Objective To investigate the application of BPDE-albumin adducts as monitoring biomarkers for coke oven workers exposed to polycyclic aromatic hydrocarbons (PAHs) and to explore possible relationship between BPDE-al...Objective To investigate the application of BPDE-albumin adducts as monitoring biomarkers for coke oven workers exposed to polycyclic aromatic hydrocarbons (PAHs) and to explore possible relationship between BPDE-albumin adducts and urinary 1-hydroxypyrene (1-OHP) levels in them. Methods Thirty-seven coke oven workers from a coke plant and 47 controls without the occupational exposure to PAHs were recruited in this study. The levels of plasma BPDE-albumin adducts and urinary 1-OHP were analyzed using high performance liquid chromatography. Results The median levels of BPDE-albumin adducts (42.10 fmol/mg albumin) and urinary 1-OHP (5.46 μmol/mol creatinine) were significantly higher in coke oven workers than in controls (14.16 fmol/mg albumin, 2.96μmol/mol creatinine, respectively; P〈0.01). Multiple logistic regression analysis showed that coke oven workers were at higher risk of having BPDE-albumin adduct levels above 25.30 μmol/mg albumin (OR=1.79, P〈0.01) and urinary 1-OHP levels above 4.13 μmol/mol creatinine (OR=2.45, P〈0.05). There was a positive correlation between the levels of BPDE-albumin adducts and urinary 1-OHP in all subjects (rs=0.349, P〈0.01). Conclusion BPDE-albumin adduct is a useful biomarker for monitoring long-term exposure to PAHs, and plasma BPDE-albumin adducts level is significantly correlated to urinary 1-OHP levels in coke oven workers.展开更多
BACKGROUND Globally,diabetic nephropathy(DN)is the primary cause of chronic kidney disease.Currently,renal function is monitored indirectly using measures of serum creatinine,estimated glomerular filtration rate(eGFR)...BACKGROUND Globally,diabetic nephropathy(DN)is the primary cause of chronic kidney disease.Currently,renal function is monitored indirectly using measures of serum creatinine,estimated glomerular filtration rate(eGFR),and proteinuria.Novel urinary biomarkers utilized in the early stages of DN have been described;these indicators can be used in the early identification of the disease,which is important for initiating treatment to halt or impediment the advance of diabetic nephropathy.AIM To estimate neutrophil gelatinase-associated lipocalin(NGAL),kidney injury molecule-1(KIM-1),and periostin(POSTN)levels as novel urinary biomarkers in DN.METHODS In this hospital based cross-sectional study,a total of 160 patients of both genders aged 18 years or more;40 healthy participants and 120 patients with diabetes mellitus(DM)were included.Patients with DM were divided into normoalbuminuria(n=40),microalbuminuria(n=40),and macroalbuminuria(n=40)groups as per urine albumin creatinine ratio(uACR).Blood urea,serum creatinine,uACR were measured.Urine NGAL,KIM-1,and POSTN were measured by enzyme linked immunosorbent assay.The eGFR was calculated and compared with urinary markers.RESULTS NGAL,KIM-1,and POSTN levels increased significantly in normo,micro,and macroalbuminuria with the highest in the macroalbuminuria group.Albumin creatinine ratio(ACR)showed a positive correlation with NGAL,KIM-1,and POSTN levels.The eGFR showed a weak negative correlation with ACR,NGAL,KIM-1,and POSTN.NGAL was significantly lower in stage 1 compared to stage 2,3,and 4 kidney disease.KIM-1 was significantly decreased in stage 1 compared to stage 4 kidney disease.POSTN was significantly decreased in stage 1 compared to stage 3 and 4 kidney disease.The receiver operator curve analysis of ACR,NGAL,KIM-1,and POSTN showed good sensitivity of 80%,75.8%,63.3%,and 80%respectively with a cut-off of 12.5 mg/g,4.5μg/L,1.5 ng/mL,and 37.5 ng/mL.CONCLUSION Urinary NGAL and POSTN are independent markers of DN.展开更多

The aims of this stu...CTX-M-producing bacteria are known as a resistant source against oxyimino-cephalosporin such as cefotaxime and ceftazidime;although laboratory diagnosis of this gene has not been properly defined. The aims of this study are determining the rates of prevalence of CTX-M and CTX-M group 1 in the Escherichia coli (E. coli) obtained from urinary tract infections (UTI), and also determining their genetic relationship in the city of Sanandaj. In current study, 180 E. coli strains isolated from urinary tract infections were used. Sensitivity to common antibiotics was studied by the disc diffusion method. Phenotypic detection of isolated ESBL-producing starins was done by the combination disc test. CTX-M and CTX-M1 genes were detected using the PCR method and finally, the possible clonal relationship between isolates was determined using the REP-PCR method. 89 samples were ESBL-positive. The PCR assay used for detecting the CTX-M gene, showed that 48 samples out of 180 samples (26.66%) contained that gene;also among these 48 samples, 23 (12.77%) had CTX-M group 1. Based on the REP-PCR assay, 48 genotypes among 48 samples were CTX-M-positive. Results from the REP-PCR assay indicated that the clonal propagation theory of one epidemic strain of Escherichia coli is not apply, i.e. all CTX-M-producing species are not originated from one single strain and the gene is spread between different isolates. Therefore, hospitals and their employees must be more hygiene and, proper disposal of hospital waste can help to prevent the spread of different resistances.展开更多
Background: Despite recent advances in perinatal and neonatal care in respiratory distress syndrome (RDS) prevention and treatment, a considerable number of these neonates suffer from acute kidney injury (AKI), and it...Background: Despite recent advances in perinatal and neonatal care in respiratory distress syndrome (RDS) prevention and treatment, a considerable number of these neonates suffer from acute kidney injury (AKI), and it is associated with poor outcome as an independent risk factor. KIM-1 mRNA and protein are expressed at a low level in normal kidney but are increased in post ischemic kidney. Aim: The aim is to detect the value of urinary KIM-1 measurement as an early predictor marker of acute kidney injury in preterm neonates with respiratory distress syndrome. Patients and methods: The study included 30 preterm newborn with (RDS) ≤36 weeks during the period from October 2014 to March 2015. Also the study included 30 apparently healthy newborn ≤36 weeks as controls. They were selected from NICU of Manshiate Elbakry hospital Cairo, Egypt. uKIM-1 along with serum creatinine levels and eGFR were assessed in days 1 of life for both groups and in day 3 for cases. Results: In day one of life, we found a significant increase in uKIM-1 levels in preterm newborn with RDS compared to their controls (2.88 ± 1.01 ng/ml and 0.95 ± 0.52 ng/ml respectively (p = 0.001)). There is no significant difference between both groups regarding serum creatinine and eGFR. In day 3 of life, preterm with RDS had significant decrease in uKIM-1 levels compared to day 1 of life with significant increase in non-survivor compared to survivor group ( 2.30 ± 1.56 ng/ml and 1.30 ± 0.90 ng/ml respectively (p = 0.03)). The sensitivity and specificity of uKIM-1 and serum creatinine was calculated (100.00%, 86.67% and 33.33%;95.00%) respectively. Conclusion: Preterm neonate with RDS is at high risk of developing AKI. Early and serial uKIM-1 measurements can be used as a non-invasive indicator of kidney injury in premature newborn with RDS.展开更多
In this editorial we comment on the article by Wei et al,published in the recent issue of the World Journal of Clinical Oncology.The authors investigated the role of Transmembrane 9 superfamily member 1(TM9SF1)protein...In this editorial we comment on the article by Wei et al,published in the recent issue of the World Journal of Clinical Oncology.The authors investigated the role of Transmembrane 9 superfamily member 1(TM9SF1)protein in bladder cancer(BC)carcinogenesis.Lentiviral vectors were used to achieve silencing or overexpression of TM9SF1 gene in three BC cell lines.These cell lines were then subject to cell counting kit 8,wound-healing assay,transwell assay,and flow cytometry.Proliferation,migration,and invasion of BC cells were increased in cell lines subjected to TM9SF1 overexpression.TM9SF1 silencing inhibited proliferation,migration and invasion of BC cells.The authors conclude that TM9SF1 may be an oncogene in bladder cancer pathogenesis.展开更多
【目的】通过检测水中待产产妇盆底组织中基质金属蛋白酶-1(MMP-1)和基质金属蛋白酶抑制物-1(TIMP-1)的表达,探讨水中待产对盆底组织是否具有保护作用。【方法】2009年6月至2011年2月,收集在中山大学孙逸仙纪念医院自愿选择水中待产的产...【目的】通过检测水中待产产妇盆底组织中基质金属蛋白酶-1(MMP-1)和基质金属蛋白酶抑制物-1(TIMP-1)的表达,探讨水中待产对盆底组织是否具有保护作用。【方法】2009年6月至2011年2月,收集在中山大学孙逸仙纪念医院自愿选择水中待产的产妇38例为试验组。同期、一般情况及产科条件相近、选择传统阴道试产的产妇70例为对照组。调查两组阴道分娩产妇孕期、产后42天压力性尿失禁症状(SUI)和盆腔器官脱垂(POP)情况。试验组和对照组最终分别有33、47例成功阴道分娩,于第三产程后取会阴侧切伤口底部组织,并进行石蜡包埋切片,用免疫组化SP染色法分别检测组织中MMP-1、TIMP-1的表达。【结果】1.两组孕期出现SUI症状的发生率差异无统计学意义(18.18%vs 31.91%,P=0.17),试验组产后42 d伴有SUI症状的发生率(2/33,6.06%)显著低于对照组(11/47,23.40%)(P=0.04)。两组阴道分娩产妇产后42 d POP发生率相近(24.24%vs 25.53%,P>0.05)。试验组盆底组织MMP-1阳性率(3/33,9.09%)显著低于对照组(32/47,68.09%)(P<0.001),试验组TIMP-1阳性率低于对照组,但差异没有统计学意义(12.12%vs 17.02%,P=0.546)。盆底组织MMP-1阳性表达与产后42 d SUI症状发生有关(OR=22.96,95%CI 2.81-187.73),盆底组织MMP-1阳性与产后42 d POP发生有关(OR=5.56,95%CI 1.88-16.42)。未发现TIMP-1与产后42 d SUI及POP的确切关系,对应OR(95%CI)分别为2.083(0.46-9.38)及3.02(0.83-11.06)。【结论】水中待产产妇盆底组织中MMP-1的阳性表达较低,可能与水中待产对盆底组织具有保护作用有关。展开更多
