The results showed that cry1A had been transmitted to progeny of transgenic maize as a single gene. Contents of cry1A insecticidal protein were significantly different among transgenic maize lines and various tissues of the same transgenic lines. High expression of cry1A protein occurred in green tissues, such as leaf and husk leaf, and low expression occurred in pith, tassel, ear pith, pollen and silk. The results also showed that the contents of cry1A insecticidal protein in leaves of transgenic maize increased with the advance of development and there was no significant difference in cry1A expression level among various generations of transgenic maize.展开更多

By Basta painting, PCR analysis and determining of the inhibitory trypsin activity, the results show that bar gene and pinII gene in rice F2 population fit the simple Mendel's low of inheritance and close linkage, but a few plants in F2 have not sufficiently expressed. The wound inducible pin II gene has an expression regulated spatially and temporally, and the signal transduction pathway is not only upward, but also downward. We examined 19-month-old NR2B transgenic mice and their littermate controls. First, we detected expression of the NR2B subunit gene, Grin2b, in the neocortex of transgenic mice using real-time PCR. Next, we used microarrays to examine differences in neocortical gene expression. Pathway and signal-net analyses identified multiple pathways altered in the transgenic mice, in-cluding the P53, Jak-STAT, Wnt, and Notch pathways, as well as regulation of the actin cytoskeleton and neuroactive ligand-receptor interactions. Further signal-net analysis highlighted the P53 and insulin-like growth factor pathways as key regulatory pathways. Our results provide new insight into understanding the molecular mechanisms of NR2B regulated age-related memory storage, normal organismal aging and age-related disease.展开更多

Here,we establi...The mammary gland provides a novel method for producing recombinant proteins in milk of transgenic animals. A key component in the technology is the construction of an efficient milk expression vector. Here,we established a simple method to construct a milk expression vector, by a combination of homologous recombination and digestion-ligation. Our methodology is expected to have the advantages of both plasmid and bacterial artificial chromosome(BAC) vectors. The BAC of mouse whey acidic protein gene(mWAP) was modified twice by homologous recombination to produce a universal expression vector, and the human lysozyme gene(hLZ) was then inserted into the vector by a digestionligation method. The final vector containing the 8.5 kb mWAP 5′ promoter, 4.8 kb h LZ genomic DNA, and 8.0 kb m WAP 3′ genomic DNA was microinjected into pronuclei of fertilized mouse embryos, to successfully generate two transgenic mouse lines that expressed recombinant human lysozyme(rhLZ) in milk. The highest expression level of rhLZ was 0.45 g$L–1, and rhLZ exhibited the same antibacterial activity as native h LZ. Our results have provided a simple approach to construct a universal milk expression vector, and demonstrated that the resulting vector regulates the expression of hLZ in milk.展开更多
目的观察C57BL/6N-Tg(1.28HBV)/Vst乙型肝炎病毒(hepatitis B virus,HBV)转基因(HBV-Tg)小鼠模型的特征,并分析HBV-Tg小鼠模型的转录组学特点。方法以10只雄性HBV-Tg小鼠为实验组,10只野生型小鼠为对照组。以血清HBV DNA、HBsAg、HBeAg...目的观察C57BL/6N-Tg(1.28HBV)/Vst乙型肝炎病毒(hepatitis B virus,HBV)转基因(HBV-Tg)小鼠模型的特征,并分析HBV-Tg小鼠模型的转录组学特点。方法以10只雄性HBV-Tg小鼠为实验组,10只野生型小鼠为对照组。以血清HBV DNA、HBsAg、HBeAg水平和肝组织HBsAg、HBcAg表达情况评价模型小鼠病毒学特征,检测血清ALT、AST,肝组织HE、天狼猩红染色及肝组织Hyp含量评估小鼠肝脏炎症及纤维化程度。两组各随机选择3例肝组织样本提取RNA进行高通量转录组测序。经R软件分析获得表达显著的差异基因,经GO和KEGG分析获得差异基因功能富集情况,再采用实时荧光定量PCR(RT-qPCR)对差异显著的基因进行验证。结果与正常组相比,模型组ALT、AST水平均有所升高,以ALT更为显著(P<0.05);模型组肝组织HE染色可见肝细胞核增大,个别肝细胞出现肿胀。天狼猩红染色结果显示,HBV转基因组汇管区及小叶间出现少量胶原沉积,呈细线状。通过筛选条件(|logFC|>2倍且P.adj<0.05)获得差异基因共计1352个,其中上调基因703个,下调基因649个。KEGG分析提示差异基因主要在PPAR信号通路、视黄醇代谢、脂肪酸降解等通路中富集(P.adj<0.05)。明显上调的差异基因主要有Cyp4a10、Cyp4a14、Acot1、Acot3、Ehhadh等,明显下调基因包括Scn5a、Apol10b、Igddc4、Cxcl1、9530077C05Rik等,经RT-qPCR验证趋势一致(P<0.05)。结论HBV-Tg小鼠具有自发纤维化的趋势,经转录组学分析,慢性乙型病毒性肝炎(chronic hepatits B,CHB)发生的潜在机制主要涉及PPAR信号通路、视黄醇代谢、脂肪酸降解、药物代谢等通路。展开更多

The kanamycin-resistant plants were obtained by Agrobacterium-mediated transformation system. It was shown that HBsAg gene was expressed in transgenic tobacco plants and their progenies by ELISA. The results showed that approximately 50% offsprings were transgenic positive. Foreign gene integrated in mouse chromosomes was intact. The hFIX expression of each mouse in the same strain was different, the content of hFIX in the milk was (43.32±5.41)μg/mL in FIX-33 transgenic strain and (1.16±0.45)μg/mL in FIX-124 transgenic strain. Meanwhile, the hFIX gene expression between the two strains was different remarkably (P<0.01). We conclude that the characteristics of inheritance and expression in the founder were able to be transferred to their offsprings stably.展开更多
