BACKGROUND Hepatocellular carcinoma(HCC)is now the most common primary liver malignancy worldwide,and multiple risk factors attribute to the occurrence and development of HCC.Recently,increasing studies suggest that u...BACKGROUND Hepatocellular carcinoma(HCC)is now the most common primary liver malignancy worldwide,and multiple risk factors attribute to the occurrence and development of HCC.Recently,increasing studies suggest that ubiquitinconjugating enzyme E2T(UBE2T)serves as a promising prognostic factor in human cancers,although the molecular mechanism of UBE2T in HCC remains unclear.AIM To investigate the clinical relevance and role of UBE2T in HCC development.METHODS UBE2T expression in HCC tissues from the TCGA database and its association with patient survival were analyzed.A lentivirus-mediated strategy was used to knock down UBE2T in HCC cells.qRT-PCR and Western blot assays were performed to check the effect of UBE2T silencing in HCC cells.Cell growth in vitro and in vivo was analyzed by multiparametric high-content screening and the xenograft tumorigenicity assay,respectively.Cell cycle distribution and apoptosis were determined by flow cytometry.The genes regulated by UBE2T were profiled by microarray assay.RESULTS UBE2T was overexpressed in HCC tissues compared with paired and non-paired normal tissues.High expression of UBE2T predicted a poor overall survival in HCC patients.In vitro,lentivirus-mediated UBE2T knockdown significantly reduced the viability of both SMMC-7721 and BEL-7404 cells.In vivo,the xenograft tumorigenesis of SMMC-7721 cells was largely attenuated by UBE2T silencing.The cell cycle was arrested at G1/S phase in SMMC-7721 and BEL-7404 cells with UBE2T knockdown.Furthermore,apoptosis was increased by UBE2T knockdown.At the molecular level,numerous genes were dysregulated after UBE2T silencing,including IL-1B,FOSL1,PTGS2,and BMP6.CONCLUSION UBE2T plays an important role in cell cycle progression,apoptosis,and HCC development.展开更多

Zhongshuang9, a new semi-winter Brassica napus variety with high resistance to Sclerotinia sclerotiorum and lodging, high-yield, double-low quality and extensive adaptability, was bred by multiple crossing and microsp...Zhongshuang9, a new semi-winter Brassica napus variety with high resistance to Sclerotinia sclerotiorum and lodging, high-yield, double-low quality and extensive adaptability, was bred by multiple crossing and microspore culture technique. It was registered and released in China in 2002. In regional trial of Hubei Province in China, Zhongshuang9 yielded 2 482. 2 kg ha-1 averagely in 2000 - 2002, 15. 33% higher than the control variety Zhongyou821. Erucic acid, glucosinolates and oil contents of Zhongshuang9 were 0.23%, 22.69 μmol g-1(in meal)and 42%, respectively. In field assessment of resistance to S. Sclerotiorum , the disease incidence and disease index of Zhongshuang9 averaged 13.31 % and 6.47, respectively, which were lower than those of Zhongyou821 by 28% and 36%, respectively. After inoculation of detached leaves with mycelia, the lesion size of Zhongshuang9 was 4. 709 cm2, which was significantly smaller than that of the mid-resistant variety Zhongyou821(5. 933 cm2). The stem lesion length of Zhongshuang9 after match-stick inoculation was 1.275 cm, which was significantly lower than that of Zhongyou821(1.943 cm). The possible mechanism of resistance to S. sclerotiorum was studied through comparing the activities of phenylalanine ammonia lyase(PAL), exo-chitinase, β-1, 3-glucanase, peroxidase(POD)and polyphenoloxidase(PPO)in Zhongshuang9 with those in other resistant, mid-resistant and susceptible cultivars.展开更多
Objective:To isolate,identify,and evaluate a new angiotensin-converting enzyme inhibitor from Peperomia pellucida(L.)Kunth herbs.Methods:A dried sample of Peperomia pellucida herb was successively macerated with n-hex...Objective:To isolate,identify,and evaluate a new angiotensin-converting enzyme inhibitor from Peperomia pellucida(L.)Kunth herbs.Methods:A dried sample of Peperomia pellucida herb was successively macerated with n-hexane and ethyl acetate.The ethyl acetate extract solution was evaporated to obtain the crude extract.Vacuum liquid column chromatography and thin layer chromatography were performed to obtain two pure compounds.Then,both compounds were elucidated and identified using the spectroscopic method.Angiotensin-converting enzyme inhibitory activity studies of both compounds were determined using angiotensin-converting enzyme kit WST-1 with spectrophotometer microplate reader 96-well at 450 nm wavelength.Results:Two bioactive compounds were successfully isolated from Peperomia pellucida herb,including a new compound of 2,3,5-trimethoxy-9-(12,14,15-trimethoxybenzyl)-1 H-indene and pellucidin A.Both compounds demonstrated angiotensin-converting enzyme inhibitory activity,with IC50 values of 72 μM(27.95 μg/mL)and 1 1μM(4.4 μg/mL),respectively.Conclusions:In the present study,two active angiotensin-converting enzyme inhibitors were successfully isolated and purified from Peperomia pellucida which is used as an antihypertensive in traditional medicine,and support its use as an angiotensin-converting enzyme-inhibiting drug.展开更多
