin resistance and abnormal glucose metabolism.It is important to obtain a more stable and sustained animal model for the efficacy evaluation of traditional Chinese medicine(TCM).METHODS To manufacture the rodent model of hyperuricemia,the theory of increasing the source of the uric acid,reducing uric acid excretion and inhibiting uricase were used.We observed the influence on the serum uric acid and other indicators of rats induced by some factors:the lipid emulsion,high purine diet,beer with sugar,beer with sugar and high purine,and so on.Then we choose one of the stable and sustained animal models,studying the effects of Plo.E(which extracted from a TCM)on modulating the level of serum uric acid and the preliminary mechanism in this abstract.RESULTS ① At the 2nd week,the level of serum UA,BUN,Cr,TC,LDL-c of rats in the lipid emulsion group raised significantly.② At the 6th weeks,the serum UA in both the high purine diet group and lipid emulsion group raised obviously.③ The effects of Plo.E on hyperuricemia rats induced by high purine diet:After 8d administration,the Plo.E(three dosages)can reduce the UA level,and the middle and low dosages can reduce the TG level.After 25 dadministration,Plo.E can reduce the plasma viscosity,UA,TG,the whole blood viscosity level,the high dosage also can reduce the TC level.CONCLUSION The rats induced by the high purine diet and lipid emulsion can raised the serum UA obviously,while the way of lipid emulsion is earlier and more stable.Plo.E has a therapeutic effect on hyperuricemia,with an activity of reducing plasma viscosity and blood lipid.The above models conform to the pathogenesis of humans,can be used to study the causes and pathogenesis of hyperuricemia complicating metabolic disorder and the related treatment drug screening. 展开更多

Although the animal models of hepatic fibrosis developed by former researchers have pathological changes of hepatic fibrosis, they do not include charac- teristics of important TCM syndromes such as stagnation of qi, deficiency of qi, liver depression, phlegm-dampness and blood stasis because of single-factor model- ing. Animal models of hepatic fibrosis and animal models integrated disease and syndrome were reviewed, and several new types of integrated disease and syndrome animal models constructed by multiple-factor modeling method were evaluated, under the guidance of etiological theory of TCM. This kind of hepatic fibrosis model animals has dual characteristics of disease and syndrome. It is consistent with pathological characteristics of hepatic fibrosis in western medicine when replicating the basic characteristics accorded with TCM syndrome. Thus, the pathogenesis and pathogenic process of clinical disease and syndrome formation is simulated more ac- curately, providing a new platform and pathway for studying hepatic fibrosis disease with integrated traditional Chinese and western medicine. 展开更多

Effect of traditional Chinese medicine on anthracycline-induced cardiotoxicity in animal models:A systematic review and metaanalysis

Objective:To evaluate the effect of traditional Chinese medicine(TCM)on anthracycline-induced cardiotoxicity(AIC)in animal models.Methods:Separate systematic searches for preclinical studies were performed in the PubMed,EMBASE,Web of Science,Chinese National Knowledge Infrastructure,Chinese Biomedical Database,Chinese Scientific Journal Database,and Wanfang Data from inception to August 2019.The primary outcomes were echocardiography,serum assays for myocardial enzymograms,histological assessments,and electrocardiograms.The secondary outcomes mainly included body weight and safety evaluations.The protocol is registered on PROSPERO(CRD42019145819).RevMan(V.5.3)was used for meta-analysis.Results:We identified 10 studies from 9 international scientific publications describing the efficacy of TCM on AIC animal models.All the included studies reported that,compared with animal model without any intervention,TCM significantly improved ventricular function,cardiac biomarkers,electrocardiograph results,and cardiac fibrosis.Improved survival rates and body mass indices were also observed with TCM.We further pooled the available data from four studies(63 animals)for the meta-analysis and the results showed that,compared with models without any intervention,TCM significantly increased the ejection fraction by 14.13%(95%CI,9.96e18.29)and fraction shortening by 8.66%(95%CI,6.05 e11.26).Creatine kinase-MB(SMD=2.49,95%CI:-3.12 to-1.85)and lactate dehydrogenase(SMD=-2.78,95%CI:-3.45 to-2.12)were also significantly decreased by TCM.Conclusions:TCM is effective in improving AIC in animal models and has tremendous potential to be translated to treat AIC in clinical practice.Additionally,the systematic review and meta-analysis of animal experiments may be valuable in enhancing and guiding animal experiments and promoting the transformation of the results. 展开更多

Alzheimer’s disease is a complex neurodegenerative ailment accompanied by cognitive problems and progressive memory loss.The main causes of the disease are indistinguishable even though aging is said to be the key risk factor,along with the hypotheses such asβ-amyloid plaques,tau-proteins,neuroinflammation,and oxidative stress.Currently,Alzheimer’s disease is not curable,and the available medicines are only used to provide symptom relief.Scientists and researchers have developed numerous models to study Alzheimer’s disease,such as in silico methods,in vitro models of cultured rat cortical neurons,in vivo rodent models involving mice and rats,and non-rodent models as well.This review provides an insight into the modality of Alzheimer’s disease treatment using medicinal plants that are natural sources with abundant bioactive substances,and traditional formulations such as an extract from Notoginseng Radix et Rhizoma,Chinese herbal formulations(i.e.,traditional Chinese medicine kidney-nourishing formula,supplements such as coenzyme Q10,alpha-lipoic acid,Ginkgo Biloba,omega-3’s,and acetyl-L-carnitine).In addition,we have discussed the evaluation parameters for experimental Alzheimer’s disease,which are used to assess the mechanism of memory and pharmacological effects of molecules for drug discovery and development.The common experimental analysis methods for Alzheimer’s disease include behavioral,biochemical,and histological studies.This work will allow researchers to access an informative reference of traditional medicines along with experimental analysis methods in one place for Alzheimer’s disease research. 展开更多

目的观察乳痛愈药膜外敷对乳腺增生症模型大鼠的作用。方法将55只大鼠按照随机数字表法分为5组,即空白组、模型组、三苯氧胺软膏组、散结乳癖贴膏组和乳痛愈药膜组,每组11只。除空白组外,其余各组建立乳腺增生模型。造模成功后,空白组... 目的观察乳痛愈药膜外敷对乳腺增生症模型大鼠的作用。方法将55只大鼠按照随机数字表法分为5组,即空白组、模型组、三苯氧胺软膏组、散结乳癖贴膏组和乳痛愈药膜组,每组11只。除空白组外,其余各组建立乳腺增生模型。造模成功后,空白组、模型组大鼠于第2对乳腺部位外用空白赋形剂0.1 g/只,三苯氧胺软膏组于第2对乳腺部位外用三苯氧胺软膏0.1 g/只,散结乳癖贴膏组予散结乳癖贴膏0.4 g/只(相当于原散结乳癖贴膏0.1 g),乳痛愈药膜组予乳痛愈药膜0.8 g/只(相当于生药0.8 g)。观察各组大鼠乳头直径、高度变化;比较各组大鼠脾脏指数、胸腺指数;比较各组大鼠血清雌二醇(E2)、孕酮(P)、催乳素(PRL)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平变化;观察各组大鼠乳腺组织病理形态;比较各组大鼠乳腺组织雌激素受体α(ERα)、ERβ和孕激素受体(PR)表达。结果与空白组比较,模型组乳头直径、乳头高度均增加(P<0.05);与模型组比较,三苯氧胺软膏组、散结乳癖贴膏组、乳痛愈药膜组乳头直径、乳头高度均减小(P<0.05);三苯氧胺软膏组、散结乳癖贴膏组、乳痛愈药膜组乳头直径、乳头高度比较差异均无统计学意义(P>0.05)。与空白组比较,模型组脾脏指数、胸腺指数均降低(P<0.05);与模型组比较,各给药组大鼠脾脏指数、胸腺指数均升高(P<0.05);三苯氧胺软膏组、散结乳癖贴膏组、乳痛愈药膜组大鼠脾脏指数、胸腺指数比较差异无统计学意义(P>0.05)。与空白组比较,模型组血清E2、PRL水平升高(P<0.05),P水平降低(P<0.05);与模型组比较,各给药组血清E2、PRL水平降低(P<0.05),P水平升高(P<0.05);散结乳癖贴膏组P水平高于三苯氧胺软膏组(P<0.05)。与空白组比较,模型组MDA水平升高(P<0.05),SOD水平降低(P<0.05);与模型组比较,各给药组血清MDA水平均降低(P<0.05),SOD水平均升高(P<0.05);与三苯氧胺软膏组比较,散结乳癖贴膏组、乳痛愈药膜组血清MDA水平均降低(P<0.05),SOD水平均升高(P<0.05)。与空白组比较,模型组大鼠乳腺组织ERα、ERβ、PR表达均升高(P<0.05);与模型组比较,各给药组大鼠乳腺组织ERα、ERβ表达均降低(P<0.05);各给药组大鼠乳腺组织ERα、ERβ、PR表达比较差异均无统计学意义(P>0.05)。结论乳痛愈药膜能改善乳腺增生症模型大鼠乳腺组织增生程度,调节血清性激素水平,调节自由基代谢和乳腺组织中雌激素受体(ERα、ERβ)表达,减弱雌激素(E2)的生物学效应。 展开更多

中医界运用现代科学方法进行相关研究已有多年历史,在病证动物模型上颇有收获。本文对脾虚证动物模型发展历程进行回顾,对动物选择、造模方法、评价指标、消化系统病证结合模型进行总结和评论。模型动物多选用以大鼠为代表的小型啮齿类... 中医界运用现代科学方法进行相关研究已有多年历史,在病证动物模型上颇有收获。本文对脾虚证动物模型发展历程进行回顾,对动物选择、造模方法、评价指标、消化系统病证结合模型进行总结和评论。模型动物多选用以大鼠为代表的小型啮齿类。目前,多基于病因病机造模,而较少使用化学药物与物理因素造模,复合因素造模法最常见。评价指标则分为中医证型指标、与“脾”功能和特性有关量化指标,以及方剂反证。功能性腹泻/消化不良为代表的脾虚证病证结合模型是目前研究的重心。脾虚证动物模型研究成绩斐然的同时仍存在许多不足,如缺乏脾虚证模型行业标准和共识,评价指标缺乏系统性和整体性,脾虚理论的现代阐释缺乏统一,学界亟需解决上述问题以促进发展。 展开更多
