Add to these the undated fault system (4) in the South-West (SW) of the South China Sea (SCS). Here we offer a unified cause for all four of these in (5), an impact in the SCS of a large, massive cosmic object, likely a comet, obliquely coming from the SW at an extremely shallow angle, striking the Sunda shelf yet unexploded with the shock of its compressed air bow wave, and causing the continual shelf and slope to collapse, resulting in the fault system (4), then traveling almost tangentially to the surface, exploding at impact with the sea surface, ejecting the tektites (1), creating the formation underlying the later atolls of Spratlies Archipelago (6), Nansha Islands in Chinese, & causing the BMR (2). An explanation of event (3) was Richard Muller’s hypothesis of planet Earth passing through an interplanetary dust cloud periodically due to ecliptic precession. Here we hypothesize this cloud actually is a belt of Australasian tektites ejected into space at super-orbital velocities that Earth encounters about every 100 ka.展开更多

The closed algebraic equations have been deduced on the basis of the physically founded normal force ?depth3/2 relation. The precise transition onset position is obtained by linear regression of the FN = kh3/2 plot, where k is the penetration resistance, which also provides the axis cuts of both polymorphs of first order phase transitions. The phase changes can be endothermic or exothermic. They are normalized per μN or mN normal load. The analyses of indentation loading curves with self-similar diamond indenters are used as validity check of the loading curves, also from calibration standards that exhibit previously undetected phase-transitions and are thus incorrect. The phase-transition energies for fused quartz are determined from the loading curves from instrument provider handbooks. The anisotropic behavior of phase transition energies is studied for the first time. Quartz is a useful test object. The reasons for the packing-dependent differences are discussed on the basis of the local crystal structure under and around the inserting tip.展开更多

Furthermore, the dichotomy method is utilized to identify the critical points for thesephase transitions. Even for pipe flow, predicting the ...Flow transition from laminar to turbulent mode (and vice versa)—that is, the initiation of turbulence—is one of the most important research subjects in the history of engineering. Even for pipe flow, predicting the onset of turbulence requires sophisticated instrumentation and/or direct numerical simulation, based on observing the instantaneous flow structure formation and evolution. In this work, a local Reynolds number equivalence c (ratio of local inertia effect to viscous effect) is seen to conform to the Universal Law of the Wall, where c = 1 represents a quantitative balance between the abovementioned two effects. This coincides with the wall layer thickness (y+= 1, where y+ is the dimensionless distance from the wall surface defined in the Universal Law of the Wall). It is found that the characteristic of how the local derivative of c against the local velocity changes with increasing velocity determines the onset of turbulence. For pipe flow, c - 25, and for plate flow, c - 151.5. These findings suggest that a certain combination of c and velocity (nonlinearity) can qualify the source of turbulence (i.e., generate turbulent energy). Similarly, a re-evaluation of the previous findings reveals that only the geometrically narrow domain can act locally as the source of turbulence, with the rest of the flow field largely being left for transporting and dissipating. This understanding will have an impact on the future large-scale modeling of turbulence.展开更多

BACKGROUND CALD1 has been discovered to be abnormally expressed in a variety of malignant tumors,including gastric cancer(GC),and is associated with tumor progression and immune infiltration;however,the roles and mech...BACKGROUND CALD1 has been discovered to be abnormally expressed in a variety of malignant tumors,including gastric cancer(GC),and is associated with tumor progression and immune infiltration;however,the roles and mechanisms of CALD1 in epithe-lial-mesenchymal transition(EMT)in GC are unknown.AIM To investigate the role and mechanism of CALD1 in GC progression,invasion,and migration.METHODS In this study,the relationship between CALD1 and GC,as well as the possible network regulatory mechanisms of CALD1,was investigated by bioinformatics and validated by experiments.CALD1-siRNA was synthesized and used to trans-fect GC cells.Cell activity was measured using the CCK-8 method,cell migration and invasive ability were measured using wound healing assay and Transwell assay,and the expression levels of relevant genes and proteins in each group of cells were measured using qRT-PCR and Western blot.A GC cell xenograft model RESULTS Bioinformatics results showed that CALD1 was highly expressed in GC tissues,and CALD1 was significantly higher in EMT-type GC tissues than in tissues of other types of GC.The prognosis of patients with high expression of CALD1 was worse than that of patients with low expression,and a prognostic model was constructed and evaluated.The experimental results were consistent with the results of the bioinformatics analysis.The expression level of CALD1 in GC cell lines was all higher than that in gastric epithelial cell line GES-1,with the strongest expression found in AGS and MKN45 cells.Cell activity was significantly reduced after CALD1-siRNA trans-fection of AGS and MKN45 cells.The ability of AGS and MKN45 cells to migrate and invade was reduced after CALD1-siRNA transfection,and the related mRNA and protein expression was altered.According to bioinfor-matics findings in GC samples,the CALD1 gene was significantly associated with the expression of members of the PI3K-AKT-mTOR signaling pathway as well as the EMT signaling pathway,and was closely related to the PI3K-Akt signaling pathway.Experimental validation revealed that upregulation of CALD1 increased the expression of PI3K,p-AKT,and p-mTOR,members of the PI3K-Akt pathway,while decreasing the expression of PTEN;PI3K-Akt inhibitor treatment decreased the expression of PI3K,p-AKT,and p-mTOR in cells overexpressing CALD1(still higher than that in the normal group),but increased the expression of PTEN(still lower than that in the normal group).CCK-8 results revealed that the effect of CALD1 on tumor cell activity was decreased by the addition of the inhibitor.Scratch and Transwell experiments showed that the effect of CALD1 on tumor cell migration and invasion was weakened by the addition of the PI3K-Akt inhibitor.The mRNA and protein levels of EMT-related genes in AGS and MKN45 cells were greatly altered by the overexpression of CALD1,whereas the effect of overex-pression of CALD1 was significantly weakened by the addition of the PI3K-Akt inhibitor.Animal experiments showed that tumour growth was slow after inhibition of CALD1,and the expression of some PI3K-Akt and EMT pathway proteins was altered.CONCLUSION Increased expression of CALD1 is a key factor in the progression,invasion,and metastasis of GC,which may be associated with regulating the PI3K-Akt pathway to promote EMT.展开更多

The possibility of using transition radiation to study the surface of substances is discussed. In addition, due to the hard radiation present in space, this research may be important for the use of light monoatomic layers as a material for satellite antennas, “solar sails” and cover layers in a future space (interstellar) mission.展开更多
The general use of aluminium as an indentation standard for the iteration of contact heights for the determination of ISO-14577 hardness and elastic modulus is challenged because of as yet not appreciated phase-change...The general use of aluminium as an indentation standard for the iteration of contact heights for the determination of ISO-14577 hardness and elastic modulus is challenged because of as yet not appreciated phase-changes in the physical force-depth standard curve that seemed to be secured by claims from 1992. The physical and mathematical analyses with closed formulas avoid the still world-wide standardized energy-law violation by not reserving 33.33% (h2 belief) (or 20% h3/2 physical law) of the loading force and thus energy for all not depth producing events but using 100% for the depth formation is a severe violation of the energy law. The not depth producing part of the indentation work cannot be done with zero energy! Both twinning and structural phase-transition onsets and normalized phase-transition energies are now calculated without iterations but with physically correct closed arithmetic equations. These are reported for Berkovich and cubecorner indentations, including their comparison on geometric grounds and an indentation standard without mechanical twinning is proposed. Characteristic data are reported. This is the first detection of the indentation twinning of aluminium at room temperature and the mechanical twinning of fused quartz is also new. Their disqualification as indentation standards is established. Also, the again found higher load phase-transitions disqualify aluminium and fused quartz as ISO-ASTM 14577 (International Standardization Organization and American Society for Testing and Materials) standards for the contact depth “hc” iterations. The incorrect and still world-wide used black-box values for H- and Er-values (the latter are still falsely called “Young’s moduli” even though they are not directional) and all mechanical properties that depend on them. They lack relation to bulk moduli from compression experiments. Experimentally obtained and so published force vs depth parabolas always follow the linear FN = kh3/2 + Fa equation, where Fa is the axis-cut before and after the phase-transition branches (never “h2” as falsely enforced and used for H, Er and giving incorrectly calculated parameters). The regression slopes k are the precise physical hardness values, which for the first time allow for precise calculation of the mechanical qualities by indentation in relation to the geometry of the indenter tip. Exactly 20% of the applied force and thus energy is not available for the indentation depth. Only these scientific k-values must be used for AI-advises at the expense of falsely iterated indentation hardness H-values. Any incorrect H-ISO-ASTM and also the iterated Er-ISO-ASTM modulus values of technical materials in artificial intelligence will be a disaster for the daily safety. The AI must be told that these are unscientific and must therefore be replaced by physical data. Iterated data (3 and 8 free parameters!) cannot be transformed into physical data. One has to start with real experimental loading curves and an absolute ZerodurR standard that must be calibrated with standard force and standard length to create absolute indentation results. .展开更多

For the enhancement of heat transfer efficiency,a novel turbulator inspired by the morphology of barchan dunes,called the mimetic barchan dune(MBD)turbulator,is designed and evaluated in the simplified gas turbine tra...For the enhancement of heat transfer efficiency,a novel turbulator inspired by the morphology of barchan dunes,called the mimetic barchan dune(MBD)turbulator,is designed and evaluated in the simplified gas turbine transition section.By using computational fluid dynamics(CFD),the numerical simulations for comparison have been carried out,concluding the smooth thermal surface,a thermal surface with riblet-shaped turbulator and a thermal surface with MBD turbulator.Then,two indicators are investigated for evaluating the coolant performance which are the heat transfer efficiency(η)on the outlet and the pressure loss(ΔP)in the coolant chamber.The numerical results show that the coolant has the best heat transfer efficiency with less pressure loss in the coolant chamber with the MBD turbulator.Then,the effect of the MBD turbulator sizes on heat transfer efficiency is investigated.When the height of the MBD turbulator(h)is set at 8 mm,the maximum amount of heat that could be transfered by the coolant is up to566.2 K and the corresponding heat transfer efficiency is 26.62%.The detail flows have been shown to elucidate the function of the MBD surface which may greatly arouse more design for solving harsh circumstance.展开更多

Firstly, we si...In this paper, we find the optimal precursors which can cause double-gyre regime transitions based on conditional nonlinear optimal perturbation (CNOP) method with Regional Ocean Modeling System (ROMS). Firstly, we simulate the multiple-equilibria regimes of double-gyre circulation under different viscosity coefficient and obtain the bifurcation diagram, then choose two equilibrium states (called jet-up state and jet-down state) as reference states respectively, propose Principal Component Analysis-based Simulated Annealing (PCASA) algorithm to solve CNOP-type initial perturbations which can induce double-gyre regime transitions between jet-up state and jet-down state. PCASA algorithm is an adjoint-free method which searches optimal solution randomly in the whole solution space. In addition, we investigate CNOP-type initial perturbations how to evolve with time. The results show:(1) the CNOP-type perturbations present a two-cell structure, and gradually evolves into a three-cell structure at predictive time;(2) by superimposing CNOP-type perturbations on the jet-up state and integrating ROMS, double-gyre circulation transfers from jet-up state to jet-down state, and vice versa, and random initial perturbations don't cause the transitions, which means CNOP-type perturbations are the optimal precursors of double-gyre regime transitions;(3) by analyzing the transition process of double-gyre regime transitions, we find that CNOP-type initial perturbations obtain energy from the background state through both barotropic and baroclinic instabilities, and barotropic instability contributes more significantly to the fast-growth of the perturbations. The optimal precursors and the dynamic mechanism of double-gyre regime transitions revealed in this paper have an important significance to enhance the predictability of double-gyre circulation.展开更多

The microinjection induced a large increase in SAMDC activity, exhaustion of the substrate SAM (S-adenosylmethionine), and execution of apoptosis at the stage called midblastula transition (MBT). By tracing GFP (green fluorescence protein)-marked apoptotic cells, we reached a conclusion that the apoptosis provides pre-blastula embryos with a fail-safe mechanism of early development. We analyzed caspase mRNAs and found that caspase-9 and -3 mRNAs are maternal mRNA and activation of caspase-9 is one of the key steps for the execution of the apoptosis. We also found that over- expression of caspase-8, and in addition p53, a tumor suppressor protein, also induces apoptosis at MBT, just like the overexpression of SAMDC and caspase-9 does. The apoptosis induced by p53 was suppressed by Xdm-2, a negative regulator of p53, and by a peptide inhibitor and a dominant-negative type mutant of caspase-9, but not by those of caspase-8. By contrast, apoptosis induced by SAMDC was suppressed by peptide inhibitors and dominant-negative mutants of both caspase-9 and caspase-8, but not by Xdm-2. Unlike caspase-9 mRNA, caspase-8 mRNA was not a maternal mRNA, but newly expressed during cleavage stage (pre-MBT stage) only in embryos overexpressed with SAMDC. In SAMDC-induced apoptotic embryos activities to process procaspase-8 and procaspase-9 appeared, whereas in p53-induced apoptotic embryos only activity to process procaspase-9 appeared. Thus, Xenopus embryos have at least two pathways to execute the maternal program of apoptosis: One induced by SAMDC overexpression through activation of caspase-9 and do novo expression of caspase-8 gene, and the other induced by p53 overexpression through activation of caspase-9 but not caspase-8. In Xenopus embryos, it has long been believed that zygotic genes are silent until MBT, but results obtained with caspase-8 may provide a novel example of gene expression before MBT.展开更多
The desert-oasis transition zone(DOTZ)serves as a buffer area between the desert and oasis.Understanding its wind field characteristics is of great significance for the prevention and control of aeolian disasters in t...The desert-oasis transition zone(DOTZ)serves as a buffer area between the desert and oasis.Understanding its wind field characteristics is of great significance for the prevention and control of aeolian disasters in the oasis.In this study,we used meteorological data during 2013–2019 from the portable meteorological stations at five sites(site A on the edge of the oasis,sites B,C,and D in the DOTZ,and site O in the desert)in Dunhuang,China to analyze the near-surface wind field characteristics and their causes,as well as to reveal the key role of the DOTZ in oasis protection.The results showed that the mean wind speed,frequency of sand-driving wind,and directional variability of wind decreased from west to east within the DOTZ,and wind speed was significantly affected by air temperature.The terrain influenced the prevailing winds in the region,mainly from northeast and southwest.Only some areas adjacent to the oasis were controlled by southeasterly wind.This indicated that the near-surface wind field characteristics of the DOTZ were caused by the combined effects of local terrain and surface hydrothermal difference.At site D,the annual drift potential(DP)was 24.95 vector units(VU),indicating a low wind energy environment,and the resultant drift direction(RDD)showed obvious seasonal differences.Additionally,the DOTZ played an important buffering role between the desert and oasis.Compared with the desert,the mean wind speed in the oasis decreased by 64.98%,and the prevailing wind direction was more concentrated.The results of this study will be useful in interpreting the aeolian activity of the DOTZ in Dunhuang.展开更多

Laser driven flyer plate technology offers improved safety and reliability for detonation of explosives in industrial applications ranging from mining and stone quarrying to the aerospace and defense industries.This s...Laser driven flyer plate technology offers improved safety and reliability for detonation of explosives in industrial applications ranging from mining and stone quarrying to the aerospace and defense industries.This study is based on developing a safer laser driven flyer plate prototype comprised of a laser initiator and a flyer plate subsystem that can be used with secondary explosives.System parameters were optimized to initiate the shock-to-detonation transition(SDT)of a secondary explosive based on the impact created by the flyer plate on the explosive surface.Rupture of the flyer was investigated at the mechanically weakened region located on the interface of these subsystems,where the product gases from the deflagration of the explosive provide the required energy.A bilayer energetic material was used,where the first layer consisted of a pyrotechnic component,zirconium potassium perchlorate(ZPP),for sustaining the ignition by the laser beam and the second layer consisted of an insensitive explosive,cyclotetramethylene-tetranitramine(HMX),for deflagration.A plexiglass interface was used to enfold the energetic material.The focal length of the laser beam from the diode was optimized to provide a homogeneous beam profile with maximum power at the surface of the ZPP.Closed bomb experiments were conducted in an internal volume of 10 cm^(3) for evaluation of performance.Dependency of the laser driven flyer plate system output on confinement,explosive density,and laser beam power were analyzed.Measurements using a high-speed camera resulted in a flyer velocity of 670±20 m/s that renders the prototype suitable as a laser detonator in applications,where controlled employment of explosives is critical.展开更多

This is shown for all kinds of solid materials, including salts, silicon, organics, polymers, composites, and superalloys. Exothermic and endothermic single and consecutive multiple phase-transitions with their surface dependence are distinguished and the results compared in 5 Tables. The sharp phase-transition onsets and the transition energies provide unprecedented most important materials’ characteristics that are indispensable for safety reasons. ISO ASTM is thus urged to thoroughly revise ISO 14577 and to work out new standards for the mechanically (also thermally) stressed materials. For example, the constancy of the first phase-transition parameters must be controlled, and materials must only be admitted for maximal forces well below the first phase-transition onset. Such onset loads can now be easily calculated. The nevertheless repeated oppositions against the physical analysis of indentations rest on incredibly poor knowledge of basic mathematics, errors that are uncovered. The safety aspects caused by the present unphysical materials’ parameters are discussed.展开更多

Th...This paper is to report the temperature dependent electrical conductivity of single crystals of radical ion salt (RIS) potassium-TCNQ (K-tetracyanoquino- dimethane) in a wide range of temperatures from 30 to 500 K. This RIS is quasi-one-dimensional in nature. These single crystals of K-TCNQ are grown by different methods like electrochemical, solution growth and diffusion method. Activation energy is determined for the sample in different temperature regions and found different values. More than one semiconductor to metal phase transition is observed in the studied samples during electrical measurements below and above room temperature. All the features observed in the studied samples are analyzed in the framework of their molecular structure as well as under different effects like disorder, impurity, Coulomb interaction, charge density wave (CDW), scattering and 3-D effects etc.展开更多
文摘Several significant events of a geological nature occurred approximately 800 ka before the present: (1) Australasian tektite fall (AA), (2) Brunhes-Matuyama geomagnetic reversal (BMR), (3) mid-Pleistocene changes in ice age cycles. Add to these the undated fault system (4) in the South-West (SW) of the South China Sea (SCS). Here we offer a unified cause for all four of these in (5), an impact in the SCS of a large, massive cosmic object, likely a comet, obliquely coming from the SW at an extremely shallow angle, striking the Sunda shelf yet unexploded with the shock of its compressed air bow wave, and causing the continual shelf and slope to collapse, resulting in the fault system (4), then traveling almost tangentially to the surface, exploding at impact with the sea surface, ejecting the tektites (1), creating the formation underlying the later atolls of Spratlies Archipelago (6), Nansha Islands in Chinese, & causing the BMR (2). An explanation of event (3) was Richard Muller’s hypothesis of planet Earth passing through an interplanetary dust cloud periodically due to ecliptic precession. Here we hypothesize this cloud actually is a belt of Australasian tektites ejected into space at super-orbital velocities that Earth encounters about every 100 ka.
文摘A transformation of the electron states—say those enclosed in a potential box—into the de Broglie waves done in the paper, enabled us to calculate the energy change between two quantum levels as a function of the specific heat and difference of the temperature between the states. In consequence, the energy difference and that of entropy between the levels could be examined in terms of the appropriate classical parameters. In the next step, the time interval necessary for the electron transition between the levels could be associated with the classical electrodynamical parameters like the electric resistance and capacitance connected with the temporary formation of the electric cell in course of the transition. The parameters characterizing the mechanical inertia of the electron were next used as a check of the electrodynamical formulae referring to transition.

文摘Ultraviolet photodetectors(UV PDs)are widely used in civilian,scientific,and military fields due to their high sensitivity and low false alarm rates.We present a temperature-dependent Lewis acid p-type doping method for transition metal dichalcogenides(TMDs),which can effectively be used to extend the optical response range.The p-type doping based on surface charge transfer involves the chemical adsorption of the Lewis acid SnCl_(4)as a light absorption layer on the surface of WS_(2),significantly enhancing its UV photodetection performance.Under 365 nm laser irradiation,WS_(2)PDs exhibit response speed of 24 ms/20 ms,responsivity of 660 mA/W,detectivity of 3.3×10^(11)Jones,and external quantum efficiency of 226%.Moreover,we successfully apply this doping method to other TMDs materials(such as MoS_(2),MoSe_(2),and WSe_(2))and fabricate WS_(2) lateral p–n heterojunction PDs.
文摘Non-iterative analysis of indentation results allows for the detection of phase transitions under load and their transition energy. The closed algebraic equations have been deduced on the basis of the physically founded normal force ?depth3/2 relation. The precise transition onset position is obtained by linear regression of the FN = kh3/2 plot, where k is the penetration resistance, which also provides the axis cuts of both polymorphs of first order phase transitions. The phase changes can be endothermic or exothermic. They are normalized per μN or mN normal load. The analyses of indentation loading curves with self-similar diamond indenters are used as validity check of the loading curves, also from calibration standards that exhibit previously undetected phase-transitions and are thus incorrect. The phase-transition energies for fused quartz are determined from the loading curves from instrument provider handbooks. The anisotropic behavior of phase transition energies is studied for the first time. Quartz is a useful test object. The reasons for the packing-dependent differences are discussed on the basis of the local crystal structure under and around the inserting tip.

文摘It is an important way to realize rural revitalization and sustainable development to guide rural settlement transition(RST)in an appropriate way.This paper uses actor network theory(ANT)to construct a theoretical framework for the study of RST.Taking two typical villages with different transition paths in rural areas of North China Plain as examples,this paper reveals the mechanism of RST and makes a comparative analysis.The results show that:1)after identifying problems and obligatory passage point,key actors recruit heterogeneous actors into the actor network by entrusting them with common interests,and realize RST under the system operation.2)Rural settlements under different transition paths have similarities in the problems to be solved,collective actions and policy factors,but there are differences in the transition process,mechanism and effect.The actor network and mechanism of RST through the path of new rural community construction are more complex and the transition effect is more thorough.In contrast,the degree of RST of retention development path is limited if there is no resource and location advantage.3)Based on the applicable conditions of different paths,this paper designs a logical framework of‘Situation-Structure-Behavior-Result’to scientifically guide the identification of RST paths under the background of rural revitalization.

文摘This paper presents a comprehensive framework for analyzing phase transitions in collective models such as theVicsek model under various noise types. The Vicsek model, focusing on understanding the collective behaviors of socialanimals, is known due to its discontinuous phase transitions under vector noise. However, its behavior under scalar noiseremains less conclusive. Renowned for its efficacy in the analysis of complex systems under both equilibrium and nonequilibriumstates, the eigen microstate method is employed here for a quantitative examination of the phase transitions inthe Vicsek model under both vector and scalar noises. The study finds that the Vicsek model exhibits discontinuous phasetransitions regardless of noise type. Furthermore, the dichotomy method is utilized to identify the critical points for thesephase transitions. A significant finding is the observed increase in the critical point for discontinuous phase transitions withescalation of population density.
文摘Flow transition from laminar to turbulent mode (and vice versa)—that is, the initiation of turbulence—is one of the most important research subjects in the history of engineering. Even for pipe flow, predicting the onset of turbulence requires sophisticated instrumentation and/or direct numerical simulation, based on observing the instantaneous flow structure formation and evolution. In this work, a local Reynolds number equivalence c (ratio of local inertia effect to viscous effect) is seen to conform to the Universal Law of the Wall, where c = 1 represents a quantitative balance between the abovementioned two effects. This coincides with the wall layer thickness (y+= 1, where y+ is the dimensionless distance from the wall surface defined in the Universal Law of the Wall). It is found that the characteristic of how the local derivative of c against the local velocity changes with increasing velocity determines the onset of turbulence. For pipe flow, c - 25, and for plate flow, c - 151.5. These findings suggest that a certain combination of c and velocity (nonlinearity) can qualify the source of turbulence (i.e., generate turbulent energy). Similarly, a re-evaluation of the previous findings reveals that only the geometrically narrow domain can act locally as the source of turbulence, with the rest of the flow field largely being left for transporting and dissipating. This understanding will have an impact on the future large-scale modeling of turbulence.

文摘The Floquet technique provides a novel anomalous topological phase for non-equilibrium phase transitions.Based on the high symmetry of the quantum anomalous Hall model,the findings suggest a one-to-one correspondence between the average spin texture and the Floquet quasi-energy spectrum.A new approach is proposed to directly measure the quasienergy spectrum,replacing previous measurements of the average spin texture.Finally,we proposed a reliable experimental scheme based on ion trap platforms.This scheme markedly reduces the measurement workload,improves the measurement fidelity,and is applicable to multiple platforms such as cold atoms and nuclear magnetic resonance.

文摘BACKGROUND CALD1 has been discovered to be abnormally expressed in a variety of malignant tumors,including gastric cancer(GC),and is associated with tumor progression and immune infiltration;however,the roles and mechanisms of CALD1 in epithe-lial-mesenchymal transition(EMT)in GC are unknown.AIM To investigate the role and mechanism of CALD1 in GC progression,invasion,and migration.METHODS In this study,the relationship between CALD1 and GC,as well as the possible network regulatory mechanisms of CALD1,was investigated by bioinformatics and validated by experiments.CALD1-siRNA was synthesized and used to trans-fect GC cells.Cell activity was measured using the CCK-8 method,cell migration and invasive ability were measured using wound healing assay and Transwell assay,and the expression levels of relevant genes and proteins in each group of cells were measured using qRT-PCR and Western blot.A GC cell xenograft model RESULTS Bioinformatics results showed that CALD1 was highly expressed in GC tissues,and CALD1 was significantly higher in EMT-type GC tissues than in tissues of other types of GC.The prognosis of patients with high expression of CALD1 was worse than that of patients with low expression,and a prognostic model was constructed and evaluated.The experimental results were consistent with the results of the bioinformatics analysis.The expression level of CALD1 in GC cell lines was all higher than that in gastric epithelial cell line GES-1,with the strongest expression found in AGS and MKN45 cells.Cell activity was significantly reduced after CALD1-siRNA trans-fection of AGS and MKN45 cells.The ability of AGS and MKN45 cells to migrate and invade was reduced after CALD1-siRNA transfection,and the related mRNA and protein expression was altered.According to bioinfor-matics findings in GC samples,the CALD1 gene was significantly associated with the expression of members of the PI3K-AKT-mTOR signaling pathway as well as the EMT signaling pathway,and was closely related to the PI3K-Akt signaling pathway.Experimental validation revealed that upregulation of CALD1 increased the expression of PI3K,p-AKT,and p-mTOR,members of the PI3K-Akt pathway,while decreasing the expression of PTEN;PI3K-Akt inhibitor treatment decreased the expression of PI3K,p-AKT,and p-mTOR in cells overexpressing CALD1(still higher than that in the normal group),but increased the expression of PTEN(still lower than that in the normal group).CCK-8 results revealed that the effect of CALD1 on tumor cell activity was decreased by the addition of the inhibitor.Scratch and Transwell experiments showed that the effect of CALD1 on tumor cell migration and invasion was weakened by the addition of the PI3K-Akt inhibitor.The mRNA and protein levels of EMT-related genes in AGS and MKN45 cells were greatly altered by the overexpression of CALD1,whereas the effect of overex-pression of CALD1 was significantly weakened by the addition of the PI3K-Akt inhibitor.Animal experiments showed that tumour growth was slow after inhibition of CALD1,and the expression of some PI3K-Akt and EMT pathway proteins was altered.CONCLUSION Increased expression of CALD1 is a key factor in the progression,invasion,and metastasis of GC,which may be associated with regulating the PI3K-Akt pathway to promote EMT.
文摘The transition radiation of a charged particle crossing the interface of two media having a monatomic impurity layer is investigated. It is shown that at sliding angles of incidence of a particle on the boundary of the media, the transition radiation is mainly determined by the properties of the surface layer. The possibility of using transition radiation to study the surface of substances is discussed. In addition, due to the hard radiation present in space, this research may be important for the use of light monoatomic layers as a material for satellite antennas, “solar sails” and cover layers in a future space (interstellar) mission.
文摘The general use of aluminium as an indentation standard for the iteration of contact heights for the determination of ISO-14577 hardness and elastic modulus is challenged because of as yet not appreciated phase-changes in the physical force-depth standard curve that seemed to be secured by claims from 1992. The physical and mathematical analyses with closed formulas avoid the still world-wide standardized energy-law violation by not reserving 33.33% (h2 belief) (or 20% h3/2 physical law) of the loading force and thus energy for all not depth producing events but using 100% for the depth formation is a severe violation of the energy law. The not depth producing part of the indentation work cannot be done with zero energy! Both twinning and structural phase-transition onsets and normalized phase-transition energies are now calculated without iterations but with physically correct closed arithmetic equations. These are reported for Berkovich and cubecorner indentations, including their comparison on geometric grounds and an indentation standard without mechanical twinning is proposed. Characteristic data are reported. This is the first detection of the indentation twinning of aluminium at room temperature and the mechanical twinning of fused quartz is also new. Their disqualification as indentation standards is established. Also, the again found higher load phase-transitions disqualify aluminium and fused quartz as ISO-ASTM 14577 (International Standardization Organization and American Society for Testing and Materials) standards for the contact depth “hc” iterations. The incorrect and still world-wide used black-box values for H- and Er-values (the latter are still falsely called “Young’s moduli” even though they are not directional) and all mechanical properties that depend on them. They lack relation to bulk moduli from compression experiments. Experimentally obtained and so published force vs depth parabolas always follow the linear FN = kh3/2 + Fa equation, where Fa is the axis-cut before and after the phase-transition branches (never “h2” as falsely enforced and used for H, Er and giving incorrectly calculated parameters). The regression slopes k are the precise physical hardness values, which for the first time allow for precise calculation of the mechanical qualities by indentation in relation to the geometry of the indenter tip. Exactly 20% of the applied force and thus energy is not available for the indentation depth. Only these scientific k-values must be used for AI-advises at the expense of falsely iterated indentation hardness H-values. Any incorrect H-ISO-ASTM and also the iterated Er-ISO-ASTM modulus values of technical materials in artificial intelligence will be a disaster for the daily safety. The AI must be told that these are unscientific and must therefore be replaced by physical data. Iterated data (3 and 8 free parameters!) cannot be transformed into physical data. One has to start with real experimental loading curves and an absolute ZerodurR standard that must be calibrated with standard force and standard length to create absolute indentation results. .

文摘For the enhancement of heat transfer efficiency,a novel turbulator inspired by the morphology of barchan dunes,called the mimetic barchan dune(MBD)turbulator,is designed and evaluated in the simplified gas turbine transition section.By using computational fluid dynamics(CFD),the numerical simulations for comparison have been carried out,concluding the smooth thermal surface,a thermal surface with riblet-shaped turbulator and a thermal surface with MBD turbulator.Then,two indicators are investigated for evaluating the coolant performance which are the heat transfer efficiency(η)on the outlet and the pressure loss(ΔP)in the coolant chamber.The numerical results show that the coolant has the best heat transfer efficiency with less pressure loss in the coolant chamber with the MBD turbulator.Then,the effect of the MBD turbulator sizes on heat transfer efficiency is investigated.When the height of the MBD turbulator(h)is set at 8 mm,the maximum amount of heat that could be transfered by the coolant is up to566.2 K and the corresponding heat transfer efficiency is 26.62%.The detail flows have been shown to elucidate the function of the MBD surface which may greatly arouse more design for solving harsh circumstance.

文摘In this paper, we find the optimal precursors which can cause double-gyre regime transitions based on conditional nonlinear optimal perturbation (CNOP) method with Regional Ocean Modeling System (ROMS). Firstly, we simulate the multiple-equilibria regimes of double-gyre circulation under different viscosity coefficient and obtain the bifurcation diagram, then choose two equilibrium states (called jet-up state and jet-down state) as reference states respectively, propose Principal Component Analysis-based Simulated Annealing (PCASA) algorithm to solve CNOP-type initial perturbations which can induce double-gyre regime transitions between jet-up state and jet-down state. PCASA algorithm is an adjoint-free method which searches optimal solution randomly in the whole solution space. In addition, we investigate CNOP-type initial perturbations how to evolve with time. The results show:(1) the CNOP-type perturbations present a two-cell structure, and gradually evolves into a three-cell structure at predictive time;(2) by superimposing CNOP-type perturbations on the jet-up state and integrating ROMS, double-gyre circulation transfers from jet-up state to jet-down state, and vice versa, and random initial perturbations don't cause the transitions, which means CNOP-type perturbations are the optimal precursors of double-gyre regime transitions;(3) by analyzing the transition process of double-gyre regime transitions, we find that CNOP-type initial perturbations obtain energy from the background state through both barotropic and baroclinic instabilities, and barotropic instability contributes more significantly to the fast-growth of the perturbations. The optimal precursors and the dynamic mechanism of double-gyre regime transitions revealed in this paper have an important significance to enhance the predictability of double-gyre circulation.
文摘To study gene control mechanisms in Xenopus embryos, we analyzed polyamines, cloned SAMDC (S-adenosylmethionine decarboxylase), a key enzyme of polyamine metabolism, and microinjected its mRNA into Xenopus fertilized eggs. The microinjection induced a large increase in SAMDC activity, exhaustion of the substrate SAM (S-adenosylmethionine), and execution of apoptosis at the stage called midblastula transition (MBT). By tracing GFP (green fluorescence protein)-marked apoptotic cells, we reached a conclusion that the apoptosis provides pre-blastula embryos with a fail-safe mechanism of early development. We analyzed caspase mRNAs and found that caspase-9 and -3 mRNAs are maternal mRNA and activation of caspase-9 is one of the key steps for the execution of the apoptosis. We also found that over- expression of caspase-8, and in addition p53, a tumor suppressor protein, also induces apoptosis at MBT, just like the overexpression of SAMDC and caspase-9 does. The apoptosis induced by p53 was suppressed by Xdm-2, a negative regulator of p53, and by a peptide inhibitor and a dominant-negative type mutant of caspase-9, but not by those of caspase-8. By contrast, apoptosis induced by SAMDC was suppressed by peptide inhibitors and dominant-negative mutants of both caspase-9 and caspase-8, but not by Xdm-2. Unlike caspase-9 mRNA, caspase-8 mRNA was not a maternal mRNA, but newly expressed during cleavage stage (pre-MBT stage) only in embryos overexpressed with SAMDC. In SAMDC-induced apoptotic embryos activities to process procaspase-8 and procaspase-9 appeared, whereas in p53-induced apoptotic embryos only activity to process procaspase-9 appeared. Thus, Xenopus embryos have at least two pathways to execute the maternal program of apoptosis: One induced by SAMDC overexpression through activation of caspase-9 and do novo expression of caspase-8 gene, and the other induced by p53 overexpression through activation of caspase-9 but not caspase-8. In Xenopus embryos, it has long been believed that zygotic genes are silent until MBT, but results obtained with caspase-8 may provide a novel example of gene expression before MBT.

文摘The desert-oasis transition zone(DOTZ)serves as a buffer area between the desert and oasis.Understanding its wind field characteristics is of great significance for the prevention and control of aeolian disasters in the oasis.In this study,we used meteorological data during 2013–2019 from the portable meteorological stations at five sites(site A on the edge of the oasis,sites B,C,and D in the DOTZ,and site O in the desert)in Dunhuang,China to analyze the near-surface wind field characteristics and their causes,as well as to reveal the key role of the DOTZ in oasis protection.The results showed that the mean wind speed,frequency of sand-driving wind,and directional variability of wind decreased from west to east within the DOTZ,and wind speed was significantly affected by air temperature.The terrain influenced the prevailing winds in the region,mainly from northeast and southwest.Only some areas adjacent to the oasis were controlled by southeasterly wind.This indicated that the near-surface wind field characteristics of the DOTZ were caused by the combined effects of local terrain and surface hydrothermal difference.At site D,the annual drift potential(DP)was 24.95 vector units(VU),indicating a low wind energy environment,and the resultant drift direction(RDD)showed obvious seasonal differences.Additionally,the DOTZ played an important buffering role between the desert and oasis.Compared with the desert,the mean wind speed in the oasis decreased by 64.98%,and the prevailing wind direction was more concentrated.The results of this study will be useful in interpreting the aeolian activity of the DOTZ in Dunhuang.

文摘Background The intestinal epithelium performs essential physiological functions,such as nutrient absorption,and acts as a barrier to prevent the entry of harmful substances.Mycotoxins are prevalent contaminants found in ani-mal feed that exert harmful effects on the health of livestock.Zearalenone(ZEA)is produced by the Fusarium genus and induces gastrointestinal dysfunction and disrupts the health and immune system of animals.Here,we evaluated the molecular mechanisms that regulate the effects of ZEA on the porcine intestinal epithelium.Results Treatment of IPEC-J2 cells with ZEA decreased the expression of E-cadherin and increased the expression of Snai1 and Vimentin,which induced Snail1-mediated epithelial-to-mesenchymal transition(EMT).In addition,ZEA induces Snail-mediated EMT through the activation of TGF-βsignaling.The treatment of IPEC-J2 cells with atractyle-nolideⅢ,which were exposed to ZEA,alleviated EMT.Conclusions Our findings provide insights into the molecular mechanisms of ZEA toxicity in porcine intestinal epi-thelial cells and ways to mitigate it.
文摘Laser driven flyer plate technology offers improved safety and reliability for detonation of explosives in industrial applications ranging from mining and stone quarrying to the aerospace and defense industries.This study is based on developing a safer laser driven flyer plate prototype comprised of a laser initiator and a flyer plate subsystem that can be used with secondary explosives.System parameters were optimized to initiate the shock-to-detonation transition(SDT)of a secondary explosive based on the impact created by the flyer plate on the explosive surface.Rupture of the flyer was investigated at the mechanically weakened region located on the interface of these subsystems,where the product gases from the deflagration of the explosive provide the required energy.A bilayer energetic material was used,where the first layer consisted of a pyrotechnic component,zirconium potassium perchlorate(ZPP),for sustaining the ignition by the laser beam and the second layer consisted of an insensitive explosive,cyclotetramethylene-tetranitramine(HMX),for deflagration.A plexiglass interface was used to enfold the energetic material.The focal length of the laser beam from the diode was optimized to provide a homogeneous beam profile with maximum power at the surface of the ZPP.Closed bomb experiments were conducted in an internal volume of 10 cm^(3) for evaluation of performance.Dependency of the laser driven flyer plate system output on confinement,explosive density,and laser beam power were analyzed.Measurements using a high-speed camera resulted in a flyer velocity of 670±20 m/s that renders the prototype suitable as a laser detonator in applications,where controlled employment of explosives is critical.
文摘-The ISO standard 14577 is challenged for its violation of the energy law, its wrong relation of normal force FN with impression depth h, and for its iterative treatments. The solution of this dilemma is the use of sacrosanct simplest calculation rules for the loading parabola (now FN = kh3/2) giving straight lines for cones, pyramids and wedges. They provide the physical penetration resistance hardness k with dimension [Nm-3/2] and allow for non-iterative calculations with closed formulas, using simple undeniable calculation rules. The physically correct FN versus h3/2 plot is universally valid. It separates out the most common surface effects and reveals gradients. It provides unmatched precision, including reliability checks of experimental data. Regression analysis of FN versus h3/2 plots reveals eventual unsteadiness kink phase-transition onset with the transition-energy. This is shown for all kinds of solid materials, including salts, silicon, organics, polymers, composites, and superalloys. Exothermic and endothermic single and consecutive multiple phase-transitions with their surface dependence are distinguished and the results compared in 5 Tables. The sharp phase-transition onsets and the transition energies provide unprecedented most important materials’ characteristics that are indispensable for safety reasons. ISO ASTM is thus urged to thoroughly revise ISO 14577 and to work out new standards for the mechanically (also thermally) stressed materials. For example, the constancy of the first phase-transition parameters must be controlled, and materials must only be admitted for maximal forces well below the first phase-transition onset. Such onset loads can now be easily calculated. The nevertheless repeated oppositions against the physical analysis of indentations rest on incredibly poor knowledge of basic mathematics, errors that are uncovered. The safety aspects caused by the present unphysical materials’ parameters are discussed.
文摘This paper is to report the temperature dependent electrical conductivity of single crystals of radical ion salt (RIS) potassium-TCNQ (K-tetracyanoquino- dimethane) in a wide range of temperatures from 30 to 500 K. This RIS is quasi-one-dimensional in nature. These single crystals of K-TCNQ are grown by different methods like electrochemical, solution growth and diffusion method. Activation energy is determined for the sample in different temperature regions and found different values. More than one semiconductor to metal phase transition is observed in the studied samples during electrical measurements below and above room temperature. All the features observed in the studied samples are analyzed in the framework of their molecular structure as well as under different effects like disorder, impurity, Coulomb interaction, charge density wave (CDW), scattering and 3-D effects etc.
