Use of random amplified polymorphic DNA （RAPD） analysis to detect jujube witches＇ broom phytoplasmas

Jujube witches＇ broom is a devastating disease of Ziziphusjujube that occurs in various jujube regions of China. Nucleic acid extracted from midribs of samples collected from three jujube varieties （＂Suanzao＂, ＂Lajiaozao＂ and ＂Langzao＂） from symptomatic and asymptomatic shoots were tested by random amplified polymorphic DNA analyses. Using 13 different 10 and 11-bp random primers the amplification of jujube DNA was achieved from all the samples; AMI4 primer provided amplification of specific DNA fragments of about 400 bp, only from samples collected from symptomatic plants. No genetic variations in these varieties were identified using the other 11 arbitrary primers; only with primer AL07 it was possible to differentiate ＂Langzao＂ from the other two varieties tested. All the experiments were repeated 2 times and the results were consistent. Compared with PCR analyses with phytoplasma-specific primers, RAPD techniques resulted to be an alternative rapid and sensitive method for detecting jujube phytoplasmas presence in different jujube varieties.

Utilization of Changyun Jujube, a Cultivar with Resistance to Witches Broom Disease

In 2021,the Shanting District Fruit Industry Service Center conducted an application test of a treatment for jujube witches broom disease using Changyun jujube.The results demonstrated that when diseased Changhong jujube plants were grafted with Changyun jujube in the spring using bark or cleft grafting,the majority of the new shoots of Changyun jujube exhibited no symptoms of witches broom disease,while a few exhibited symptoms of the disease.With the growth of new shoots,the symptoms of witches broom disease gradually abated,returning to normal growth and development.Similarly,the symptoms of witches broom disease on the rootstock below the grafting mouth also gradually abated,returning to normal.The Changyun jujube rootstock was utilized as the intermediate rootstock to grafting the jujube cultivars Qiyuexian and Fucuimi.The two cultivars were subsequently affixed with branch bark from the witches broom disease.The two cultivars did not exhibit any symptoms of witches broom disease,thus providing an opportunity to investigate potential treatments for this disease in jujube.Finally,the cultivation techniques of the Changyun jujube were presented.

Spatial pattern of paulownia witches' broom and its effects on the growth of Paulownia elongata in Shaanxi Province,China

The spatial pattern of paulownia witches＇ broom was investigated using an Iwao＇s （m^＊-m） model in Shaanxi Province, China. Iwao＇s （m^＊-m） model revealed that spatial pattern of paulownia witches＇ broom exhibited a uniform distribution in sample plots. In this model, characteristics of contagiousness coefficient （a） justified the probability of infection by virus-transmission vectors. For tree age less than six, susceptible individuals were aggregated as clusters in the plots and then mutually repelled over age. A preliminary survey of stand volume show that from age five to six, stand volumes of disease grade II showed excessive vegetative growth and returned to normal level between ages ten and 11. Loss rate of volume increment with disease grade II decreased, and then returned to raising trend both at grades III and IV. This phenomenon is possibly caused by an over-compensation effect and by environmental factors. The redundancy of growth hormones in branches, shoots, leaves, buds and roots increased the stem volume of entire trees.

Transgenic Paulownia Expressing shiva-1 Gene Has Increased Resistance to Paulownia Witches＇ Broom Disease

Stem segments from diseased Paulownia tomentosaXP, fortunei and leaves from healthy control were transformed with the expression vector p438PRSI via Agrobacterium tumefaciens. The p438PRSI vector contained shiva-1 gene, which encodes an antibacterial peptide under the control of a CaMV35S promoter. The regenerated plants from transformed explants were planted in a greenhouse and nursery. PCR and Southern blotting analysis showed that the shiva-1 gene was successfully integrated into the Paulownia genome. Transcription of the integrated shiva-1 gene was confirmed by RT-PCR. Bioassay in the green house and phytoplasma DNA-dot blotting demonstrated that resistance to Paulownia witch＇s broom disease （PWB） increased significantly in shiva-l-transgenic Paulownia. Further investigations indicated that higher Shiva- 1 expression correlated with fewer phytoplasma and less symptoms in diseased transgenic Paulownia. Together, our findings strongly suggest that breeding shiva-1-Paulownia is an effective strategy to control PWB disease.

ZERO-CURRENT AND ZERO-VOLTAGE SWITCHING PWM BOOST FULL-BRIDGE CON VERTER

This paper proposes a zer o current and zero voltage switching (ZCZVS) PWM Boost full bridge (FB) conve rter. With series inductors, the leading switches can realize zero current swit ching (ZCS) in a wide load range using the energy of the output capacitor. Ma king use of parasitic capacitors of the lagging switches and parallel auxiliary i nductance with the primary winding of the transformer, the lagging switches can realize zero voltage switching (ZVS) under any load. Compared with the ZCS PWM Boost FB converter, the new converter has no current duty cycle loss. Operat ional principle and parameter design are analyzed. Experimental results verify the effectiveness of the proposed converter.

Technical Studies of Controlling Jujube Witches Broom Disease with Resistant Rootstock

[Objectives]The paper was to screen and identify the control effect of resistant rootstock on jujube witches broom disease.[Methods]The seeds or branches of disease-free plants were collected from wild jujube bushes infected by witches broom disease,and 5434 seedlings were obtained by sowing or cutting.The virus was inoculated by grafting diseased bark.After 6 years of preliminary screening,40 plants with resistance to jujube witches broom disease were obtained.Taking these resistant plants as the rootstocks,the susceptible variety‘long red jujube’was grafted at a height of 40 cm.When the plants grew to appropriate thickness,diseased bark was grafted to‘long red jujube’and rootstock to transmit the virus.After 18 years of observation and judgment,4 rootstock strains with high resistance or immunity to jujube witches broom disease were finally determined,namely 90801,90803,90806 and 908011.The susceptible variety‘long red jujube’was grafted to 4 rootstocks and inoculated the virus through diseased bark grafting method.[Results]The resistant rootstock had obvious disease resistance effect.When the branches of 90801 and 90803 rootstocks were grafted to the trunk of diseased trees,the symptoms of jujube witches broom disease gradually alleviated till disappeared.[Conclusions]Using germplasm with high resistance or immunity to jujube witches broom disease as rootstock can effectively prevent the spread of jujube witches broom disease,and bridging the trunk of diseased trees with branches from resistant rootstock has certain therapeutic effect.

国际气候谈判中,美国等发达国家把中国承诺强制减排目标作为其加入全球减排协定的前提,那么如果我国加入全球碳交易市场,将会对我国产生怎样的影响呢?本文基于Ramsey最优增长模型和博弈论思想,构建世界诱导技术变化混合(WITCH)模型,模拟评估全球碳交易市场对我国经济-能源-气候系统的影响,结果表明:12014年(GCM2014情景)和2020年(GCM2020情景)加入全球碳交易市场,到2100年我国GDP比常规情景(BAU情景)下分别减少6.42%和10.22%,说明加入全球碳市场将会对我国经济增长产生较大的负面影响,越早加入负面影响越小;2BAU、GCM2014和GCM2020三种情景下,2100年我国总投资分别达到98.48亿美元、85.59亿美元和85.13亿美元,其中我国能源研发投资预计分别为32.9亿美元、82.5亿美元和96亿美元,说明对我国总投资的影响差异不是很大,但对能源技术研发投资却有快速而显著的促进作用;3BAU、GCM2014和GCM2020三种情景下,2100年我国一次能源消费分为270 EJ、247 EJ和254 EJ,说明全球碳交易市场可以减少我国一次能源的总消费,但总量减少不是太显著,却能显著减少我国的煤炭等化石能源的消费量,因为GCM2014和GCM2020情景下,2100年煤炭消费仅占到我国一次能源消费的10%左右,而BAU情景下煤炭仍占59%;4全球碳交易市场可以比较显著的降低我国能源强度和碳排放强度;5全球碳交易市场的建立能有效降低全球气温,但不能将气温升幅控制在2℃范围以内。因此,我国要结合本国国情和实际,先建立国内碳交易市场,避免在国际压力下盲目承诺加入全球碳交易市场,至少2020年前我国不应该加入全球碳交易市场。

Identification of the SERK Gene Family in Paulownia fortunei and Its Involvement in the Response to Biotic and Abiotic Stresses

Somatic embryogenesis receptor-like kinases(SERKs)are receptor-like proteins that contain leucine-rich repeats and are involved in various signaling pathways.This study identified SERK family members in the Paulownia fortunei genome and analyzed their characteristics and expression profiles using bioinformatics methods.We identified 12 SERK genes with relatively conserved gene structures and motifs that were distributed unevenly on eight Paulownia chromosomes.The gene promoters contained various cis-acting elements that regulated the expression of the PfSERK genes in response to hormones and abiotic stresses.Synteny analysis indicated that 10 segmental duplication events had occurred during evolution of the PfSERK family.The expression profile of PfSERKs in various tissues of Paulownia fortunei was detected by quantitative real-time PCR(qRT-PCR).Many PfSERK genes can respond to drought and salt stress.Combined with RNA-seq and protein interaction network,it is speculated that PfSERK3/11 may participate in the occurrence of Paulownia witches’broom(PaWB)by regulating the plant height of Paulownia.

The Malleus Maleficarum(1486)by Henry Institoris and Jacques Sprenger was written in order to help inquisitors identify,persecute,and prosecute witches.The book is well-known but not studied as much as one could think,and is sometimes confused with another treatise written in 1669,which includes works by demonologists,such as Jean Nider,Bernard Basin,and Bartolomeo de Spina.This can be explained by the fact that several successive editions were published,which changed the essence of the first essay,until the 17th century.My argument is that the first edition was paradoxical and not as strict as one could imagine.The book was a“bible”for the inquisition about the subjugation of witches,but in the same way,the book seemed to be far too rationalistic when confronted with satanism and superstition.Indeed,we believe that the thesis of the Malleus Maleficarum was also a new way of conceiving of knowledge,fighting against superstition by using the tools of early rationalism at the end of the 15th century.The witches’knowledge that is investigated in the book is confronted with theology,as well as with reason.There is significant evidence of the writers’rationalistic view about occultism and belief.

In his Antichrist and Nymphomaniac, Lars von Trier seems to defy norms on sex and women＇s desires. However he actually reproduces Christianity＇s perspective that women are sinful by their dispositions. According to the classical creation story, Eve falls into temptation and then, convinces Adam to commit the same crime. All Abrahamic religions but especially Christianity identifies women with the body and bodily desires, rather than the mind which comes from nonmaterial and divine substance. Accordingly, women can easily be deceived by Satan since they contain a lesser amount of mental substance compared to men. In Antichrist, Trier reiterates the story of Adam and Eve in a different context. The woman writes her dissertation on women who are burnt with the accusation of being a witch. But she realizes that her body is sinful and deserving of punishment, too. She finds her salvation when she cuts her clitoris. In Nymphomaniac, the main character gets her clitoris whipped. While she resists traditional and bourgeois morality, she defines herself as morally evil because of her unmanageable sexual desires. Thus, reputed as critical of all social structures and morality, Trier in fact simply rehashes the classical sexist and misogynist perspective.

The Lady of Shalott is considered to be one of Alfred Lord Tennyson’s masterpieces.The poem conversely demonstrates art and life,the aesthetic and the political,are fully interwoven:the involvement in the social world which is symbolically the destination of the Lady in the poem.The Lady who is the emblem of such an artwork can be ascribed qualities,for instance,self-containment,objectified otherness,removal from the fluidity of life,and participation in a higher order of existence,for which it is autonomy.Kermode points out that“the Romantic artwork often identifies itself with an emblematic image of autonomy and femininity”,inextricably interlinked.Her mirror shows her only“Shadows of the world”.The Lady’s web flies“Out”and floats“wide”when she“turns round”with“that desire of making an impression upon another mind”,when she seeks to address Lancelot.The mirror reconnects the Lady’s art to the real world beyond her window.Though the artist is unable to see it during the process of production,her artistic web,once out in a wider space than the tower and freed from the loom,is a true representation of outward things,the actions,and events of Camelot.This paper examines that the traditional referentiality of Tennyson’s‘The Lady of Shalott’can be better understood by supplementing the poet’s medieval sources,of little more than tangential relevance to the poem,with Homeric influences.The poem is similar to a fairytale,however,I have given an alternate reality of the Lady of Shallot in my version of a fairytale,where the characters remain the same but the fairytale takes a happy ending rather than a tragic death.My version is different from Tennyson’s since I believe the lady deserves to be jovial.Sir Lancelot was unaware of her fascination and feelings for him,as she could hardly express her feelings due to her imprisonment.She is so taken by him that she stops her work and looks at Camelot,though she risks the mysterious curse.The mirror breaks and her web magically floats out of the window on its own.She realizes that she is cursed.
