In terms of special geological conditions of the Western Route of South-to-North Water Transfer Project, the classification method for surrounding rocks is discussed by combining with the construction method of tunnel boring machine (TBM). The classification standard of surrounding rocks is put forward on the basis of physical simulations and engineering practices. Damage, deformation and evolution of surrounding rocks induced by TBM excavation are discussed. Meanwhile, the long-term deformation mechanisms and stability of surrounding rocks are also studied. On this basis, a three-dimensional constitutive model for interbedded sandstone slate and a fiat shell-joint element-foundation system for calculating internal forces of segment lining are established. The deformation features of surrounding rocks of deep and steep interbedded sandstone slate and their influences on internal forces of segment lining are presented. Finally, the design methods of segment lining constructed in deep and steep flysch are proposed. 展开更多

Estimating minimum ecological water required of the western route first stage project of China's south-to-north water transfer scheme for water exporting rivers

It is well known that there is abundant water resources in basin of the Yangtze River, the first largest river in China, which is mainly located in Southern China. However, water resources is very scarce in the basin of the Yellow River, which is mainly located in Northern China. So the western route project of south-north water transfer scheme (WRP-SNWTS) aims to transfer water from the Yangtze River to the Yellow River. The area of WRP-SNWT, located in the upper reaches of the Yangtze River and the main areas of Sichuan and the marginal areas of the Qinghai-Tibet Plateau, has sufficient water resources but fragile ecology and environment. Therefore, it is necessary for WRP-SNWT to analyze the ecology water required. Based on the planning principles of from low elevation to high elevation, from small to large, from short to long and from easy to difficulty, the WRP-SNWT will be constructed through three stage projects. The western route first stage project of the south-north water transfer scheme (WRFST-SNWTS) is planned to transfer 4×10^9m^3/a from six tributaries of the Yalong river and from Dadu river to Jiaqu of Yellow River.. Daqu river and Niqu river are the branches of Xianshui river. Sequ river, Duke river, Make river and Ake river are the branches of Dadu river, which account for 65-70% of the total river run-off. It need more research and the rest run-off can satisfy channel ecology water required. According to analysis ecological water required which mainly satisfy for aquicolous biology in water-exporting region, such as low air temperature. Fish and aquicolous biology main living from May to August, and rivers are iced up from December to March of next year, ecology water required mainly for fish and aquicolous biology. The flow criterion of Tennant method is modified. The ecology water required of WRFSP-SNWTS is estimated by the flow data of Zhuwo gauging station, Zhuba gauging station, Chuosijia gauging station and Zumuzu gauging station. The result show that the ecology water required calculated by modified Tennant less 1 l percent than that of Tennant. This estimating result can supply more water resources for transferring to Yellow River. Meanwhile, this can supply gist for research transferring water of WRFSP-SNWTS. 展开更多

目的探讨快、慢转法及不同滤膜和显色检测法在Western blotting中的应用及各实验环节分析。方法采用MDA-MB-231细胞制备细胞蛋白,应用不同印迹膜[硝酸纤维素膜、聚偏乙烯二氟(PVDF)膜和阳离子尼龙膜]分别采用快转法和慢转法进行增强化... 目的探讨快、慢转法及不同滤膜和显色检测法在Western blotting中的应用及各实验环节分析。方法采用MDA-MB-231细胞制备细胞蛋白,应用不同印迹膜[硝酸纤维素膜、聚偏乙烯二氟(PVDF)膜和阳离子尼龙膜]分别采用快转法和慢转法进行增强化学发光法(ECL)和DAB化学显色法检测,并对Western blotting的各实验环节进行了分析。结果(1)在快、慢转法中硝纤膜的蛋白预染marker条带略强于尼龙膜,而尼龙膜又略强于PVDF膜;PVDF膜和尼龙膜的正反面易于混淆,而硝纤膜的正反面不易混淆。(2)在快、慢转法中,PVDF膜的DAB化学显色法图像略强于尼龙膜和硝纤膜,而尼龙膜和硝纤膜无明显差异;与慢转法相比,快转的硝纤膜和尼龙膜中的蛋白条带略呈波浪状。(3)在慢转法中三种膜化学发光法的图像无明显背景,但在快转法中尼龙膜的背景较明显,而硝纤膜和PVDF膜亦无明显背景。结论应根据实验需要选用不同的印迹膜;慢转法通常优于快转法,增强化学发光法优于DAB化学显色法;增强化学发光法特异胜强、灵敏度高,是分析蛋白质表达较为理想的方法。 展开更多

南水北调西线水源区大型底栖动物的适宜水文条件

为探索南水北调西线工程水源区大型底栖动物群落结构及其适宜的水文条件,于2022年7月(汛期)及11月(非汛期)在南水北调西线工程水源区开展了2次大型底栖动物采样调查,共记录大型底栖动物50个分类单元。结果表明:西线水源区底栖动物群落... 为探索南水北调西线工程水源区大型底栖动物群落结构及其适宜的水文条件,于2022年7月(汛期)及11月(非汛期)在南水北调西线工程水源区开展了2次大型底栖动物采样调查,共记录大型底栖动物50个分类单元。结果表明:西线水源区底栖动物群落组成以节肢动物昆虫纲(Insecta)为绝对优势,占总分类单元数的90.20%。其中,汛期以扁蜉(Heptageniidae sp.,优势度Y=19.9%)为第一优势种,非汛期以襀(Perlidae sp.,优势度Y=13.8%)为第一优势种。时间上,汛期生物密度更高,但非汛期生物多样性及物种均匀度更高;空间上,西线工程上线方案调水河段的生物多样性及生物密度明显高于下线方案。在0~1.0 m/s的流速范围内,不同流速区间的物种数及Margalef多样性指数具有显著差异;且无论是汛期还是非汛期,流速在0.5~1.0 m/s的底栖动物物种数及各项生物多样性指数均处于最高水平。在0~0.6 m的水深范围内,不同水深区间的生物量在汛期或非汛期均具有显著差异,且底栖动物密度整体在0~0.2 m区间内的平均值最高。针对水源区8个优势物种,利用栖境使用模拟法,得到西线工程水源区河流大型底栖动物优势物种的适宜水文条件为水深0.2~0.4 m、流速0.3~0.6 m/s。 展开更多

Dynamic optimization is utilized to calculate the growth rate of the Chinese economy on the optimal growth path, and backward integration is utilized to simulate the evolvement of some key indicators, including relative GDP per capita of Western China to Eastern China, regional capital growth rate and regional GDP growth rate. The calibrations of this model after 1992 are conducted. The calibration results provide evidence about the speed of Western China's relative development, and also shed some light on the future regional convergence process under the Western Development Programme launched in 2000. 展开更多

Western blotting is a popular technique for examining expression levels of proteins using gel-based electrophoretic fractionation followed by blotting and antibody reactions. Although this is a mature technique, one of the major limitations is the need to prepare an individual electrophoretic gel for each of the protein species to be analyzed. Since most analyses require the detection of multiple protein species, a procedure that allows utilization of a single gel for detecting multiple protein species should significantly save time and resources. In this paper, we developed a novel multiprotein detection device, which enabled simultaneous detection of several proteins species from a single electrophoretic gel. In this device, a protein transfer unit utilized a multi-anode plate that generated a non-uniform voltage profile. This voltage profile enabled uniform transfer regardless of molecular mass of proteins. In vitro experiments using samples, isolated from boneforming osteoblast cells, showed that the expression levels of 5 - 7 different proteins were detectable in the presence and absence of mechanical stimulation that activated genes necessary for bone formation. The result supports the notion that through simultaneous detection of multiple protein species, the described device contributes to reduction in procedural time and sample amounts, as well as a removal of variations among multiple gels. 展开更多

Improved Protein Transfer Efficiency and Signal Intensity in BlotMan Using Pulse Width Modulation

BlotMan is a protein blotting device that allows generating multiple membranes from a single polyacrylamide gel. To transfer all proteins uniformly with the same efficiency regardless of protein size, BlotMan employs pulse-width-modulated (PWM) voltage that applies a higher average voltage to a larger protein species. BlotMan can be controlled not only by its custom-made interface but also by a smart phone via Bluetooth technology. In this study, we examined effects of PWM signals (50%, 60%, and 80% duty cycle) on transfer efficiency and signal intensity in comparison to a constant voltage signal (100% duty cycle). The result revealed that in response to the same average voltage of 150 V, a lower duty cycle with a higher maximum voltage increased transfer efficiency as well as sharpness of transferred proteins. We validated BlotMan’s capability using a chondrosarcoma cell line (SW1353 cells) and a breast cancer cell line (MDA-MB231 cells) in response to antitumor chemical agents. BlotMan successfully generated 5 membranes from a single gel and detected 5 protein species such as c-Src, eukaryotic translation initiation factor 2 alpha (eIF2), phosphorylated eIF2, lamin B, and actin. Collectively, we demonstrated herein that BlotMan reduces an amount of protein samples by generating multiple membranes from a single gel and improving signal intensity with PWM voltage signals. 展开更多

The Western Route of the South-to-North Water Transfer Project will divert water from the upper Yangtze River and its tributaries, the Dadu River and Yalong River, to the upper Yellow River. The project may ease the water shortage in the Yellow River Basin. However, it may also have some effects on the ecosystem in the upper Yangtze River Basin. Benthic invertebrates play an important role in the river ecosystem, particularly in the circulation of materials and nutrition. Benthic invertebrates are widely used to quickly assess river ecosystems because of their rapid response to changes in the water environment. The diversity of benthic invertebrates is closely associated with the aquatic habitat area. This study examined this interaction by sampling the benthic invertebrates in an expanding area. The conclusions are that the diversity of benthic invertebrates begins to decrease when the aquatic habitat area is reduced to 45% of the original area, and decreases dramatically when the aquatic habitat area is reduced to 10% of the original area. The aquatic habitat area should be kept at more than 45% of the original area in order to maintain the significant diversity of benthic invertebrates. 展开更多

本文主要对1993年至2006年期间我国东中西部地区间工业产业转移的趋势和特征进行了初步分析,并探讨了我国东中西部地区间产业转移的趋势和特征的形成以及与传统的国际产业转移理论出现背离的原因。本文统计分析结果表明:截止到2006年,... 本文主要对1993年至2006年期间我国东中西部地区间工业产业转移的趋势和特征进行了初步分析,并探讨了我国东中西部地区间产业转移的趋势和特征的形成以及与传统的国际产业转移理论出现背离的原因。本文统计分析结果表明:截止到2006年,我国东中西部地区间只发生了部分的相对产业转移,绝对产业转移的现象尚未发生,我国东中西部地区间的产业转移还不显著;在我国东中西部地区间的产业转移过程中,出现了部分技术密集型产业先于劳动型密集型产业转移和许多产业越过中部地区直接转移到西部地区的现象。本文研究结论的重要启示是:同一国度内区域间的产业转移与国际产业转移的条件有着很大的不同,因此在分析我国地区间产业转移时不能机械照搬国际产业转移理论;目前我国东中西部地区间大规模产业转移的条件还不成熟,因而在相当长一个时期内,试图通过东中西部地区间的产业转移来实现我国的区域协调发展以及解决国际金融危机背景下我国产业结构调整与扩大就业之间的矛盾还是相当困难的。 展开更多

"水少、沙多、水沙调控能力不足"是治黄历史矛盾。新时期治黄矛盾发生了重大变化,"沙多"得到了大幅度控制,"水沙调控能力不足"有了一定程度的缓解,"水少"已经演化成为主要矛盾。面向新时期黄河流域生态保护和高质量发展目标,应对治黄矛盾的思路在于"增水、减沙、提高水沙调控能力",促进流域水沙关系协调。实施西部调水是破解治黄矛盾的战略性举措,不仅可以保障黄河长治久安,支撑流域高质量发展用水需求,还能够有效提升黄淮海平原生态环境质量。基于治黄矛盾及其发展变化规律,提出了新时期治黄五大方略,即坚持生态优先的高质量发展、以水为脉的系统治理保护、应对极端灾害风险的综合预防、流域自然-社会水循环协同管控、流域和区域统筹的五大平衡控制。 展开更多

水网在一定程度上决定了国家或区域的经济社会发展格局,形成密切联系的水域生态经济带,并辐射和带动周边区域发展。我国从东部沿海经济带和长江经济带为主轴的“江-海”T字型水网经济带,逐步演变为东部沿海带、长江经济带和黄河生态经... 水网在一定程度上决定了国家或区域的经济社会发展格局,形成密切联系的水域生态经济带,并辐射和带动周边区域发展。我国从东部沿海经济带和长江经济带为主轴的“江-海”T字型水网经济带,逐步演变为东部沿海带、长江经济带和黄河生态经济带为主轴“江-河-海”Π字型水网经济格局。为应对南北方经济发展差距拉大和东西部发展不均衡的问题,提出“双T”型水网经济布局构想,“东部沿海经济带-长江经济带”为核心的南方水网经济带(ST)与“黄河生态经济带-西部经济带”为核心的北方水网经济带(NT),可形成互联、互通、互济的水网经济格局。构建“双T”型水网经济格局,对于支撑黄河流域生态保护与高质量发展重大国家战略,带动西部崛起,促进我国南北方、东西部均衡协同发展,促进民族和文化大融合,均具有十分重要的意义。实施西部调水、完善国家水网,是构建“双T”型水网经济格局的重要基础。 展开更多
