The objective of this study was to determine the effectiveness of a novel redox cycling agent, vitamin K3, and various peroxides including hydrogen peroxide, calcium peroxide and magnesium peroxide in alleviating the effects of hypoxia in bean seedlings grown in nutrient culture. All the anti-hypoxic agents including vitamin K3 had a positive impact on the overall growth of bean seedlings under hypoxic conditions, but their responses were variable depending on the concentration. With regard to shoot growth, vitamin K3 (5 μM) increased the leaf area significantly, by more than 58% over the hypoxic control plants and produced the highest stem fresh weight similar to calcium peroxide (20 μM) and magnesium peroxide (10 μM). In addition, the use of vitamin K3 resulted in the highest accumulation of chlorophyll (chla + chlb) in the leaves, an increase of nearly two-fold over the hypoxic control plants. Furthermore under hypoxia, calcium peroxide (20 μM) and magnesium peroxide (10 μM) produced the highest leaf biomass (FW) followed by vitamin K3. Vitamin K3 (1 μM) also favored root growth in bean seedlings under hypoxia;it produced the largest increase in root length and root biomass (DW) similar to calcium peroxide and magnesium peroxide. Based on the overall shoot and root growth response of bean seedlings to various anti-hypoxic substances under hypoxic conditions, calcium peroxide, magnesium peroxide and vitamin K3 performed better than hydrogen peroxide. These findings show that vitamin K3 and peroxide salts are effective in alleviating hypoxic stress in bean seedlings and also, further highlight their potential for dealing with hypoxia in wide ranging situations.展开更多
Objective:Acupuncture can relieve pain by acting on the mitogen-activated protein kinase(MAPK)signal pathway,which plays a critical role in the balance between hyperalgesia and inflammation.Our previous studies have s...Objective:Acupuncture can relieve pain by acting on the mitogen-activated protein kinase(MAPK)signal pathway,which plays a critical role in the balance between hyperalgesia and inflammation.Our previous studies have suggested that acupoint injection of Vitamin K3(Vit K3)had an intensive analgesic effect on primary dyspareunia.However,the mechanism by which Vit K3 worked on nerve cells has not been elucidated.Methods:Cell apoptosis,mitochondrial membrane potential(MMP),and reactive oxygen species(ROS)changes of PC-12 cells with Vit K3 treatment,for which the concentration gradient was 0,5,10,20,40,and 60μmol/L,were quantified by flow cytometry.The expression and phosphorylation of c-Jun N-terminal kinase,p38,and extracellular-regulated protein kinase(ERK),the three critical molecules of the MAPK pathway,were further assessed using Western blotting.Results:The level of ROS first decreased and then increased with Vit K3 at 20μmol/L,but no change in neither apoptosis nor MMP was evident.In addition,only ERK level decreased at 20μmol/L and the relative phosphorylation level increased.Changes in ROS were negatively correlated with the expression of ERK.Conclusions:The rapid analgesic effect of Vit K3 acupoint injection may be through the reduction of ROS in nerve cells with a small dose of Vit K3 or by influencing the expression of ERK but without damaging the nerve cells themselves.展开更多
Exponentially growing cultures of human bladder tumor cells (RT4 and T24) were treated with Vitamin C (VC) alone, Vitamin K3 (VK3) alone, or with a VC:VK3 combination continuously for 5 days or treated with vitamins f...Exponentially growing cultures of human bladder tumor cells (RT4 and T24) were treated with Vitamin C (VC) alone, Vitamin K3 (VK3) alone, or with a VC:VK3 combination continuously for 5 days or treated with vitamins for 1 h, washed with PBS and then incubated in culture medium for 5 days. Co-administration of the vitamins enhanced the antitumor activity 12- to 24-fold for the RT-4 cells and 6- to 41-fold for the T24 cells. Flow cytometry of RT4 cells exposed to the vitamins revealed a growth arrested population and a population undergoing cell death. Growth arrested cells were blocked near the G0/G1-S-phase interface, while cell death was due to autoschizis. Catalase treatment abrogated both cell cycle arrest and cell death which implicated hydrogen peroxide (H2O2) in these processes. The H2O2 production resulted in a moderate increase in lipid peroxidation and depletion of cell thiol levels. Analysis of cellular ATP levels revealed a transient increase in ATP production for VC and the VC:VK3 combination, but decreased ATP levels following VK3 treatment. Lipid peroxidation, thiol depletion and ATP modulation occurred at a 17-fold lower concentration in the vitamin combination than with either vitamin alone. These results suggested that the increased cytotoxicity of the vitamin combination was due to redox cycling and increased oxidative stress.展开更多

This method was based on that vitamin K3 was reduced at a glassy carbon electrode, and its product with characteristic fluorescence at 420 nm was determined with excitation wavelength at 309 nm. Under optimized electrochemical reaction conditions and fluorescent experiment parameters, the fluorescence intensity was proportional to the concentration of vitamin K3 in a range from 3.50×10^-7 to 1.05×10^-5 mol/L with a correlation coefficient of 0.9991, and detection limit was estimated to be 7.50× 10^-8 mol/L at a signal/noise ra- tio of 3. The relative standard deviation was less than 4.3%(n=5) and the recovery was in a range of 97%-105% for the determination of vitamin K3 in pharmaceutical preparations. The result is satisfactory for the determination of vitamin K3 as comparison to that from HPLC method.展开更多

It has been reported in the recent ten or more years that intramuscular injection of vitamin K3 (Vit K3) could relieve pain induced by smooth muscle spasm. In order to evaluate the effect of Vit K3 administered by acupoint injection in relieving pelvic pain, 180 patients were treated and observed from April 1997 to April 1999 in our hospital, and good therapeutic effect was obtained.
